RNA干渉およびsiNA核酸分子
RNA干渉は、短鎖干渉RNA(siRNA)によって誘導される動物における配列特異的転写後遺伝子サイレンシングのプロセスを指す(Zamore et al., 2000, Cell, 101, 25-33、Fire et al., 1998, Nature, 391, 806、Hamilton et al., 1999, Science, 286, 950-951、Lin et al., 1999, Nature, 402, 128-129、Sharp, 1999, Genes & Dev., 13:139-141およびStrauss, 1999, Science, 286, 886)。植物における対応するプロセス(Heifetz et al., PCT国際公開WO 99/61631)は、転写後遺伝子サイレンシング(PTGS)またはRNAサイレンシングとしばしば称される。転写後遺伝子サイレンシングのプロセスは、外来遺伝子の発現を防止するのに用いられる進化的に保存された細胞防衛機構であると考えられている(Fire et al., 1999, Trends Genet., 15, 358)。外来遺伝子の発現からのかかる保護は、ウイルス感染、または宿主ゲノムへのトランスポゾン要素のランダムな統合に由来する二本鎖RNA(dsRNA)の産生に対応して、相同な一本鎖RNAまたはウイルスゲノムRNAを特異的に破壊する細胞反応を介して進化したと考えられる。細胞におけるdsRNAの存在は、まだ完全に特徴づけられていないメカニズムによって、RNAi反応を引き起こす。このメカニズムは、二本鎖RNAに特異的なリボヌクレアーゼを伴う他の既知のメカニズム、例えば、プロテインキナーゼPKRおよび2’,5’−オリゴアデニル酸合成酵素の活性化によって生じ、リボヌクレアーゼLによるmRNAの非特異的切断をもたらすインターフェロン反応などとは異なるようである(例えば、米国特許第6,107,094号、第5,898,031号、Clemens et al., 1997, J. Interferon & Cytokine Res., 17, 503-524、Adah et al., 2001, Curr. Med. Chem., 8, 1189参照)。

本明細書に開示される核酸分子(例えばsiNA分子)は、核酸分子に存在するヌクレオチドの総数のパーセンテージとしての、修飾ヌクレオチドを含んでもよい。このように、核酸分子は、約5%〜約100%の修飾ヌクレオチド(例えば約5%、10%、15%、20%、25%、30%、35%、40%、45%、50%、55%、60%、65%、70%、75%、80%、85%、90%、95%または100%の修飾ヌクレオチド)を含んでもよい。所定の核酸分子に存在する修飾ヌクレオチドの実際のパーセンテージは、核酸に存在するヌクレオチドの総数に依存する。核酸分子が一本鎖の場合、パーセント修飾は一本鎖核酸分子に存在するヌクレオチドの総数に基づくことができる。同様に、核酸分子が二本鎖の場合、パーセント修飾はセンス鎖、アンチセンス鎖またはセンス鎖およびアンチセンス鎖の両方に存在するヌクレオチドの総数に基づくことができる。
例示的なパターンを以下でさらに詳細に提供するが、本明細書に開示される、および、当該技術分野において既知の、核酸分子の全ての可能な特徴(例えば、特徴は、限定されずに、センス鎖の長さ、アンチセンス鎖の長さ、二重鎖領域の長さ、オーバーハングの長さ、二本鎖核酸分子の一方または両方の末端が平滑であるか、または、オーバーハングを有するか、修飾核酸の数、修飾の種類、ダブルオーバーハング核酸分子が、各側のオーバーハングに同じか、または、異なる数のヌクレオチドを有するか、核酸分子において用いられている修飾は1種類か、または1種類を超えるか、および、連続する修飾/非修飾ヌクレオチドの数を含む)を有する全てのパターンの置換が企図される。以下で提供される全ての詳細な例に関して、二重鎖領域は19ヌクレオチドであることが示されているが、二重鎖領域の各々の鎖は独立して17〜49ヌクレオチドの長さであることができるため、本明細書において提供される核酸分子は1〜49ヌクレオチドの長さの範囲の二重鎖領域を有することができる。例示的なパターンを本明細書において提供する。
本明細書において提供される核酸分子(例えばsiNA分子)は、ニックまたはギャップを有する鎖、好ましくはセンス鎖を有してもよい。このように、核酸分子は、例えば、国際特許出願PCT/US07/081836に開示されている部分二重鎖RNA(mdRNA)のような、3本または4本以上の鎖を有してもよい。ニックまたはギャップを有する鎖を有する核酸分子は、約1〜49ヌクレオチドであってもよく、または、RISC長(例えば約15〜25ヌクレオチド)またはダイサー基質長(例えば約25〜30ヌクレオチド)、例えば本明細書に開示されているものなどであってもよい。
一部の態様において、本明細書において提供される核酸分子(例えばsiNA分子)は、例えばRossi、米国特許出願20050244858に記載されているような、in vivoでプロセシングされ、活性な核酸分子を生成する前駆体「ダイサー基質」分子、例えば二本鎖核酸であってもよい。一部の条件および状況において、これらの比較的長いdsRNA siNA種、例えば約25〜約30ヌクレオチドのものが、効力および作用持続時間に関して予想外に効果的な結果をもたらすことができることが見出されている。いかなる特定の理論に束縛されることを望むものではないが、より長いdsRNA種は、細胞の細胞質における酵素ダイサーの基質として用いられると考えられる。二本鎖核酸をより短いセグメントに切断することに加えて、ダイサーは、切断されたdsRNAに由来する一本鎖切断産物の、標的遺伝子に由来する細胞質内RNAの破壊に関与するRNA誘導サイレンシング複合体(RISC複合体)への組込みを容易にすることができる。
熱ショックタンパク質47(HSP47)は、コラーゲン特異的分子シャペロンであり、小胞体に存在する。これは、フォールディング、組立ておよび小胞体からの輸送プロセスの間、プロコラーゲンと相互作用する(Nagata Trends Biochem Sci 1996; 21:22-6、Razzaque et al. 2005; Contrib Nephrol 2005; 148: 57-69、Koide et al. 2006 J. Biol. Chem.; 281: 3432-38、Leivo et al. Dev. Biol. 1980; 76:100-114、Masuda et al. J. Clin. Invest. 1994; 94:2481-2488、Masuda et al. Cell Stress Chaperones 1998; 3:256-264)。HSP47は種々の組織線維症(Koide et al. 広範なコラーゲン型とのHSP47の特異的な結合は、HSP47を線維症の処置の標的とする。hsp47発現の阻害は、細胞外コラーゲンI分泌を防止することができる。Sato et al.(Nat Biotechnol 2008; 26:431-442)は、hsp47発現の阻害のためにsiRNAを用い、ラットにおいて肝線維症の進行を防止するによって、この可能性を探った。同様に、Chen et al.(Br J Dermatol 2007; 156: 1188-1195)およびWang et al.(Plast. Reconstr Surg 2003; 111: 1980-7)は、RNA干渉技術によるhsp47発現阻害を調査した。
線維性疾患は、一般的に、細胞外マトリックス中の線維状物質の過剰な堆積を特徴とし、それは組織構造の異常な変化の一因となり、正常な器官の機能を妨げる。
眼の手術に起因する瘢痕組織の収縮は頻繁に生じ得る。新たな排出路を形成するための緑内障手術は組織の瘢痕化と収縮のために失敗することが多く、形成した排出システムが閉塞することがあり、さらなる外科的介入が必要となる。現行の抗収縮レジメン(マイトマイシンCまたは5FU)は、関連する合併症(例えば失明)によって制限されている(例えば、Cordeiro MF, et al., Human anti-transforming growth factor-beta2 antibody: a new glaucoma anti-scarring agent Invest Ophthalmol Vis Sci. 1999 Sep;40(10):2225-34参照)。角膜外傷または角膜手術(例えば組織の収縮が不正確な結果をもたらすことがある近視または屈折異常のためのレーザーまたは外科的処置)の後に形成される瘢痕組織の収縮もあり得る。瘢痕組織は、例えば硝子体液または網膜の上/内部に形成されることがあり、一部の糖尿病患者においては最終的に失明を生じ得、網膜剥離手術後に形成されることがあり、増殖性硝子体網膜症(PVR)と呼ばれている。PVRは網膜剥離後の最もよく見られる合併症であり、網膜孔または網膜裂孔と関係している。PVRは、網膜色素上皮細胞を含む、硝子体腔内および網膜前面および後面上の細胞膜の増殖を指す。本質的に瘢痕組織であるこれらの膜は網膜を牽引し、当初は成功した網膜剥離治療の後でさえ、網膜剥離の再発をもたらすことがある。
肝線維症(LF)は、複数の病因の肝損傷の一般的に不可逆的な結果である。欧米諸国において、主な病因学的カテゴリーは、アルコール性肝疾患(30〜50%)、ウイルス性肝炎(30%)、胆管疾患(5〜10%)、原発性ヘモクロマトーシス(5%)、ならびに薬剤性肝硬変および病因不明の特発性肝硬変(10〜15%)である。ウイルソン病、α1アンチトリプシン欠損症および他の奇病も、肝線維症を症状の1つとして有する。肝線維症の末期である肝硬変は、肝移植を要することが多く、欧米諸国における死因の上位10位以内に入っている。

慢性腎不全は、老廃物を排出し、尿を濃縮し、電解質を維持する腎臓の能力の漸次的かつ進行性の喪失である。CRFは、ゆっくりと進行する。それは最も多くの場合、腎機能の漸次的な喪失をもたらす任意の疾患から生じ、線維症はCRFを生じさせる主な病態である。
糖尿病性腎障害(その特徴は糸球体硬化症および尿細管間質性線維症である)は、現代社会における末期腎疾患の唯一のもっとも頻度の高い原因であり、糖尿病患者は透析を受ける最大の人口を構成する。かかる治療は効果であり、決して最適なものではない。移植はより良い結果を提供するが、提供者の深刻な不足に悩まされている。
慢性腎臓病(CKD)は、世界的な公衆衛生上の問題であり、心臓血管疾患および慢性腎不全(CRF)の増大した危険性と関連している、よく見られる状態であると認識されている。
ステージ1:正常または亢進したGFR(>90mL/分/1.73m2)を伴う腎臓損傷
ステージ2:GFRの軽度低下(60〜89mL/分/1.73m2)
ステージ3:GFRの中程度低下(30〜59mL/分/1.73m2)

約100万個のネフロンが各々の腎臓に存在し、各々が総GFRに寄与する。腎損傷の病因論にかかわりなく、ネフロンの漸進性破壊を受けると、腎臓は、残存する健康なネフロンの過剰ろ過および代償性肥大によって、GFRを維持することができる。このネフロンの適応性により、血漿溶質の継続的な正常なクリアランスが可能となり、総GFRが50%に低下した後に、腎臓の余力が使い果たされて初めて、尿素およびクレアチニンなどの物質の血漿レベルが顕著な上昇を示し始める。血漿クレアチニン値は、GFRの50%の低下でほぼ2倍になる。したがって、患者における、0.6mg/dLのベースライン値から1.2mg/dLへの血漿クレアチニンの倍加は、機能するネフロン数の50%の消失を実質的に表す。
間質性肺線維症(IPF)は、鉱物粒子、有機ダストおよびオキシダントガスを含む種々の吸入物質によって、または未知の理由(特発性肺線維症)に起因する肺の瘢痕形成である。この疾患は世界中で何百万もの人を苦しめており、効果的治療手段がない。有用な処置を欠く主な理由は、治療のための適切な標的を設計するために十分に明らかにされた疾患の分子機構がほとんどないことである(Lasky JA., Brody AR. (2000), “Interstitial fibrosis and growth factors”, Environ Health Perspect.;108 Suppl 4:751-62)。
心不全のみの有病率が高まる一方、他の状態は顕著に減少しているという点で、心不全は、心血管障害の中でユニークな存在である。その一部の原因は、米国および欧州の人口の高齢化に帰することができる。心筋の損傷を有する患者を救助する能力もまた主な要因であるが、それは、これらの患者が心臓の有害なリモデリングによる左室機能不全の進行を起こすことがあるためである。
特に線維性疾患で起こり得る特有の問題は、組織の収縮、例えば瘢痕の収縮である。細胞外マトリックス成分を含む組織の収縮、特にコラーゲン含有組織の収縮は、多くの異なる病的状態、および外科的または美容的手法に関連して生じることがある。例えば、瘢痕の収縮は、医学的処置が必要となり得るの身体的な問題を引き起こすことがあり、または、純粋に美容的性質の問題を引き起こすことがある。コラーゲンは、瘢痕および他の収縮した組織の主成分であり、したがって、考慮すべき最も重要な構造的成分である。とはいえ、瘢痕および他の収縮した組織はまた、他の構造的成分、特に他の細胞外マトリックス成分、例えばエラスチンを含み、これもまた組織の収縮に関与し得る。
核酸分子は、線維性の(例えば、肝、腎、腹膜、肺)疾患、形質、状態および/または障害および/または、細胞または組織におけるhsp47のレベルに関連する、または、反応する他のあらゆる形質、疾患、障害または状態を、単独でまたは他の治療法と組み合わせて、予防または処置することに使用するために適応させることができる。核酸分子は、リポソームを含む、対象に投与するための送達ビヒクル、担体および希釈剤およびその塩を含んでいてもよく、および/または、薬学的に許容し得る製剤中に存在してもよい。
投与すべき有用な投与量および具体的な投与方法は、当業者に直ちに明らかであるように、細胞種、または、in vivoでの使用については、年齢、体重、および処置する動物およびその領域、使用する具体的な核酸と送達方法、考慮される治療的もしくは診断的用途、および製剤の剤形(例えば、懸濁剤、エマルション、ミセルもしくはリポソーム)などの要因に応じて異なる。典型的には、投薬量はより低いレベルで投与され、所望の効果が達成されるまで増量する。
また提供されるのは、本明細書において提供されるhsp47の発現を低減するための核酸分子(例えばsiNA分子)を含む、該核酸分子を患者に投与または分配するための組成物、キット、容器および製剤である。キットは、少なくとも1個の容器と少なくとも1枚のラベルとを含んでもよい。好適な容器は、例えば、ボトル、バイアル、シリンジ、および試験管を含む。容器は、種々の材料、例えばガラス、金属またはプラスチックから形成されていてもよい。容器は、1または2以上のアミノ酸配列、1または2以上の小分子、1または2以上の核酸配列、1または2以上の細胞集団および/または1または2以上の抗体を保持していてもよい。一態様において、容器は、細胞のmRNA発現プロファイルを検査するためのポリヌクレオチドを、この目的のために用いる試薬と共に保持する。別の態様において、容器は、細胞および組織におけるhsp47タンパク質の発現の評価に用いるための、または関連する実験室用途、予後判定用途、診断用途、予防用途および治療用途のための抗体、その結合断片または特異的結合タンパク質を含む。これらの用途のための指示および/または使用法が、かかる容器上またはかかる容器と共に含まれていてもよい。これらの目的のために用いる試薬および他の組成物またはツールが、同様に含まれていてもよい。キットは、関連する指示および/または使用法をさらに含んでもよく、かかる目的のために用いる試薬および他の組成物またはツールが含まれていてもよい。
本明細書に開示される核酸分子は、hsp47に関連する疾患、状態または障害、例えば肝線維症、肝硬変、肺線維症、腎線維症、腹膜線維症、慢性肝損傷および原線維形成など、ならびに、細胞または組織におけるhsp47のレベルに関係するか、または反応するあらゆる他の疾患または状態を、単独でまたは他の治療法と組み合わせて処置するのに用いることができる。したがって、本明細書に開示される組成物、キットおよび方法は、ラベルまたは添付文書を含む、本明細書に開示される核酸分子の包装を含んでもよい。ラベルは、核酸分子の使用法、例えば、肝線維症、腹膜線維症、腎線維症および肺線維症、ならびに、細胞または組織におけるhsp47のレベルに関係するか、または反応するあらゆる他の疾患または状態の、単独での、または他の治療法と組み合わせての処置または予防のための使用法を含んでもよい。ラベルは、hsp47の発現の低減における使用法を含んでもよい。「添付文書」は、治療用製品の商用パッケージに通例含まれる指示を指すのに用い、それは、適応症、用法、用量、投与、禁忌、包装された製品と併用すべき他の治療用製品および/またはかかる治療用製品の使用に関する警告などに関する情報を含む。
hsp47核酸分子配列の選択
Hsp47に対する核酸分子(例えば、25ヌクレオチド以下のsiNA)は、siRNA at Whitehead(Whitehead Institute for Biomedical Research)、IDT siRNA Design(Integrated DNA Technologies)、BLOCK-iT RNAi Designer(Invitrogen)、siDESIGN Center(Dharmacon)、およびBIOPREDsi(Friedrich Miescher Institute for Biomedical Research、Novartis Research Foundationの一部、http://www.biopredsi.org/start.htmlで利用可能)を含む複数のコンピュータプログラムを用いて設計した。これらのプログラムから最高のスコアを得たsiRNAの配列を比較し、アルゴリズムならびにヒトとラットとの配列相同性に基づいて選択した(表1参照)。候補配列は、in vitroノックダウンアッセイにより有効性を確認した。

ヒトおよびラットhsp47遺伝子の両方に対する種々のsiNA分子の有効性をスクリーニングするために、293、HT1080、ヒトHSC系hTERT、またはNRK細胞系へのヒトHSP47 cDNA−緑蛍光タンパク質(GFP)またはラットGP46 cDNA−GFP構築物のレンチウイルスによる誘導により、種々のレポーター細胞系を樹立した。これらの細胞系は、GFPに対するsiRNAによってさらに評価した。残存する蛍光シグナルを測定し、スクランブルsiRNA(Ambion)に対して正規化し、次いで細胞生存率に対して正規化した。結果は、GFPに対するsiRNAが、異なる細胞系において異なる程度で蛍光をノックダウンすることを示した(図1)。293_HSP47−GFPおよび293_GP46−GFP細胞系を、そのトランスフェクションの容易性と、蛍光ノックダウンに対する感度から、siHsp47のスクリーニングのために選択した。
細胞は、96穴組織培養プレートにて、Lipofectamine RNAiMAX(Invitrogen)を用い、リバーストランスフェクション法により、1ウェルあたりGFPに対するsiNA1.5pmolでトランスフェクションした。細胞は、1ウェルあたり6,000個を播種し、siNA複合体と混合した。蛍光の読み取りは、72時間のインキュベーションの後、Synergy 2 Multi-Mode Microplate Reader(BioTek)で行った。
siNA有りまたは無しで処理した細胞の生存率を、72時間のインキュベーションの後、CellTiter-Glo Luminescent Cell Viability Assay Kit(Promega)用い、マニュアルに従って測定した。測定値は、スクランブルsiNA分子で処理したサンプルに対して正規化した。
レポーター細胞系におけるhsp47発現に対するsiHsp47の阻害効率の評価
hsp47に対するsiNAは、レポーターGFPからの蛍光シグナルの変化を評価することによって、293_HSP47−GFPおよび293_GP46−GFP細胞系におけるその阻害効率について評価した。実験は、例2に記載したとおりに行った。蛍光シグナルは、対照として用いた、スクランブルsiRNA(Ambion)で処理した細胞からの蛍光シグナルに対して正規化した。結果は、試験したhsp47 siNA分子が、両方の細胞系においてhsp47 mRNAを阻害するのに効果的だったことを示す。しかしながら、GP46 mRNAに対するsiNA(2008年のSatoらの論文で公表されたもの)は、293_GP46−GFP細胞系においてのみ有効だった。結果を図2A〜Bに示す。
TaqMan (R) qPCRによるhsp47 mRNAに対するsiHsp47の阻害効果の評価
例3では、レポーター細胞系におけるsiHsp47のノックダウン効率を、蛍光シグナルの変化によって評価した。結果をmRNAレベルで確認するために、内因性のhsp47を標的とするsiRNAを、例2に記載のとおり、Lipofectamine RNAiMAX(Invitrogen)を用い、リバーストランスフェクション法により、ヒトHSC細胞系hTERT細胞にトランスフェクションした。
siHsp47のノックダウン効果のタンパク質レベルでの確認
hsp47 mRNA発現に対する種々のHsp47 siNA分子(siHsp47)の阻害効果を、種々のsiHsp47をトランスフェクションしたhTERT細胞におけるHSP47を測ることにより、タンパク質レベルで確認した。種々のsiHsp47のhTERT細胞へのトランスフェクションは、例2に記載のとおりに行った。トランスフェクションしたhTERT細胞は溶解し、細胞溶解物を遠心分離により清澄化した。清澄化した細胞溶解物中のタンパク質を、SDSポリアクリルアミドゲル電気泳動によって分割した。細胞溶解物中のHSP47タンパク質のレベルを、抗HSP47抗体(Assay Designs)を一次抗体として、HRPとコンジュゲートしたヤギ抗マウスIgG(Millipore)を二次抗体として用い、その後Supersignal West Pico Chemiluminescence kit(Pierce)で検出して決定した。抗アクチン抗体(Abcam)を、タンパク質負荷対照として用いた。結果は、siHsp47−C、siHsp47−2dのみ、または、siHsp47−1とsiHsp47−2dとの組合せで処理した細胞における、Hsp47タンパク質のレベルの顕著な減少を示した。
hsp47 siRNAによるコラーゲンI発現の下方調節
コラーゲンI発現レベルに対するsiHsp47の効果を決定するために、hsp47に対する種々のsiRNAで処理したhTERT細胞におけるコラーゲンI mRNAレベルを測定した。簡潔に述べると、hTERT細胞に種々のsiHsp47を例2に記載のとおりにトランスフェクションした。細胞を72時間後に溶解し、mRNAをRNeasy mini kitを用い、マニュアル(Qiagen)に従って単離した。コラーゲンI mRNAのレベルは、TaqMan(R)プローブを用いた定量PCRと組み合わせた逆転写により決定した。簡潔に述べると、cDNA合成を、High-Capacity cDNA Reverse Transcription Kit(ABI)を用い、メーカーの指示に従って行い、TaqMan Gene Expression Assay(ABI、COL1A1アッセイID Hs01076780_g1)に供した。コラーゲンI mRNAのレベルは、メーカーの指示(ABI)に従って、GAPDH mRNAのレベルに対して正規化した。シグナルは、スクランブルsiNAをトランスフェクションした細胞から得られるシグナルに対して正規化した。結果は、図4に示すとおり、コラーゲンI mRNAレベルが、候補のいくつか、siHsp47−2、siHsp47−2d、およびこれらとsiHsp47−1との組み合せで処理した細胞において顕著に低下したことを示した。
hsp47 siRNAで処理したhTERT細胞の免疫蛍光染色
siHsp47有りまたは無しでトランスフェクションしたhTERT細胞における2種の線維症マーカー、コラーゲンIおよびα平滑筋アクチン(SMA)の発現を視覚化するために、細胞をウサギ抗コラーゲンI抗体(Abcam)およびマウス抗アルファSMA抗体(Sigma)で染色した。Alexa Fluor 594ヤギ抗マウスIgGおよびAlexa Fluor 488ヤギ抗ウサギIgG(Invitrogen(Molecular Probes))を、コラーゲンI(緑)およびアルファSMA(赤)を視覚化する二次抗体として用いた。Hoeschtを、核(青い)を視覚化するために用いた。結果は、標的遺伝子の一部のsiRNAによるノックダウンと、コラーゲン/SMA発現との間の相関関係を示す。

独自のアルゴリズムと標的遺伝子の既知の配列を用いて、多くの潜在的siRNAの配列を生成した。この方法を用いて生成した配列は、対応するmRNA配列に相補的である。
ヒトおよびラットSERPINH1遺伝子に対するsiRNAオリゴのためのロースループットスクリーニング(LTS)
細胞系:ヒト前立腺腺癌PC3細胞(ATCC, Cat# CRL-1435)は10%のFBSおよび2mMのL−グルタミンを添加したRPMI培地で生育し、ヒト上皮性子宮頸癌HeLa細胞(ATCC, Cat#CCL-2)は10%のFBS、2mMのL−グルタミンを添加したダルベッコ改変イーグル培地(DMEM)で維持した。細胞は、37℃にて5%CO2で維持した。
SERPINH1遺伝子を内因性に発現する約2×105個のヒトPC−3細胞を、24時間後に30〜50%コンフルエントに達するよう、1.5mlの増殖培地に接種した。細胞に、SERPINH1二本鎖RNA化合物(すなわちSERPINH1_2、4、6、12、13、18、45、51、58、88)を、0.0029〜100nMの範囲の最終的なトランスフェクション濃縮に達するよう、LipofectamineTM2000試薬でトランスフェクションした。陰性対照として、細胞をLipofectamineTM2000試薬または20〜100nMの終濃度の合成ランダム化配列の非標的化siRNAで処理する。Cy3標識siRNAをトランスフェクションした細胞を、トランスフェクション効率についての陽性対照として用いた。

SERPINH1遺伝子を内因性に発現するヒトPC−3細胞を、1ウェルあたり約2×105個で6ウェルプレートに接種し、約24時間生育させて30〜50%コンフルエントにした。細胞に、試験するsiRNAを種々の濃度で、LipofectamineTM2000試薬(Invitrogen)を用いてトランスフェクションした。細胞を、5%CO2インキュベーター内で37℃にて、48時間または72時間インキュベーションした。トランスフェクションの48〜72時間後、細胞を採取し、細胞のRNAを抽出した。Cy3で標識したsiRNA二重鎖を、トランスフェクション効率についての陽性対照として用いた。LipofectamineTM2000試薬で処理したmock細胞を「対照非活性サンプル」(陰性対照)と定義し、5nMの終濃度の既知の活性なsiRNA(HSP47−C)で処理した細胞を「対照活性サンプル」(陽性対照)と定義した。Z’および対照倍数(controls fold){倍数=平均(陰性)/平均(陽性)}は、アッセイ効率を記述する手段である。
psiCHECKシステムは、ガイド鎖(GS)(アンチセンス)およびパッセンジャー鎖(PS)(センス鎖)が、標的化(オンターゲット)効果およびオフターゲット効果を引き起こすかを、その標的配列の発現レベルの変化をモニターすることによって評価することができる。各々の試験分子のGSおよびPS鎖の標的活性と、生じ得るオフターゲット活性の評価のために、4個のpsiCHECKTM-2に基づく(Promega)構築物を調製した。各々の構築物において、試験分子のGSおよびPSの完全な標的配列またはシード(seed)標的配列のいずれかの1コピーまたは3コピーを、3’−UTR領域のウミシイタケルシフェラーゼの翻訳停止コドンの下流に位置するマルチクローニングサイトにクローニングした。

本発明による修飾化合物を、ヒト血清またはヒト組織抽出物中の二重鎖の安定性について以下のように試験する:
7μMの終濃度のsiRNA分子を、100%のヒト血清(Sigma Cat# H4522)中、37℃でインキュベーションする。(100μMのsiRNA原液を、ヒト血清または種々の組織種からのヒト組織抽出物に1:14.29で希釈)。5μlを、異なる時点(例えば、0、30分、1時間、3時間、6時間、8時間、10時間、16時間および24時間)で、15μlの1.5×TBEローディングバッファーに添加する。サンプルを、直ちに液体窒素中で凍結させ、−20℃に維持した。
各々のサンプルを、当該技術分野において既知の方法によって調製した非変性の20%アクリルアミドゲルに負荷する。オリゴを、紫外線下、エチジウムブロマイドで視覚化した。
核酸分子に対する3’非ヌクレオチド部分の安定化効果を調査するため、センス鎖、アンチセンス鎖およびアニーリングしたsiRNA二重鎖を、種々の細胞種から調製した細胞質抽出物中でインキュベーションする。
抽出物:HCT116細胞質抽出物(12mg/ml)。
抽出物バッファー:25mMのHepes 37℃でpH7.3、8mMのMgCl、1mMのDTTを含む150mMのNaClを、使用の直前に新たに加えた。
方法:3.5mlの試験siRNA(100mM)を、120mgのHCT116細胞質抽出物を含む46.5mlと混合した。この46.5mlは、12mlのHCT116抽出物と、DTTおよびプロテアーゼインヒビターカクテル/100(Calbiochem, setIII-539134)を添加した34.5mlの抽出バッファーとからなる。インキュベーションチューブ中のsiRNAの終濃度は7mMである。サンプルを37℃でインキュベーションし、示した時点で、5mlを新たなチューブに移し、15mlの1×TBE−50%グリセロールローディングバッファーと混合し、液体窒素で急速凍結した。ローディングバッファー中のsiRNAの終濃度は、1.75mM(21ng siRNA/ml)である。非変性PAGEおよびEtBr染色による分析については、レーンあたり50ngを負荷する。ノーザン分析については、レーンあたり1ngの試験siRNAを負荷した。
新鮮なヒト血液を(RTで)、無菌の0.9%NaClとRTにて1:1の比率で混合し、フィコール(Lymphoprep, Axis-Shield cat# 1114547)に穏やかに負荷した(1:2の比率)。サンプルを、スイング式遠心機でRTにて(22℃、800g)30分間遠心分離し、RPMI1640培地で洗浄し、10分間遠心分離した(RT、250g)。細胞を計数し、増殖培地(なRPMI1640+10%FBS+2mM L−グルタミン+1%ペニシリン−ストレプトマイシン)中に1.5×106個/mlの終濃度で播種し、siRNA処理の前に37℃で1時間インキュベーションした。
次いで、細胞を、LipofectamineTM2000試薬(Invitrogen)を用い、メーカーの指示に従って、種々の濃度の試験するsiRNAで処理し、5%CO2インキュベーター内で37℃にて24時間インキュベーションした。

線維性状態のモデル系
本発明の活性なsiRNAの試験は、予測動物モデルで行うことができる。腎線維症のラット糖尿病およびエイジングモデルは、Zucker糖尿病肥満(ZDF)ラット、老齢fa/fa(肥満Zucker)ラット、老齢Sprague-Dawley(SD)ラットおよびGoto Kakizaki(GK)ラットを含む。GKラットは、Wisterラットに由来し、NIDDM(II型糖尿病)の自然発生について選択された近交系である。腎線維症の誘導モデルは、健康な非糖尿病動物に生じる急性間質性線維症のモデルである、恒常的片側性尿管閉塞(UUO)モデルを含み、腎線維症は、閉塞後で数日のうちに発症する。腎線維症の別の誘導モデルは、5/6腎摘出術である。
本発明の活性なsiRNAの緑内障の処置または予防に関する試験は、例えば、Maeda, K. et al., “A Novel Neuroprotectant against Retinal Ganglion Cell Damage in a Glaucoma Model and an Optic Nerve Crush Model in the rat”, Investigative Ophthalmology and visual Science (IOVS), March 2004, 45(3)851に記載の、視神経挫滅ラット動物モデルで行う。具体的には、視神経切断のために、麻酔下ラットの眼窩の視神経(ON)を眼窩上アプローチで露出し、髄膜を切断し、篩板から2mmのON内の全ての軸索を、10秒間鉗子で挫滅することにより切断する。

Claims (20)
- 下記の構造(A2):

ここで、N2、NおよびN’の各々は、非修飾リボヌクレオチドまたは修飾リボヌクレオチドまたは非定型部分であり、
(N)xおよび(N’)yの各々は、各々の連続するNまたはN’が隣接するNまたはN’に共有結合によって連結したオリゴヌクレオチドであり、
xとyの各々は、独立して17〜39の間の整数であり、
(N’)yの配列は(N)xの配列に相補性を有し、(N)xは、hsp47をコードするmRNAの連続する配列に相補性を有し、
N1は(N)xに共有結合的に結合しており、hsp47をコードするmRNAに対してミスマッチであるか、または、hsp47をコードするmRNAに対する相補的DNA部分であり、
N1は、天然または修飾ウリジン、デオキシリボウリジン、リボチミジン、デオキシリボチミジン、アデノシンまたはデオキシアデノシンからなる群から選択される部分であり、
z’’は存在しても、または不在であってもよいが、存在する場合はN2−(N’)yの5’末端に共有結合的に付着したキャッピング部分であり、
ZおよびZ’の各々は、独立して、存在するか、または不在であるが、存在する場合は、独立して、それが存在する鎖の3’末端に共有結合的に付着した、1〜5個の連続するヌクレオチド、連続する非ヌクレオチド部分またはその組合せであり、
センス鎖は配列番号101に記載の配列を含み、アンチセンス鎖は配列番号168に記載の配列を含む
を有する、二本鎖核酸化合物。 - x=y=18である、請求項1に記載の二本鎖核酸化合物。
- 構造
A、C、GおよびUの各々は、独立して、非修飾もしくは修飾リボヌクレオチド、または非定型部分であり、
A、C、GおよびUの各々は、隣接するA、C、GまたはUに共有結合で連結しており、
ZおよびZ’の各々は、独立して、存在するか、または不在であるが、存在する場合は、独立して、それが存在する鎖の3’末端に共有結合的に付着した、1〜5個の連続するヌクレオチド、または非ヌクレオチド部分またはその組合せであり、
z’’は存在しても、または不在であってもよいが、存在する場合はセンス鎖の5’末端に共有結合的に付着したキャッピング部分である、請求項1または2に記載の二本鎖核酸化合物。 - A、C、GおよびUの各々は、非修飾リボヌクレオチドであり、z’’、ZおよびZ’は不在である、請求項3に記載の二本鎖核酸化合物。
- ZおよびZ’の各々が存在する、請求項3に記載の二本鎖核酸化合物。
- ZおよびZ’の各々が、独立して、ヌクレオチド部分または非ヌクレオチド部分を含む、請求項5に記載の二本鎖核酸化合物。
- Z’がC3PiまたはC3OHを含み、ZがC3Pi−C3OHを含む、請求項5または6に記載の二本鎖核酸化合物。
- アンチセンス鎖が、1、8および15位(5’>3’)に2’−O−メチル糖修飾リボヌクレオチドを含む、請求項3、5〜7のいずれか一項に記載の二本鎖核酸化合物。
- アンチセンス鎖が、5、6、7または8位(5’>3’)に2’5’リボヌクレオチドを含む、請求項3、5〜8のいずれか一項に記載の二本鎖核酸化合物。
- センス鎖が、9または10位(5’>3’)に2’5’リボヌクレオチドを含む、請求項3、5〜9のいずれか一項に記載の二本鎖核酸化合物。
- センス鎖が、4、11、13および17位(5’>3’)に2’−O−メチル糖修飾リボヌクレオチドを含む、請求項3、5〜10のいずれか一項に記載の二本鎖核酸化合物。
- z’’が存在し、キャッピング部分が、無塩基リボース部分およびその修飾体、逆位無塩基リボース部分およびその修飾体、逆位無塩基デオキシリボース部分およびその修飾体、無塩基デオキシリボース部分およびその修飾体、C6−イミノ−Pi、ミラーヌクレオチド、5’OMeヌクレオチド、4’,5’−メチレンヌクレオチド、1−(β−D−エリスロフラノシル)ヌクレオチド、4’チオヌクレオチド、炭素環式ヌクレオチド、5’−アミノ−アルキルホスフェート、1,3−ジアミノ−2−プロピルホスフェート、3−アミノプロピルホスフェート、6−アミノヘキシルホスフェート、12−アミノドデシルホスフェート、ヒドロキシプロピルホスフェート、1,5−アンヒドロヘキシトールヌクレオチド、アルファ−ヌクレオチド、トレオ−ペントフラノシルヌクレオチド、非環式3’,4’−セコヌクレオチド、3,4−ジヒドロキシブチルヌクレオチド、3,5−ジヒドロキシペンチルヌクレオチド、5’−5’−逆位無塩基部分、1,4−ブタンジオールホスフェート、5’−アミノおよび架橋または非架橋メチルホスホネートおよび5’−メルカプト部分から選択される、請求項3、5〜11のいずれか一項に記載の二本鎖核酸化合物。
- A、C、GおよびUの各々を隣接するA、C、GまたはUに連結する共有結合が、ホスホジエステル結合である、請求項3〜12のいずれか一項に記載の二本鎖核酸化合物。
- センス鎖が、4、11、13および17位(5’>3’)における2’−O−メチル糖修飾リボヌクレオチド、3’末端に共有結合的に付着したC3PiまたはC3OH非ヌクレオチド部分、および5’末端に共有結合的に付着した逆位無塩基キャッピング部分を含み、アンチセンス鎖が、
a)1、8および15位(5’>3’)における2’−O−メチル糖修飾リボヌクレオチド、6または7位(5’>3’)における2’5’リボヌクレオチド、3’末端に共有結合的に付着したC3Pi−C3OH非ヌクレオチド部分、または、
b)1、4、8、13および15位(5’>3’)における2’−O−メチル糖修飾リボヌクレオチド、6または7位(5’>3’)における2’5’リボヌクレオチド、3’末端に共有結合的に付着したC3Pi−C3OH非ヌクレオチド部分、または、
c)1、4、8、11および15位(5’>3’)における2’−O−メチル糖修飾リボヌクレオチド、6位(5’>3’)における2’5’リボヌクレオチド、3’末端に共有結合的に付着したC3Pi−C3OH非ヌクレオチド部分、または、
d)1、3、8、12、13および15位(5’>3’)における2’−O−メチル糖修飾リボヌクレオチド、6位(5’>3’)における2’5’リボヌクレオチド、3’末端に共有結合的に付着したC3Pi−C3OH非ヌクレオチド部分
を含む、請求項3に記載の二本鎖核酸化合物。 - センス鎖が、9または10位(5’>3’)における2’5’リボヌクレオチドをさらに含む、請求項14に記載の二本鎖核酸化合物。
- センス鎖が、4、11、13および17位(5’>3’)における2’−O−メチル糖修飾リボヌクレオチド、9位(5’>3’)における2’5’リボヌクレオチド、3’末端に共有結合的に付着したC3OH非ヌクレオチド部分、および5’末端に共有結合的に付着した逆位無塩基キャッピング部分を含み、アンチセンス鎖が、(5’>3’)1、4、8、11および15位(5’>3’)における2’−O−メチル糖修飾リボヌクレオチド、6位(5’>3’)における2’5’リボヌクレオチド、3’末端に共有結合的に付着したC3Pi−C3OH非ヌクレオチド部分を含む、請求項15に記載の二本鎖核酸化合物。
- 請求項1〜16のいずれか一項に記載の二本鎖核酸化合物を含む組成物。
- 請求項1〜16のいずれか一項に記載の二本鎖核酸化合物を含む、線維症を処置するための組成物。
- 線維症が、肝線維症、肝硬変、肺線維症、腎線維症、腹膜線維症、慢性肝損傷、心線維症、網膜線維症、眼窩線維症、涙腺線維化、骨髄線維症、腸管線維症、声帯粘膜線維症、喉頭線維症、腎性全身性線維症、先天性肝線維症および口腔粘膜下線維症からなる群から選択される、請求項18に記載の組成物。
- 線維症が、皮膚、腹膜、肝臓、膵臓、腎臓、心臓、肺、骨髄、眼、腸管、声帯および/または脈管の線維症である、請求項18に記載の組成物。
