本特許願は、米国特許願第09/119,666号(1998年7月21日提出)の部分継続であり、本出願と相容れる程度まで該特許願を本明細書中に全体的に取り入れる。
血液成分の汚染物除去についての溶媒界面活性剤法は、HIVのようなウイルスを包囲しているリン脂質膜を溶かすことにより効果を示し、血液のタンパク質成は損なわないが、もし血球が存在すると、このような方法は細胞膜に損傷を与えるので使用できない。
(a)無毒性、有効量の内因性光増感剤あるいは内因性を基本とする誘導光増感剤を流体と混合し;
(b)光増感剤を活性化するのに十分な光放射線に流体を暴露することにより微生物の少なくとも若干を不活性化する。
例1.7,8−ジメチル−10−リビチル−イソアロキサジンの吸光度プロフィル
7,8−ジメチル−10−リビチルイソアロキサジン(純度98%)の試料はSigma Chemical Companyから得た。この試料の一部を、走査UV分光光度計を用いて分析にかけた。調べた範囲は200から900nmの領域をカバーした。分析のため、試料を蒸留水に溶かした。この分析から得た試料スペクトルを図1に示す。
Isolyte S媒質中の7,8−ジメチル−10−リビチルイソアロオキサジンの最高溶解度を次のように決定した。
濃度(373)=110μM=42μg/ml
濃度(447)=109μM=40.9μg/ml
ACD−A抗凝固剤を用いて上記の同じ手順を繰り返した。これらの測定から得られた値は次の通りであった。
濃度(373)=166μM=63μg/ml
濃度(447)=160μM=60.3μg/ml
Sigma ACD−A中の7,8−ジメチル−10−リビチルイソアロオキサジンの溶液を63μg/mlの濃度で調製した。この調製物をガラスピペット中に採り、UV光源(320nm以下の光を除去するフィルターを用い、365nm λmax)の光路に置いた。懸濁液を特定の時間間隔で照射し、この間隔で一定部分を取り出して分光法による分析に供した。溶解した7,8−ジメチル−10−リビチルイソアロオキサジンの吸光度を、各時間間隔で373nmおよび447nmでモニターした。結果を図3と表1に示す。
現存スペクトルキュベットはポリカーボネートから構成されている。このキュベットの光透過の性質は、UV分光光度計の光通路にそのキュベットを置くことにより373nmおよび447nmで測定された。得られた値は以下のとおりであった:
流動システムが実施可能であるために、サンプルはビーム路にそれが存在する間に適当な光束が提供されなければならない。もし提案されたスペクトルキュベットがこの目的に役に立つとしたら、そのとき、以下のとおりにしてキュベットを通過する流速の関数として光束要件を予測することが可能である:キュベットの照射帯域中に存在する溶液の容量は約0.375mlである。キュベットのこの領域における細胞についての走行時間は以下の方程式から決定されることが出来る:
100ml/分で、走行時間(T)は0.00375分=0.225秒であろう。
図2は吸光度が血小板の濃度とともにどのように変化するかを示している。
UV光が赤血球を浸透出来る程度およびサンプルの厚さの効果および光浸透の区域上のヘマトクリット(赤血球容積率)の効果を評価するために、吸収光が化学反応に作用する能力および程度を測定することにより、化学光量測定、光源から発する光強度の実際量を測定する方法を用いて幾つかの予備実験が行なわれた。これらの研究において、水について観察された強度に対しての光源強度を測定するためにフェリオキサレート(ferrioxalate)溶液が使用された。化学反応およびサンプル調製のために使用される方法の詳細はGordon,A.J.およびFord,R.Aによる“The Chemist’s Companion:A Handbook of Pratical Data,Techniques and References”(John Wiley & Sons発行)第362頁〜第368頁(1972)において教示されている。
Lambertの法則から吸光度=Log(1/透過率)。もし濃度(C)をサンプルのヘマトクリットと等しくさせたならばそしてb=0.3cm(スペクトルキュベットの通路長さ)なので、ヘマトクリットかける通路長さの積に対して赤血球サンプルについての吸光度値をプロットすることによりサンプルについての擬似吸光係数(ε’)を決定することが可能である。サンプルについての吸光係数はこの線の勾配により表わされる。
A=εbC (但し、A=1(入射光線の90%吸収、ε=0.08661、C=サンプルヘマトクリット、b=通路の長さ)。
血小板インビトロパラメータへのウイルス不活化処理の影響:
血小板インビトロパラメータへのウイルス不活化処理の影響を評価した。血小板製剤はUV光と組み合わせて7,8−ジメチル−10−リビチルイソアロキサジンで処理された。種々のインビトロパラメータが、処理条件により誘導される変化の程度を決定するために血小板機能のモニターとして用いられた。UV光露光のエネルギー水準、使用された7,8−ジメチル−10−リビチルイソアロキサジンの量およびサンプル処理条件のようなファクターは血小板品質後処理上へのそれらの影響について調べられた。この研究からの結果は、血小板の機能と妥協することなしにHIV−1の不活化のための適当な処理の窓口を確立するために用いられる。
* 合計エネルギー露光の水準は表4の変換表に従って放射線照射室を通過する懸濁液の流速により決定された。
** 媒体が70:30(媒体:血漿)に希釈されるので、血漿と混合する前の媒体単独中の7,8−ジメチル−10−リビチルイソアロキサジンの原料濃度は適当に調節された。これは1.43、71.4、および143μMの、Isolyte S中の出発濃度を必要とした。
血漿中非処理サンプル(ヒストリカル対照);
+流動−UV−7,8−ジメチル−10−リビチルイソアロキサジン。
正常な供与者の血小板アフェレーシス製品がAABB認定血液銀行機関から得られた。標準のSpectra LRS法を用いてサンプルを収集した。下記のすべての操作または処理手順は標準の実験室の安全な処理手順および方法を用いて行なわれた。ユニット番号および血液タイプが記録された。すべてのサンプルは収集の24時間以内に用いられた。すべてのサンプル移送および処理工程のために無菌での処理手順に従った。
1.)10μMを超える増感剤濃度および1.5E+06/μl以上の血小板濃度で、2日目にサンプルpHにおける低下があった。そのpHは、貯蔵の2日目をこえて着実に下降し、貯蔵の3日目に許容出来ない水準(<6.5)に到達した。すべての他のインビトロパラメータはサンプルpHとともに観察されたパターンに従った。
2.)サンプルpHにおけるこの減少はサンプルをUV光に露光したかまたは露光しなかったかに無関係に起った。
3.)5.4E+05/μlの血小板濃度で、100μMまで研究された任意の増感剤濃度で延長された貯蔵後にサンプルpHにおける低下はなかった。
4.)10μMまでの増感剤濃度、1.5E+06/μl以上の血小板濃度および10J/cm2までのUVA水準で測定された血小板の性質は対照の非処理細胞に匹敵出来た。これらは、処理後貯蔵の5日またはそれ以上の後に対照水準に匹敵出来る状態のままであった。
流速およびサンプルヘマトクリットの関数としての赤血球上への剪断応力の測定
高いヘマトクリットでの赤血球サンプル中へのUV光浸透の低い水準は、光路の狭い開口を赤血球が通過することの効果を理解することの必要性を高めた。光路におけるサンプルの厚さの減少は、高いサンプルヘマトクリットでUV線量の発送を増大させるはずである。この方法を確認するために、寸法を変化させての開口を用いて幾つかの圧力低下測定が行なわれた。狭くされた開口からの上流および下流の両方にぜん動性(波動性:peristaltic)ポンプを有する配管中に圧力ゲージを置いた。変化させてのヘマトクリットの全血を制御された流速で開口に通過させた。両方の位置での圧力読み取りにおける差は開口を横切る圧力低下の直接の測定を可能にした。この値および開口の寸法を用いて、以下の方程式を用いて、赤血球が狭くされたセルを通過するときに赤血球により受ける剪断応力を調べることが可能であった:
血小板濃縮物を20:80の血小板濃縮物:Isolyte Sの比で血小板添加剤溶液Isolyte Sと混合した。血小板濃縮物と血小板添加剤溶液との混合物は本明細書において“媒体”中として言及される。添加剤溶液なしの血小板濃縮物を本明細書において“血漿”中として言及される。両方はListeria monocytogenesでスパイクされた。次に、ビタミンK5が300μg/mlの量で各々に加えられた。次に図7のキュベット装置中で、各各はUV光、可視光または室内光に露光され、結果が表6に示される。
ビタミンK5を含有する上に記載されたとおりの媒体および血漿をバクテリアでスパイクしそして表7において示されるように室内光のみ(K5−光)を照射し、即ちそれに露光しそしてインキュベーションの3日後に増殖を評価した。放射線照射なしで幾つかの種の不活化が見られた。
70:30のIsolyte S:血小板濃縮物の比で、例8において記載されたとおりの血小板濃縮物およびIsolyte Sを用いて造られそして300μg/mlのビタミンKを含有する媒体が幾つかの種のバクテリアでスパイクされそして30および60J/cm2のエネルギー水準で放射線照射された。放射線照射のエネルギーの関数としての不活化が表8および図8に記載されている。
例8において記載されたとおりの血小板濃縮物にそして例10において記載されたとおりの70:30媒体に、10μMの7,8−ジメチル−10−リビチルイソアロキサジンを加えた。血小板濃縮物および媒体はS.aureusまたはS.epidermidisでスパイクされそして80J/cm2および30J/cm2で放射線照射されそして不活化が上記のとおりにして測定された。結果が図9に示される。
標準の血液バッグ中に含有されている例8において記載されたとおりの血漿濃縮物に、粉末形での25μMの7,8−ジメチル−10−リビチルイソアロキサジンを加えた。そのバッグを表9において示されるとおりのバクテリアでスパイクし、かき混ぜそして120J/cm2放射線に露光した。不活化の結果を表9に記載する。
例8において記載されたとおりの血小板濃縮物に7,8−ジメチル−10−リビチルイソアロキサジン、アロキサジンモノヌクレオチドまたは7,8−ジメチルアロキサジンを加え、次にS.aureusまたはS.epidermidisでスパイクしそして80J/cm2で放射線照射した。不活化結果を表10に示す。
例8の血小板濃縮物に10μMの7,8−ジメチル−10−リビチルイソアロキサジンを加えた。分別液は添加剤なしかあるいは“クエンチャー(quencher)”、即ち酸化防止剤としての、10mMのアスコルビン酸塩を含有するかまたは10mMのKIを含有した。それらの溶液をHSV−2、ΦX174、S.epidermidisまたはS.aureusでスパイクしそして80J/cm2で照射した。結果を図10に示す。
例8の血小板濃縮物に、変化させての濃度の7,8−ジメチル−10−リビチルイソアロキサジンを加えた。これらの溶液を単純ヘルペスウイルスタイプII(HSV−II),二本鎖DNAエンベロープウイルスでスパイクした。80J/cm2で放射線照射を行なった。その実験を3回繰り返した。すべての3回の試行において、完全な不活化が達成された。結果を図11に示す。
40、80および120J/cm2の照射のエネルギーで、HSV−IIの代りにS.epidermidisを用いて例15の実験計画に従った。不活化の結果を図12に示す。
ΦX174,一本鎖DNAバクテリオファージを用い、変化させての濃度の7,8−ジメチル−10−リビチルイソアロキサジンおよび変化させてのエネルギーの照射で例15の実験計画に従った。不活化の結果を図13に示す。
例8の血小板濃縮物に10μMの7,8−ジメチル−10−リビチルイソアロキサジンを加えた。これらはS.aureusまたはΦX174でスパイクされそして可視光と紫外光との50:50の混合光を用いての照射の変化させてのエネルギーで放射線照射された。不活化の結果を図14に示す。
微生物としてS.epidermidisおよびHSV−IIを用いて例18の実験計画に従った。紫外光と可視光との50:50混合光は、DYMAX光源により供給された。不活化の結果を図15に示す。
例8の血小板濃縮物に10μMの7,8−ジメチル−10−リビチルイソアロキサジンを粉末形で加えた。アスコルビン酸塩を加えての試験およびアスコルビン酸塩を加えないでの試験が行なわれた。150mlの試験溶液をSpectraTM血液バッグに入れそして振り混ぜそして50:50の可視光:紫外光を用いての照射の変化させていくエネルギーに露出させた。40J/cm2を受容した後に、バッグのスパイクポートに残っていた可能性がある微生物に起因する誤差を避けるために、各々のバッグの内容物を新しいバッグに移した。不活化の結果を図16に示す。下方の矢印は検出することが可能であった水準(2.5log力価)までの不活化を示す。
例8の血小板濃縮物そして30:70の血小板濃縮物:Isolyte SでのIsolyte S中の血小板濃縮物に、20μMの7,8−ジメチル−10−リビチルイソアロキサジンを加えた。これらはワクシニアウイルス,二本鎖DNAエンベロープウイルスでスパイクされそして30分間DYMAX2000UV光源を用いて、60J/cm2での可視光あるいは混合された(50:50)可視光および紫外光に露光された。検出の限界は1.5logであった。不活化の結果を図17に示す。光増感剤なし、Isolyte S媒体単独中の光増感剤、Isolyte S媒体中血小板、7,8−ジメチル−10−リビチルイソアロキサジンの代りに8−メトキシソラレンを用いてのIsolyte S媒体中血小板および(30:70の)Isolyte媒体中血小板濃縮物を用いて比較が行なわれた。
10μMの7,8−ジメチル−10−リビチルイソアロキサジンを有するかまたはそれを有しない、30:70のIsolyte S中の血小板濃縮物のサンプルをワクシニアウイルスでスパイクしそして変化させての時間期間にわたって50:50の可視光:紫外光を用いての60J/cm2で照射しそして不活化の結果を図18において示されるとおりに比較した。
例8において記載されたとおりの血小板濃縮物のサンプルに5μMまたは50μMの7,8−ジメチル−10−リビチルイソアロキサジンを加えた。サンプルをHIVIでスパイクした。図7に示されるキュベット流動のセルを使用して、EFOS光システムを用いる変化させてのエネルギーで50:50の可視光:UV光でサンプルを照射した。不活化の結果を図19に示す。
HIV感染ACH−2細胞を例8において記載された血小板濃縮物のサンプルに加えた。5μMまたは50μMの7,8−ジメチル−10−リビチルイソアロキサジンを該サンプルに加えた。例23の実験計画に従いそして不活化の結果を図20に示す。HIVの存在を試験細胞上へのその細胞変性効果により分析検定した。
例24の実験計画に従いそしてP24抗原生産の水準を定量することによりHIVの存在を分析検定した。不活化の結果を図21に示す。
例8に記載されたとおりの血小板濃縮物および30%血小板濃縮物と70%PAS IIITM媒体とを含有する媒体のサンプルに6mMのアスコルビン酸塩および14μMの7,8−ジメチル−10−リビチルイソアロキサジンを加えた。サンプルをHIV−IIでスパイクした。不活化の結果を図22および表11に示す。
1.たんぱく質、血液および血液成分からなる群から選ばれた1種またはそれ以上の成分を含有する液体を、その中に存在する可能性がある微生物を不活化するために処理するための方法であって、その方法が、
(a)前記液体に、内因性光増感剤または内因的に基づく誘導体光増感剤の不活化に有効な、実質的に非毒性の量を加え;
(b)該光増感剤を活性化するのに十分な光放射線に工程(a)の液体を露光し、それにより前記微生物が不活化される、
ことからなる、前記方法。

ことを含む、前記方法。
80.前記混合光が50:50の紫外光:可視光である、79の方法。
81.前記光増感剤が非毒性の内因性光増感剤または内因的に基づく誘導体光増感剤である、79の方法。
82.液体の中に存在する可能性がある白血球を不活化するために前記液体を処理するための方法であって、その方法が、
(a)前記液体に、内因性光増感剤または内因的に基づく誘導体光増感剤の不活化に有効な、実質的に非毒性の量を加え;
(b)該光増感剤を活性化するのに十分な光放射線に、工程(a)の該液体を露光し、それにより前記白血球が不活化される;
ことを含む、前記方法。
83.前記液体が血液または血液成分を含む、請求項82の方法。

Claims (6)
- 内因性アロキサジンまたは内因性を基本とするアロキサジン誘導体から選ばれる内因性光増感剤を有効成分として含む、微生物を複製から完全にまたは部分的に防止するための水性血小板添加剤溶液。
- 生理的食塩水;および緩衝剤を含む、請求項1に記載の血小板添加剤溶液。
- 塩化マグネシウムをまた含む、請求項1または2に記載の血小板添加剤溶液。
- グルコン酸ナトリウムをさらに含む、請求項1〜3のいずれか1項に記載の血小板添加剤溶液。
- 前記光増感剤が7,8−ジメチル−10−リビチルイソアロキサジンである、請求項1〜4のいずれか1項に記載の血小板添加剤溶液。
- 内因性光増感剤の濃度が1μM〜10μMである請求項1〜5のいずれか1項に記載の血小板添加剤溶液。
