Description
ヒドロゲルを脱水する。最終の乾燥工程は、通常オーブン中、好ましくは、低温度の真空オーブン中で行う。米国特許第6,271,278号は、各種のSPHの調製について詳しく示している。SPHについては、また、チェエンほかがJ. Biomed. Mater. Res. 44:53-62(1999)で示している。
迅速に膨張する。ヒトにそれらを用いるとき、低pH条件で2cm以上の直径まで膨張する超多孔性ヒドロゲルが望ましい。なぜなら、その後それらは幽門括約筋を通過することができず、胃の中で長く滞留し上部消化管を通して薬を良く吸収するからである。
Claims (25)
- 次の各工程を備えるヒドロゲルの形成方法。
(a)少なくとも一つのエチレン性不飽和モノマー(炭素−炭素二重結合を備えるモノマー)、架橋剤、および1または2以上のカチオンをもつイオン性多糖類を組み合わせて混合物を形成する工程
(b)その混合物を重合条件にさらし、ヒドロゲルを形成する工程
(c)そのヒドロゲルを平衡条件下で1または2以上のカチオンを含む溶液と接触させる工程
この(c)工程において、(i)少なくとも一つのカチオンとして工程(a)で用いなかったものを用いるか、あるいは、(ii)工程(a)および(c)で複数のカチオンの同じ混合物を用いるときには、それらの工程で用いる複数のカチオンの割合を異ならせる。 - 前記混合物が、希釈剤、発泡安定剤、発泡助剤、還元剤(リダクタント)、酸化剤(オキシダント)および膨張剤(発泡剤)から構成されるグループの中の1または2以上のものを含む、請求項1の方法。
- 前記混合物が、希釈剤、発泡安定剤、発泡助剤、還元剤(リダクタント)、酸化剤(オキシダント)および膨張剤(発泡剤)を含む、請求項2の方法。
- 前記ヒドロゲルが超多孔性ヒドロゲルである、請求項1の方法。
- 前記工程(a)で用いるカチオンの少なくとも一つが1価であり、しかも、前記工程(c)で用いるカチオンの少なくとも一つが2価である、請求項1の方法。
- 前記(c)工程で形成したヒドロゲルを平衡条件下で1または2以上のカチオンと反応させる付加的な工程を備え、
その付加的な工程において、(i)少なくとも一つのカチオンとして工程(a)で用いなかったものを用いるか、あるいは、(ii)工程(a)および(c)で複数のカチオンの同じ混合物を用いるときには、それらの工程で用いる複数のカチオンの割合を異ならせる、請求項1の方法。 - 前記(c)工程で形成したヒドロゲルと反応させるカチオンの少なくとも一つが3価である、請求項6の方法。
- 前記多糖類が、カルボキシメチルセルロース、アルジネート、ヒアルロン酸、スターチグリコール酸塩(starch glycolate)、カルボキシメチルスターチ、硫酸デキストラン、ペクチネート(pectinate)、キサンタン、カラギーナン、あるいはキトサンである、請求項1の方法。
- 前記多糖類が、カルボキシメチルセルロースである、請求項8の方法。
- 前記エチレン性不飽和モノマーが、アクリルアミド(AM)、N−イソプロピルアクリルアミド(NIPAM)、2−ヒドロキシエチルアクリレイト(HEA)、2−ヒドロキシエチル メタクリレイト(HEMA)、 、N−ビニル ピロリドン(VP)、アクリル酸(AA)、アクリル酸のナトリウム/カリウム/アンモニウム塩、メタアクリル酸およびその塩、N,N−ジメチルアミノエチル アクリレイト、2−アクリルアミド−2−メチル−1−プロパンスルフォン酸(AMPS)、3−スルフォプロピル アクリレイト カリウム塩(SPAK)、3−スルフォプロピル メタアクリレイト カリウム塩(SPMAK)、あるいは2−(アクリロイルエチル)トリメチル−硫酸メチルアンモニウム(ATMS)である、請求項1の方法。
- 前記エチレン性不飽和モノマーが、2−ヒドロキシエチルアクリレイトである、請求項10の方法。
- 前記架橋剤が、N,N’−メチレンビスアクリルアミド、エチレングリコール ジ(メタ)アクリレイト、ポリ(エチレングリコール)ジ(メタ)アクリレイト、トリメチロールプロパン トリアクリレイト(TMPTA)、ピペラジン ジアクリルアミド、グルタルアルデヒド、エピクロロヒドリン、1または2以上の1,2−ジオール構造を含む架橋剤、1または2以上の機能性ペプチドを含む架橋剤、あるいは1または2以上の機能性タンパク質を含む架橋剤である、請求項1の方法。
- 前記架橋剤が、ポリ(エチレングリコール)ジアクリレイトである、請求項12の方法。
- 前記工程(c)で用いるカチオンが、Na+ ,K+ ,NH4 + ,Ca2+ ,Ba2+ ,Mg2+ ,Cu2+ ,Zn2+ ,Mn2+ ,Fe2+ ,Fe3+ ,Al3+ ,Cr3+ ,あるいはCe4+ である、請求項1の方法。
- 前記付加的な工程で、前記ヒドロゲルとCa2+ ,Cu2+,Zn2+ ,Ba2+ ,Mg2+ ,Mn2+ ,Fe2+ ,Fe3+ ,Cr3+ ,Al3+ ,あるいはCe4+ との反応がある、請求項6の方法。
- 前記請求項1の形成方法で作るヒドロゲルを凍結乾燥することによって、0.005〜0.1の水/ヒドロゲルの重量比を得る、ヒドロゲルの乾燥方法。
- 前記ヒドロゲルが超多孔性ヒドロゲルである、請求項16の方法。
- 次の各工程を備える、請求項17の方法。
(a)ヒドロゲルを3℃/1時間の冷却速度で−10℃の温度に凍結する工程
(b)そのヒドロゲルを16〜24時間−10℃に保つ工程
(c)そのヒドロゲルを−10℃で、しかも0.2Torr(トル)以下で60〜80時間凍結乾燥する工程
(d)そのヒドロゲルの温度を3℃/1時間の速度で10℃まで温度上昇させる工程
(e)そのヒドロゲルを10℃で、しかも200mTorr以下で少なくとも12時間保つ工程 - 前記請求項16における、0.005〜0.1の水/ヒドロゲルの重量比を得るヒドロゲルの乾燥方法であって、次の各工程を備える、ヒドロゲルの乾燥方法。
(a)前記ヒドロゲルに含有する水を非水溶媒、水混和性溶媒あるいは溶媒混合物と置換する工程
(b)前記の非水溶媒、水混和性溶媒あるいは溶媒混合物を50Torr以下の圧力によって、または/および前記ヒドロゲルを加熱することによって除去する工程 - 前記ヒドロゲルが超多孔性ヒドロゲルである、請求項19の方法。
- 前記非水溶媒が、メタノール、エタノール、1−プロパノール、2−プロパノール、テトラヒドロフラン、ジオキサン、蟻酸、酢酸、アセトニトリル、ニトロメタン、アセトン、あるいは2−ブタノンである、請求項19の方法。
- 前記非水溶媒がエタノールを含む、請求項19の方法。
- 前記超多孔性ヒドロゲルが、薬剤、栄養物質、あるいは受精媒介物を含む、請求項17あるいは請求項20のいずれかの方法で製造した超多孔性ヒドロゲル。
- 前記超多孔性ヒドロゲルが、さらに徐放性組織を含む、請求項23の超多孔性ヒドロゲル。
- 前記超多孔性ヒドロゲルが、フィルム、シート、粒子、細粒、繊維、ロッド、あるいはチューブである、請求項17あるいは請求項20のいずれかの方法で製造した超多孔性ヒドロゲル。
