Description
本出願は、2002年10月4日に出願された米国特許出願第10/264,890号の一部継続出願であり、それは、2000年4月24日に出願された米国特許出願第09/530,171号の分割出願であり、それは、1998年12月21日に出願された国際特許出願第PCT/US98/27203の米国国内段階であり、それは、1997年12月22日に出願された(現在は、放棄されている)米国特許出願第08/996,280号の一部継続出願であり、またこれらの出願の各々から優先権メリットを請求している。本出願は、2003年6月20日に出願された米国特許仮出願第60/480,298号にも優先権を主張している。これらの出願の各々の内容はここに参照により組み入れられる。
本発明は、流体中の検体の濃度を測定するときに使用する測定方法および装置に関するものである。さらに、詳細には、本発明は、排他的ではないが、血液中のグルコースの濃度を測定するために使用される方法および装置に関するものである。
図1は、本発明によるシステムおよび方法において使用に適した好ましい実施形態の励起を示しており、一般的に100で示され、その中で、DC励起およびAC励起の4周波数が使用されている。図1は、励起が、適当な試薬と混合された全血液のサンプルに印加される際の、励起に対する典型的な応答をも図示しており、応答は一般的に102示される。相対的に高い周波数の信号が、時刻101で開始して印加される。好ましい実施形態においては、周波数は、約10kHzから約20kHzのあいだであり、また約12.4mVと約56.6mVとのあいだの振幅を持っている。20kHzの周波数は、図1の例で使用されている。当業者らは、これらの値が、セル形状および特定のセル化学などの種々のパラメータに対して最適化されることを理解するであろう。
少なくとも、印加されたDCおよびAC成分のいくつかは、同時に印加できることが理解されよう。図2は、本発明によるシステムおよび方法において使用するために適した励起信号を図示しており、そこでは、ACおよびDC成分のいくつかは、同時に印加され、一般的に200で示され、図1中に対応して番号を付される対応するイベントを持っている(たとえば、信号200は、時刻201に開始されまた、ストリップは、時刻210において挿入されるなど)。信号100の場合のように、信号200は、約10から20kHzの周波数を持ち、かつ約12.4から56.6mVの振幅を持っている。しかし、前記ストリップが、投与された後には、時刻220に示されるように、DCオフセットが、270において示されるように、重ねあわされる。典型的なACおよびDC応答が図2に示されている。ACおよびDC応答は同時に測定され、かつ数学的に解析され、またインピーダンス(アドミタンスの大きさおよび位相)および電流的あるいは電量的応答を決定するために使用される。
実施例1で行われた測定は、図3Aおよび3B中に図示され、かつ300で一般的に示されているテストストリップを用いて行った。テストストリップ300は、ここで参照することにより組み入れられた米国特許第5,997,817号中に記載されているように、相対的に厚い膜試薬を含む毛細管充填スペース、および作用電極および対極を含んでいる。このテストストリップ300は、ロッシュ・ディアグノスティクス コーポレーション(インディアナ州、インディアナポリス市)からComfort Curve(登録商標)として市販されているものである。使用されたフェリシアナイド試薬は、表IおよびII中に記載された組成を持っていた。
実施例2で行われた測定も、図3Aおよび3B中に図示され、300で一般的に示されているテストストリップを用いて行った。上述したように、テストストリップ300は、ここで参照することにより組み入れられた米国特許第5,997,817号中に記載されているように、相対的に厚い膜試薬を含む毛細管充填スペース、および作用電極および対極を含んでいる。
実施例3で行われた測定も、図3Aおよび3B中に図示され、300において一般的に示されているテストストリップを用いて行った。上述したように、テストストリップ300は、ここで言及することにより組み入れられた米国特許第5,997,817号中に記載されているように、相対的に厚い膜試薬を含む毛細管充填スペース、および作用電極および対極を含んでいる。毛細管血液サンプルからのヘマトクリットレベルが、典型的には、30%から50%の間でのみ変化するため、20%から70%からのヘマトクリット範囲を持つスパイクされた静脈血液サンプルが、本実施例3のために使用された。5つのレベルのグルコース、温度(14,21,27,36および42℃)およびヘマトクリット(20,30,45,60および70%)は、独立して変化し、125のサンプルについて、共分散性研究が実現した。
実施例4で行われた測定も、図3Aおよび3B中に図示され、300で一般的に示されているテストストリップを用いて行った。上述したように、テストストリップ300は、ここで参照することにより組み入れられた米国特許第5,997,817号中に記載されているように、相対的に厚い膜試薬を含む毛細管充填スペース、および作用電極および対極を含んでいる。しかし、テストストリップは、異なった試薬を使用することにより、米国特許第5997817号中に記載されているテストストリップから改質された。使用されたニトロソアニリン試薬は、表IIIおよびIV中に記載されている組成物を持っている。
本発明の測定方法が、他のテストストリップ設計の場合にも同様に有用であることが見出された。実施例5は、図17Aおよび17B中に図示され、1700で一般的に示されているテストストリップ設計を用いて行った。図17Aを参照して、テストストリップ1700は、50nmの導電性(金)層(たとえば、スパタリングあるいは蒸着)で被覆された350μm厚みのポリエステル(好ましい実施形態においては、これは、デュポン社から入手可能なメリネックス329である。)の不透明な断片から形成されている底箔層1702を含んでいる。電極および接続トレースは、その後レーザ切除プロセスにより、導電性層にパターン形成され、示されているように、作用電極、対極および用量充足電極(以下にさらに詳細に記載されている)を形成する。このレーザ切除プロセスは、石英上クロムマスクを経由して通過するエキシマレーザ手段により行われる。マスクパターンは、レーザフィールドの一部の反射を生じ、一方フィールドの他の部分の通貨を生じ、レーザ光により接触された表面から放出されたパターンを金上に形成する。
実施例5に対して上述した同一のテストストリップ1700および試薬を用いて、図24中に示された励起プロファイルを、全テスト時間を減少させるために利用した。図5に対して記述されたように、20kHzおよび10kHzにおける位相角は、ヘマトクリット推定値と最も密接に相関していることが決定された。したがって、全テスト時間を減少させるために、実施例6中のこれらの2つの周波数に対して、AC部分の励起を限定することが決められた。全テスト時間をさらに減少させるために、10kHzAC励起をDC信号(すなわち、DCオフセットを持ったAC信号)に同時に印加され、またこの理論は、この組み合わされたモードが、DC電流、AC位相およびACアドミタンスに対する同時結果の収集を可能にし、最速の可能な結果を提供する。したがって、20kHz信号は、0.9秒に対して印加された。その後、10kHzおよびDC信号が、0.1秒間隔後1.0秒間同時に印加された。
検体測定プロセスに対する品質制御の追加の方策を提供するために、特に、このテストシステムが、素人の最終使用者により使用されることになるときには、ミス投与された(2回投与されるなど)センサー(テストストリップ)、以前使用されたセンサー、あるいは劣化した酵素を持っていたセンサー(あまりにも多湿の環境に貯蔵されたり、あまりにも古かったりなど)を検出することが、望ましい。これらの状態は、集約的には「不正使用センサー」として見なされる。不正使用センサーがテスト計中に挿入される場合には、検体測定プロセスを中断する(あるいは、テスト結果が正確ではないということを、少なくとも使用者に警告する)テストを考案することが望ましい。
信頼システム中のバイオセンサーのコットレル応答は、次の式により与えられる。

いくつかの実施形態は、測定シーケンス中に2つのDCブロックを含み得るため、改良されたコットレルフェイルセーフ比(MCFR)は、次のように公式化できる。
以上記述したように、正確なサンプル測定は、サンプルによる測定電極の十分なカバーが必要とすることが確認された。先行技術において、サンプル体積の不充分さを検出するために、種々の方法が使用されてきた。たとえば、ロッシュ・ダイアグノスティクス社、インディアナポリス、インディアナ州により販売されているアキュチェック(登録商標)アドバンテージ(登録商標)グルコーステスト計は、非コットレル電流減衰が、単一測定電極対により検出された場合に、サンプル体積の不足の可能性を使用者に警告した。使用者は、特定の時間割り当て以内に、テストストリップの再投与を支持された。
・毛細管充填スペースを形成する異なった壁の表面エネルギー間の不均衡、
・テストストリップ製造施設内の材料あるいは完成品の汚染、
・毛細管充填スペースの壁を構成する単一要素からの汚染物の非意図的導入(たとえば、放出ライナーが使用される製造工程で一般的な離型剤(典型的にはシリコン)がある)。
・毛細管充填スペース壁の異質な表面の荒れ、
・寸法アスペクト比、
・毛細管充填スペース内の汚染されたメッシュ材料、および
・毛細管充填スペース内のメッシュ材料上への界面活性剤の不均一な適用が、挙げられる。
毛細管充填スペースを用いるテストストリップ中の用量充足を適切に表示する電極システムを設計するために、毛細管充填スペースを経由してサンプルが、進行するにしたがって、サンプルのリード端における流頭形状を試験する実験が行われた。両面接着テープで結合された2枚の透明なポリカーボネートシートからなるテスト備品が、使用され、そこでは、毛細管充填スペースは、両面接着テープ中の経路を切断することにより形成された。ポリカーボネートの上シートおよび下シートの使用により、サンプルの流頭が毛細管充填スペースを経由して流れるためビデオテープに録画できる。

毛細管充填スペース中の凹状流頭に関連した問題は、図32Aから図32Cに図示されている。各々の図中では、テストストリップは、作用電極3200と、参照電極3202と、および測定電極3200あるいは3202の1つと併せて作動する下流の用量充足電極3204とを含む。上述した測定電極の1つと併せて、用量充足電極3204の使用に関連する測定ゾーンの化学量論的な問題に加えて、図32Aから図32Cは、凹形状を示すサンプル流頭が、またバイアスされた測定結果を生じることを示している。各図において、サンプルの移動方向が、矢印により示されている。図32Aにおいては、毛細管壁に隣接したサンプルの部分が、用量充足電極3204到達し、これにより用量充足性決定を行うためのテスト計によりモニターされる測定電極対の1つとこの電極のあいだのDC回路を電気的に完結させる。テスト計は、このとき測定するために充分なサンプルがあることを結論づけることになるが、サンプルは、参照電極3202にかろうじて到達し、このとき得られた測定結果は、高度にバイアスされる。
Claims (8)
- 生物学的流体テストサンプル内の検体濃度を測定する方法であって、
AC成分を有する第1信号を、前記テストサンプルに印加する工程、
前記テストサンプルのAC成分を有する第1電気的応答を測定する工程、
第2信号を前記テストサンプルに印加する工程、
前記テストサンプルの第2電気的応答を測定する工程、および
前記第1電気的応答の一部または全部と前記第2電気的応答に基づいて、前記テストサンプルの検体濃度を表示する値を決定する工程、前記テストサンプルの検体濃度を表示する値を決定する工程が前記第1電気的応答の一部または全部に基づいて、第2電気的応答を補正する工程、
前記第1信号および前記第2信号を、一部または全部が共通する時間のあいだ生物学的流体に印加する工程を
含むことを特徴とする方法。 - 第3信号を、血液サンプルに対して印加し、第3信号がAC成分を有すること、
前記第3信号に対する第3応答を測定する工程、
前記テストサンプルの検体濃度を表示する値を決定する工程が前記第1応答の一部または全部、第2応答および第3応答に基づくことであることを特徴とする請求項1記載の方法。 - 前記第1応答が、大きさおよび位相角情報を含むことを特徴とする請求項1または2記載の方法。
- 前記第1応答が、アドミタンス値を含むことを特徴とする請求項1〜3のいずれか1項に記載の方法。
- 前記第1信号のAC成分が、約1Hz以上で、かつ約20kHz以下である周波数を持つことを特徴とする請求項1〜4のいずれか1項に記載の方法。
- 前記第2テスト信号が、DC信号であることを特徴とする請求項1〜5のいずれか1項に記載の方法。
- 前記テストサンプルの検体濃度を表示する値を決定する工程が、前記第1信号の一部または全部に対する前記第1応答および前記第3信号に対する第3応答に基づいて、前記第2信号に対する第2応答を補正する工程を含むことを特徴とする請求項2〜6のいずれか1項に記載の方法。
- 前記テストサンプルの温度を測定する工程、およびAC応答および温度を用いて血液サンプルのヘマトクリット値を決定する工程をさらに含むことを特徴とする請求項1〜7のいずれか1項に記載の方法。
