〔1〕
製紙工程における微生物の侵入を識別する方法であって、
製紙工程に関する品目上の欠陥に注目することと、
前記品目から取った少なくとも一つのサンプルに対して、少なくとも1種の微生物に関連することが知られる核酸配列を標的にしたプライマーを利用したPCR分析を少なくとも一つ行うことと、
肯定的な結果を示す場合、微生物の測定した濃度が所定の閾値を超えるか否かを決定することと、
測定した前記濃度が閾値を超える場合、少なくとも一つの更なるPCR分析を行い、サンプルに存在する特定の微生物を決定することと
を含んでおり、
検出された前記微生物のそれぞれの測定した前記濃度が所定の閾値を超える場合、前記欠陥は前記微生物の侵入に少なくとも一部起因する、
方法。
〔2〕
前記品目がフェルトであり、前記欠陥が前記フェルト内の一つ以上の詰まりである、項目1に記載の方法。
〔3〕
前記品目が製紙工程によって製造される紙シートであり、前記欠陥が紙シート上の一つ以上の孔、変色、線条、斑点、半透明の斑点、及び任意のこれらの組み合わせである、項目1に記載の方法。
〔4〕
識別した前記微生物を、保存及び/又は送信することができる形式に記録する工程を更に含む、項目1の方法。
〔5〕
識別した前記微生物の改善措置に関連する殺生物性プログラムを行う工程を更に含む、項目1に記載の方法。

前記PCR分析がqPCR分析である、項目1に記載の方法。
〔7〕
前記PCR分析の閾値が、1mL当たりに10 4 セル、又は1グラム当たりに10 4 セルである、項目1に記載の方法。

前記品目上に、欠陥を生じたであろう生きた微生物がほとんどいないか又はいないように前記品目を乾燥させる、項目1に記載の方法。

前記品目の状態が、製紙工程の間に前記品目が遭遇する流体とは大きく異なっており、前記品目に住む微生物が前記流体内の微生物とは異なっており、前記流体内に住む微生物の決定は、欠陥を生じている品目上の微生物の誤った識別を生ずる、項目1に記載の方法。

前記閾値を上回る任意の微生物の存在を決定することができるよう、充分な種類のプライマーを品目のサンプルに適用する工程を更に含む、項目1に記載の方法。
〔11〕
前記PCR分析が前記閾値を超えたいかなる微生物も示さない場合、前記欠陥が非生物学に基づくものであると識別する工程を更に含む、項目10に記載の方法。
〔12〕
非生物学的化学的混入物の改善措置を製紙工程に適用する工程を更に含む、項目11に記載の方法。
〔13〕
前記PCR分析がサンプルにはびこる微生物量を決定する、項目1に記載の方法。
〔14〕
欠陥に注目する前に、前記品目を製紙工程の加熱部又は乾燥部に通して、欠陥を生じた微生物を殺す、項目1に記載の方法。
〔15〕
製紙工程における微生物の侵入を識別する方法であって、
製紙工程に関する品目上の欠陥に注目することと、
品目から取った少なくとも一つのサンプルに対して、品目にそのような欠点を生じることが知られる少なくとも一つの微生物に関連することが知られる核酸配列を標的にしたプライマーを利用したPCR分析を少なくとも一つ行うことと、
少なくとも一つの微生物について肯定的な結果が示された場合、前記微生物の測定した濃度が前記微生物についての所定の閾値を超えているかどうかを決定することと、
測定した前記濃度が前記閾値を超えている場合、前記欠陥が前記微生物の侵入に少なくとも一部起因することを識別することと
を含む、方法。
Claims (10)
- 製紙工程における微生物の侵入を識別する方法であって、
製紙工程に関する品目上の欠陥に注目することと、
前記品目から取った少なくとも一つのサンプルに対して、少なくとも1種の前記品目に住むことができる微生物に関連することが知られる核酸配列を標的にしたプライマーを利用した第一のqPCR分析を少なくとも一つ行うことと、
肯定的な結果を示す場合、微生物の測定した濃度が所定の閾値を超えるか否かを決定することと、
測定した前記微生物の前記濃度が所定の閾値を超える場合、少なくとも一つの更なる第二のqPCR分析を行い、サンプルに存在する特定の微生物を決定することであって、前記第二のqPCR分析は、サンプル中の微生物がサンプルに存在する合計微生物量の割合として表される多様性指数を与え、
多様性指数として検出された前記微生物のそれぞれの測定した前記濃度が所定の閾値を超える場合、前記欠陥は前記微生物の侵入に少なくとも一部起因すると判断することと
を含む、方法。 - 前記品目がフェルトであり、前記欠陥が前記フェルト内の一つ以上の詰まりである、請求項1に記載の方法。
- 前記品目が製紙工程によって製造される紙シートであり、前記欠陥が紙シート上の一つ以上の孔、変色、線条、斑点、半透明の斑点、及び任意のこれらの組み合わせである、請求項1に記載の方法。
- 識別した前記微生物に関する情報を、保存及び/又は送信することができる形式に記録する工程を更に含む、請求項1の方法。
- 識別した前記微生物の改善措置に関連する殺生物性プログラムを行う工程を更に含む、請求項1に記載の方法。
- 前記欠陥に注目する工程の前に、前記品目上に、欠陥を生じたであろう生きた微生物がほとんどいないか又はいないように前記品目を乾燥させる工程を更に含む、請求項1に記載の方法。
- 前記閾値を上回る任意の微生物の存在を決定することができるよう、充分な種類のプライマーを品目のサンプルに適用する工程を更に含む、請求項1に記載の方法。
- 前記第二のqPCR分析が前記閾値を超えたいかなる微生物も示さない場合、前記欠陥が非生物学に基づくものであると識別する工程を更に含む、請求項7に記載の方法。
- 非生物学的化学的混入物の改善措置を製紙工程に適用する工程を更に含む、請求項8に記載の方法。
- 前記欠陥に注目する工程の前に、前記品目を製紙工程の加熱部又は乾燥部に通して、欠陥を生じた微生物を殺す工程を更に含む、請求項1に記載の方法。
