Description
薬物および担体材料(線状ポリマー、架橋ポリマー、および、例えばシクロデキストリンなどの複合化剤(complexing agent)から選択される)を含む薬物/担体封入複合体(drug/carrier inclusion composite)は、共粉砕(co-grinding)プロセスをメカノケミカル活性化(mechanochemical activation)へ転換するように適切に改変した流体ジェットミルを使用する高エネルギー共粉砕プロセスによって調製される。このようにして得られた活性化された(activated)薬物/担体複合体は、難溶性薬物の場合において、「そのまま」の薬物と比較して大幅に改善された化学物理的(chemico-physical)および生物薬剤学的特性:高い割合の非晶質相、ナノ結晶性、高い可溶化キネティクス(solubilisation kinetics)およびより良いバイオアベイラビリティを示す。
現在開発中の薬物の約40%は、水に難溶性である(Lipinski、2002;Merisko-Liversidge & Liversidge、2008);言い換えれば、それらは、API(医薬品有効成分)のための生物薬剤学分類システム(Biopharmaceutical Classification System)(BCS)のクラスIIに属する。このクラスは、例えば、低いバイオアベイラビリティおよびおそらく遅い吸収などの経口吸収の無視できない問題、患者間のより大きい変動性、ならびに食物摂取によって生じる影響の潜在的に大きい可能性を有する薬物を含む(Amidonら、1995)。
上記で報告したように、空気または他のガス状の流体のジェットを用いて作動するミルの使用は、難溶性薬物の生物薬剤学的特性の改善を目的とした薬物/担体共粉砕プロセスへの現在のアプローチの中にはない。なぜなら、今日までに使用された操作手順およびプロセスパラメータが、担体材料中の薬物の分子分散を誘導するために必要なエネルギーレベルに達することを妨げていたからである。
上記されたように改変された流体ジェットミルの概略図を、古典的な微粉化プロセスをメカノケミカル活性化プロセスへ変換することを実現し得る種々の可能な改変の一例として、図1に示す。
共粉砕チャンバ内でのダイナミックグレーダ(4)の存在により、回転速度を変化させることにより予め決められたサイズを有する粒子が通過し得る。
− 薬物の残留結晶のエンタルピーを決定し、その結果として到達した非晶質化のレベルを決定する示差走査熱量測定。
− 残留結晶の融点を決定し、ナノ結晶の識別を可能にする示差走査熱量測定。
− 多形体(polymorphs)の形成を引き起こす可能性のある任意の残留結晶の結晶格子の内部構造の分析のためのX線回折。

Claims (10)
- 粉砕チャンバ、グレーダ、複数の粉末用および加圧ガス用の入口を含みグレーダ出口と粉砕チャンバとの間の再循環パイプによって特徴付けられる流体ジェットミル中で、薬物および担体の粉末の混合物を共粉砕にかけることを含む、薬物/担体封入複合体を調製する方法。
- 粉末のメカノケミカル活性化を起こすのに十分な時間、共粉砕チャンバ中に薬物/担体粉末混合物が滞在し得るようにミルが改変された、請求項1に記載の方法。
- 薬物が特徴付けられる、請求項1または2に記載の方法。
- 薬物が、タキソール、ケトプロフェン、イブプロフェン、ピロキシカム、メゲストロールアセテート、デヒドロエピアンドロステロン、ラロキシフェン塩酸塩、シンバスタチン、アトルバスタチン、カンデサルタン、セレコキシブ、フェロジピン、ブデソニド、シスプラチン、リトナビル、トリアゾラム、サキナビル、シクロスポリン、エストラジオール、アミオダロン塩酸塩、イスラジピンおよびマレイン酸ロシグリタゾンから選択される、請求項3に記載の方法。
- 担体が、ポリビニルピロリドン、ヒアルロン酸、キトサン、キサンタン、アルギン酸ナトリウム、ポリビニルアセテート、デンプングリコール酸ナトリウム、カルボキシメチルセルロースナトリウム、ポリビニルピロリドン/ポリビニルアセテート、ポリビニルカプロラクタム/ポリビニルアセテート/ポリエチレングリコール、ポリビニルピロリドン/ポリビニルアルコール、ポリビニルアルコール/ポリエチレングリコール、架橋ポリビニルピロリドン、架橋カルボキシメチルセルロースナトリウム、β−シクロデキストリン、α−シクロデキストリン、ヒドロキシプロピル−β−シクロデキストリン、微孔シリカゲル、ポリオルガノシロキサン、医薬用クレイ、硫酸バリウムから選択される請求項1から4のいずれかに記載の方法。
- 薬剤が高水溶性によって特徴付けられる、請求項1または2に記載の方法。
- 薬剤が、メトプロロール、シメチジン、テオフィリン、イソニアジド、ピンドロール、プロプラノロール、ヒドロクロロチアジド、アテノロール、ベラパミル、ジルチアゼム、アシクロビル、ラニチジン、メトホルミンから選択される、請求項6に記載の方法。
- 担体が、ポリメタクリレート、ポリメチルメタクリレート、メチルセルロース、アクリレートコポリマー、アンモニウムメタクリレートコポリマー、アミノエチルメタクリレートコポリマー、疎水性シリカゲルから選択される、請求項6または7に記載の方法。
- 薬剤と担体との間の重量比が、1/0.1w/wから1/100w/wの範囲である、請求項1〜8のいずれかに記載の方法。
- 粉砕チャンバ、グレーダ、複数の粉末用および加圧ガス用の入口を含み、グレーダ出口
と粉砕チャンバとの間の再循環パイプによって特徴付けられる、請求項1から9のいずれかに記載の方法における使用のためのジェットミル。
