Description
リポソーム薬物送達テクノロジーは、薬物送達の面で数々の新しい利点をもつ安定なナノ粒子を作製するためにポリマー化学テクノロジーを取込んだより進歩した薬物送達システムにゆっくりと凌駕されつつある。例えば、高分子ナノ粒子は、長期間の体内利用性、疾病細胞のターゲティング、および、生体応答性の薬物制御放出が可能である(17)。薬物は、吸着、閉じ込め、および共有結合によって高分子ナノ粒子に充填され、脱着、拡散、ポリマーエロージョンまたはこれらのメカニズムのいずれかまたは全部の組み合わせによってナノ粒子から放出される。高度に枝分かれした反応性の三次元ナノ粒子であるデンドリマーは、高度な分子均一性、狭い分子量分布、内部ボイドおよびキャビティのような特定のサイズの興味深い構造特性、ならびに高度に官能性の末端表面を有するので生物医学用途に好適である。本発明のデンドリマーは、ポリカチオン性ポリマー(ポリ(L−リシン),PLL)と、生物分解性ポリマー(ポリ(L−乳酸),PLLA)と、熱応答性ポリマー(ポリ(N−イソプロピルアクリルアミド),PNIPAAM)とからなる。C6のような疎水性薬物をPLLAとの疎水性−疎水性相互作用によって1000mg/mlまでの濃度でデンドリマーに充填する。生物分解性ポリマーに応答性ポリマーを組込むと、温度のような生理的刺激に応答して薬物が持続放出されるという利点が得られる。
材料および細胞培養物
卵ホスファチジルコリン(EPC)、ジオレオイルホスファチジルエタノールアミン(DOPE)、ジオレオイルホスファチジルコリン(DOPC)、コレステロール(CH)、ポリエチレングリコール(2000〜5000)−ジステアロイルホスファチジルエタノールアミン(PEG−DSPE)、D−エリスロ−ヘキサノイル−スフィンゴシン(C6−セラミド)、ポリエチレングリコール−750−C8−セラミド(PEG−C8)、ジオレオイル−1,2−ジアシル−3−トリメチル−アンモニウム−プロパン(DOTAP)はAvanti Polar Lipids(Alabaster,AL)から購入した。ジ−ヒドロ−エリスロヘキサノイル−スフィンゴシン(DHC6)はBiomol(Plymouth Meeting,PA)から購入した。[3H]−C6はARC(St.Louis,MO)から入手し、[3H]−チミジンはICN(Costa Mesa,CA)から購入し、コレステリル−1,2−3H(N)ヘキサデシルエーテル([3H]−CHE)はPerkinElmer(Boston,MA)から入手した。シリカゲル60薄層クロマトグラフィープレートは、EMD Chemicals(Gibbstown,NJ)から購入した。formvar/カーボンコートした400メッシュの銅グリッドは、Electron Microscopy Sciences(Fort Washington,Pa)から購入し、ポリ−L−リシンはSigma(StLouis,MO)から入手した。
脂質を配合し、薬物送達小胞にC6を取込むその能力を試験した。簡単に説明すると、クロロホルム(CHCl3)に溶解した脂質を特定のモル比で組合せ、脂質の転移温度よりも高温の窒素流下で乾燥し、滅菌したリン酸塩緩衝生理的塩類溶液(PBS)で水和した。得られた溶液を2分間音波処理し、次いで、100nmのポリカーボネート膜から押出した。取込み効率は、微量の[3H]C6を配合物に組込んで、構成脂質をCHCl3/MeOH(2:1)に抽出し、シンチレーションカウンターを使用して押出前と押出後との放射能を比較することによって測定した。in vivo投与用の配合物は、DSPC:DOPE:DSPE−PEG(5000):C8−セラミド−PEG(750):C6−セラミド(3.75:1.75:0.75:0.75:3.0、モル比)から構成した。PEG(750)−C8を添加すると30モル%までのC6−セラミドが許容される。これらのPEG修飾配合物の生物活性を410.4乳腺癌細胞で確認した。配合したリポソームの組成は、構成脂質をクロロホルム/メタノール(2:1)に抽出し、次いで予熱したシリカゲル60薄層クロマトグラフィー(TLC)でCHCl3/MeOH/ddH2O(60:25:4)溶媒系を使用して分解することによって検証した。脂質をヨウ素室で可視化した。透過型電子顕微鏡法(TEM)を使用して配合リポソームのサイズおよび形態のキャラクタリゼーションを行った。

リポソーム送達の量を非リポソーム投与に比較して定量するために微量の[3H]C6をリポソーム配合物に組込んだ。ヒトMDA−MB−231(MDA)乳腺癌細胞を、24−ウェルプレートに3.5×104細胞/ウェルで播種し、10%FBSを含有する培地で一夜増殖させた。次に、1%FBSを補給した培地中、微量の[3H]C6または[3H]CHEを含有するリポソームC6または非リポソームC6で細胞を様々な時間間隔で処理した。リポソームC6は細胞培地に直接添加し、非リポソームC6はジメチルスルホキジト(DMSO)ビヒクル中に最終濃度≦0.1%(v/v)として添加した。指定の時点毎に、培地を取り出し、細胞を低温のPBSで一回洗浄してリポソーム/膜−非特異的相互作用を解離した。次に、細胞を1%SDSで可溶化し、MDA細胞への[3H]C6または[3H]CHEの蓄積をシンチレーションカウンターで測定した。
短鎖セラミドをMDA細胞に送達する慣用の脂質配合物の有効性を判定するために、[3H]チミジン増殖アッセイを実施した。簡単に説明すると、MDA細胞を24−ウェルプレートに3.5×104細胞/ウェルで播種し、24時間の血清飢餓を行う前に一夜増殖させた。血清飢餓の12時間目に、細胞をリポソームC6または非リポソームC6で処理し、残りの血清飢餓を続行した。血清飢餓の後、培地にFBS(10%最終濃度)を補給して更に12時間維持し、処理の最後の4時間に0.5mCi/mlの[3H]チミジンを添加して細胞増殖を検定した。細胞を低温のPBSで一回、次いで10%のトリクロロ酢酸で10分間ずつ二回洗浄した。細胞を0.3NのNaOHで可溶化し、酸不溶性DNA内への[3H]チミジンの取込みをシンチレーションカウンターで測定した。
in vivo試験に使用したPEG修飾配合物のin vitro有効性を測定するために、これらの配合物をネズミの410.4乳腺癌細胞で試験した。410.4細胞を96−ウェルのプレートで平板培養し、FBSを1%まで補給した培養培地中でPEG修飾リポソームC6または遊離C6によって24時間処理した。Promega Cell Titer Proliferation Kit(Promega)を製造業者の指示通りに使用して細胞毒性を測定した。PEG修飾リポソーム−C6送達は、細胞毒性を増強した。リポソーム−C6配合物の投与は、DMSOビヒクル中のC6の遊離投与に比べてC6のIC50を減少させた(図5)。これらのデータは、PEG−C8リポソーム配合物によって送達されたときにC6セラミドのIC50が35〜40%減少することを表す。処理は、1%FBSの存在下で24時間行った。
アポトーシスはカスパーゼ活性の正の調節に付随する。従って、MDA細胞をPEG修飾リポソームで処理した後のカスパーゼ−3/7活性を測定した。簡単に説明すると、MDA細胞を96−ウェルプレートに6.0×103細胞/ウェルの密度で播種し、10%FBSを含有している培養培地で48時間増殖させた。次に細胞を、1%FBSを含有している培地中でリポソームC6または非リポソームC6で24時間処理した。カスパーゼ−3/7の酵素活性レベルを、Apo−ONE均質カスパーゼ−3/7アッセイ(Promega,Madison,WI)を使用し当業界で公知の標準プロトコルに従って測定した。
C6依存性増殖阻害がアポトーシスの増進に相関関係を有しているか否かを判断するための試験を行った。C6の送達がMDA細胞のアポトーシスに導くことを確認するために、細胞アポトーシスの指標となる開裂DNAを染色するTUNEL分析(Upstate Biotechnology,Lake Placid,NY)を行った(図7A)。
C6のリポソーム配合物または非リポソーム配合物で処理したMDA細胞中のセラミド調節Aktシグナル伝達経路をウェスタンブロット分析を使用して調べた。簡単に説明すると、MDA細胞を60mmプレートに4.0×105細胞/ウェルで播種し、一夜増殖後に血清飢餓状態に24時間維持した。血清飢餓の16時間目に細胞をリポソームC6または非リポソームC6で処理し、残り時間の血清飢餓を維持した。24時間の血清飢餓の後、細胞培地にIGF−1(20ng/ml)を加えて15分間維持した。細胞を低温のPBSで一回洗浄し、次いで氷上で150μlの低温溶解バッファ(1%のトリトンX−100、20mMのトリス、150mMのNaCl、1mMのEDTA、1mMのEGTA、2.5mMのNa4P2O7、1mMのβグリセロホスフェート、1mMのNa3VO4、ddH2O中に1μg/mlのロイペプチン、pH7.5)を加えた。細胞を氷上で15分間溶解させ、細胞溶解液を収集し、15,000×Gで15分間遠心した。35μgのタンパク質を4%〜10%のプレキャストSDS−PAGE勾配ゲルに充填し、pAktをプローブした。均等な充填を証明するためにブロットを剥ぎ取ってAkt−1,2,3を再度プローブした。ECLケミルミネセンスを使用してタンパク質バンドを可視化し、密度測定法によって定量した。結果は、PEG修飾リポソームC6が非リポソームセラミドよりもIGF−1−刺激pAktレベルを有効に低下させることを示した。他方、ゴースト配合物はpAktレベルに全く効果を有していなかった(図8Aおよび8B)。リポソームジヒドロ−エリスロ−ヘキサノイル−スフィンゴシン(DHC6)も非リポソームDHC6に比較してIGF−1刺激Aktリン酸化の阻害を示した。8時間というC6処理時間を選択したが、この時点がMDA細胞中のC6の蓄積がほぼ最大になる時点に対応するからである。この試験は、リポソームC6がAktシグナル伝達カスケードの長期間阻害を介して細胞増殖阻害およびアポトーシスを誘導したことを裏付けた。
410.4ネズミ乳腺癌細胞内のC6の蓄積を実証するために、リポソーム−C6配合物をC6マーカーとなる10モル%NBD−C6と共に投与した。参照用に細胞をDAPI(核)およびMitroTrakerレッド(ミトコンドリア)でカウンター染色した。倍率60×のコンフォーカル顕微鏡法によってC6送達を評価した。リポソーム小胞から細胞へのNBD−C6送達のコンフォーカル顕微鏡像(図9A)。NBD−C6(グリーン)はミトコンドリア(Miro Trakerレッド)と共に局在した;ブルーの染色は、DAPI−染色された核を表す。
全細胞中および小嚢富化脂質ラフト中のC6の時間依存的細胞内蓄積を測定するために、リポソーム配合物に微量の[3H]−C6を組込んだ。これらの脂質ラフトはセラミドも含めた多重経路のシグナル伝達を促進すると考えられているので、リポソーム−C6を投与すると脂質ラフトでC6の蓄積が促進されるはずである。この現象を実証するために、細胞溶解液のショ糖勾配(5%、35%および45%ショ糖)処理を行って、小嚢富化脂質ラフトを単離した。勾配の画分(合計12の画分)から各1mlの等量アリコートを取り出してシンチレーションカウンターでカウントした。全C6用マーカーとして[3H]−C6を使用すると、リポソーム送達の結果として小嚢脂質のシグナル伝達ラフトにセラミドが時間依存的に蓄積した(図9B)。セラミドは、ショ糖勾配の画分No.4および5に蓄積したが、これはカベオリン−1富化脂質ラフト(小嚢)を表していた(図9B)。
in vivoマウス乳腺癌モデル系を使用し、固形腫瘍治療用のC6の全身送達方法を確立した。in vivoデータは、410.4腫瘍保有BALB/cマウスに対するPEG修飾リポソーム配合物の有望な抗癌活性を示唆する(図10A〜B)。これらのin vivo結果は、空のゴーストリポソームに比較してリポソームC6による腫瘍体積の用量応答的縮小を示す。これは、in vivoの腫瘍形成モデルで全身投与C6配合物の有効性が証明された最初の試験である。更に、in vivoの薬物動態解析は、リポソームC6の全身送達の結果として腫瘍組織でC6が定常状態の生物活性濃度に到達し24時間以上この濃度に維持されることを証明した(図11A〜B)。C6が血液および主要な初期通過器官から速やかにクリアランスされるにもかかわらずこの定常状態生物活性濃度は維持されていた。総合的に考察すると、全身投与C6配合物は、in vitroおよびin vivoの双方で有利な薬物動態を伴う有効性を示すことが判明した。更に、スイスウェブスターマウスに使用したPEG修飾リポソーム−C6配合物は、100mg/kgまで静注後に毒性副作用を全く示さなかったが、DMSO中の遊離C6の注入は10mg/kgの量でマウスの50%を致死させた。
全身性リポソーム−C6送達のin vivo効果を測定するために、5×106の410.0細胞をBalb/Cマウスの右後脇腹に皮下注射した。410.4細胞注入の4日後にマウスにリポソーム−C6、空のリポソーム(ゴースト)または0.9%NaClを静注(i.v.)した。マウスを隔日に処理した。処理直前に、マウスを計量し、腫瘍を測定した。腫瘍のサイズはカリパスで測定し、腫瘍の体積は半楕円体に関する式:V=π/6×L×W2を使用して算出した。式中、V=腫瘍体積、L=長さ、W=幅。
C6送達のマーカーとして[3H]−C6を使用し、腫瘍保有マウスに10および40mg/kgのリポソーム−C6を注射し、選択した時点に血液、腫瘍、脾臓、腎臓、肝臓および心臓の組織を採取した。組織を計量し、可溶化し、シンチレーションカウンターでカウントした。組織1mg(または血液1ml)あたりの全C6の質量を採取組織毎に計算し、薬物動態プロフィルを実証した。C6の分布に対するリポソームビヒクルの送達を追跡するために、送達ビヒクルのマーカーとして[3H]−CHEをリポソーム配合物に組込んだ。
デンドリマーの合成
ポリ(L−リシン)(PLL)デンドロンとPLLAをグラフトしたPNIPAAMとをコンジュゲートすることによってデンドリマーを合成した。PLLAをグラフトしたPNIPAAMは、フリーラジカル重合によって合成した(図12)。
UV−vis顕微鏡法(Perkin Elmer Lamda 25,Shelton,CT)を使用し、PBS(pH=7.4)中、500nmのデンドリマーの透過率を1℃/30分で温度を上昇させながら種々の濃度で試験した(図13A)。デンドリマーは熱応答性であり、1,0.5,0.1および0.05mg.ml−1の濃度で下部臨界溶液濃度(LCST)(最大透過率の95%になる温度と定義)はそれぞれ31,32,34および39℃であった。デンドリマーの濃度を低下させるとLCSTが不明になった。LCSTよりも高温では、ポリマーの相互作用の増加が原因で透過率の値が濃度の増加に伴って減少した。PNIPAAMおよびPLLAグラフトPNIPAAMのLCSTは、対数濃度に伴って直線状に低下し、PLLAが疎水性なのでどの濃度でも後者は前者よりも2℃低い値であった。しかしながら、PLLAグラフトPNIPAAMの両端にPLLをコンジュゲートすると、デンドリマーのLCSTは対数濃度と非直線状の関係を示し、他の2種類のポリマーに比較して最も高い値になった。その理由はPLLの正電荷および疎水性である。
LCSTよりも低温および高温であるそれぞれ25℃および37℃におけるPBS(pH=7.4)中の1mg.ml−1のデンドリマーの動的分解を、MALDI−TOFを使用してデンドリマーのモル質量の経時的変化を測定することによってプローブした。デンドリマーの数モル質量(Mn)は、一ヶ月まで経時的に減少し、25℃のときよりも37℃のときのほうが迅速に減少し、双方の温度で19日後に比較的安定な値に到達した。注目すべきは、19日後の安定なMnが約2700g.mol−1であり、初期Mn(約4200g.mol−1)との差が約1500g.mol−1というPLLAの値に等しい値だったことである。この結果は、デンドリマーが分解したこと、また、その分解の原因がデンドリマーのPLLA成分の加水分解による分解であるらしいことを示唆する。この所見の裏付けをとるために、デンドリマーのFTIRスペクトルおよび粘度をそれぞれ経時的に測定した(データ示さず)。PLLAのエステルC=O伸縮に帰属する〜1760cm−1のピーク強度は明らかに経時的に低下し、19日後に消失することが観察された。PNIPAAMおよびPLLのアミドC=O伸縮に帰属する〜1660cm−1のピークは比較的安定だったので、〜1760cm−1のピーク強度を正規化する標準ピークとしてこれらを使用した。得られたピーク高パーセンテージは経時的に減少し、19日後に0になった(データ示さず)。更に、デンドリマーの粘度(Cannon−Ubbelohdeタイプ粘度計によってASTM D 445およびISO 3104の手順に従って測定)は時間に伴って減少し、25℃のときよりも37℃のときのほうが減少が迅速であり、19日後に安定値に到達した(データ示さず)。従って、FTIRの結果は、粘度計およびMALDI−TOF結果と同様に、設計したデンドリマーがPLLA成分の加水分解による分解が原因で生物分解性であることを強力に示唆した。
蒸留水、エタノールおよびN−ジメチルホルムアミド(DMF)(蒸留水/エタノール/DMF{5:5:3 v/v/v})からなる溶媒系でC6とデンドリマーとを3:1の割合(C6:デンドリマー、w/w)で1mg/mlの濃度に混合し、シールし、室温で7時間保存した。C6/デンドリマー溶液をセルロース膜(MWCO−3500)に入れ、エタノール(50ml)に透析して、膜内部から遊離C6を除去した。セルロース膜の内部および外部のC6の量をMALDI−TOFマススペクトロホトメトリーで測定した。膜の内部および外部の双方の溶液を2,5−ジヒドロキシ安息香酸のマトリックス溶液と1:9(サンプル:マトリックス)に混合した。C16−セラミド(C16)を内部標準材料として使用し、各溶液に添加した。時間の経過(2,4,6および10時間)に伴うC6の量を、それぞれ424および562m/zにおけるC6およびC16の質量ピークの相対強度によって計算した。3:1(C6:デンドリマー)の割合でデンドリマーにC6を充填したときの充填効率は約35.9±1.2%であった。
C6とデンドリマーとの相互作用を測定するためには、デンドリマーによるC6放出キネティクスを評価する必要がある。C6の放出率(Mt/Wω、Mtは時刻tのC6の放出量、WωはC6の最大放出量)はデンドリマーの加水分解による分解によって経時的に増加した。デンドリマー−C6複合体を滅菌PBS(pH=7.4)に溶解し、セルロース膜(MWCO=3500)に入れ、0.5%(w/v)のドデシル硫酸ナトリウム(SDS)を含有している滅菌PBS(pH=7.4)(50ml)に透析した。C6が極めて疎水性なので、SDSのような界面活性剤の存在下で透析を行うことが必須であった。デンドリマーの最終濃度を0.02、0.05、0.1および0.5mg/mlとし、LCSTよりも低温(25℃)および高温(37℃)で連続磁気攪拌を用いた。放出されたC6の濃度を測定するために、選択した時間間隔(0から30日の間)毎に1mlのバッファを除去し、新しいバッファに交換した。デンドリマーによるC6放出を定量するために、セルロース膜の内外のC6の量をMALDI−TOFスペクトロホトメトリーでC16を内部標準として測定した。デンドリマーのLCSTよりも高温の37℃では、デンドリマーの疎水性が進んでおり、従って、C6充填デンドリマーからのC6の放出プロフィルがゆるやかであった(図13B)。
前述のように重合ナノ粒子がリポソームテクノロジーに比べていくつもの利点をもつので、我々は、温度誘導送達戦略を使用して固形腫瘍をターゲットするために温度感受性デンドリマーにC6を充填した。我々の特許デンドリマーナノ粒子は、PLLAとPLLとPNIMPAMとからなる。前述同様にUV−vis顕微鏡法を使用して温度上昇に伴うデンドリマー溶液の透過率をモニターすると明らかな変化が観察され、これらのデンドリマーが真に熱応答性であることを確認した。これらの原型デンドリマーのLCSTの概数値は100μg/mlで約34℃であると判明した。MALDI−TOFマススペクトロホトメトリーを使用してデンドリマーのモル質量が経時的に減少することを証明し、これらもまた生物分解性であることを立証した(データ示さず)。コンフォーカル顕微鏡法で分析すると、デンドリマーはLCSTを下回る温度(25℃)のときよりも上回る温度(37℃)のときに優先的にMDA細胞に蓄積したが、その理由は恐らく疎水性が進んだためであろう。更に、フローサイトメトリーを使用してFITC−標識デンドリマーの細胞内蓄積および取込みを定量し、LCSTを下回る温度(25℃)のときよりも上回る温度(37℃)のときに有意に多い量のデンドリマーがMDA細胞に取込まれることを証明した(図14B)。もっと重要な知見は、これらのC6充填デンドリマーでMDA細胞を処理すると有意な増殖阻害/細胞毒性が生じたが、デンドリマー単独は細胞毒性を全く示さなかったことである(図15A)。C6富化デンドリマーは増殖抑制の誘導に加えてMDA細胞のアポトーシスを誘導した(図15B)。総合的に考察すると、我々の予備データは、デンドリマーが抗癌薬C6の放出を効果的にコントロールすることを証明している。この実施態様の最適な実現は、生理的加温薬物送達を可能にするために生理的温度よりもやや高いLCSTをもつポリマーデンドリマーの設計を含む。
第一の目的は、C6と複合体を形成でき、静注によって投与でき、血流中で長時間可溶性であり、固形腫瘍にターゲット指向性および持続的な治療薬送達を果たし得る生物分解性および温度感受性の樹状ナノ粒子を製造することである。デンドリマーを固形腫瘍に熱ターゲティングする約40℃のLCSTをもつデンドリマーを創製するためには、NIPAAMと疎水性PLLAと親水性PLLとの相対的モル比が極めて重要な役割をもつ。ホモポリマーPNIPAAMが32℃のLCSTを有していることは公知である{Eeckman,2001 #52}。このLCSTは疎水性成分または親水性成分の組込み量を増加することによってそれぞれ低下または上昇するであろう{Eeckman,2001 #52}。
以下の実施例でより詳細に説明する。
非イオン性界面活性剤ポリ(オキシエチレン)ノニルフェニルエーテル(Igepal CO−520、Aldrich Chemical Co.)をそれ以上精製しないで使用して逆ミセルを調製した。シクロヘキサン、脱イオン水およびC6セラミドは入手したものをそのまま合成に使用した。
熱応答性および生物分解性という双方の特性をもつ9種類の多機能ヒドロゲルを合成し特性決定した。ヒドロゲルは、熱応答性成分であるN−イソプロピルアクリルアミド(NIPAAM)と加水分解的に分解性の疎水性成分であるポリ(L−乳酸)(PLLA)と酵素分解的に分解性の親水性成分であるデキストランとから構成された共重合ネットワークである。多機能特性を有しているので、設計したヒドロゲルは薬物送達および組織工学のような生物医学用途に適している(図16)。

- 治療用化合物をその送達が必要な動物またはヒトに送達するための組成物であって、治療用化合物の全身送達を可能にするナノ粒子の吸収性を与えるカルシウムホスホ−シリケートシェルを有しているナノ粒子を含み、前記シェルが増殖抑制性で、プロアポトーシスの脂質由来生物活性化合物を含有し、前記の増殖抑制性で、プロアポトーシスの脂質由来生物活性化合物がセラミドまたはC2〜24セラミドを含む、組成物。
