c) Ensayo de sangre entera humana
La sangre entera humana se recoge de múltiples donantes sanos utilizando Refludan® (Bayer Pittsburgh, PA) como anticoagulante. La concentración final de Refludan® en sangre entera es de 10 μg/mL. Una mezcla de estimulación 5 de rhuIL-23 o cyno IL-23 (concentración final 1 ng/mL) + rhuIL-18 (concentración final 20 ng/mL) + rhuIL-2 (concentración final 5 ng/mL) en RPMI 1640 + 10% FBS, a una placa de 96 pozos, volumen final 20 µL/pozo. Se adicionan anticuerpos de IL-23 diluidos en serie (11 puntos de diluciones en serie 1:3 a partir de 3 μg/mL) a 20 μl/pozos y se incuba con la mezcla de estimulación durante 30 minutos a temperatura ambiente. A continuación, se adiciona sangre entera (120 μL/pozo) y el volumen final se ajusta a 200 μL/pozo con RPMI 1640 + 10% de FBS. La
10 concentración final de sangre total es del 60%. Las placas se incuban durante 24 horas a 37ºC, CO2 al 5%. Se recogen los sobrenadantes libres de células y se miden los niveles de IFNγ a partir de los sobrenadantes mediante IFNγ ELISA (R&D Systems) de acuerdo con las instrucciones del fabricante.
El análisis estadístico se puede realizar utilizando el software GraphPad PRISM. Los resultados se pueden expresar como la desviación estándar ± media (SD).

d) Ensayo de IL-22
Se sabe que la IL-23 es un potente inductor de citocinas proinflamatorias. IL-23 actúa sobre células T activadas y de memoria y promueve la supervivencia y expansión de células Th17 que producen citoquinas proinflamatorias 5 incluyendo IL-22. En este ensayo, se utiliza sangre entera humana para evaluar la capacidad de los anticuerpos de IL-23 para inhibir la producción de IL-22 inducida por IL-23.
Se lleva a cabo un ensayo de sangre entera de la misma manera que se ha descrito anteriormente con la modificación de la utilización de rhuIL-23 o cynoIL-23 a 1 ng/mL y rhuIL-18 a 10 ng/mL para inducir la producción de IL-22. La concentración de IL-22 se determina mediante IL-22 ELISA (R&D Systems, Minneapolis, MN).
10 Como se ve en la Tabla 8, los anticuerpos inhibieron potentemente la producción de IL-22 inducida por rhuIL-23 e inducida por cyno IL-23 en células de sangre entera de una manera dependiente de la dosis. Los anticuerpos tenían valores de IC50 en el intervalo picomolar.

Determinación de la constante de disociación de equilibrio (KD) para los anticuerpos anti-IL-23 utilizando la tecnología KinExA
La afinidad de unión de rhuIL-23 a anticuerpos de IL-23 se evalúa utilizando un ensayo de exclusión cinética (ensayo
20 KinExA, Sapidyne Instruments, Inc., Boise, ID). Se prerecubrieron perlas de flujo rápido de Sepharose 4 activadas con suero humano normal (NHS) (Amersham Biosciences, parte de GE Healthcare, Uppsala, Suecia) con rhuIL-23 y se bloquearon con solución reguladora Tris 1 m con 10 mg/mL de BSA. 50 pM del anticuerpo de IL-23 se incuba con rhuIL-23 (12 puntos de diluciones 1:2 a partir de 800 pM) a temperatura ambiente, durante 72 horas antes de que se ejecute a través de las perlas de Sepharose revestidas con rhuIL-23. La cantidad del anticuerpo unido a perlas se
25 cuantificó mediante anticuerpo anti-Fc-humano de cabra marcado con fluorescencia (Cy5) (Jackson Immuno Research, West Grove, Pa.). La señal de unión es proporcional a la cantidad del anticuerpo libre en equilibrio.
La constante de equilibrio de disociación (KD) y la velocidad de asociación (Kon) se obtienen a partir de la adaptación de curvas utilizando el software KinExA Pro. La velocidad de disociación (Koff) se deriva de: KD = Koff/Kon
Como se observa en la Tabla 9, los anticuerpos tienen alta afinidad por la unión a IL-23 humana. Todos tenían 30 valores de KD en el intervalo bajo a sub pM.

Los contactos del residuo en el paratopo (la porción del anticuerpo que reconoce el antígeno) y la porción del antígeno que se une enlazada por el paratopo en un complejo IL-23-Fab del anticuerpo B humano y en un complejo IL-23-Fab del anticuerpo E se determinó utilizando diferencias de área de superficie accesibles con solvente. Las "áreas unidas" se restaron después de las "áreas libres" resultando en la "diferencia de superficie expuesta al solvente" para cada residuo en el conjunto. Los residuos Fab del anticuerpo B que no tuvieron ningún cambio en el 15 área superficial, o una diferencia cero, no tuvieron contacto con los residuos del antígeno IL-23 cuando estaban complejados. Los residuos Fab del anticuerpo B que tenían un valor de diferencia ≥ 10 Å2 se consideraron en contacto significativo con residuos en el antígeno de IL-23 de tal manera que estos residuos de Fab del anticuerpo B estaban al menos parcialmente ocluidos completamente cuando el Fab del anticuerpo B estaba unido a IL-23 humana. Este conjunto de residuos Fab del anticuerpo B forman el "parche cubierto", los residuos implicados en la 20 estructura de la interfaz cuando el Fab del anticuerpo B está unido a IL-23 humana, véanse las Tablas 10 y 11.
