Description
本発明の一つの実施形態による組成物に使用される前記玄参(Scrophularia buergeriana)は、玄参科の多年草植物であって、重台、玄台、鬼蔵、逐馬とも呼ばれる。漢方では、根を玄参と言い、解熱剤として咽喉炎、腫れ物、リンパ節炎に使用する。
まず、玄参、人参、丹参、黄耆、蔓参、苦参及び沙参のうち1種以上を麹発酵して、本発明の一つの実施形態による組成物を製造することができる。
本発明の玄参、人参、丹参、黄耆、蔓参、苦参及び沙参の7種の薬参抽出物は下記の方法によって製造した。前記それぞれの薬参を流水で洗浄した後、250メッシュ(mesh)のフィルターで濾過して50℃で24時間乾燥する。乾燥したそれぞれの薬参50gに、70%のエタノールを1L添加し、7種の乾燥した薬参混合物350g(各50g)には、70%のエタノールを10L添加して、4℃の温度条件下で48時間還流抽出した。その後、250メッシュのフィルターで濾過して沈澱させた後、再び0.5μmのフィルターで濾過してそれぞれの薬参抽出物及び混合抽出物を得た。
本発明で使用した組成物は、前記薬参の麹発酵抽出物またはその混合抽出物を有効成分として含み、下記のような方法によって製造した。7種の薬参(玄参、人参、丹参、黄耆、蔓参、苦参、及び沙参)を流水で洗浄した後、250メッシュ(mesh)のフィルターで濾過して50℃で24時間乾燥する。麻布風呂敷で前記乾燥したそれぞれの薬参50gと7種の乾燥した薬参混合物350g(各50g)を包んで、麹とご飯が1:1で混合された混合物が入っているカメに入れた。35℃で3日間発酵させた後、麻布風呂敷を取り出した。発酵した薬参の重量の20倍の精製水を入れ、80℃で24時間熱水抽出して麹発酵抽出物を得た。
前記製造例1の抽出物を使用して比較例1〜9を、前記製造例2の麹発酵抽出物を使用して実施例1〜8(参照例1〜7、実施例8)を下記表1の組成によって製造した。
毛髪を構成するケラチンタンパク質は、毛根部のケラチン形成細胞(keratinocyte)で生成され、このケラチン形成細胞は、毛乳頭細胞から分化する。本発明による組成物の毛乳頭細胞の活性を評価するために、本発明では、DP6(rat immortalized dermal papilla cell)細胞株を使用した〔Wendy Filsell, Journal of Cell Science 107, 1761−1772(1994)〕。本毛乳頭細胞株は、雄PVG ratのひげの毛根でmicrodissection方法で分離して培養した細胞株であり、10%のウシ胎仔血清(Fetal bovine serum、FBS)が含まれたDMEM(Dulbecco’s modified Eagle’s medium;Gibco BRL, Gaithersburg, MD, USA)培地で5%のCO2、37℃が維持される培養器(incubator)で24時間培養した。
脱毛治療剤であるミノキシジルは、潜在的なミトコンドリアKATP channel opener(カリウムイオンチャンネルオープナー)として知られており、アンドロゲン性脱毛の治療に使用される代表的な薬物である。このようなミノキシジルのメカニズムを評価するために、頭皮の真皮を構成する線維芽細胞で、KATP channelを塞ぐトルブタミド(Tolbutamide, SIGMA AlDRICH, T0891)を処理して細胞増殖を抑制し、再びカリウムイオンチャンネルを開いて細胞増殖が回復する試験法を使用した。
前記製造例1の薬参抽出物を使用して比較例21〜31を、前記製造例2の麹発酵抽出物を使用して実施例17〜24(参照例17〜23、実施例24)を、下記表4及び表5の組成によって製造した。
本組成物の抗酸化効果を評価するために、DPPH抗酸化評価法を使用した。体内で起こる酸化反応は、主に自由ラジカルによって起こり、このような作用は、細胞の老化及び癌の発生を招くと知られている。したがって、体内の自由ラジカルの生成抑制は、結局、体内抗酸化、抗老化に関連し、このような抗酸化効果は、体内の自由ラジカルを対象として実験し難いため、DPPH(2,2−diphenyl−1−picrylhydrazyl)ラジカルを普遍的に使用している。DPPHラジカルは、濃い紫色を帯びているが、抗酸化効能の物質と出合って反応すると、薄い黄色に変わり、517nmの波長で吸光度を測定して抗酸化能を評価する。物質の抗酸化能が高ければ高いほど、薄い黄色を帯び、吸光度が低下する。
Claims (4)
- 玄参、人参、丹参、黄耆、蔓参、苦参及び沙参からなる薬参混合物の麹発酵抽出物を有効成分として含む脱毛防止又は毛髪成長促進用組成物。
- 前記薬参混合物の麹発酵抽出物は、組成物の総質量に対して0.01〜10重量%含まれることを特徴とする請求項1に記載の脱毛防止又は毛髪成長促進用組成物。
- 前記組成物は、血行を促進することを特徴とする請求項1に記載の脱毛防止又は毛髪成長促進用組成物。
- 前記組成物は、頭皮抗酸化用であることを特徴とする請求項1に記載の脱毛防止又は毛髪成長促進用組成物。
