本発明はまた、本明細書に開示するものなどの医学的障害の防止または療法的治療のた
めの、IL-23抗原結合タンパク質の使用にも関する。IL-23抗原結合タンパク質は、根底にある疾患または障害に寄与する際に、IL-23が関与するかまたは役割を果たすか、あるいは別の方式で負の症状に寄与する、多様な状態を治療するのに有用である。

またはより好ましくは、5~12mg/m2の範囲である。小動物、例えばイヌまたはネコに関しては、適切な用量は0.4mg/kgである。IL-23抗原結合タンパク質(好ましくはレシピエント種に由来する遺伝子から構築される)を、注射または他の適切な経路によって、週あたり1回またはそれより多く、動物の状態が改善されるまで投与するか、あるいは無期限に投与してもよい。
ヒトIL-23抗体の生成
XenoMouseTM技術(Amgen、カリフォルニア州サウザンドオークス)を用いて、天然ヒトIL-23活性を認識し、そして阻害する一方、ヒトIL-12を残す、ヒトモノクローナル抗体を開発した。抗体はまた、組換えカニクイザル(cynomologous)IL-23も認識し、そして阻害するが、ネズミまたはラットIL-23を認識しない。
機能アッセイ
a)STAT-ルシフェラーゼ・アッセイ
IL-23は、ヘテロ二量体受容体に結合し、そしてJAK2およびTyk2を通じてシグナル伝達して、STAT1、3、4および5を活性化することが知られる。このアッセイにおいて、STAT/ルシフェラーゼ・レポーター遺伝子でトランスフェクションした細胞を用いて、IL-23抗体がIL-23誘導性生体活性を阻害する能力を評価する。
IL-23がナチュラルキラー細胞に作用して、炎症促進性サイトカイン、例えばインターフェロンγ(IFNγ)の発現を誘導することが知られる。このアッセイにおいて、ヒト初代ナチュラルキラー(NK)細胞を用いて、ヒトIL-23の天然受容体を発現している細胞において、IL-23抗体がIL-23誘導性IFNγ活性を阻害する能力を評価する。
-23またはcyno IL-23(10ng/ml)および組換えヒトIL-18(20ng/ml、R&D Systems、ミネソタ州ミネアポリス)で24時間刺激する。製造者の指示にしたがって、IFNγ ELISA(R&D Systems、ミネソタ州ミネアポリス)によって、上清中のIFNγレベルを測定する。
表6でわかるように、すべての抗体は、用量依存方式で、NK細胞におけるrhuIL-23およびcyno IL-23誘導性IFNγ発現を強力に阻害した。抗体はすべて、ピコモル範囲のIC50値を有した。天然ヒトIL-23(30μg/ml、調製法を実施例1に記載する)およびrhuIL-18(40ng/ml、R&D Systems)を用いて、抗体サブセットに対してアッセイを行い、そして表6に示す結果を得た。IL-23特異的抗体に関する選択と一致して、これらの抗IL-23抗体は、上記アッセイを用いたNK細胞におけるIL-12が刺激するIFNγ産生に影響を及ぼさない一方、IL-12p35特異的中和抗体、mAb219(R&D Systems、ミネソタ州ミネアポリス)は、組換えヒトIL-12を強力に阻害した。
抗凝固剤として、Refludan(登録商標)(Bayer、ペンシルバニア州ピッツバーグ)を用いて、多数の健康なドナーからヒト全血を収集する。全血中のRefludan(登録商標)の最終濃度は10μg/mlである。RPMI1640+10%FBS中のrhuIL-23またはcyno IL-23(最終濃度1ng/ml)+rhuIL-18(最終濃度20ng/ml)+rhuIL-2(最終濃度5ng/ml)の刺激混合物を96ウェルプレートに最終体積20μl/ml添加する。連続希釈IL-23抗体(3μg/mlで始めて1:3連続希釈の11ポイント)を20μl/ウェルで添加して、そして室温で30分間、刺激混合物とインキュベーションする。次いで、全血を添加し(120μl/ウェル)、そしてRPMI1640+10%FBSで最終濃度を200μl/ウェルに調整する。全血の最終濃度は60%である。プレートを37℃、5%CO2で24時間インキュベーションする。細胞不含上清を採取し、そして製造者の指示にしたがって、IFNγ ELISA(R&D Systems)によって、上清由来のIFNγレベルを測定する。
表7でわかるように、すべての抗体は、全血球中のrhuIL-23誘導性およびcyno-IL-23誘導性IFNγ発現を用量依存方式で強力に阻害した。抗体はすべて、ピコモル範囲のIC50値を有した。
IL-23は、炎症促進性サイトカインの強力な誘導因子であることが知られる。IL-23は、活性化およびメモリーT細胞に作用し、そしてIL-22を含む炎症促進性サイトカインを産生するTh17細胞の生存および拡大を促進する。このアッセイにおいて、ヒト全血を用いて、IL-23抗体がIL-23誘導性IL-22産生を阻害する能力を評価する。
o IL-23誘導性IL-22産生を、用量依存方式で、強力に阻害した。抗体はすべて、ピコモル範囲のIC50値を有した。
KinExA技術を用いた、抗IL-23抗体に関する平衡解離定数(K D )の決定
動力学的排除アッセイ(KinExAアッセイ、Sapidyne Instruments, Inc.、アイダホ州ボイズ)を用いて、IL-23抗体に対するrhuIL-23の結合アフィニティを評価する。正常ヒト血清(NHS)活性化Sepharose 4迅速流動ビーズ(Amersham Biosciences、GE Healthcareの一部、スウェーデン・ウプサラ)をrhuIL-23であらかじめコーティングし、そして10mg/mL BSAを含む1M Tris緩衝液でブロッキングする。50pMのIL-23抗体を、rhuIL-23(800pMで始めて1:2希釈の12ポイント)と室温で72時間インキュベーションした後、rhuIL-23でコーティングしたSepharoseビーズに通す。蛍光(Cy5)標識ヤギ抗ヒトFc抗体(Jackson ImmunoResearch、ペンシルバニア州ウェストグローブ)によって、ビーズに結合した抗体の量を定量化した。結合シグナルは、平衡時の未結合抗体の量に比例する。
X線結晶学を用いた構造決定
抗体-抗原複合体の構造を決定する1つの方法は、X線結晶学を用いることにより、例
えば、HarlowおよびLane Antibodies: A Laboratory Manual Cold Spring Harbor Laboratory Press, ニューヨーク州コールドスプリングハーバー(1990), p.23を参照されたい。IL-23の結晶構造は決定されてきており(LupardusおよびGarcia, J Mol Biol, 2008, 382:931-941を参照されたい)、そしてIL-23/Fab複合体の結晶構造が開示されてきている(Beyerら J Mol Biol, 2008. 382(4):942-55を参照されたい)。本明細書に請求する抗体のFab断片を含むIL-23の構造決定は、X線結晶学を用いて得られた。
組換え的に得られたヒトIL-23ヘテロ二量体を結晶化研究に用いた(Beyerら、上記を参照されたい)。ヒトp19サブユニットの配列は、配列番号145の残基20~189、配列番号154のシグナル配列および配列番号155のC末端6-Hisタグで構成される。配列番号147の222位でのグリコシル化を防止するため、ヒトp40サブユニットの配列をこの位でアスパラギンからグルタミンに突然変異させた(Beyerら、上記)。
複合体形成および結晶化
2Xモル過剰の抗体B Fabと上述のヒト・ヘテロ二量体IL-23を混合することによって、IL-23-抗体B Fab複合体を作製した。サイズ排除クロマトグラフィーによって複合体を精製して、過剰な抗体B Fabを除去し、そして結晶化のため、~12mg/mlに濃縮した。IL-23-抗体B Fab複合体は、0.1M Hepes pH7、8%PEG8000中で結晶化した。
IL-23-抗体B Fab結晶は、P21空間群、単位格子寸法(unit cell dimensions)a=70.93、b=71.27、c=107.37Å、β=104.98°で成長し、そして2.0Å分解能で回折する。出発検索モデルとしてIL-23構造(Beyerら、上記)を用いて、プログラムMOLREP(CCP4、The CCP4 suite: programs for protein crystallography. Acta Crystallogr D Biol Crystallogr, 1994. 50(Pt 5): p.760-3)での分子置換によって、IL-23-抗体B Fab構造を解析した。IL-23溶液を固定して維持しつつ、抗体可変ドメインを検索モデルとして用いた。IL-23抗体可変ドメイン溶液を固定して維持しつつ、抗体定常ドメインを検索モデルとして用いた。Quantaを用いた多数周期のモデル構築およびcnxでの精密化を用いて、完全構造を改善した(Brungerら, Acta Crystallogr D Biol Crystallogr, 1998, 54(Pt 5):p. 905-21)。
IL-23ヘテロ二量体から4Åまたはそれ未満の原子を含む抗体B Fab重鎖アミノ酸残基には、配列番号46のGly32、Gly33、Tyr34、Tyr35、His54、Asn58、Thr59、Tyr60、Lys66、Arg101、Gly102、Phe103、Tyr104およびTyr105が含まれる。IL-23ヘテロ二量体から≦5Åの原子を含む抗体B Fab重鎖アミノ酸残基には、配列番号46のSer31、Gly32、Gly33、Tyr34、Tyr35、His54、Ser56、Asn58、Thr59、Tyr60、Lys66、Arg101、Gly102、Phe103、Tyr104およびTyr105が含まれる。

溶媒アクセス可能表面積相違を通じた、IL-23-抗体複合体接触残基の決定
溶媒アクセス可能表面積相違を用いて、パラトープ(抗原を認識する抗体部分)における残基接触、ならびにヒトIL-23-抗体B Fab複合体において、そしてヒトIL-23-抗体E Fab複合体において、パラトープが結合する抗原の、パラトープによって結合される部分を決定した。Molecular Operating Environment(Chemical Computing Group、ケベック州モントリオール)を用いて、溶媒アクセス可能表面積計算を行った。
次いで、「結合面積」を「未結合面積」から減じ、セット中の各残基に関する「溶媒曝露表面積相違」を生じた。表面積が変化しない、またはゼロ相違である抗体B Fab残基は、複合体化した際、IL-23抗原の残基と接触しなかった。≧10Å2の相違値を有する抗体B Fab残基は、抗体B FabがヒトIL-23に結合した際に、これらの抗体B Fab残基が少なくとも部分的から完全に塞がれるように、IL-23抗原中の残基との有意な接触にあると見なされた。抗体B Fab残基のこのセットは、抗体B
FabがヒトIL-23に結合した際に界面の構造に関与する残基である「カバードパッチ」を構成する。表10および11を参照されたい。このカバードパッチ中の抗体B Fab残基は、IL-23抗原の残基との結合相互作用に関与していない可能性もあるが、カバードパッチ内の任意の単一残基の突然変異は、ヒトIL-23への抗体B Fabの結合に影響を及ぼすであろうエネルギー相違を導入しうる。Tyr49を例外として、すべての残基は、抗体B Fab軽鎖および重鎖のCDR領域中に位置する。これらの残基はまた、実施例4に記載するように、抗体B Fabに結合した際、IL-23抗原の5Åまたはそれ未満の範囲内であった。
Fab残基のこのセットは、抗体E FabがヒトIL-23に結合した際に、界面構造に関与する残基であるカバードパッチを構成する。表12および13を参照されたい。このカバードパッチ中の抗体E Fab残基は、IL-23抗原残基との結合相互作用に関与していない可能性もあるが、カバードパッチ内の単一残基いずれかの突然変異は、ヒトIL-23への抗体E Fabの結合に影響を及ぼすエネルギー相違を導入しうる。大部分に関して、これらのカバードパッチ残基は、抗体E Fab重鎖および軽鎖のCDR領域内に位置した。これらの残基はまた、実施例4に記載するように、抗体E Fabに結合した際、IL-23抗原の5Åまたはそれ未満内であった。
Fabのパラトープが結合するIL-23ヘテロ二量体部分の溶媒アクセス可能表面積相違を計算した。抗体B Fab-IL-23複合体に関して実施例4で得た構造情報を用い、そして抗体B Fabの存在下で、IL-23ヘテロ二量体のアミノ酸残基の残基溶媒アクセス可能表面積を計算し、そしてこれは該セットに関する結合面積に相当する。

- 請求項54記載の単離抗原結合タンパク質であって、前記抗原結合タンパク質がヒトIL-23に結合した際に形成されるカバードパッチが、配列番号147に記載するようなヒトIL-23p40サブユニットの残基121~125内の残基接触をさらに含み、前記残基接触が、溶媒曝露表面積によって決定されるように10Å2より大きいかまたは10Å2に等しい差分値を有する、前記単離抗原結合タンパク質。
- ヒトIL-23に結合する単離抗原結合タンパク質であって、前記抗原結合タンパク質がヒトIL-23に結合した際、X線結晶学によって決定されるように、前記抗原結合タンパク質が、配列番号145に記載するようなヒトIL-23p19サブユニットの残基46~58内の残基、残基112~123内の残基、および残基155~163内の残基から5Åまたはそれ未満である、前記単離抗原結合タンパク質。
- 請求項56記載の単離抗原結合タンパク質であって、前記抗原結合タンパク質がヒトIL-23に結合した際、X線結晶学によって決定されるように、前記抗原結合タンパク質が、配列番号147に記載するようなヒトIL-23p40サブユニットの残基121~125内の残基から5Åまたはそれ未満である、前記単離抗原結合タンパク質。
- a)ヒトIL-23活性を減少させる特性;
b)炎症促進性サイトカインの産生を減少させる特性;
c)≦5x10-8MのKDでヒトIL-23に結合する特性;
d)≦5x10-6 1/sのKoff速度を有する特性;および
e)≦400pMのIC50を有する特性;
からなる群より選択される少なくとも1つの特性を有する、請求項1、4、7、10、19、20、26~31、33、35、37、54または56の単離抗原結合タンパク質。 - 請求項1、4、7、10、19、20または26の少なくとも1つの抗原結合タンパク質および薬学的に許容されうる賦形剤を含む、薬学的組成物。
- 標識基またはエフェクター基をさらに含む、請求項59の薬学的組成物。
- 前記標識基が、同位体標識、磁気標識、レドックス活性部分、光学色素、ビオチン化基および二次レポーターによって認識されることがあらかじめ決定されたポリペプチド・エピトープからなる群より選択される、請求項60の薬学的組成物。
- 前記エフェクター基が、放射性同位体、放射性核種、毒素、療法基および化学療法基からなる群より選択される、請求項60の薬学的組成物。
- 前記抗原結合タンパク質が、標識基にカップリングしている、請求項60の単離抗原結合タンパク質。
- 患者において、IL-23に関連する状態を治療するかまたは防止するための方法であって、こうした方法を必要とする患者に、請求項1、4、7、10、19、20または26の少なくとも1つの単離抗原結合タンパク質の有効量を投与する工程を含む、前記方法。
- 状態が、炎症性障害、リウマチ性障害、自己免疫障害、腫瘍学的障害および胃腸障害からなる群より選択される、請求項64の方法。
- 状態が、多発性硬化症、関節リウマチ、癌、乾癬、炎症性腸疾患、クローン病、潰瘍性大腸炎、全身性エリテマトーデス、乾癬性関節炎、自己免疫心筋炎;1型糖尿病および強直性脊椎炎からなる群より選択される、請求項65の方法。
- 単離抗原結合タンパク質が、単独で、または併用療法として投与される、請求項64の方法。
- 請求項1、4、7、10、19、20または26の少なくとも1つの抗原結合タンパク質の有効量を投与する工程を含む、患者においてIL-23活性を減少させる方法。
- 前記IL-23活性が炎症促進性サイトカインの産生を誘導することである、請求項68のIL-23活性を減少させる方法。
