Description
各バッチにおいて120kgの杜仲生葉を、送帯蒸機において蒸熱処理し、葉打機で攪拌および揉圧しながら乾燥し、揉捻機で杜仲葉中の水分を均一化した。その後、葉打機で攪拌および揉圧しながら乾燥し、その後実施例2においてはさらに揉捻機で杜仲葉中の水分を均一化した。このようにして処理した杜仲葉を再乾機により乾燥してリパーゼ阻害剤用杜仲葉を得た。各工程における条件を以下に示す:
送帯蒸機:蒸気量140L/分、蒸熱時間80秒、蒸気温度100〜110℃。
実施例で得られた実施例1〜3の杜仲葉30kgを、株式会社ホーライ製、UGC−280型で、2.8mm〜1.4mmに粉砕した。次に遠赤外焙煎機を用いて、通過速度を45秒としてセラミックヒーターを上段、下段それぞれ1灯照射し、約200℃で焙煎を行い、杜仲葉遠赤焙煎葉26kgを得た。実施例1で得られた杜仲葉の色調は、株式会社島津製作所製、分光色彩計CLR−7100Fの反射式で測定するとL値が25.89、a値が−4.97、b値が7.91であった。また、当該杜仲葉3gを100mLの熱水にて2分間抽出して得られる杜仲葉抽出液は、株式会社島津製作所製、分光色彩計CLR−7100Fの透過光式で色調を測定するとL値が37.51、a値が−7.06、b値が19.14の緑黄色を呈していた。
実施例1の杜仲葉を使用して実施例4で得られた杜仲葉遠赤焙煎葉26kgを、槇野産業株式会社製、コロプレックス250Z型により75μmに粉砕した。次に、ジェットミルにて粉砕圧力を0.6Mpaに固定して、原料供給量を4kg/時間で微粉砕した。このジェットミルに粉砕する時に、圧縮空気を雰囲気温度で通気し、杜仲葉粉末をそれぞれ25kg得た。
比較例として、杜仲生葉を水分率が11%以下となるまで天日乾燥し、粉砕機((株)ホーライ UGC−280)により12番篩以下となるまで粉砕し、その後、焙煎機(横山製作所 品温自動排出式排気乾燥火入れ機80kg)を用いて140℃〜170℃で20分間焙煎し、得られた焙煎物1kgを90℃の熱水15kgに投入し、90℃で30分間抽出した。その後150メッシュのフィルターを用いて濾過し、濾液を5℃に冷却し一晩放置した。上澄み液を取り出し、減圧下濾液を濃縮した。濃縮液を遠心分離器で処理し、濾過により沈殿物を除去し、得られた上澄み液を加熱殺菌(85℃、2時間)し、杜仲葉水抽出エキス(950g)を得た。
被験物質の調製にあたっては、実施例5で得た粉末または比較例1で得たエキスを0.1M NaClを含む0.1M TES緩衝液(pH7.0)で各最終濃度が1、5、10、20mg/mLになるようそれぞれ調整し、サンプル液とした。また基質溶液の調製には、トリオレイン80mg、ホスファチジルコリン10mg及びタウロコール酸ナトリウム5mgを9mLの0.1M緩衝液中で超音波処理したものを用い、これを基質溶液とした。酵素溶液として、ブタ膵臓由来リパーゼを最終濃度4μg/mLに0.1M TES緩衝液で調整したものを用いた。リパーゼ活性の測定は、基質溶液100μLに酵素溶液50μL及びサンプル溶液50μLを加え、37℃で30分間反応させた後、遊離する脂肪酸量を市販のキット(和光純薬、NEFA C−テスト ワコー)を用いて定量した。
ICR系雌性マウスに、牛脂と杜仲葉粉末を配合した高脂肪食を摂取させて、血液中の中性脂肪量、脂肪組織重量、及び体重に対する影響を調べた。なお、実施例5で得た杜仲葉粉末は7.5%(w/w)となるように飼料に添加し、摂取させた。
Claims (2)
- 杜仲生葉を蒸熱する工程;杜仲葉を攪拌および/または揉圧しながら乾燥する工程;杜仲葉を乾燥する工程;および、杜仲葉を粉砕する工程を含む製造方法により得られる杜仲葉粉砕物を含むリパーゼ阻害剤。
- 杜仲葉粉砕物が緑色である請求項1に記載のリパーゼ阻害剤。
