Description
細胞試料を準備するステップと、
デジタルホログラフィ顕微鏡法(DHM)によって細胞試料からホログラフィ情報を得るステップと、
ホログラフィ情報から少なくとも1つの細胞パラメーターを導出するステップと、
細胞試料の細胞を分類するステップと、
を含み、
分類は、細胞パラメーターに基づいて、細胞試料の細胞にスコアリングファクターを指定することにより行われることを特徴とする。
Sc=pN+pNc+pR+pG+pV
ただし、pxは細胞を悪性であると規定するようにファクターの重要度に関連付けられる考量ファクターであり、
pNは細胞の核サイズに関連する。細胞の核サイズと細胞質サイズとの比が正常である場合には、pNは0に等しく、そうでない場合には、pNは1に等しい。
pNcは細胞の核と細胞質との比に関連する。この比が0.5に等しい場合には、pNcは1に等しく、そうでない場合には、pNcは0に等しい。
pRは細胞の形状に関連する。この形状が正常である場合には、pRは0に等しく、そうでない場合には、pRは1に等しい。
pGはクロマチンの粒度分布に関連する。この粒度分布が均質である場合には、pGは0に等しく、そうでない場合には、pGは1に等しい。
pVは細胞の核サイズのばらつきに関連する。このばらつきが正常である場合には、pVは0に等しく、そうでない場合には、1に等しい。
上記スコアリングファクターは、DHMによって評価される細胞ごとに決定される。
Sc=pN+pNc+pR+pG+pV+pH
ただし、pHは上記細胞の光学的核高に関連する。光学的高さが正常である場合には、pHは0であり、そうでない場合には、pHは1である。
Scが0に等しい場合には、その分析された細胞は良性と見なされる。
Scが1又は2に等しい場合には、その細胞は未決定と見なされ、最後に、
そのScが3以上である場合には、その細胞は形成異常又は悪性と見なされる。
照明手段と、干渉計と、サーバーに接続されるデジタル記録デバイスとを備える、デジタルホログラフィ顕微鏡(DHM)と、
細胞試料を含む少なくとも1つの交換可能な試料バイアル又は試料キャリアと、
上記細胞試料に関連するデジタル報告を与えることができるコンピューター又はプリンターと、
を備えることが好ましい。
試料に結び付けられるホログラフィ情報を得るステップであって、そのホログラフィ情報はデジタルホログラフィ顕微鏡(DHM)を用いて得られることを特徴とする、得るステップと、
上記ホログラフィ情報から少なくとも1つのパラメーターを導出するステップと、
上記パラメーターをデータベース内に記憶される上記しきい値と比較するステップと、
上記パラメーターと上記しきい値との上記比較に基づいてスコアリングファクターを計算するステップと、
上記スコアリングファクターを実務者に報告するステップと、
上記スコアリングファクターに関して上記実務者のフィードバックを得るステップと、
上記フィードバックに基づいて上記データベースを更新するステップと、
を含む。
デジタルホログラフィ顕微鏡を用いて物体を含む試料から得られたホログラフィ情報及び/又はそこから導出されたパラメーターと、
上記ホログラフィ情報及び/又はパラメーターの分析に用いるしきい値、及びそのしきい値に関連付けられるクエリーと、
上記ホログラフィ情報及び/又はパラメーターから導出されるスコアリングファクターと、
画像識別子と、
識別情報と、
を含み、
データベースの上記しきい値及びクエリーは、第三者からのフィードバック情報を受信することに基づいて更新されることを特徴とする。
Claims (29)
- 液状細胞試料内のがん性細胞を検出し、及び/又は細胞を分類する方法であって、
細胞試料を準備するステップと、
デジタルホログラフィ顕微鏡法によって前記細胞試料からホログラフィ情報を得るステップと、
前記ホログラフィ情報から少なくとも1つの細胞パラメーターを導出するステップと、
細胞試料の前記細胞を分類するステップと、
を含み、
前記分類は、前記細胞パラメーターに基づいて、細胞試料の前記細胞にスコアリングファクターを指定することにより行われることを特徴とする、方法。 - 得られたホログラフィ情報から導出された前記少なくとも1つの細胞パラメーターは光学的核高を含むことを特徴とする、請求項1に記載の方法。
- 得られたホログラフィ情報から導出された前記少なくとも1つの細胞パラメーターは、細胞核直径、クロマチンテクスチャ、細胞サイズ、細胞形及び細胞形態を含むことを特徴とする、請求項1又は2に記載の方法。
- 画像識別子が前記細胞パラメーター及びそこから導出されたスコアリングファクターに結び付けられることを特徴とする、請求項1〜3のいずれか一項に記載の方法。
- 前記指定されたスコアリングファクターは、前記少なくとも1つの細胞パラメーターとしきい値データベースとの比較に基づくことを特徴とする、請求項1〜4のいずれか一項に記載の方法。
- 前記しきい値データベースは内部サーバー又は外部サーバー上に記憶されることを特徴とする、請求項5に記載の方法。
- 前記スコアリングファクターは前記サーバー上でクエリーを使用することにより指定されることを特徴とする、請求項6に記載の方法。
- 実務者に、スコアリングファクターと、細胞試料内の前記細胞の分類とを含むデジタル報告を提供するステップを更に含む、請求項5〜7のいずれか一項に記載の方法。
- 前記デジタル報告において前記細胞試料の2次元画像及び3次元画像を与えるステップを更に含む、請求項8に記載の方法。
- 細胞の前記分類前に、前記試料内の前記細胞の細胞タイプを識別するステップを更に含む、請求項1〜9のいずれか一項に記載の方法。
- 細胞試料内の細胞タイプの前記識別は前記細胞パラメーターの細胞サイズに基づいて行われる、請求項10に記載の方法。
- 細胞のサブセットのみを分類し、細胞の前記サブセットは前記細胞試料において識別された特定の細胞タイプに関連付けられることを特徴とする、請求項11に記載の方法。
- 前記細胞試料が子宮頚部試料であることを特徴とする、請求項1〜12のいずれか一項に記載の方法。
- 細胞試料における前記細胞が、表在性扁平上皮細胞、介在性扁平上皮細胞、基底細胞、傍基底細胞、赤血球、マクロファージ、リンパ球及び微生物を含むことを特徴とする、請求項13に記載の方法。
- 前記表在性扁平上皮細胞、介在性扁平上皮細胞、基底細胞及び傍基底細胞に対して、スコアリングファクターが指定されることを特徴とする、請求項14に記載の方法。
- 請求項1〜15のいずれか一項に記載の方法を利用して、細胞試料内のがん性細胞を検出し、及び/又は細胞を分類するシステムであって、
照明手段と、干渉計と、サーバーに接続されるデジタル記録デバイスとを備える、デジタルホログラフィ顕微鏡と、
液状細胞試料を含む少なくとも1つの交換可能な試料バイアル又は試料キャリアと、
前記液状細胞試料に関連するデジタル報告を与えることができるコンピューター又はプリンターと、
を備える、システム。 - 前記サーバーは内部サーバー又は外部サーバーである、請求項16に記載のシステム。
- 前記サーバーは、前記細胞パラメーターをしきい値データベースと比較するアルゴリズムを与えられる、請求項17に記載のシステム。
- 前記交換可能な試料バイアル又は試料キャリアは識別用指示を備える、請求項16〜18のいずれか一項に記載のシステム。
- 前記識別用指示はRFIDを含む、請求項19に記載のシステム。
- ホログラフィ情報に結び付けられるしきい値を含むデータベースを更新し、及び/又は改善する方法であって、
試料に結び付けられるホログラフィ情報を得るステップであって、前記ホログラフィ情報はデジタルホログラフィ顕微鏡を用いて得られることを特徴とする、得るステップと、
前記ホログラフィ情報から少なくとも1つのパラメーターを導出するステップと、
前記パラメーターをデータベース内に記憶される前記しきい値と比較するステップと、
前記パラメーターと前記しきい値との比較に基づいてスコアリングファクターを計算するステップと、
前記スコアリングファクターを実務者に報告するステップと、
前記スコアリングファクターに関して前記実務者のフィードバックを得るステップと、
前記フィードバックに基づいて前記データベースを更新するステップと、
を含む、方法。 - スコアリングファクターの前記計算の際に、前記導出されたパラメーターを外部サーバーに送信する、請求項21に記載の方法。
- 少なくとも1つのパラメーターの前記導出、及び/又はスコアリングファクターの前記計算の際に外部サーバーを通してクエリーを与える、請求項22に記載の方法。
- 前記試料に結び付けられる識別情報を記憶する、請求項21〜23のいずれか一項に記載の方法。
- 前記試料は細胞試料若しくは液状細胞試料である、請求項24に記載の方法。
- 前記計算されたスコアリングファクターに基づいて前記細胞試料内に存在する細胞を識別し、及び/又は分類する、請求項25に記載の方法。
- 物体のデータベースが、
デジタルホログラフィ顕微鏡を用いて前記物体を含む試料から得られたホログラフィ情報及び/又はそこから導出されたパラメーターと、
前記ホログラフィ情報及び/又は前記パラメーターの分析に用いるしきい値、及び該しきい値に関連付けられるクエリーと、
画像識別子と、
識別情報と、を含み、
物体のデータベースが前記ホログラフィ情報及び/又は前記パラメーターから導出されるスコアリングファクターを含み、及びそのことによりデータベースの前記しきい値及び前記クエリーは、第三者からのフィードバック情報を受信することに基づいて更新されることを特徴とする、物体のデータベース。 - 前記試料は細胞試料若しくは液状細胞試料である、請求項27に記載のデータベース。
- 前記ホログラフィ情報及び/又はそこから導出されたパラメーターは識別情報及び/又は画像識別子に結び付けられる、請求項27又は28に記載のデータベース。
