Description
本発明は、組換え型タンパク質発現の一般的分野、HCV感染の診断、HCV感染の治療又は予防そして慢性肝炎を患う個体の治療の臨床的効率の予後診断/監視又は自然疾病の予後診断/監視に関する。
C型肝炎ウイルス(HCV)感染は、開発国及び開発途上国の両方において大きな健康上の問題である。世界の人口の約1〜5%がこのウイルスに冒されていると推定されている。HCV感染は、輸血による肝炎の最も重要な原因であると考えられ、往々にして慢性肝臓障害へと進行する。その上、肝細胞ガン腫の誘発にHCVが関与することを示す証拠が存在している。その結果として、信頼性の高い診断方法及び有効な治療剤に対する需要は高い。同様に、HCV感染した血液製剤の高感度かつ特異的スクリーニング方法及びHCVを培養するための改善された方法も必要とされている。
S.cerevisiae(野生型及びmnn9突然変異体)コアオリゴ糖の特徴は、末端のα1,3−結合されたマンノース残基の存在にある(Montesinoら(1998))。P.pastoris又はS.cerevisiae och1mnn1の中で発現されたタンパク質のN−グリコシル化部位に結合されたオリゴ糖には、このような末端のα1,3−結合されたマンノースが欠けている(Gellissenら(2000))。末端のα1,3結合されたマンノースは、アレルギー誘発性であるとみなされている(Jenkinsら(1996))。従って、そのオリゴ糖上に末端のα1,3−結合されたマンノース残基を担持しているタンパク質は、診断又は治療目的では適切でない。
本発明の第1の態様は、少なくとも1つのN−グリコシル化部位を含み、真核細胞の発現産物であること、さらにはN−グリコシル化部位の最高80%がコアグリコシル化されていることを特徴とする、単離されたHCVエンベロープタンパク質又はそのフラグメントに関する。特に、前記コアグリコシル化された部位の70%超がMan(8〜10)−GlcNAc(2)により定義された構造をもつオリゴマンノースでグリコシル化されている。さらに、構造Man(8)−GlcNAc(2)オリゴマンノースに対する構造Man(7)−GlcNAc(2)をもつオリゴマンノースの比は、0.45以下である。より詳細には、前記オリゴマンノースは10%未満の末端α1,3マンノースを含有する。前記単離されたS.cerevisiae又はその一部分を発現する真核細胞は、Hansenula細胞のごとき酵母細胞でありうる。
CL−[(A1)a−(PS1)b−(A2)c]−HCVENV−[(A3)d−(PS2)e−(A4)f]
という構造を特徴とするタンパク質から誘導され、式中
CLは、鳥類リゾチームリーダーペプチド又はその機能的等価物であり、
A1、A2、A3及びA4は、異なるもの又は同じものでありうるアダプタペプチドであり、
PS1及びPS2は、異なるもの又は同じものでありうるプロセッシング部位であり、
HCVENVは、HCVエンベロープタンパク質又はその一部分であり、
a、b、c、d、e及びfは0又は1であり、
ここで、A1及び/又はA2はPS1の一部分であってもよく、かつ/又はA3及び/又はA4はPS2の一部分であってもよい。
(i)前記HCVエンベロープタンパク質又はその一部分と前記抗−HCV抗体の複合体化を可能にする条件下で前記試料と請求項1〜15のいずれか1項に記載のHCVエンベロープタンパク質又はその一部分を接触させる段階;
(ii)(i)で形成された複合体を検出する段階;及び
(iii)前記試料中の前記抗−HCV抗体の存在を(ii)から推論する段階、を含んで成る方法である。
本発明に導く研究作業においては、Saccharomyces cerevisiae、Pichia pastoris及びHansenula plymorpha内のグリコシル化されたHCVエンベロープタンパク質の発現が、前記HCVエンベロープタンパク質に結合されたシグナルペプチド配列を含むタンパク質としての前記HCVエンベロープタンパク質の発現によって可能であるということが観察された。しかしながらこれら3つの酵母種の中で発現されたHCVエンベロープタンパク質のグリコシル化パターンは、非常に異なっていた(実施例6、10、13及び25参照)。より詳細には、S.cerevisiae(グリコシル化欠損突然変異体)及びH.polymorphaにより発現されたHCVエンベロープタンパク質は、コアグリコシル化に類似した要領でグリコシル化された。Pichia pastoris内で発現されたHCVエンベロープタンパク質は、この酵母内で発現されたタンパク質が通常、過剰グリコシル化されないという以前の報告(Gellisenら、2000,Sugrueら、1997)にも関わらず、過剰グリコシル化されていた。

1.例えばスルホン化及びチオエステル化によるようなシステイニルを可逆的に修飾する修飾剤による:
スルホン化は、ジスルフィド架橋に関与するチオール又はシステインがS−スルホネートに修飾される反応である:
RSH→RS−SO3 −(Darbre,A.1986)又はRS−SR→2RS−SO3 −(亜硫酸分解: Kumar,N.ら、1986)。スルホン化のための試薬は、例えばNa2SO3又はテトラチオン酸ナトリウムである。後者のスルホン化試薬は、10〜200mM、より好ましくは50〜200mMの濃度で使用される。任意にはスルホン化は、例えばCu2+(100μM〜1mM)又はシステイン(1〜10mM)のごとき触媒の存在下で実施可能である。
反応は、タンパク質変性ならびに未変性条件下で実施可能である(Kumar,N.ら、1985;Kumar,N.ら、1986)
チオエステル結合形成又はチオエステル化は、以下の式によって特徴づけされる:
2.例えば重金属、特にZn2+、Cd2+、モノ−、シチオ及びジスルフィド−化合物(例えばアリル−及びアルキルメタンチオスルホネート、ジチオピリジン、ジチオモルフォリン、ジヒドロリポアミン、エルマン試薬、アルドロチオールTM(アルドリッチ(Aldrich))(Rein,A.ら、1996)、ジチオカルバメート)、又はチオール化剤(例えばグルタチオン、N−アセチルシステイン、システインアミン)などにより本発明のシステイニルを可逆的に修飾する修飾用作用物質による。ジチオカルバメートは、スルフヒドリル基と反応する能力を付与するR1、R2NC(S)SR3官能基を有する広範なクラスの分子を含んで成る。などにより本発明のシステイニルを可逆的に修飾する修飾用作用物質による。チオール含有化合物が、好ましくは0.1〜50mM、より好ましくは1〜50mM、さらに一層好ましくは10〜50mMの濃度で使用される。
3.10μM〜10mM、より好ましくは1〜10mMの濃度範囲内での例えばDTT、ジヒドロアスコルベート、ビタミンS及び誘導体、マンニトール、アミノ酸、ペプチド及び誘導体(例えばヒスチジン、エルゴチオネイン、カルノシン、メチオニン)、没食子酸塩、ヒドロキシアニソール、ヒドロキシトルエン、ハイドロキノン、ヒドロキシメチルフェノール及びその誘導体のごとき、チオール状態を保存する(安定化させる)修飾用作用物質、特に酸化防止剤の存在による。
4.例えば(i)金属イオン(Zn2+、Mg2+)、ATPのごとき補因子、(ii)pH制御(例えば、タンパク質については大部分のケースにおいてpH〜5又はpHは好ましくはチオールpKa−2;例えば逆相クロマトグラフィにより精製されたペプチドについてはpH〜2に)のごときチオール安定化条件による。
− 特に1〜200mM、より好ましくは50〜200mMの濃度での還元剤、特にDTT、DTE、−2−メルカプトエタノール、ジチオナイト、SnCl2、水素化ホウ酸ナトリウム、ヒドロキシルアミン、TCEP;
− 例えばpH増大によるチオール安定化条件又は作用物質の除去;
− 特に0.01〜5μM、より一層詳細には0.1〜5μMの濃度範囲内の、特にチオエステラーゼ、グルタレドキシン、チオレドキシンのごとき酵素;
− 上述の化学的及び/又は酵素条件の組合せ。
(i)前記HCVエンベロープタンパク質又はその一部分と前記抗−HCV抗体の複合体化を可能にする条件下で前記試料と本発明によるHCVエンベロープタンパク質又はその一部分を接触させる段階;
(ii)(i)で形成された複合体を検出する段階;及び
(iii)前記試料中の前記抗−HCV抗体の存在を(ii)から推論する段階、を含んで成る方法に関する。特定の実施形態においては、段階(i)での接触は競合的条件で起こる。前記方法に対するもう1つの特定の実施形態においては、前記HCVエンベロープタンパク質又はその一部分は固体支持体に結合させられている。さらなる一実施形態においては、抗−HCV抗体を含むことが疑われている前記試料は生体試料である。
(i) HCV E1及び/又はHCV E2タンパク質又はその任意の一部分をコードするエンベロープ遺伝子で形質転換されたHansenula又はSaccharomycesグリコシル化マイナス菌株を適切な培地で増殖させる段階;
(ii) 前記HCV E1及び/又はHCV E2 遺伝子又はその任意の一部分の発現をひき起こす段階、及び

PFPMT−MFα−E1−H6シャトルベクターの構築
Hansenula polymorpha形質転換のためのプラスミドを以下のように構築した。pFPMT−MFα−E1−H6シャトルベクターを多工程手順で構築した。最初に、HCV E1sタンパク質をコードする核酸配列CSN21をCHHリーダー配列(CHH=Carcinus maenas血糖上昇ホルモン)に従ってクローニングし、これをその後MFαリーダー配列に変更した(MFα=Saccharomyces cererisiaeα−交接因子)。
プラスミドPGEMTE1sH6(配列番号6、図1)からPCRにより、[6−His−残基で伸長されたE1sのアミノ酸192〜326から成るHCV1bE1s型タンパク質についてコードする(配列番号5)]E1−H6DNAフラグメントを単離した。さらに、以下のプライマーを使用した。
−CHHE1−F:

pCHH−Hirプラスミド(配列番号9;図2)からPCRにより受容体プラスミドを製造し、これは、Hirコーディング配列がPCR産物内には存在しないという点を除いて、ほぼ完全なpCHH−Hirプラスミドで構成されている。このPCRのためには、以下のプライマーを使用した。

上述の通りPCRにより生成されたpCHH−Hir由来の受容体プラスミドとE1s−H6−コーディングDNAフラグメントを、供給業者の仕様書に従って、PCR産物精製キット(キアゲン(Qiagen))を用いて精製した。その後、精製されたフラグメントをEam1104Iで別々に消化した。その後、E1s−H6DNAフラグメントを供給業者の仕様書に従って、T4リガーセ(ベーリンガー)を用いてpCHH−Hir由来の受容体プラスミド内に連結させた。
pCHHE1kのEcoRI/BamHIフラグメントをEcoRI/BamHIで消化したベクターpFPMT121(配列番号12:図3)と連結させた。供給業者の指示に従ってT4リガーゼ(ベーリンガー)を使用した。E.ColiDH5αF細胞を形質転換するために連結混合物を使用した。プラスミドDNAの制限パターン上でいくつかの形質転換体を分析し、陽性クローンを保留し、これをpFPMT−CHH−E1H6と呼称した(配列番号13:図4)。
最後に、以下のような3つのフラグメントの連結により、シャトルベクターpFMPT−MFα−E1−H6を生成した。
1. 6.961kbのEcoRI/BamHIで消化されたpFPMT121(配列番号12:図3)

Hansenula polymorphaの形質転換のためのプロスミドを以下のように構築した。pFPMT−CL−E1−H6シャトルベクターを、pFPMT−MFα−E1−H6(配列番号16、図5)から出発して3工程で構築した。

pFPMT−MFα−E2−H6及びpMPT−MFα−E2−H6シャトルベクターの構築
Hansenula polymorphaの形質転換用プラスミドを以下の通りに構築した。MFα−E2s(HCV E2アミノ酸384−673)−VIEGR−His6(配列番号5)をコードするDNA配列を、プラスミドpSP72E2H6(配列番号22、図9)から1.331kbのEcoRI/BglIIフラグメントとして単離した。このフラグメントを、供給業者の推奨事項に従って、T4DNAリガーゼ(ベーリンガーマンハイム(Boehringer Mannheim))を用いてEcoRI/BglIIで消化されたpFPMT121(配列番号12、図C+2)又はpMPT121(配列番号23、図10)のいずれかと連結させた。E.coliを形質転換させ制限酵素消化により異なる形質転換体から単離されたプラスミドDNAを検査した後、陽性クローンを保留し、結果として得られたシャトルベクターをそれぞれpFPMT−MFα−E2−H6(配列番号22、図11)及びpMPT−MFα−E2−H6(配列番号23、図12)と呼称した。
pFPMT−CL−E2−H6シャトルベクターの構築
3工程手順で、シャトルベクターpFPMT−CL−E2−H6を組み立てた。E2コーディング配列がSchwanniomyces occidentalisのα−アミラーゼのシグナル配列の後ろでクローニングさせた。これは、シームレスクローニング方法(パジェット(Padgett),K.A.および、ゾルゲ(Sorge)J.A,1996)により行われた。
最初に、E2−H6をコードするDNA配列(リンカーペプチド「VIEGR」及び6His残基、配列番号5で伸長されたHCV E2のアミノ酸384−673)をpSP72E2H6プラスミド(配列番号24、図11)から、PCRにより増幅させた。使用したプライマーは、MF30E2/F及びMF30E/Rと記され、以下の配列を有している。

第2のフラグメントはプラスミドpMF30(配列番号28、図13)に由来し、アンプリコンは、S.occidentalisの成熟α−アミラーゼのコドンを除いて、完全なpMF30プラスミドであり、クローニングに関連する修飾をプライマー設計により導入した。以下のプライマーセットを使用した。
−プライマーMF30−Links;
PCRにより得たpMF30由来の受容体プラスミド及びE2s−H6コーディングDNAフラグメントを1%のアガロースゲル上のゲル電気泳動によりそのそれぞれのサイズに基づき検査した。供給業者の指示に従って、PCR産物精製キット(キアゲン)でPCR産物を精製した。その後、精製したフラグメントをEam11004Iで別々に消化した。供給業者の推奨事項に従ってT4リガーゼ(ベーリンガー(Boehringer))を用いることで、pMF30由来の受容体プラスミドとのE2s−H6フラグメントの連結を実施した。連結混合物を用いて、E.coliDH5αF’細胞を形質転換し、複数のクローンのプラスミドDNAをEcoRI/Bam HI消化により分析した。陽性クローンを選択し、そのプラスミドをさらにpAMY−E2と呼称し、以下に記載するようなさらなる修飾のために利用した。
α−アミラーゼシグナル配列のコドンを鳥類リゾチームプレ配列のコドンと置換する目的で、pAMY−E2をPCR誘導突然変異誘発に付した。これは、さらに「CL」と呼称され、鳥類リゾチームORFの最初の18個のアミノ酸(配列番号1)に対応している。この突然変異誘発のためには、以下のプライマーが用いられた:

(CL−E2(384−673)−VIEGR−H6を包含する)pUC18−CL−E2−H6から0.966kbのEcoRI/Bam HIフラグメントを単離し、EcoRI/Bam HIで消化されたpFPMT121(配列番号12、図3)の中に連結させた。反応のためには、供給業者の条件に従って、T4リガーゼ(ベーリンガー)を使用した。連結試料をエタノールで沈降させ、10μLの水中で溶解させた。これを用いて、E.coli DH5αF’細胞を形質転換し、制限分析の後に陽性クローンを保留し、それぞれのプラスミドをpFPMT−CL−E2−H6(配列番号32、図4)と呼称した。
pFPMT−CL−K−H6−E1シャトルベクターの構築
シャトルベクターの構築は、2工程から成る。

Hansenula polymorphaの形質転換及び形質転換体の選択
H.polymorpha菌株RB11を、実施例1〜5に記載されている通りの異なる親シャトルベクターでの修飾(Roggenkamp,R.)ら、1986)を用いて、形質転換させた(基本的にKlebe, R.J.)ら、1983)に記載の通りのPEG媒介DNA摂取プロトコル)。各々の形質転換について、72ウラシル原栄養コロニーを選択し、以下の手順により菌株生成のために使用した。各コロニーについて、2mLの液体培養を、48時間試験管の中で(37℃;160rpm;角度45℃)、選択培地(YNB/グルコース、Difco)内に接種し増殖させた。この工程は、第1継代工程として定義づけされる。第1の継代工程の培養の150μLのアリコートを用いて、2mLの新鮮なYNB/グルコース培地に接種した。ここでも又、上述のように培養をインキュベートした(第2の継代工程)。一緒にかかる継代工程を8回実施した。2mLの非選択的YPD培地(Difco)を接種するために、第3及び第8の継代工程の実施後の培養アリコートを使用した。37℃で48時間のインキュベーション(160rpm;角度45℃;いわゆる第1の安定化工程)の後、これらのYPD培養の150μLのアリコートを用いて、上述のとおりにインキュベートした新鮮な2mLのYPD培養を接種した(第2の安定化工程)。その後第2の安定化工程の培養のアリコートを、選択的YNB/寒天を含有する平板上で、画線培養した。これらの平板を、肉眼的コロニーが目に見えるようになるまで4時間インキュベートした。各分離の明確な単一のコロニーを菌株として定義し、さらなる発現分析のために使用した。
pSY1aMFElsH6aベクターの構築
S.cerevisiae 発現プラスミドを以下の通りに構築した。E1コーディング配列をpGEMT−E1sH6(配列番号6、図1)からのNsI1/Eco52Iフラグメントとして単離し、これを(T4DNAポリメラーゼを用いて)平滑末端化し、供給業者の仕様に従ってT4DNAリガーゼ(ベーリンガー)を用いてpYlG5ベクター(配列番号41、図19)内でクローニングした。クローニングは、E1s−H6コーディングフラグメントが直接的にかつaMFコーディング配列と同一枠内で接合されるようなものであった。連結混合物をE.coli DH5αF’細胞内で形質転換させた。その後、複数のアンピシリン耐性クローンのプラスミドDNAを制限消化により分析し、陽性クローンを保留し、pYIG5E1H6(ICCG3470;配列番号42、図20)と呼称した。
pSYYIGSE2H6ベクターの構築
S.cerevisiae 発現プラスミドpSYYIGSE2H6を以下の通りに構築した。E2コーディング配列をpBSK−E2sH6(配列番号45、図23)からのSalI/KpnIフラグメントとして単離し、これを(T4DNAポリメラーゼを用いて)平滑末端化し、その後、供給業者の仕様に従ってT4DNAリガーゼ(ベーリンガー)を用いてpYlG5ベクター(配列番号41、図19)内でクローニングした。クローニングは、E2−H6コーディングフラグメントが直接的にかつaMFコーディング配列と同一枠内で接合されるようなものであった。次に、連結混合物をE.coli DH5αF’細胞内で形質転換させ、複数のアンピシリン耐性クローンのプラスミドDNAを制限消化により分析し、陽性クローンを保留し、pYIG5HCCL−22aH6(ICCG2424;配列番号46、図24)と呼称した。
pSY1Y1G7E1sベクターの構築
S.cerevisiae 発現プラスミドpSY1YIG7E1sを以下の通りに構築した。E1コーディング配列をpGEMT−E1s(配列番号6、図1)からのNsI1/Eco52Iフラグメントとして単離し、これを平滑末端化し、供給業者の仕様に従ってT4DNAリガーゼ(ベーリンガー)を用いてpYlG7ベクター(配列番号48、図26)内でクローニングした。クローニングは、E1コーディングフラグメントが直接的にかつaMFコーディング配列と同一枠内で接合されるようなものであった。連結混合物をE.coli DH5αF’細胞へと形質転換させ、複数のアンピシリン耐性クローンのプラスミドDNAを制限消化により分析し、陽性クローンを保留し、pYIG7E1(配列番号49、図27)と呼称した。
Saccharomyces cerevisiaeの形質転換及び形質転換体の選択
Saccharomyces cerevisiae内で過剰発現されたタンパク質について報告されることの多い過剰グリコシル化の問題を克服するために、突然変異体スクリーニングをセットした。このスクリーニングは、自然発生的劣性オルトバナジウム酸塩耐性突然変異体を選択するBallouの方法(Ballou L.ら、1991)に基づいていた。初期菌株選択は、未変性ゲル電気泳動の後に見られるようなインベルターゼのグリコシル化パターンに基づいて実施された。グリコシル化能力が低減した菌株を、さらなる組換え型タンパク質発現実験のために保留し、これは菌株IYCC155より優勢であった。突然変異の性質についてはこれ以上研究しなかった。
pPICZalphaD’ElsH6及びpPICZalphaE’E1sH6ベクターの構築
pPICZalphaAベクター(Invitrogen;配列番号51、図29)から出発して、シャトルベクターpPICZalphaE’E1sH6を構築した。第1工程では、それぞれKEX2又はSTE13処理用プロテアーゼの開裂部位のすぐ後ろでE1コーディング配列のクローニングを可能にするように、前記ベクターを適合させた。従って、XhoI及びNotIでpPIC ZalphaAを消化した。消化物を1%のアガロースゲル上で分離させ、3519kbのフラグメント(ベクターの大部分)を、ゲル抽出キット(キアゲン)を用いて単離させ精製した。このFGを、その後、供給業者の条件に従って特異的オリゴヌクレオチドの存在下でT4ポリメラーゼ(ベーリンガー)を用いて連結させて、pPIC Zalpha D’(配列番号52、図30)又はpPIC ZalphaE’(配列番号53、図31)を生成した。

実施例11及び12に記載されている通りのP.pastorisシャトルプラスミドを、供給業者の条件(Invitrogen)に従ってP.pastoris細胞へと形質転換させた。E1−及びE2−産生菌株をさらなる特徴づけのために保留した。

Saccharomyces cerevisiae
セルバンキング
選択された組換え型クローンから、マスターセルバンク及びワーキングセルバンクを調製した。対数増殖中期の振とうフラスコ培養(発酵種培養の場合と同様のインキュベーション条件、以下参照)からクリオ−バイアル(cryo−vials)を調製した。凍結防止剤としてグリセロール(最終濃度50%)を添加した。
低温保存されたワーキングセルバンクから種培養を開始し、48時間200rpm、37℃で2L入り振とう三角フラスコの中で500mLの培地(2%のスクロールを補足したYNB、Difco)内でこれを増殖させた。
セルバンキング
選択された組換え型クローンから、マスターセルバンク及びワーキングセルバンクを調製した。対数増殖中期の振とうフラスコ培養(発酵種培養の場合と同様のインキュベーション条件、以下参照)からクリオ−バイアルを調製した。凍結防止剤としてグリセロール(最終濃度50%)を添加した。
低温保存された(−70℃)ワーキングセルバンクから種培養を開始し、48時間200rpm、37℃で2L入り三角振とうフラスコの中で500mLの培地(YPD、Difco)内でこれを増殖させた。
振とうフラスコ培養中に、組換え型Pichia pastorisでの小規模タンパク質産生実験をセットした。YPD培地(Difco)内で一晩、種培養を増殖させた。初期培地のpHを4.5に補正した。回転振とう機上で37℃、200〜250rpmで振とうフラスコをインキュベートした。
選択された酵母細胞内で発現されたMFa−E1−H6及びMFa−E2−H6タンパク質からのリーダーペプチドの除去
S.Cerevisiaeのa−交接因子(aMF)リーダー配列を伴うHCV E1及びE2タンパク質構築物のHansenula polymorpha及びSaccharomyces cerevisiaeグリコシル化マイナス菌株の中の発現産物をさらに分析した。両方の遺伝子型1bHCV E1s(aa192−326)及びHCV E2s(VIEGR(配列番号69)−配列で伸長されたaa383−673)は、共にC末端higタグ付けされた(H6、HHHHHH、配列番号63;このHCVタンパク質は本例ではさらにMF−E1−H6及びMF−E2−H6と記される)タンパク質として発現されたため、酵母細胞の塩化グアニジニラム(GuHCl)可溶化後の発現済み産物の迅速かつ効率の良い精製がNi−IDA(Ni−イミノジ酢酸)上で実施された。簡単に言うと、細胞ペレットを50mMのリン酸塩、6MのGuHCl、pH7.4(9vol/gの細胞)中に再懸濁させた。320mM(4%w/v)の亜硫酸ナトリウム及び65mM(2%w/v)の4チオン酸ナトリウムの存在下で室温(RTで一晩、タンパク質をスルホン化した。溶解物を、遠心分離(10.000g、30分、4℃)による凍結−解凍サイクルの後に清澄させ、エンピゲン(アイブライト&ウィルソン(Aibright&Wilson)、英国)及びイミタゾールを、それぞれ1%(w/v)及び20mMの最終濃度まで上清に添加した。試料をろ過し(0.22μM)、Ni−IDAセファロースFFカラム上に投入し、50mMのリン酸塩、6MのGuHCl、1%のエンピゲン(緩衝液A)に20mMのイミダゾールを補足したものを用いてこれを平衡化した。その後、カラムをそれぞれ20mM及び50mMのイミダゾールを含有する緩衝液Aを用いて、280nmという達成基線レベルまで洗浄した。緩衝液D、50mMリン酸塩、6MのGuHCl、0.2%(E1について)又は1%(E2について)エンピゲン、200mMイミダゾールを適用することによって、hisタグ付生成物を溶出させた。溶出した材料を、E1(IGH201)、又はE2(IGH212)に対し向けられた特異的モノクローナル抗体を用いたSDS−PAGE及びウエスタンブロットにより分析した。
大規模産生及び精製に適した酵母中のE1構築物の発現
S.Cerevisiae aMFリーダーペプチドに置き換えるため、CHH(Carcinus maenas高血糖ホルモンのリーダー配列)、Amg1(S.Occidentalisからのアミラーゼのリーダー配列)、Gam1(S.Occidentalisからのグルコアミラーゼのリーダー配列)、Phy5(真菌性フィターゼからのリーダー配列)、pho1(Pichia pastorisからのリーダー配列)及びCL(鳥類リゾチームC、1,4−ベータ−N−アセチルムラミダーゼCのリーダー)を含むその他の複数のリーダー配列を使用し、E1−H6(すなわちC末端his−タグを伴うE1)に連結させた。全ての構築物を、Hansenula polymorpha内で発現させ、結果として得られた細胞溶解物の各々をウエスタンブロット分析に付した。これによりすでに、CLがリーダーペプチドとして使用される構築物を除いて、リーダー又はシグナル配列又はペプチドの除去程度がきわめて低いという結論を下すことができた。このことは、Ni−IDAで精製された材料のEdman分解により、CHH−E1−H6構築物について確認された:複数の異なる配列が回収されたものの、適正に開裂された産物を検出することはできなかった(表4参照)。
CL−E2−H6コーディング構築物からのHansenula polymorpha内で発現されたHCV E2タンパク質の精製及び生化学的特徴づけ
Hansenula polymorpha内で発現されたCL−E2−VIEGR−H6(本例では以下「CL−E2−H6」と記す)タンパク質からのCLリーダーペプチドの除去効率を分析した。HCV E2s(aa383−673)はhis−タグ付けされたタンパク質として発現されたことから、Ni−IDAについて、収集済み細胞のGuHCl可溶化後の発現済みタンパク質の効率の良い高速精製を行った。簡単に説明すると、30mMのリン酸塩、6MのGuHCl、pH7.2の中に細胞ペレットを再懸濁させた(細胞1gあたり9mLの緩衝液)。タンパク質を、320mM(4%w/v)の亜硫酸ナトリウム及び65mM(2%w/v)の四チオン酸ナトリウムの存在下で室温で一晩、スルホン化させた。遠心分離(10.000g、30分、4℃/による凍結−解凍サイクルの後、溶解物を清澄させた。それぞれ1%(w/v)及び20mMの最終濃度までエンピゲンBB(アイブライト&ウィルソン)及びイミダゾールを添加した。さらなるクロマトグラフィー工程はすべて、AktaFPLCワークステーション(Pharmacia)上で実施した。0.22μmの孔径の膜(酢酸セルロース)を通して試料をろ過し、Ni−IDAカラム(Ni2+と共に投入されるキレート化セファロースFF)上に投入し、これを20mMのイミダゾールで補足された1%エンピゲンBB、pH7.2(緩衝液A)、6MのGuHCl、50mMのリン酸塩で平衡化した。カラムをそれぞれ20mM及び50mMのイミダゾールを含有する緩衝液Aで、280nmでの吸光度が基線レベルに達するまで洗浄した。緩衝液D、50mMのリン酸塩、6MのGHCl、0.2%のエンピゲンBB(pH7.2)、200mMのイミダゾールを適用することにより、his−タグ付けされた産物を溶出した。E2に対し向けられた特異的モノクローナル抗体E2(IGH212)を用いてSDS−PAGE及びウエスタンブロットにより、精製された材料を分析した(図41)。IMACで精製されたE2−H6タンパク質を同様に、Edman分解によるN末端配列決定に付した。さらに、タンパク質をN−グリコシダーゼF(ロシュ(Roche))(0.2U/μgのE2、PBS/3%エンピゲンBB中で37℃で1時間のインキュベーション)で処理するか、又は未処理のまま放置した。グリコシル化及び脱グリコシル化されたE2−H6タンパク質をSDS−PAGEに付し、アミノ酸配列決定のためPVDF膜上でブロットした(分析は、アプライドバイオシステム(Applied Biosystems)のPROCISETM492タンパク質シーケンサ、上で実施した。この工程で、SDS−PAGEは幾分かの分解産物を顕示したため、サイズ排除クロマトグラフィーによるさらなる分画を実施した。これに関して、Ni−IDA溶出液を限外ろ過によって濃縮させ(MWCO10kDa、centriplus、ミリポアアミコン(Amicon、Millipore))、PBS、1%のエンピゲンBB中でSuperdex G200(ファーマシア(Pharmacia))上に投入した。SDS−PAGE上の移動に基づき、〜30kDaと〜70kDaの間のMrをもつ無傷のE2s関連産物を主として含有する溶出画分をプールし、場合によってアルキル化させた(37℃で30分間5mMのDTTでのインキュベーションとそれに続く37℃で30分間20mMのヨードアセタミドでのインキュベーション)。IMAC精製の後に分解産物が存在する可能性は、かくしてサイズ排除クロマトグラフィーを用いた無傷の産物のさらなる分画によって克服できる。予想外に優れた結果を得ることができた。N末端配列決定に基づいて、CLリーダーペプチドが除去されているE2産物の量を推定することができた。タンパク質産物の合計量をEdman分解により回収されたピークの強度に基づいて、タンパク質のpmolとして計算する。その後、各々の特異的タンパク質(すなわち各々の「検出されたN末端」)について、合計に対するモル%を推定する。現行の実験では、E2−H6の適正なN末端のみが検出され、E2タンパク質内に含まれていないN末端アミノ酸を含有するか又はE2タンパク質のE2−H6欠如アミノ酸のその他の変異体は存在しなかった。結論としてはH.polymorphaによりCL−E2−H6タンパク質として発現されたE2−H6タンパク質は、95%以上の適正に開裂されたタンパク質としてそれ以上のいかなるin vitro処理も行なわずに、単離された。これは、単離されたタンパク質の25%中に発生すると推定された、E2−H6タンパク質に対するMF−E2−H6タンパク質のH.polymorphaによるリーダーペプチド除去の忠実度と好対照をなすものである(表3参照)。
CL−H6−K−E1コーディング構築物からのHansenula polymorpha内で発現されたHCV E1タンパク質の精製及び生化学的特徴づけ及びH6含有タンパク質のinvitro処理
H.Polymorpha内で発現されたCL−H6−K−E1タンパク質からCLリーダーペプチドの除去の効率、ならびにH6(his−タグ)−アダプタペタペプチド及びEndo Lys−Cプロセッシング部位を除去するためのその後のin vitroプロセッシングの効率を分析した。HCV E1s(aa192−326)は、N末端His−K−タグ付けされたタンパク質CL−H6−K−E1として発現されたことから、実施例17で記載された通り高速かつ高効率の精製を実施することができた。H6−K−E1(そして場合によってはCL−H6−K−E1)タンパク質のIMAC−クロマトグラフィー精製の溶出プロフィールは、図42に示されている。ゲルのSDS−PAGE及び銀染色及びE1に対して向けられた特異的モノクローナル抗体(IGH201)(図43)、組換え型E1s産物を含有する溶出画分(63−69)をプールし(「IMACプール」)、一晩エンドプロテイナーセLys−C(ロシュ)処理(酵素/基質比1/50(w/w)、37℃)に付してH6−K−融合テールを除去した。未処理の融合産物の除去をNi−IDAカラム上での頁のIMACクロマトグラフィー工程によって実施し、かくしてフロースルー画分内でEndo−Lys−C処理されたタンパク質を収集する。さらに、10mMのNaH2PO4・3H2O;1%(v/v)のエンピゲンB、pH7.2(緩衝液B)での10倍希釈の後、Ni−IDAカラム上にエンドブロテイナーゼLys−C消化されたタンパク質試料を適用し、その後、280nmでの吸光度が基線レベルに達するまで緩衝液Bで洗浄した。フロースルーを異なる画分(1−40)の形で収集し、これらの画分をE1s産物の存在についてスクリーニングした(図44)。(SDS−PAGE上の遊走とそれに続く銀染色又はE1に対して向けられた特異的モノクローナル抗体(IGH201)を用いたウエスタンブロット分析に基づき〜15kDaと〜30kDaの間のMrでの)N末端H6−K(そして場合によっては残留CL−H6−K)テールが除去されている無傷のE1を含有する画分(7〜28)をプールし、アルキル化した(37℃で30分間5mMのDTTでのインキュベーションとそれに続く37℃で30分間20mMのヨードアセタミドでのインキュベーション)。
ヘパリンによるHCV E1の低グリコシル化形態の特異的除去
酵母細胞由来のHCVウイルス様粒子のための特異的精製工程を発見するために、ヘパリンとの結合を評価した。ヘパリンは、複数のウイルスに結合することがわかっており、その結果、HCVウイルス様粒子に対する結合がすでに示唆されてきた(Garson J.A.ら、1999)。この潜在的結合を分析するため、ヘパリンをビオチニル化し、H.Polymophaからのスルホン化されたHCV E1、H.Polymorpha由来のアルキル化されたHCV E1(共に実施例16に記載されている通りに産生される)及びワクシニア発現ベクターでのトランスフェクションを受けた哺乳動物細胞の培養由来のアルキル化されたHCV E1のいずれかでコーティングされたマイクロタイタープレートの中で、HCV E1との相互作用を分析した。驚くべきことに、強い結合は、H.Polymorpha由来のスルホン化されたHCV E1でのみ観察され、一方哺乳動物の細胞培養由来のHCV E1での結合は全く存在しなかった。ウエスタンブロットを用いて、我々は、この結合が、低グリコシル化された成熟HCV E1い対応するHCV E1sタンパク質混合物のさらに低い分子量のバンドに特異的であること(図45)を示すことができた。図45は同様に、スルホン化を除去した時点でもなお結合が低分子量のE1について観察される(レーン4)ことから、このスルホン化がヘパリン結合にとって必須でないということをも顕示している。あるいはまた、アルキル化がこの結合を実質的に低減させているが、これは、この例で用いられている特異的なアルキル化剤(ヨード−アセタミド)によりひき起こされ得る。驚くべきことに、高度のグリコシル化を有する(すなわちより多くのグリコシル化部位が占有されている)HCV E1タンパク質を伴う調製物を得るためにHCV E1調製物を豊富化することが特異的に可能であることから、この発見事実はさらに、酵母のためのCL−HCV−エンベロープ発現カセットの工業的な利用可能性を実証した。
ウイルス様の粒子(VLP)の形成と分析
H.Polymorpha内で発現されたHCV E1及びE2タンパク質(実施例16〜18)のVLPへの変換は、基本的に、WO99/67285中でDeplaら、によって又WO01/30815中でボスマン(Bosman)ら、によって記載されている通りに行なわれた。簡単に言うと、HCVウイルス様粒子が発現された形質転換済みH.Polymorpha細胞の培養の後、細胞を収穫し、実施例17で記載された通りにGuHCl中で溶解させスルホン化させた。His−タグ付けされたタンパク質をその後IMACによって精製し、実施例17で記載されているように、限外ろ過により濃縮した。
単離手順中にスルホン化された濃縮HCVウイルス様粒子を還元処理に付さずPBS、1%(v/v)のエンピゲンで平衡化されたサイズ排除クロマトグラフィーカラム(Superdex G200、pharmacia)上に投入した。溶出した画分をSDS−PAGE及びウエスタンブロット法で分析した。相対Mr〜29−〜15kD(SDS−PAGE移動に基づき)をもつ画分をプールし、濃縮し、Superdex G200上に投入し、PBS、3%(w/v)ベタインで平衡化して、ウイルス様粒子(VLP)を形成させた。画分でプールし、濃縮し、PBS、0.5%(w/v)ベタインに対し脱塩した。
単離手順中にスルホン化された濃縮HCVウイルス様粒子を還元処理(PBS中の5mMのDTTの存在下でのインキュベーション)に対して、スルホン化されたCys−チオール基を遊離Cys−チオール基に変換した。(i)20mMのヨートアセタミドの存在下での30分間のインキュベーション又は(ii)5mMのN−エチルマレイミド(NEM)及び15mMのビオチン−N−エチルマレイミドの存在下での30分間のインキュベーションにより、不可逆的Cys−チオール修飾を実施した。その後、ヨードアセタミド遮断の場合にはPBS、1%(v/v)エンピゲン、又はNEM及びビオチン−NEMでの遮断の場合にはPBS、0.2%CHAPSで平衡化されたサイズ排除クロマトグラフィーカラム(Superdex G200、Pharmacia)上に、タンパク質を投入した。溶出した画分をSDS−PAGE及びウエスタンブロット法で分析した。〜29−〜15kD(SDS−PAGE移動に基づく)の相対Mrをもつ画分をプールし、濃縮し、ウイルス様粒子(VLP)を形成させるために、PBS、3%(w/v)ベタインで平衡化された、Superdex G200上に投入した。画分をプールし、濃縮し、ヨードアセタミド遮断の場合にはPBS、0.5%(w/v)ベタインに対して、又NEM及びビオテン−NEMでの遮断の場合にはPBS、0.05%のCHAPSで脱塩した。
単離手順中にスルホン化された濃縮HCVウイルス様粒子を還元処理(PBS中の5mMのDTTの存在下でのインキュベーション)に付して、スルホン化されたCys−チオール基を遊離Cys−チオール基に変換した。ジチオジピリジン(DTDP)、ジチオカルバメート(DTC)又はシステインの存在下での30分間のインキュベーションにより、可逆的Cys−チオール修飾を実施した。その後、PBS、1%(v/v)エンピゲンで平衡化されたサイズ排除クロマトグラフィーカラム(Superdex G200、Pharmacia)上に、タンパク質を投入した。溶出した画分をSDS−PAGE及びウエスタンブロット法で分析した。〜29−〜15kD(SDS−PAGE移動に基づき)の相対Mrをもつ画分をプールし、濃縮し、ウイルス様粒子(VLP)を形成させるために、PBS、3%(w/v)ベタインで平衡化された、Superdex G200上に投入した。画分をプールし、濃縮し、PBS、0.5%(w/v)ベタインに対して脱塩した。
動的光散乱によりVLP粒度を決定した。光散乱実験のためには、光子相関分光法(PCS、ソフトウエアにより制御される粒度分析装置(Zetasizer 1000 HS型、マルバーンインストラメント社(Malvern Instruments Ltd.,)ウスターマルバーン(Malvern,Worcester)英国)を使用した。光子相関分光法又は動的光散乱(DLS)は、ブラウン運動を測定しそれを粒度に関係づけする光学的方法である。連続的な可視レーザービームからの光は、懸濁状態にありブラウン運動に基づいて移動する巨大分子又は粒子全体を通して導かれる。レーザー光の一部分は、粒子によって散乱され、この散乱光は、光電子増倍管によって測定される。散乱光の強度の変動は、相関装置内に供給される。こうして適切なデータ解析が実施されるコンピュータへと渡される自動相関関数が生成される。使用されたレーザーは、633nmの固定波長をもつ10mwの単色コヒーレントHe−Neレーザーであった。各試料について、3〜6個の連続的測定が行なわれた。これらの実験の結果は表5にまとめられている。
Hansenulaが産生したHCV E1−H6及びワクシニア感染した哺乳動物細胞が産生したHCV E1の抗原的等価性
HCV慢性キャリヤからの血清とHansenula産生したHCV E1−H6との反応性を、WO99/67285でデプラ(Depla)ら、により記載されている通りに、HCV組換え型ワクシニアウイルス感染哺乳動物細胞が産生したHCV E1の反応性と比較した。テスト対象の両方のHCV−E1調製物は共に、HCV E1タンパク質がNEM及びヒオチン−NEMでアルキル化されたVLPで構成されていた。HCV慢性キャリヤからの血清との両方のHCV E1 VLP調製物の反応性は、ELISAによって決定された。結果は表6にまとめられている。表6から導出できるように、HCV組換え型ワクシニアウイルス感染哺乳動物細胞内で発現されたHCV E1と、H.polymorphaの中で発現されたHCV E1の間には反応性の違いは全く認められなかった。
Hansenulaが産生したHCV E1−H6及びワクシニア感染した哺乳動物細胞により産生されたHCV E1の免疫原性等価性
Hansenulaが産生したHCV E1−H6の免疫原性を、WO99/67285でデプラら、が記載している通りにHCV組換え型ワクシニア感染哺乳動物細胞により産生されたHCV E1の免疫原性と比較した。テスト対象の両方のHCV−E1調製物は共に、HCV E1タンパク質がヨードアセタミドでアルキル化されたVLPで構成されていた。VLP調製物は両方共ミョウバンと共に処方され、Balb/cマウスの体内に注入された(各々3週間の間隔をおいて、各々0.13%のAlhydrogel、Superfos、デンマークを含有する125μl中5μgのE1から成る三回の筋内/皮下注射)。3回目の免疫化の後10日目にマウスを放血させた。
スルホン化されているHansenula産生HCV E1−H6の抗原性及び免疫原生
HCV慢性キャリヤの血清とHansenula産生HCV E1−H6血清の反応性をWO99/67285内のDeplaら、により記載されている通りに、HCV組換え型ワクシニアウイルス感染哺乳動物細胞により産生されたHCV E1の反応性と比較した。テスト対象のHCV−E1調製物は両方共、Hansenulaにより産生されたHCV E1タンパク質がスルホン化され哺乳動物細胞により産生されたHCV E1がアルキル化されたVLPで構成されていた。結果は、表7に示されている。全体的(平均的)反応性は、同一であったが、個々の血清についていくつかの主要な差異が指摘された。このことは、スルホン化された材料が、アルキル化されたHCV E1とは異なる形でそのエピトープの少なくともいくつかを提示するということを暗に意味する。
ワクチン接種を受けたチンパンジーからの血清との、Hansenulaで産生されたHCV E1−H6及びワクシニア感染した哺乳動物細胞によって産生されたHCV E1の同一の抗原反応性
HCV組換え型ワクシニアウイルス感染哺乳動物細胞により産生されたE1及びHansenulaにより産生されたE1−H6(両方共アルキル化されている)の、ワクチン接種を受けたチンパンジーからの血清及びモノクローナル抗体との反応性を比較した。さらに、前記E1タンパク質をELISA内のプレートに直接コーティングしその後カゼインでプレートを飽和させた。ELISAプレートにコーティングされたE1タンパク質を結合させる抗体の終点力価を、全て哺乳動物細胞によって産生されたE1で免疫化された動物から得られた特異的マウスモノクローナル抗体及びチンパンジー血清について決定した。終点力価決定は、実施例22に記載されている通りに行なわれた。使用されたマウスモノクローナル抗体はIGH201(実施例15を参照)、IGH198(IGH198=WO96/04385内のメルテンスら、中の23C12)、IGH203(IGH203=WO96/04385内のメルテンスら、中の15G6)及びIGH202(IGH202=WO99/50301内のメルテンスら、中の3F3)であった。
蛍光体援用型炭水化物電気泳動(FACE)によるHCV E1の糖プロファイリング
グリコシル化プロフィールをWO99/67285でデプラら、によって記載されているように、HCV組換え型ワクシニアウイルスに感染した哺乳動物細胞により産生されたHCV E1及びHansenula産生HCV E1について比較した。これは、蛍光体援用型炭水化物電気泳動(FACE)を用いて行なわれた。さらに、オリゴ糖を、ペプチド−N−グリュシダーゼ(タンパク質Gase F)により哺乳動物細胞又はHansenulaにより産生されたE1sから遊離させ、ANTSで標識づけした(PNGase F消化に先立ち、ヨートアセタミドでE1タンパク質をアルキル化した)。2−3時間4℃で15mAの電流にて21%のポリアクリルアミド上でPAGEにより、ANTS標識づけされたオリゴ糖を分離した。図54から哺乳動物細胞により産生されたE1及びHansenulaにより産生されたE1−H6上のオリゴ糖が単糖7〜11個という重合度でオリゴマルトースのように移動するということが結論づけされた。このことはすなわち、Hansenula発現系が驚くべきことに、過剰グリコシル化されずかつ哺乳動物細胞中で産生されたE1タンパク質に添加された糖鎖と類似の長さをもつ糖鎖を有するE1タンパク質を導く、ということを表わしている。
Saccharomyces及びHansenulaで産生されたE1s及びHCV組換え型ワクシニアウイルスに感染した哺乳動物細胞により産生されたE1sから誘導されたN−結合したオリゴ糖の配列決定
PBS、0.5%ベタイン中に存在しSaccharomyces又はHansenula培養(実施例15−16参照)又はHCV細胞え型ワクシニアウイルスに感染したRK13細胞の培養(MaertensらW096/04385明細書参照)から精製されたE1s(225μg)を、40μg/mlという最終濃度まで、脱グリコシル化インキュベーション緩衝液(50mM、Na2HPO4、0.75%のNonidet P−40)で希釈した。溶液のpHを濃H3PO4でpH5.5に調整した。この溶液に対し、2UのPNGaseF(Flavobacterium meningosepticumのペプチド−N4−(アセチル−ベータ−グルコサアニル)−アスパラギンアミダーゼ;EC3.5.1.52;Roche社より入手)を添加し、試料を37℃で一晩インキュベートした。一晩インキュベートした後、溶液のpHを濃H3PO4でpH5.5に調整した。タンパク質及びオリゴ糖をその後、4体積のアセトン(−20℃で)を添加することで沈殿させ、混合物を15分間−20℃でインキュベートした。試料を4℃で5分間1300rpmで遠心分離させた。アセトン上清を廃棄し、150μLの氷冷60%メタノールを添加した後1時間−20℃でペレットをインキュベートした。遊離されたオリゴ糖を含有するメタノール上清を収集し、ロータリーエバポレーション(Speed Vac)により乾燥させた。
組換え型HCV E1内のN−グリコシル化部位の占有
占有されたN−グリコシル化部位の量に応じて、E1sは、SDS−PAGE分析上で異なる泳動挙動を示す。この特性に基づいて、E1産物内の占有されたN−グリコシル化部位の平均的量を推定することができた。この点に関し、精製されたE1産物の試料を、SDS−PAGE及びクーマシーブリリアントブルー染色に付し(図67)、Image Master 1 D Prime Softwareパケット(Pharmacia)を用いてさらに分析した。簡単に説明すると、ゲルをスキャンし、各々の特定のタンパク質バンドについて、出現%(全てのバンドの合計を100%にする異なるバンドの合計強度との関係におけるその強度)を推定した(表11)。各々の特定のタンパク質バンドが、同じ占有されたN−グリコシル化部位数をもつE1s−分子を表わしているということに留意すべきである。
組換え型HCV E2内のN−グリコシル化部位の占有
Hansenulaにより産生された200μgのE2−H6タンパク質をPNGase下で脱グリコシル化した。脱グリコシル化されたE2s−H6をミニ・ゲル(10μg/レーン)上に投入した。タンパク質バンドをトリプシン及びエンドAsp−Nで消化した。結果として得られたペプチドの質量をMaldi−MS(乾燥液滴及び薄層法)で決定した。
Saccharomyces又はHansenulaにより産生されたHCV E1との血液ドナー血清の反応性
(α−MFリーダーで発現された)Saccharomycesにより産生されたE1s−H6及び(CLリーダで発現された)Hansenulaにより産生されたH1s−H6を両方共、実施例15及び16の中で記載されている通りに精製し、実施例20中で記載されている通りに最終的にアルキル化及びVLP形成に付した。両方のタンパク質を直接0.5μg/mLのマイクロタイタープレート上に吸着させ(1時間37℃)、プレートを遮断した後(PBS−0.1%カゼイン、1時間37℃)、HCVスクリーニングを受け陰性の血液ドナーからの血清を1/20の希釈度(PBS−0.5%カゼイン、10%(w/v)スクロース、0.2%(v/v)Triton−X−705、1時間、37℃)でインキュベートした。最終的に、1/50000の希釈度で(PBS−0.1%カゼイン、1時間、RT)ペルオキシダーゼ(Dako、Denmark)にカップリングされた二次的ウサギ抗ヒトIgG−Fc特異的抗血清を用いて結合を検出し、その後発色させた。全ての段階の間でプレートをPBS−0.05%(w/v)Tween−20で3回洗浄した。比較のため、Deplaら、がW099/67285明細書中で記載した通りに産生され精製された哺乳動物細胞由来のE1sについても同一の要領で血清を分析した。

Claims (33)
- 少なくとも1つのN−グリコシル化部位を含み、酵母細胞の発現産物であり、さらにはN−グリコシル化部位の80%までが占有されており、該占有されたN−グリコシル化部位がコアグリコシル化されており、該コアグリコシル化が10%未満の末端α1,3マンノースを含有することを特徴とする、単離されたHCVエンベロープタンパク質。
- 前記コアグリコシル化された部位の70%よりも多くが8〜10個のマンノースを含むオリゴマンノースでグリコシル化されている、請求項1に記載の単離されたHCVエンベロープタンパク質。
- Man(8)−GlcNAc(2)で定義される構造をもつオリゴマンノースによりコアグリコシル化された部位に対するMan(7)−GlcNAc(2)で定義される構造をもつオリゴマンノースによりコアグリコシル化された部位の比が、0.45以下である、請求項1または2に記載の単離されたHCVエンベロープタンパク質。
- 前記酵母細胞がHansenula細胞である、請求項1から3のいずれか1項に記載の単離されたHCVエンベロープタンパク質。
- 前記HCVエンベロープタンパク質に結合されたアミノ酸配列MRSLLILVLCFLPLAALG(配列番号99)により定義されるニワトリリゾチームリーダーペプチド又はその機能的変異体を含むタンパク質から誘導される、請求項1〜4のいずれか1項に記載の単離されたHCVエンベロープタンパク質であって、前記機能的変異体は、前記ニワトリリゾチームリーダーペプチドのアミノ酸配列において1または数個のアミノ酸の欠失、置換、又は付加を有し、かつプレタンパク質を粗面小胞体にターゲティングする能力を有する、タンパク質。
- CL−[(A1)a−(PS1)b−(A2)c]−HCVENV−[(A3)d−(PS2)e−(A4)f]
という構造を特徴とするタンパク質から誘導され、式中:
CLは、アミノ酸配列MRSLLILVLCFLPLAALG(配列番号99)により定義されるニワトリリゾチームリーダーペプチド又はその機能的等価物であり、
A1、A2、A3及びA4は、異なるもの又は同じものでありうるアダプタペプチドであり、
PS1及びPS2は、異なるもの又は同じものでありうるプロセッシング部位であり、
HCVENVは、HCVエンベロープタンパク質又はその一部分であり、
a、b、c、d、e及びfは0又は1であり、
ここで、前記機能的等価物は、前記ニワトリリゾチームリーダーペプチドのアミノ酸配列において1または数個のアミノ酸の欠失、置換、又は付加を有し、かつプレタンパク質を粗面小胞体にターゲティングする能力を有し、A1及び/又はA2はPS1の一部分であってもよく、かつ/又はA3及び/又はA4はPS2の一部分であってもよい、
請求項5に記載の単離されたHCVエンベロープタンパク質。 - Aが配列番号63−65、70−72及び74−82から選択されるアミノ酸配列からなり、PSが配列番号66−68及び83−84から選択されるアミノ酸配列からなるか、又はPSが二塩基部位又は一塩基部位であり、かつHCVENVが配列番号85−98から選択されるアミノ酸配列からなる、請求項6の単離されたHCVエンベロープタンパク質。
- PSがLys−Lys、Arg−Arg、Lys−Arg及びArg−Lysから選択される二塩基部位である、請求項7に記載の単離されたHCVエンベロープタンパク質。
- PSが一塩基部位Lysである、請求項7に記載の単離されたHCVエンベロープタンパク質。
- 単量体、ホモダイマー、ヘテロダイマー、ホモオリゴマー及びヘテロオリゴマーからなるグループの中から選択される構造の中に含まれている、請求項1〜9のいずれか1項に記載の単離されたHCVエンベロープタンパク質。
- ウイルス様粒子内に含まれている、請求項1〜9のいずれか1項に記載の単離されたHCVエンベロープタンパク質。
- システインのチオール基が化学的に修飾されている請求項1〜11のいずれか1項に記載の単離されたHCVエンベロープタンパク質。
- 抗原性である、請求項1〜12のいずれか1項に記載の単離されたHCVエンベロープタンパク質。
- 免疫原性である、請求項1〜12のいずれか1項に記載のHCVエンベロープタンパク質。
- T細胞エピトープを含んで成る、請求項1〜12のいずれか1項に記載の単離されたHCVエンベロープタンパク質。
- 請求項1〜15のいずれか1項に記載の単離されたHCVエンベロープタンパク質を含んで成る組成物。
- 薬学的に受容可能な担体をさらに含み、薬剤である、請求項16に記載の組成物。
- 薬学的に受容可能な担体をさらに含み、ワクチンである、請求項16に記載の組成物。
- 請求項1〜15のいずれか1項に記載の単離されたHCVエンベロープタンパク質を産生するための方法。
- 抗−HCV抗体を含むことが疑われている試料中の抗−HCV抗体の存在を検出する方法であって、
(i)前記HCVエンベロープタンパク質と前記抗−HCV抗体の複合体化を可能にする条件下で前記試料と請求項1〜15のいずれか1項に記載のHCVエンベロープタンパク質を接触させる段階;
(ii)(i)で形成された複合体を検出する段階;及び
(iii)前記試料中の前記抗−HCV抗体の存在を(ii)から推論する段階、
を含んで成る方法。 - 段階(i)での前記接触が競合的条件で起こる、請求項20に記載の方法。
- 前記HCVエンベロープタンパク質が固体支持体に結合させられている、請求項20に記載の方法。
- 抗−HCV抗体を含むことが疑われている試料中の抗−HCV抗体の存在を検出するための診断用キットであって、請求項1〜15のいずれか1項に記載のHCVエンベロープタンパク質を含んで成るキット。
- 前記HCVエンベロープタンパク質が固体支持体に結合させられている、請求項23に記載の診断用キット。
