Description
本発明は、新規アミドに関する。本発明の化合物は、アデノシン受容体に対して生物学的にテストされ、アデノシン受容体へ結合可能であることを驚くことに示し、機能的アッセイにおいて活性を有し、生体内で効果を示した。
従って、本発明の一つの観点は、アデノシン受容体に対して活性を示す低分子量化合物を提供することである。さらなる観点は、経口投与の後取りこまれ、かつ、脳血液関門を経て充分に通過可能な化合物を提供することである。
Zは、1、2、3、4または5、好ましくは1または2、最も好ましくは2つの炭素原子を有する飽和または不飽和、直鎖または分岐の鎖状の非環式炭化水素基であり;
R2は、アミノ、置換アミノ[例えば、アルキルアミノまたはジアルキルアミノ(例えば、メチルアミノ、ジメチルアミノ、エチルアミノ、ジエチルアミノ)]またはグアニジノ基から選択され;
R3、R4およびR5は、同一または異なっていてもよく、水素;ハロゲン;トリハロアルキル;1、2、3、4もしくは5の炭素原子を有するアルキル;1、2、3、4もしくは5の炭素原子を有するアルコキシ、トリハロアルコキシ、ヒドロキシまたはアミノから選択される電子供与性基;およびシアノ、スルホン、ニトロもしくはアミドから選択される電子受容性基から、独立して選択され;
R6、R7およびR8は、同一または異なっていてもよく、水素;ハロゲン;トリハロアルキル;1、2、3、4もしくは5の炭素原子を有するアルキル;1、2、3、4もしくは5の炭素原子を有するアルコキシ、トリハロアルコキシ、ヒドロキシまたはアミノから選択される電子供与性基;およびシアノ、スルホン、ニトロもしくはアミドから選択される電子受容性基から、独立して選択され;ならびに、
R9、R10およびR11は、同一または異なっていてもよく、水素;ハロゲン;トリハロアルキル;1、2、3、4または5の炭素原子を有するアルキル;1、2、3、4または5の炭素原子を有するアルコキシ、トリハロアルコキシ、ヒドロキシまたはアミノから選択される電子供与性基;およびシアノ、スルホン、ニトロまたはアミドから選択される電子受容性基から、独立して選択される。
別の観点では、本発明の化合物は、以下に示す合成スキームに従い製造することができる。
一般式(I)の化合物は、以下の方法により製造することができる。
以下の実施例は、本発明を説明することを意図しており、挙げられた化合物が特定の目的に特に興味深いものであったとしても、その範囲を限定するものではない。これらの化合物は、番号コードa:bにより呼ばれており、aはその化合物の製造を説明した実施例の番号を意味し、bは、その実施例に従い製造された化合物の順番を意味する。従って、実施例1:2は、実施例1に説明した方法に従い同様に製造された第2の化合物を意味する。
3−ブロモ−ベンズアルデヒド(1当量)、2−(1H−インドール−3−イル)−エチルアミン(1当量)、N−Boc−3−アミノプロパン酸(1当量)およびシクロヘキシルイソシアニド(1当量)を、15mlのメタノール中に溶解させた。反応物を室温で18時間攪拌させた。そのときまでに白色沈殿物が生じた。溶媒を蒸発させ、その沈殿物をメタノールで洗浄し、粗生成物を78%収率で得た。粗生成物をHCl/メタノールの混合物中1時間、40℃で加熱することによりBoc基を除去した。CH2Cl2:MeOH=6:1でシリカクロマトグラフィーを行うことにより純粋な生成物を得た。融点209℃。
[実施例2−アデノシンへの親和性]
この実施例は、式(I)の化合物およびその治療的に活性な酸付加塩の効力を示す。アデノシンA2A受容体に対する親和性および機能性は、Varaniら、表1により説明されたように本質的に行った(Varani, Pharmacological and biochemical characterization of purified A2A adenosine receptors in human platelet membranes by [3H]CGS21680 binding, Br.J. Pharmacol. 117:1693-1701, 1996)。
[滑膜線維芽細胞]
研究デザイン:抗原誘導関節炎を有するラットから、過増殖性滑膜、パンヌスを、疾患発症から4日に炎症膝から取り出した。そのパンヌス組織を、PEST(100IU ペニシリン、100μg/mlストレプトマイシン)およびフンギソン(Fungizone)(2.5μg/ml)(すべてスェーデンのインビトロジェン(InVitrogen)より)を含むPBS中で小片に切断し、その後、コラゲナーゼ(400U/ml、米国、ワージントン)中で3時間37℃、5%のCO2でインキュベーションした。細胞を遠心分離し(8分、室温、1100rpm.)、10%FCS(スェーデンのインビトロジェン)、PESTおよびフンギゾンを追加したRPMI 1640中に懸濁させ、25cm2フラスコ中に37℃、5%CO2で接種した。次の日、細胞を培地でゆすいで、さらにインンキュベートした。融合(confluent)したとき、細胞を1分間トリプシン化し(EDTAを含む0.25%トリプシン、スェーデンのインビトロジェン)、計数し、96ウェル・プレートに10000細胞/ウェル/200μlで接種した。
[軟骨外植]
IL−1誘導軟骨におけるNO放出に対する化合物の効果を以下に説明するようにして測定した。
皮膚を剥したウシ鼻(雌牛、18〜24ヶ月齢から)を、ヘービー(Hoerby)屠殺場(スェーデン、チーム・ウグラップ(Team Ugglarp))で集めた。軟骨をPEST(100IUペニシリン、100mg/mlストレプトマイシン)および2.5μg/mlフンギゾン(全てスェーデンのインビトロジェン)を含むPBS中に2時間室温で入れた後、鼻内部隔壁を切断して除去し、粘膜および軟骨膜を除去した。2mm片を軟骨の外に穿孔した。10μg/mlBSA、25mg/mlアスコルビン酸塩(両方ともスェーデンのシグマから)、PEST(100IUペニシリン、100mg/mlストレプトマイシン)および2.5μg/mlフンギゾンを追加した1mlの細胞培養培地、HAMs F12(スェーデンのインビトロジェン)を含む24ウェル細胞培養プレート(スェーデンのファルコン(Falcon))中に各片を入れた。24時間後、培地を取り替え、軟骨片をヒト組み換えIL−1a、10ng/ml(スェーデンのロッシュ)で誘導した。テスト化合物を5〜1000nMの適切な濃度で三重にテストした。軟骨組織を別途6日間インキュベートし、培地を3日ごとに交換した。毎回、培地をNO測定のため集めた(グリース反応)。
[抗炎症効果]
[コントロール]
メスBALB/cマウス(重量20〜22g)を0.5%の2,4−ジニトロフルオロベンゼン(DNFB)30μlで剃毛した腹を処理することにより、感作させた。4日後、これらのマウスの足に0.3%のDNFB10μlを感作させた。感作させていないマウス足をコントロールとして用いた。最後の感作から24時間後、足重量の相違を炎症(足浮腫)の指標として測定した。
マウスはコントロールとして処理したが、ただし、感作前2時間(0日)にプレドニゾロン(20mg/kg)を腹膜内へ注射(i.p.)または皮下注射(s.c.)をさらに行い、同じ量を4連続日間の間、感作後、繰返し投与した。足浮腫の阻害を前記のようにして測定した。
マウスをコントロールとして処理したが、ただし、感作前2時間(0日)に様々な量(0.05、0.15または0.25、0.375、0.5、0.75および最後のほうの研究では、1.5、3および時々6mg/kg)の各化合物を腹膜内へ注射i.p.または皮下注射(s.c.)をさらに行い、同じ量を4連続日間の間、感作後、繰返し投与した。足浮腫阻害は、前記のとおり。少なくとも10マウスを含むそれぞれの群を全ての実験に用いた。
[抗原誘導関節炎(AIA)]
ラットにおける抗原誘導関節炎(AIA)は、良好に再現可能な単独関節炎モデルである。感作動物の膝関節中への抗原メチル化ウシ血清アルブミン(mBSA)の関節内注射により、炎症応答が誘発される。滑膜を侵害するパンヌス組織の形成は、関節の軟骨全体に広がり、骨内に成長し、組織びらんおよびリモデリングを生じる。
1:16は、抗原誘導関節炎後、膝の膨張を低下させた。
[コラーゲン誘発性関節炎(CIA)]
マウスのコラーゲン誘発性関節炎(CIA)は、ヒト疾患と幾つかの特徴が共通する、リュウマチ様関節炎に対する最も一般的な実験モデルである。完全フロイントアジュバントにおいて乳化された自己由来または異種のコラーゲン・タイプII(CII)は、滑液膜組織の浮腫、滑膜細胞増殖、炎症性細胞浸潤ならびに軟骨および骨のびらんを伴う多発性関節炎を誘発する。テスト化合物は、全身に投与されるべきである。多発性関節炎の特徴は、関節炎の症候のスコア、組織学的分析および血清バイオマーカーを測定することにより評価できる。骨無機質含量と密度は、マウス・デンシトメトリー(PIXIMUS)により分析してもよい。

- 一般式(I)の化合物、もしくは互変異性体、ジアステレオマー、鏡像異性体、もしくはそれらの混合物、または無機酸もしくは有機酸との生理学的に許容される塩。
Zは、1、2、3、4または5の炭素原子を有する、飽和または不飽和、直鎖または分岐の鎖状の非環式炭化水素基であり、
R2は、アミノ、アルキルアミノ、ジアルキルアミノまたグアニジン基から選択され、
R3、R4およびR5は、同一または異なっており、水素;ハロゲン;トリハロアルキル;1、2、3、4または5の炭素原子を有するアルキル;1、2、3、4もしくは5の炭素原子を有するアルコキシ、トリハロアルコキシ、ヒドロキシまたはアミノから選択される電子供与性基;シアノ、スルホン、ニトロまたはアミドから選択される電子受容性基から独立して選択され、
R6、R7およびR8は、同一または異なっていてもよく、水素;ハロゲン;トリハロアルキル;1、2、3、4または5の炭素原子を有するアルキル;1、2、3、4または5の炭素原子を有するアルコキシ、トリハロアルコキシ、ヒドロキシまたはアミノから選択される電子供与性基;シアノ、スルホン、ニトロまたはアミドから選択される電子受容性基から選択され、

- 標識または毒物をさらに含む請求項1〜9のいずれかに記載の化合物。

- 炎症、関節炎状態、リュウマチ様関節炎、変形性関節炎もしくは、精神障害の治療のための薬剤、または、中枢神経再生を誘導するための薬剤として使用されるための、請求項1〜9のいずれかに記載の化合物。
- 医薬的に許容される補助剤、担体、希釈剤または賦形剤の1以上と共に、請求項1〜9、11および12のいずれかに記載の化合物を含む医薬組成物。
- 炎症、関節炎状態、リュウマチ様関節炎、変形性関節炎、もしくは精神障害の治療のための薬剤または、中枢神経再生を誘導するための薬剤の製造における、請求項1〜9、11および12のいずれかに記載の化合物の使用。
