Description
4℃で12日間保存した前記第1試薬(R1−1)78μLに、同様に4℃で12日間保存した前記第2試薬(R2−1)26μLを添加し、37℃で5分間インキュベートした。そして、第2試薬添加直前の反応液における波長658nmの吸光度(A0)と、第2試薬の添加5分後の反応液における波長658nmの吸光度(A5)とを、生化学自動分析装置(商品名JCA−BM8:日本電子社製、以下同じ)によって測定し、その差(A5−A0)を求めた。これらの結果を下記表2に示す。
第2試薬(R2−3A,R2−3B)をそれぞれ5℃で2日間保存した。前記第1試薬(R1−3)78μLと精製水13μLとを混合し、37℃で5分間インキュベートした。この混合液に、さらに保存した前記第2試薬19.5μLを添加し、37℃で5分間インキュベートして発色反応を行った。そして、第2試薬添加直前の反応液における波長658nmの吸光度(A0)と、第2試薬の添加5分後の反応液における波長658nmの吸光度(A5)とを、生化学自動分析装置(商品名JCA−BM8)によって測定し、その差(A5−A0)を求めた。第2試薬としてR2−3Aを使用した結果が実施例3であり、R2−3Bを使用した結果が比較例3である。また、実施例3と比較例3の結果の差を求めた。これらの結果を下記表6にあわせて示す。
Claims (10)
- メチレンブルー系発色剤、炭素数12以上の炭化水素鎖を有する第四級アンモニウム又はその塩、及び、液体媒体を含む、液系の発色試薬であることを特徴とする液体発色試薬(但し、その用途が酸素インジケーター用組成物である場合を除く)。
- メチレンブルー系発色剤、炭素数12以上の炭化水素鎖を有する第四級アンモニウム又はその塩、及び、液体媒体を含み、前記メチレンブルー系発色剤の発色反応を吸光度により測定するための液系の発色試薬であることを特徴とする液体発色試薬。
- 前記メチレンブルー系発色剤が、10−(カルボキシメチルアミノカルボニル)−3,7−ビス(ジメチルアミノ)フェノチアジン、又は、10−(アセチルアミノカルボニル)−3,7−ビス(ジメチルアミノ)フェノチアジンである、請求項1又は2に記載の液体発色試薬。
- 前記第四級アンモニウムの少なくとも1つの基が、炭素数が12以上である炭化水素基である請求項1〜3のいずれか一項に記載の液体発色試薬。
- 前記炭素数が、14〜18の範囲である請求項1〜4のいずれか一項に記載の液体発色試薬。
- 調製後、少なくとも3日経過した請求項1〜5のいずれか一項に記載の液体発色試薬。
- 前記第四級アンモニウムは、塩化ベンゼトニウム、塩化ステアリルトリメチルアンモニウム、臭化セチルトリメチルアンモニウム、塩化セチルトリメチルアンモニウム、臭化ヘキサデシルトリメチルアンモニウム、塩化ヘキサデシルトリメチルアンモニウム、塩化ベンザルコニウム、塩化ベンジルジメチルテトラデシルアンモニウム、臭化ミリスチルトリメチルアンモニウム、塩化ミスチリルトリメチルアンモニウム、ココナットアミンアセテート、及び塩化ラウリルトリメチルアンモニウムからなる群から選択される請求項1〜6のいずれか一項に記載の液体発色試薬。
- メチレンブルー系発色剤を含み、前記メチレンブルー系発色剤の発色反応を吸光度により測定するための液体発色試薬の安定化方法であって、液体媒体中で前記発色剤と炭素数12以上の炭化水素鎖を有する第四級アンモニウム又はその塩とを共存させることを特徴とする安定化方法。
- 前記共存を少なくとも3日間続ける請求項8記載の方法。
- メチレンブルー系発色剤の発色反応を吸光度の測定により検出及び/又は測定するためのキットであって、
メチレンブルー系発色剤と、
炭素数12以上の炭化水素鎖を有する第四級アンモニウム又はその塩と、
メチレンブルー系発色剤を発色させる反応に用いる試薬と、
請求項8又は9に記載の安定化方法を行うための取扱説明書を含むキット。
