Description
「硬化性」は、例えば加熱、化学的架橋、放射線重合または架橋などにより組成物をキュアさせ、または凝固させることができることを意味する。
前述のように、本発明の硬化性歯科用組成物は、場合によっては重合性成分を含む。重合性成分は、場合によっては、酸官能基の有無にかかわらないエチレン性不飽和化合物とすることができる。
重合性成分は、存在する場合、酸官能基を有するエチレン性不飽和化合物を場合によっては含む。好ましくは、酸官能基には、炭素、硫黄、リン、またはホウ素のオキシ酸(すなわち、酸素含有酸)が含まれる。
ラジカル重合(例えば、硬化)では、開始反応系は、放射線、熱、またはレドックス/自己キュア化学反応により重合を開始する系から選択することができる。ラジカルとして活性な官能基の重合を開始することができる開始剤のクラスには、光増感剤または促進剤と場合によっては組み合わせてラジカルを発生させる光開始剤が含まれる。このような開始剤は、通常は200〜800nmの波長を有する光エネルギーに曝露させて付加重合するためのラジカルを発生することができる。
本発明の組成物には、少なくとも1つのポリ酸が含まれ、非硬化性もしくは非重合性のポリ酸、または硬化性もしくは重合性のポリ酸(例えば、樹脂改質ポリ酸)とすることができる。通常は、ポリ酸は、複数の酸性の繰返し単位および複数の重合性基を有するポリマーである。代替実施例では、ポリ酸は、重合性基を実質的に含まない場合がある。ポリ酸は、完全に水溶性である必要はないが、他の水性成分と組み合わせた場合に実質的な沈降を起こさないように少なくとも十分に水混和性であるべきである。適切なポリ酸は、(特許文献41)(ウィルソン(Wilson)ら)、第2欄62行から第3欄6行に記載されている。ポリ酸は、良好な貯蔵、取扱い、および混合特性を提供するのに十分な分子量を有するべきである。典型的な重量平均分子量は、ゲルパーミエーションクロマトグラフィーを使用してポリスチレン標準物質に比べて評価して、5,000〜100,000である。
適切な酸反応性フィラーには、金属酸化物、ガラス、および金属塩が含まれる。典型的な金属酸化物には、酸化バリウム、酸化カルシウム、酸化マグネシウム、および酸化亜鉛が含まれる。典型的なガラスには、ホウ酸塩ガラス、リン酸塩ガラス、およびフルオロアルミノケイ酸(「FAS」)ガラスが含まれる。FASガラスが特に好ましい。FASガラスは、通常は十分な溶離可能カチオンを含み、したがってガラスを硬化性組成物の成分と混合すると硬化した歯科用組成物が形成する。ガラスは、通常は十分な溶離可能フッ化物イオンも含み、したがって硬化した組成物が齲蝕特性を有する。ガラスは、FASガラス製造技術分野の当業者によく知られている技法を使用して、フッ化物、アルミナ、および他のガラス形成材料を含む溶融物から作製することができる。FASガラスは、通常は十分に微細化された粒子の形であり、したがって他のセメント成分と都合よく混合することができ、得られた混合物を口中で使用するとうまく機能する。
本発明の組成物は、酸反応性でもよくまたは酸反応性でなくてもよい1つまたは複数のナノフィラーを含有する。このようなナノフィラーは、通常は200ナノメートル以下、より典型的には100ナノメートル以下の平均粒径を有する。このようなナノフィラーは、通常は少なくとも2ナノメートル、より典型的には少なくとも5ナノメートルの平均粒径を有する。通常は、ナノフィラーは、シリカ;ジルコニア;チタン、アルミニウム、セリウム、スズ、イットリウム、ストロンチウム、バリウム、ランタン、亜鉛、イッテルビウム、ビスマス、鉄、およびアンチモンの酸化物;ならびにその組合せから選択されたナノ粒子を含む。より典型的には、ナノフィラーは、シリカ;ジルコニア;チタンの酸化物;およびその組合せから選択されたナノ粒子を含む。いくつかの実施形態では、ナノフィラーは、ナノクラスター、通常は少なくとも80重量パーセントのナノクラスターの形である。より典型的には、ナノクラスターには、シリカクラスター、シリカ−ジルコニアクラスター、およびその組合せが含まれる。他の実施形態では、ナノフィラーは、ナノ粒子とナノクラスターの組合せの形である。ナノフィラーの表面の一部分をシラン処理し、またはその他の方法で化学的に処理して、1つまたは複数の所望の物理的諸特性を提供する場合が多い。
酸反応性フィラーおよびナノフィラー成分に加えて、本発明の組成物は、場合によっては1つまたは複数の他のフィラーも含むことができる。このようなフィラーは、歯科用および/または歯科矯正用の組成物での使用に適した広範囲の材料の1つまたは複数から選択することができる。
本発明の組成物は水を含有する。水は、蒸留水、脱イオン水、または淡水の水道水とすることができる。通常は、脱イオン水を使用する。
場合によっては、硬化性組成物は、他の溶媒、共溶媒(例えば、アルコール)、または希釈剤を含有することができる。必要に応じて、本発明の硬化性組成物は、指示剤、染料、顔料、阻害剤、促進剤、粘度調整剤、湿潤剤、酒石酸、キレート化剤、界面活性剤、緩衝剤、安定剤、および当業者に自明である他の同様な材料などの添加剤を含有することができる。さらに、医薬品または他の治療物質を、歯科用組成物に場合によっては添加することができる。例としては、歯科用組成物でしばしば使用されるタイプのフッ化物供給源、ホワイトニング剤、抗カリエス剤(例えば、キシリトール)、再石灰化剤(例えば、リン酸カルシウム化合物)、酵素、ブレスフレッシュナー、麻酔剤、凝固剤、酸中和剤、化学療法剤、免疫応答調節剤、チキソトロープ剤、ポリオール、抗炎症剤、抗菌剤、抗真菌剤、口内乾燥症の治療剤、減感剤などが挙げられるが、これらに限定されない。上記の添加剤のいずれの組合せでも使用することができる。いずれか1つのこのような添加剤の選択および量は、過度の実験なしに所望の結果が実現されるように当業者によって選択される。
本発明の硬化性歯科用組成物は、通常の混合技法を使用して様々な成分をすべて組み合わせることによって調製することができる。上述のように、酸反応性フィラーとポリ酸のイオン反応により、組成物を部分または完全硬化させることができる。場合によっては、組成物は、重合性成分および光開始剤を含有し、光開始反応により硬化することができ、あるいは組成物が例えば酸化剤および還元剤を含めてラジカル開始剤系を含むレドックスキュア系などの化学重合により部分または完全硬化することができる。あるいは、硬化性組成物は、様々な開始剤系を含有することができ、したがって組成物を、光重合性組成物と化学重合性組成物、ならびにイオン性硬化性組成物となり得る。
粒径決定試験方法
粒径分析器による平均粒径:コールター(Coulter)LS 230粒径分析器(コールターコーポレーション、米国フロリダ州ハイアリーア(Coulter Corporation,Hialeah,FL))を使用して、粒径(クラスターサイズを含めて)分布(体積パーセントに基づいて)を決定した。分析器は、偏光強度示差走査(PIDS)ソフトウェアを装備した。300mgのフィラー試料を、フィラーをすべて湿潤させるのに十分なMICRO−90界面活性剤(コールパーマー、米国ニューヨーク州バーノンヒルズ(Cole−Parmer,Vernon Hills,NY))の入っているガラス製バイアルに添加した。カルゴン溶液(0.20gのフッ化ナトリウム、4.00gのピロリン酸ナトリウム、40.00 gのヘキサメタリン酸ナトリウム、8.00gのMICRO−90界面活性剤、および3948mlの脱イオン水を完全に混合することによって作製)の30mlを添加し、得られた混合物を15分間振盪し、プローブソニケーター(ヒートシステムズ−ウルトラソニックス、モデルW−225ソニケーター、米国ニューヨーク州ファーミンデール(Heat Systems−Ultrasonics,Farmingdale,NY))で出力制御ノブ設定を9にして6分間音波処理した。コールター(Coulter)LS 230粒子特性評価ソフトウェア、バージョン3.01を使用して、粒子分析を実施した。試験条件は、ランレングス(Run Length)90秒、ウェイトレングス(Wait Length)0秒であり、PIDSの読みが45%〜55%になるまで試験試料を試料オリフィス中に滴下した。1試料に付き3組のデータを平均して、平均粒径を得た。
象牙質接着(DA):象牙質の前処理を使用せず、20秒の光キュア露光時間を使用した点以外は(特許文献40)(ミトラ(Mitra)ら)に記載の手順に従って、象牙質接着を測定した。さらに、試料を湿度チャンバ中、37℃および相対湿度90%で20分間状態調節し、次いで脱イオン水中、37℃で24時間貯蔵した。
エナメル質接着(EA):象牙質接着について上記に記載するのと同じキュア時間および状態調節シーケンスを用いた点以外は(特許文献40)(ミトラ(Mitra)ら)に記載の手順に従って、エナメル質接着を測定した。
まず、ペースト−ペースト混合試験試料を内径4mmのガラス管に注入することによって、圧縮強さを評価した。ガラス管の両端をシリコーン栓で塞いだ。充填した管を、0.275メガパスカル(MPa)圧力に5分間かけ、XL 1500キュアリングランプ(3M社(3M Company))で60秒間照射し、クルツアーユニックス(KULZER UniXS)(クルツアーインク、ドイツ(Kulzer,Inc.,Germany))ライトボックスに90秒間配置した。このようなキュアした試料5個を8mmの長さに切断し、37℃の水に1日配置した。ISO規格7489に従って、1ミリメートル/分(mm/分)のクロスヘッド速度で操作したインストロン(INSTRON)万能試験機(インストロンコープ、米国マサチューセッツ州カントン(Instron Corp.,Canton,MA))を使用して、圧縮強さを決定した。結果は、5回の反復実験の平均として報告した。
試料を2mmの長さに切断したということを使用した点以外は上記のCS手順を使用して、間接引張強さを測定した。結果は、7回の反復実験の平均として報告した。
ペースト−ペースト混合試験試料を調製し、直径20mm×高さ1mmの円柱状型に配置し、2枚のプラスチックシートで蓋をして、C字形クランプを使用して中圧で締め付けることによって、フッ化物放出性をin vitro評価した。試料をXL 1500キュアリングランプ(3M社(3M Company))で各側面から60秒間光キュアし、次いで湿度チャンバ中に、37℃および相対湿度90%で1時間貯蔵した。試料をチャンバから取り出し、各試料を、37℃のオーブン中、25mlの脱イオン水を含む試験体バイアルに別々に様々な時間にわたって浸した。それぞれ測定間隔をおいて、試験体バイアルをオーブンから取り出し、試験体バイアルから10mlの水を量り取り、10mlのTISAB II全イオン強度調整緩衝液(シグマ−アルドリッチ(Sigma−Aldrich))と組み合わせた。得られた溶液を撹拌し、フッ化物イオン選択性電極を使用して測定して、適用測定時間の試験試料1グラム当たり浸出したフッ化物の累積量(マイクログラム)を、3個の試料の平均を使用して決定した。試験体バイアルに新しい脱イオン水を補充し、次の測定時間までオーブンに戻した。
円盤形状の(厚さ1mm×直径15mm)ペースト試料を、VISILUX(ビジラックス)2キュアリングランプ(3M Co.、米国ミネソタ州セントポール(St.Paul,MN))からの照明に、6mmの距離をおいて円盤の各側面で60秒間露光することによってキュアした。硬化した試料について、マクベス(MacBeth)(マクベス、米国ニューヨーク州ニューバーグ(MacBeth,Newburgh,NY))から入手可能な、可視光フィルターを装備したマクベス(MacBeth)透過濃度計モデルTD−903を使用して、円盤の厚さを通り抜ける光の透過率を測定することによって、直接光透過率を測定した。マクベス値が低いほど、視覚不透明性が低く透光性が高い材料であることを示唆している。報告された値は、3個の測定値の平均である。
円盤形状の(厚さ1mm×直径15mm)ペースト試験試料を、VISILUX(ビジラックス)2(3M社(3M Company))キュアリングランプからの照明に、6mmの距離をおいて円盤の各側面で60秒間露光することによってキュアした。次いで、キュアした試料について、放射線不透過性を以下の通り評価した。
硬化した試料の艶出保持を下記の方法により測定した。長方形の形状のペースト−ペースト混合試料(長さ20mm×幅9mm×厚さ3mm)をVISILUX(ビジラックス)2装置(3M社(3M Company))で60秒間キュアした。直ちに、光キュアした試料を、湿度チャンバ中に、37℃および90%の相対湿度で1時間配置した。次いで、試料を、オーブン中、脱イオン水に37℃で24時間配置した。試料を、両面接着テープ(スコッチブランドテープ、コアシリーズ2−1300、米国ミネソタ州セントポール(Scotch Brand Tape,Core series 2−1300,St.Paul,MN))でホルダーに搭載し、表1に示すように逐次的に実施される下記の一連のステップに従って艶出しを行った。オートメット2研磨ヘッド(AUTOMET 2 Polishing Head)を有するビューラーのエコメット4研磨機(Buehler ECOMET 4 Polisher)を、時計回りで使用した。
ダビッドソン磨耗試験機(Davidson Wear Tester)モデル2 (ACTA、オランダ アムステルダム(Amsterdam))装置を使用したin−vitro3体磨耗試験により、キュアしたペースト−ペースト試験試料の磨耗速度を決定した。ダビッドソン磨耗試験機(ダビッドソン磨耗試験機(Davidson Wear Tester))を、磨耗痕がホイール面に垂直になるように較正した。未キュアのペースト−ペースト混合試料(第1体を構成する)を、ダビッドソン磨耗試験機(Davidson Wear Tester)の直径47.75mmの磨耗ホイール上の10mm×4mmのスロットにロードした。試料を、VISILUX(ビジラックス)2キュアリングランプ(3M社(3M Company))を使用して60秒間キュアした。キュアした試料を搭載して、磨耗ホイールの直径を測定すると、50.80〜53.34mmであった。カーター・ダイアモンド・ツールカーター・ダイアモンド・ツール(Carter Diamond Tool)装置(S−2192 SYN、カーター・ダイアモンド・ツール・コープ、米国オハイオ州ウィロビー(Carter Diamond Tool Corp.,Willoughby,Ohio))を900rpm回転で使用して、磨耗ホイール上のキュアした試料を滑らかに機械加工した。機械加工中、ホイール上に多量の水を注ぎ、ダストを制御し、熱を散逸させた。機械加工中、磨耗ホイールをできる限り湿潤に維持した。
調製例1A:
シラン処理したナノジルコニア
ジルコニアゾル(800.0g;184gのジルコニア)およびMEEAA(72.08g)を1リットルの丸底フラスコに加えた。水および酸をロータリーエバポレーションで除去して、さらに強制空気オーブン中(90℃)で乾燥した粉末(291.36g)が、乾燥した粉末(282.49g)をもたらすことができた。脱イオン(DI)水(501.0g)を添加し、粉末を再分散した。得られた分散液を2リットルのビーカーに加え、続いて撹拌しながら、1−メトキシ−2−プロパノール(783g;シグマ−アルドリッチ(Sigma−Aldrich))、シルクエスト(SILQUEST)A−174(83.7g)、およびシルクエスト(SILQUEST)A−1230(56.3g)を添加した。得られた混合物を室温で30分間撹拌し、次いで2個のクオートジャーに分け入れ、シールした。ジャーを90℃に4.0時間加熱し、内容物をロータリーエバポレーションで濃縮して、液体濃縮物(621g)を得た。
シラン処理したナノジルコニア
ジルコニア粉末(50g)を、ネジ蓋付きのクオートジャーに加えた。脱イオン(DI)水(67.2g)および1−メトキシ−2−プロパノール(99.5g)を添加し、粉末を分散した。得られた分散液に、撹拌しながら、シルクエスト(SILQUEST)A−174(10.23g)およびシルクエスト(SILQUEST)A−1230(6.87g)を添加した。得られた混合物を室温で10分間撹拌し、次いでクオートジャーをシールした。ジャーを90℃に4.0時間加熱した。
ペーストA組成物
表2Aおよび2Bに列挙する材料(重量部で示す)を組み合わせることによって、9個の第1のペースト組成物(文字Aを用いてA1〜A9と称される)を調製した。フィラーI(ナノジルコニア)を、組成物に樹脂A(HEMAおよびPEGDMA−400;調製例1Aを参照のこと)中80%のナノジルコニアからなるペーストとして添加し、乾燥重量ベースで表に報告する。樹脂A成分は、表中のHEMAおよびPEGDMA−400成分の一部分として含まれている。
ペーストB組成物
表3Aおよび3Bに列挙する材料(重量部で示す)を組み合わせることによって、9個の第2のペースト組成物(文字Bを用いてB1〜B9と称される)を調製した。
ペーストA−ペーストB組成物
第1のペーストを等体積の第2のペーストと25秒間スパチュラで混練することによって、硬化性組成物(実施例1〜9および比較例1〜2)を調製した。利用するペーストの相対重量部および組成物中の成分の重量部を表4Aおよび4Bに示す。
ビトゥレマー(VITREMER)グラスアイオノマー修復材
市販の粉末−液体ビトゥレマー(VITREMER)樹脂改質グラスアイオノマー修復材製品(3M社(3M Company))を、製造者の指示書に従って計量分配し、手で混合し、得られた材料について、本明細書に記載する試験方法に従って圧縮強さ(DS)、間接引張強さ(DTS)、象牙質接着(DA)、エナメル質接着(EA)、視覚不透明性、放射線不透過性、フッ化物放出性、艶出保持性、および3体磨耗性を評価した。結果を表6に報告する。
Claims (6)
- 硬化性歯科用組成物であって、
(a)ポリ酸;
(b)酸反応性フィラー;
(c)少なくとも10重量パーセントの、平均粒径200ナノメートル以下のナノフィラーまたはその組合せ;および
(d)水を含む組成物。 - さらに重合性成分を含む、請求項1に記載の組成物。
- 前記酸反応性フィラーが、フルオロアルミノケイ酸(FAS)ガラスを含む、請求項1または2に記載の組成物。
- 多剤型硬化性歯科用組成物であって、
(a)ポリ酸を含む第1剤;
(b)酸反応性フィラーを含む第2剤;
(c)少なくとも1つのナノフィラーが前記第1剤、前記第2剤、または両方の剤に存在する、少なくとも10重量パーセントの、平均粒径200ナノメートル以下のナノフィラーまたはその組合せ;および
(d)前記第1剤、前記第2剤、または両方の剤に存在する水を含む組成物。 - 歯科用物品を調製する方法であって、
(a)請求項1に記載の歯科用組成物を提供するステップと、
(b)前記歯科用組成物を硬化して、前記歯科用物品を形成するステップとを含む方法。 - 前記歯科用物品が、歯科用ミルブランク、歯冠、歯科用充填物、義歯、および歯科矯正用装置からなる群から選択される、請求項5に記載の組成物。
