Claims (6)
- コリネバクテリウム・グルタミクムに、(i)グリセロールファシリテーター(glycerol facilitator)をコードする遺伝子、(ii)グリセロールキナーゼをコードする遺伝子、グリセロールデヒドロゲナーゼをコードする遺伝子、(iii)グリセロールデヒドラターゼ(glycerol dehydratase)をコードする遺伝子、(iv)グリセロールレアクティバーゼ(glycerol reactivase)をコードする遺伝子、及び(v)1,3−PDOオキシドレダクターゼをコードする遺伝子が導入されており、そしてコリネバクテリウム・グルタミクムからアルデヒドデヒドロゲナーゼをコードする遺伝子が欠失又は弱化しており、3−HP生産能が欠失又は弱化し、そしてグリセロールから1,3−PDOを生成する変異微生物。
- 前記アルデヒドデヒドロゲナーゼをコードする遺伝子は、NCgl0049、NCgl0157、Ncgl0437、NCgl0463、NCgl0521、NCgl0523、NCgl0900、NCgl2272、NCgl2578、NCgl2619及びNCgl2698からなる群から選ばれることを特徴とする、請求項1に記載の変異微生物。
- 前記グリセロールファシリテーター(glycerol facilitator)をコードする遺伝子、グリセロールキナーゼをコードする遺伝子、及びグリセロールデヒドロゲナーゼをコードする遺伝子はそれぞれ、glpF、glpK、及びglpDであることを特徴とする、請求項1に記載の変異微生物。
- 前記グリセロールデヒドラターゼをコードする遺伝子及びグリセロールレアクティバーゼ(glycerol reactivase)をコードする遺伝子は、pduCDEGHクラスターであり、1,3−PDOオキシドレダクターゼをコードする遺伝子は、yqhDであることを特徴とする、請求項1に記載の変異微生物。
- 前記導入される遺伝子は、tac,trc及びtufからなる群から選ばれる強いプロモーターによって過発現することを特徴とする、請求項1に記載の変異微生物。
- 次の段階を含むグリセロールから1,3−PDOを製造する方法:
(a)請求項1〜5のいずれか一項に記載の変異微生物をグリセロール含有培地で培養して1,3−PDOを生成させる段階;及び
(b)前記生成された1,3−PDOを得る段階。
