Description
本発明の種々の側面および実施形態を例示するために、以下に実施例を提供する。これらの実施例は、開示された発明を限定することを意図するものでは全くなく、特許請求の範囲によってのみ限定される。
初めに、ゲノムDNAライブラリーをChromobacterium sp.USM2から単離した。Chromobacterium sp.USM2を50mlの富栄養培地(1%ペプトン、1%肉エキス、0.5%酵母エキス、pH7.0)において30℃で一晩培養し、その後、ゲノムDNAを標準法を用いて微生物から得た。
初めに、ApaI−SalI高分子合成酵素遺伝子を、あらかじめ同じ制限酵素を用いて切断したクローニングベクターpGEM−(Promega)に挿入した。その後、断片をApaIおよびSalI制限酵素を用いて再度消化し、得られたApaI−SalI高分子合成酵素遺伝子断片を、Cupriavidus属に属する微生物において発現可能な組換えベクターpBBR1MCS−2内に挿入した。得られた組換えプラスミドを接合伝達法によって、Cupriavidus necator PHB−4(DSM 541)(高分子を合成する能力を欠いた株)に形質転換した。
それぞれのC.necatorH16、C.necator形質転換体、およびPHB−4を、1ml/Lの微量元素を含む50mlの無機培地(3.32g/Lのリン酸水素二ナトリウム、2.8g/Lのリン酸二水素カリウム、0.54g/Lの尿素)に植え付け、フラスコ内において30℃でインキュベートした。50mg/LのカナマイシンをC.necator形質転換体の培地に添加し、微生物を48および72時間培養した。
吉草酸ナトリウムを培養24時間の時点で添加した。

aPHA生合成培地において72時間30℃、初期pH7.0、200rpmでインキュベート。

Claims (7)
- ポリ(3−ヒドロキシブチレート−co−3−ヒドロキシバレレート−co−3−ヒドロキシヘキサノエート)ランダム共重合体[P(3HB−co−3HV−co−3HHx)]合成酵素活性を有するSEQ ID NO:1に記載のアミノ酸配列を含むポリペプチドをコードする、単離されたポリヌクレオチド。
- SEQ ID NO:2に記載のヌクレオチド配列を含む請求項1に記載の単離されたポリヌクレオチド。
- TはUによって置換されるSEQ ID NO:2に記載のヌクレオチド配列を含む請求項1に記載の単離されたポリヌクレオチド。
- 請求項1に記載の単離されたポリヌクレオチドを含む組換えベクター。
- 請求項4に記載の組換えベクターであって、それはプラスミドまたはファージである組換えベクター。
- 請求項5に記載のベクターによって形質転換された形質転換体。
- P(3HB−co−3HV−co−3HHx)を産生するための方法であって、
重合性材料を含む培地において請求項6に記載の形質転換体を培養する段階と、
培養培地からP(3HB−co−3HV−co−3HHx)を回収する段階とを含む方法。
