ナノ粒子薬物コンジュゲート(NDC)は、抗体薬物コンジュゲート(ADC)などのその他の薬物担体プラットフォームの欠点の多くを解決する可能性を提供する。例えば、超小型サイズ(例えば、20nmまたはそれよりも小さい直径)のNDCは、ADCなどのその他のプラットフォームを使用すると不可能な、破壊された血液脳関門を通過しかつ治療物質を脳腫瘍に送達することができる。超小型ナノ粒子は、治療分子の腫瘍に対する深い腫瘍浸透を示すことができかつ均質な送達をもたらすことができ、一方、ADCなどのその他の薬物担体プラットフォームは、遅い腫瘍内拡散に起因して、限定された腫瘍浸透を有する。NDCは、ADCなどの従来の薬物送達プラットフォームよりも著しく多くの薬物分子を運ぶこともでき、それによってNDCは、比較的より多くの量の薬物をがん細胞に送達することが可能になる。このことは、十分な量の薬物の送達を困難にする、受容体発現が低いがん細胞を標的とする場合に、特に有用である。NDCの別の利点は、有機ポリマー層でナノ粒子をコーティングする、例えばナノ粒子表面をPEG基の層でコーティングする可能性であり、生理学的環境において(例えば、対象において)血清タンパク質のナノ粒子への吸着を防止することができ、効率的な尿排泄を容易にし、かつナノ粒子の凝集を減少させ得る(例えば、Burns et al. ”Fluorescent silica nanoparticles with efficient urinary excretion for nanomedicine”, Nano Letters (2009) 9(1):442-448参照)。

したがって、ナノ粒子を官能化しかつ分子とナノ粒子、例えば薬物分子または標的化リガンドとのコンジュゲーションを可能にするための方法に、未だ満たされないニーズがある。
本開示は、ナノ粒子を官能化する方法を特徴とする。方法は、化合物をナノ粒子表面(例えば、がん標的化リガンドなどの標的化リガンド、例えば、葉酸受容体標的化リガンド、例えば葉酸;および細胞毒性化合物などの治療剤、例えばエキサテカン)にコンジュゲートして、ナノ粒子薬物コンジュゲート(NDC)を形成するのに適したナノ粒子の調製を含むことができる。得られたNDCは、コンジュゲートされた分子とナノ粒子との非常に安定した連結を示し、また標的化された生体系における、例えばがん細胞における有効な薬物放出をもたらす。
選択された化学的定義
本明細書で使用される場合、「アルキル」という用語は、1から18個の炭素原子、例えば1から約12個の炭素原子、または1から約6個の炭素原子を含んでいてもよい1価の脂肪族炭化水素基(「C1~18アルキル」)を指す。アルキル基は、直鎖、分岐鎖、単環式部分、または多環式部分、またはこれらの組合せとすることができる。アルキル基の例には、メチル、エチル、プロピル、イソプロピル、ブチル、iso-ブチル、sec-ブチル、tert-ブチル、ペンチル、ヘキシル、シクロプロピル、シクロブチル、シクロペンチル、シクロヘキシル、ノルボルニル、および同様のものが含まれる。アルキル基のそれぞれの場合は、独立して、必要に応じて置換されてもよく、即ち、置換されなくても(「非置換アルキル」)または1個または複数の置換基で、例えば1から5個の置換基、1から3個の置換基、または1個の置換基で置換されても(「置換アルキル」)よい。
本明細書に開示される方法は、任意の適切なナノ粒子を官能化するのに使用されてもよい。例えば、シリカナノ粒子は、本明細書に開示される方法を使用して、官能化することができる(シリカから部分的に構成される、またはシリカから完全に構成されるナノ粒子を含む)。ナノ粒子は、約0.5nmから約100nm、例えば約0.1nmから約50nm、約0.5nmから約25nm、約1nmから約20nm、約0.8nmから約15nm、約1nmから約10nm、または約1nmから約8nm、例えば約1nm、約2nm、約3nm、約4nm、約5nm、約6nm、約7nm、約8nm、約9nm、約10nm、約11nm、または約12nmの直径を有することができる。

材料および方法
全ての材料は、受け取ったままの状態で使用した。全ての非水性反応は、火炎乾燥したガラス器具で、アルゴンの陽圧下で実行した。無水溶媒は、商業上の供給元(RANKEM)から購入した。フラッシュクロマトグラフィーを、指示される溶媒系により230~400メッシュのシリカゲル上で行った。プロトン核磁気共鳴スペクトルを、重水素化クロロホルムまたはDMSOを溶媒として使用して、Bruker分光計で、400MHZで記録した。ピーク位置は、内部標準としてのテトラメチルシランから百万分率単位で低磁場に与えられた。J値は、ヘルツを単位として表される。質量分析は、エレクトロスプレー(ES)技法を使用する(Agilent/Shimadzu)分光計で行った。HPLC分析は、(Agilent/Waters)、Gemini C-18(1000×4.6mm;5u)を備えたPDA-UV検出器で行い、試験した全ての化合物は、この方法を使用して>95%純粋であることを決定した。本明細書に記述される手順に従い調製された化合物は、分取HPLC法によって単離されてもよい。シアニン5マレイミドは、Lumiprobeから購入した。DBCO-PEG4-マレイミド、葉酸-PEG-アジド、およびエキサテカン-リンカーコンジュゲートは、WuXi AppTec、Tianjin、Chinaによるカスタム合成から得た。(3-メルカプトプロピル)トリメトキシシラン、2-[メトキシ(ポリエチレンオキシ)6-9プロピル]トリメトキシシラン、および(3-シクロペンタジエニルプロピル)トリエトキシシラン前駆体を、Gelestから購入した。10×PBSを、Thermo Fisher Scientificから購入した。ヒトカテプシン-Bは、Millipore Sigmaから購入した。ヒトおよびマウス血清は、BIOIVT(New York)から購入した。全てのその他の化学物質は、Millipore Sigmaから購入した。Cy5-C’ドットは、参照によりその全体が本明細書に組み込まれるMa, K. et al. DBCO-PEG-シランを使用した例示的な官能化ナノ粒子の合成。

有機ポリマーコーティング(例えば、PEG層)の下のナノ粒子の表面のシラノール基は、DBCO-PEG-シランなどの嵩高い分子に対して容易にアクセス可能ではない。結果は、DBCO-PEG-シランを二官能性前駆体として使用した場合(例えば、実施例1におけるように)、DBCO-PEG-シランのナノ粒子に対する反応比が高い場合であっても(図2参照)、1個または2個のDBCO基しか各ナノ粒子(例えば、C’ドット)に付着できない。したがって、代替の2ステップ反応が設計され、スキーム4に示されるように高い拡散係数をもつ比較的小さいシラン分子を用いた。
コンジュゲーション化学合成のスクリーニング
実施例2に記述されるアミン-シラン前駆体を使用することの欠点を克服するために、コンジュゲーション化学のライブラリーをスクリーニングし、これを表1にまとめる。

ジエン系DBCO-C’ドットの合成
Diels-Alder反応の使用は、アミン-エステル反応の適切な置換えとして実施例3で特定された。例えば、実施例2に記述される方法で使用したアミン-シランは、シクロペンタジエン基でC’ドットを最初に官能化するために(3-シクロペンタジエニルプロピル)トリエトキシシラン(「ジエン-シラン」)と置き換えることができ、実施例2の方法で使用したDBCO-PEG-NHSエステルは、DBCO-PEG-マレイミドと置き換えることができる。この方法をスキーム5に示し、DBCO-PEG-マレイミド上のマレイミド基とC’ドット上のジエン基との間のDiels-Alder反応を使用して、DBCO(リンカーを介して)をナノ粒子に付着することができる。
FA-CDCの合成
本明細書に開示される方法によって生成されたアルキン部分(例えば、DBCO)を含むNDCを、反応性基、例えばアルキン反応性基(例えば、アジド)を含む化合物とさらにコンジュゲートすることができる。これらの化合物は、例えば合成実施例1に記述されるようにペイロード-リンカーコンジュゲート前駆体、および/または例えば合成実施例2に記述されるように葉酸塩標的化リガンド前駆体を含むことができる。

食塩水、PBS、ヒトおよびマウス血清におけるNDCの安定性
本明細書で生成されたNDCの安定性を、0.9%の食塩水、PBS、ヒト血漿(10%)、およびマウス血漿(10%)中、37℃で、振盪乾燥浴内で種々の期間にわたりインキュベーションした後、評価した。分析前に、試料中の血漿タンパク質を、等体積の冷アセトニトリルの添加を経た沈殿によって除去し、その後、Eppendorf 5425微量遠心分離機で10000rpmで遠心分離した。遠心分離後、透明な上澄みを、遠心管から透明な完全回収HPLCバイアルに移した。目に見えるいかなる凝集もない上澄みを、等体積の脱イオン水で希釈して、HPLC分離の出発条件に一致するようにかつ感度の損失が回避されるように、試料マトリクスを調節した。次いで各試料の純度および不純物を、上述のようにRP-HPLCにより定量した。実施例2および実施例4の方法により生成されたNDCのヒトおよびマウス血清における安定性を、図8A~8Bに提示する。

Claims (33)
- ナノ粒子を官能化する方法であって、
ナノ粒子と第1の二官能性前駆体とを接触させるステップであって、前記第1の二官能性前駆体がシラン部分およびジエン部分を含み、前記ナノ粒子が、シランに反応性のある表面(例えば、シリカ表面)を含み、および前記接触させるステップが、前記シラン部分と前記ナノ粒子表面との間の反応に適した条件下にある、ステップ、ならびに
前記シラン部分と前記ナノ粒子の表面との間に共有結合を形成し、それによってジエン部分で官能化されたナノ粒子を形成するステップ
を含む、方法。 - ジエン部分で官能化された前記ナノ粒子と、第2の二官能性前駆体とを接触させるステップであって、前記第2の二官能性前駆体が、アルキン部分と、ジエン部分に反応性のある基(例えばジエノフィル、例えばマレイミド)とを含み、前記接触させるステップが、ジエン部分に反応性のある前記基と前記ジエン部分との間の反応に適した条件下にある、ステップ、および
前記ナノ粒子の前記ジエン部分と前記第2の二官能性前駆体とを反応させ、それによってアルキン部分で官能化されたナノ粒子を形成するステップ

nは、0から12の整数であり、および
mは、0から5の整数である、

Yは、2価のリンカー(例えば、置換または非置換ヘテロアルキレン基)であり、
R3およびR4はそれぞれ独立して、アルキル、アルケニル、アルキニル、ヘテロアルキル、シクロアルキル、ヘテロシクリル、アリール、ヘテロアリール、ハロ、-ORA、-NRBRC、-NO2、-CNであり、ここで各アルキル、アルケニル、アルキニル、ヘテロアルキル、シクロアルキル、ヘテロシクリル、アリール、およびヘテロアリールは、置換されても置換されなくてもよく、

請求項8に記載の方法。 - 前記2価のリンカーが、アルキレンまたはヘテロアルキレン基であり、前記アルキレンまたはヘテロアルキレン基は、置換されていないかまたは置換されている(例えば、1個または複数のオキソ基で置換されたアルキレンまたはヘテロアルキレン)、請求項8または9に記載の方法。
- アルキン反応性基を含む前記化合物が、ペイロード部分(例えば、細胞毒性薬物、例えばエキサテカン)または標的化リガンド(例えば、葉酸受容体(FR)標的化リガンド、例えば葉酸)を含む、請求項3から13のいずれか一項に記載の方法。
- 前記ナノ粒子が、シリカナノ粒子(例えば、コア-シェルシリカナノ粒子)である、前記請求項のいずれか一項に記載の方法。
- 前記ナノ粒子(例えば、前記ナノ粒子の表面)が、有機ポリマー(例えば、ポリエチレングリコール)でコーティングされる、前記請求項のいずれか一項に記載の方法。
- 前記第1の二官能性前駆体の前記シラン部分と、前記ナノ粒子の表面との間に前記共有結合を形成するステップが、前記第1の二官能性前駆体を、前記有機ポリマーの有機ポリマー分子間(例えば、PEG分子間)の間隙空間内に挿入することを含む、請求項20に記載の方法。
- 複数のジエン部分で官能化されたナノ粒子が形成される、前記請求項のいずれか一項に記載の方法。
- 複数のアルキン部分で官能化されたナノ粒子が形成される、請求項2に記載の方法。
- (a)前記複数のアルキン部分の第1の部分と、アルキン反応性基を含む第1の化合物とを反応させるステップ、および
(b)前記複数のアルキン部分の第2の部分と、アルキン反応性基を含む第2の化合物とを反応させ、それによって、前記第1の化合物および前記第2の化合物で官能化されたナノ粒子を形成するステップ
をさらに含み、
前記第1の化合物および前記第2の化合物が化学的に異なる、

(i)シリカナノ粒子を第1の二官能性前駆体と接触させるステップであって、前記第1の二官能性前駆体が、シラン部分およびシクロペンタジエン部分を含み、前記シリカナノ粒子が、前記シラン部分に反応性のある表面(例えば、シリカ表面)を含み、および前記接触させるステップが、前記シラン部分と前記シリカナノ粒子表面との間の反応に適した条件下にあり、それによって、前記シラン部分と前記ナノ粒子の表面との間に共有結合を形成し、シクロペンタジエン部分で官能化されたナノ粒子を提供する、ステップ、
(ii)シクロペンタジエン部分で官能化された前記ナノ粒子と、第2の二官能性前駆体とを接触させるステップであって、前記第2の二官能性前駆体が、アルキン部分(例えば、DBCO)およびジエノフィル(例えば、マレイミド)を含み、前記接触させるステップが、前記ジエノフィルと前記シクロペンタジエン部分との間の反応に適した条件下にあり、それによって、前記ナノ粒子のジエン部分と前記第2の二官能性前駆体とを反応させ、アルキン部分で官能化されたナノ粒子を提供する、ステップ、ならびに
(iii)前記アルキン部分と、アジド部分を含む化合物とを、前記アルキン部分とアジド部分との間の反応に適した条件(例えば、クリックケミストリー条件)下で接触させ、それによって、前記ナノ粒子の前記アルキン部分と前記アジド部分とを反応させ、前記化合物(例えば、ペイロードまたは標的化リガンドを含む化合物)で官能化されたナノ粒子を提供するステップ

を含む、方法。
