Description
、ろ過、活性炭などによる脱色、脱臭、エタノール沈殿などの処理をして用いても良い。更には、抽出や精製した溶液を濃縮乾固、噴霧乾燥、凍結乾燥などの処理を行い、乾燥物として用いても良い。
黒霊芝子実体の乾燥物100gに2Lの精製水を加え、95〜100℃で2時間抽出した後、濾過し、その濾液を濃縮後、凍結乾燥して、黒霊芝の熱水抽出物を5.4g得た。
黒霊芝子実体の乾燥物250kgに5kLの精製水を加え、95〜100℃で2時間抽出した後、濾過して固形分濃度を測定した。該固形分に相当するデキストリンを加えて濃縮し、スプレー乾燥して黒霊芝の熱水抽出物を25kg得た。
黒霊芝子実体の乾燥物100gに精製水1L及びエタノール1Lを加え、常温で7日間抽出した後、濾過し、その濾液を濃縮し、凍結乾燥して黒霊芝の50%エタノール抽出物を2.9g得た。
黒霊芝子実体の乾燥物1kgにエタノール15Lを加え、常温で7日間抽出した後、濾過し、その濾液を濃縮乾固して、黒霊芝のエタノール抽出物を20g得た。
製造例4の黒霊芝のエタノール抽出物3.0gをエタノール500mLに溶解し、同容量の精製水とヘキサンを加えて抽出した。水層部分を1/2に濃縮し、同容量の酢酸エチルを加えて抽出した後、濃縮乾固し、酢酸エチル画分0.84gを得た。
製造例1の黒霊芝の熱水抽出物10.0gを精製水に溶解し、スチレン−ジビニルベンゼン系の合成吸着樹脂500mLを充填したカラムに通液した。次いで、精製水、10、20、50、80、90、100%エタノール各1Lで溶出させた後、それぞれを濃縮乾固した。80%以上のエタノール画分をまとめて、黒霊芝のカラム精製物を約0.40g得た。
赤霊芝の乾燥物100gに精製水2Lを加え、95〜100℃で2時間抽出した後、濾過し、その濾液を濃縮し、凍結乾燥して赤霊芝の熱水抽出物を5.0g得た。

[製法]成分1〜3を混合し、散剤とする。
処方 配合量
1.黒霊芝の50%エタノール抽出物(製造例3) 5部
2.乾燥コーンスターチ 25
3.カルボキシメチルセルロースカルシウム 20
4.微結晶セルロース 40
5.ポリビニルピロリドン 7
6.タルク 3
[製法]成分1〜5を混合し、次いで10%の水を結合剤として加えて、押出し造粒後乾燥する。成形した顆粒に成分6を加えて混合し打錠する。1錠0.52gとする。
処方 配合量
1.黒霊芝のエタノール抽出物(製造例4) 1.5部
2.乾燥コーンスターチ 50.0
3.エリスリトール 40.0
4.クエン酸 5.0
5.ショ糖脂肪酸エステル 3.4
6.香料 0.1
[製法]成分1〜4を混合し、10%の水を結合剤として加え流動層造粒する。成形した顆粒に成分5及び6を加えて混合し打錠する。1粒1.0gとする。

1.黒霊芝の熱水抽出物2(製造例2) 1.0部
2.赤霊芝の熱水抽出物(比較製造例1) 0.3

4.マルチトール 3.6
5.グルコン酸液(50%) 0.6

[製法]成分1〜6を成分7の一部の水に撹拌溶解する。次いで、成分7の残りの水を加えて混合する。
処方 配合量
1.黒霊芝の熱水抽出物2(製造例2) 10.0部
2.デキストリン 89.0
3.微粒子二酸化ケイ素 1.0
[製法]成分1〜3を混合して米飯添加物を得る。使用に当たっては、加水時に米1合に対して該添加物を2.0g加えて炊飯する。
肝臓細胞におけるLDLレセプター合成に対する効果について、mRNAの発現量を指標として評価した。
4週齢の雄性ICRマウス(日本SLC)を1週間予備飼育し、5週齢時に血中コレステロールをコレステロールE-テストワコー(和光純薬)により測定して、値が均等となるよう4群に分けた。1群の例数は6匹とした。ノーマル群には、粉末飼料MF(オリエンタルバイオ製)を、コントロール群には粉末MFに1%コレステロール、0.5%コール酸を添加した高コレステロール食飼料(以下、HC飼料)を与えた。試料の投与は、赤霊芝の熱水抽出物(比較製造例1)または黒霊芝の熱水抽出物1(製造例1)を1%添加したHC飼料を調製し、それを自由摂取させることにより行った。飼育開始から1ヶ月後に、1晩絶食させたマウスより採血した。遠心分離により得られた血漿を試料として、コレステロールE−テストワコー(和光純薬)により血中コレステロール値を測定した。測定結果は平均±標準誤差で表し、群間の比較についてはTukeyの多重比較検定を用いた。
Claims (4)
- 黒霊芝の抽出物を含有することを特徴とする、LDLレセプター合成促進剤(食品を除く)。
- 請求項1記載のLDLレセプター合成促進剤を配合したことを特徴とするコレステロール低下剤(食品を除く)。
- 請求項1記載のLDLレセプター合成促進剤又は請求項2記載のコレステロール低下剤を配合したことを特徴とする経口投与剤(食品を除く)。
- 黒霊芝の抽出物が溶媒分画及び/又は吸着による精製物であることを特徴とする請求項1から3の何れかに記載されたLDLレセプター合成促進剤、コレステロール低下剤、又は経口投与剤(食品を除く)。
