Description
本実施例で用いた微生物は、アエロモナス・キャビエ由来のPHA合成酵素群遺伝子を導入したアルカリゲネス・ユウトロファス AC32(受託番号FERM BP−6038(平成8年8月12日に寄託された原寄託(FERM P−15786)より移管)(平成9年8月7日、独立行政法人産業技術総合研究所 特許生物寄託センター、あて名;日本国茨城県つくば市東1丁目1番地1 中央第6))である。これを、J.Bacteriol., 179, 4821-4830頁(1997)に記載の方法で、炭素源としてパーム油を用いて培養した。培養は、撹拌機を備えた3Lジャーにて、温度35℃、撹拌数500rpm、培養時間60時間で行ない、Mw100万のPHBHを約60重量%含有する菌体を得た。つぎに、この培養液を遠心分離(5000rpm、10分間)することによってペースト状菌体を得、これに水を加えて50g菌体/Lの水性分散液とした。
炭素源としてヤシ油を用いた以外は実施例1と同様にして培養を行ない、固形分濃度20重量%の水性分散液を得た。この水性分散液中のPHBHの平均粒子径は1.69μmであった。この水性分散液を減圧下で乾燥して得たパウダーの、3HH組成は10モル%、Mwは76万であった。また、実施例1と同様にして曲げ弾性率、塗膜フィルムの破壊伸びを測定し、フィルムの折り畳み部のクラック、および生分解性を観察した。曲げ弾性率は500MPaであった。塗膜フィルムの破壊伸びは400%であった。結果を表1に示す。
炭素源としてヤシ油を用い、培養温度を30℃とした以外は実施例1と同様にして培養を行ない、固形分濃度20重量%の水性分散液を得た。この水性分散液中のPHBHの平均粒子径は1.95μmであった。この水性分散液を減圧下で乾燥して得たパウダーの、3HH組成は15モル%、Mwは96万であった。また、実施例1と同様にして曲げ弾性率、塗膜フィルムの破壊伸びを測定し、フィルムの折り畳み部のクラック、および生分解性を観察した。曲げ弾性率は300MPaであった。塗膜フィルムの破壊伸びは760%であった。結果を表1に示す。
実施例1において、得られた固形分濃度20重量%の水性分散液に乳化剤として部分鹸化ポリビニルアルコール(クラレ社製、クラレポバールPVA205)を3重量部添加し、撹拌機にて30分間撹拌して乳化剤を含有する水性分散液を得た。実施例1と本実施例で得られた水性分散液を室温で3ヵ月間放置して放置安定性を比べたところ、実施例1の水性分散液はわずかに沈殿物が見られたが、本実施例の水性分散液は沈殿物が見られず変化しなかった。
実施例3で得られた水性分散液を撹拌機を備えた1Lの容器に入れ、撹拌数500rpmで1時間撹拌した。得られた水性分散液中のPHBHの平均粒子径は、1.02μmとなり、もとの平均粒子径である1.95μmよりさらに微粒化されていた。
酪酸を炭素源として用い、培養温度を30℃とした以外は実施例1と同様にして、固形分濃度20重量%のPHBの水性分散液を得た。この水性分散液中のPHBの平均粒径は1.12μmであった。この水性分散液を減圧下で乾燥して得たパウダーのMwは60万であった。また、実施例1と同様にして曲げ弾性率、塗膜フィルムの破壊伸びを測定し、フィルムの折り畳み部のクラック、および生分解性を観察した。曲げ弾性率は2600MPaであった。塗膜フィルムの破壊伸びは8%であった。結果を表1に示す。
酪酸および吉草酸の混合比が、酪酸/吉草酸=92/8である炭素源を用い、培養温度を30℃とした以外は実施例1と同様にして、固形分濃度20重量%のPHBVの水性分散液を得た。この水性分散液中のPHBVの平均粒径は1.35μmであった。この水性分散液を減圧下で乾燥して得たパウダーの、Mwは59万、3HV組成は10モル%であった。また、実施例1と同様にして曲げ弾性率、塗膜フィルムの破壊伸びを測定し、フィルムの折り畳み部のクラック、および生分解性を観察した。曲げ弾性率は1800MPaであった。塗膜フィルムの破壊伸びは20%であった。結果を表1に示す。
酪酸および吉草酸の混合比が、酪酸/吉草酸=83/17である炭素源を用い、培養温度を30℃とした以外は実施例1と同様にして、固形分濃度20重量%のPHBVの水性分散液を得た。この水性分散液中のPHBVの平均粒径は1.54μmであった。この水性分散液を減圧下で乾燥して得たパウダーの、Mwは57万、3HV組成は20モル%であった。また、実施例1と同様にして曲げ弾性率、塗膜フィルムの破壊伸びを測定し、フィルムの折り畳み部のクラック、および生分解性を観察した。曲げ弾性率は1600MPaであった。塗膜フィルムの破壊伸びは50%であった。結果を表1に示す。
Claims (5)
- 曲げ弾性率が100〜1500MPaであり、重量平均分子量が5万〜300万である、3−ヒドロキシブチレートと3−ヒドロキシヘキサノエートとの共重合体からなり、水性分散液中の該共重合体の平均粒径が0.1〜50μmであり、3−ヒドロキシブチレート/3−ヒドロキシヘキサノエート比が97〜75/3〜25モル%である生分解性ポリエステル水性分散液。
- 前記水性分散液中の共重合体の固形分濃度が5〜70重量%である請求項1記載の生分解性ポリエステル水性分散液。
- 前記水性分散液中に乳化剤を含有する請求項1または2記載の生分解性ポリエステル水性分散液。
- 前記共重合体が微生物から産生されるものであって、該共重合体を含有する微生物菌体を水性分散体の状態で破砕して菌体内の該共重合体を分離する工程を含む請求項1、2または3記載の生分解性ポリエステル水性分散液の製造方法。
- 前記水性分散液に機械的剪断を与え、一部凝集した前記共重合体の粒子を相互に分離させる工程を含む請求項4記載の生分解性ポリエステル水性分散液の製造方法。
