そこで本発明は、疎水性表面を有する基材でさえ親水性コーティング可能なグリセロールデンドロン共重合体、当該グリセロールデンドロン共重合体を用いたコーティング方法、および当該グリセロールデンドロン共重合体を合成するためのグリセロールデンドロン単量体を提供することを目的とする。

ADGMAの濃度を0.1M、BMAの濃度を0.9Mに変更し、沈殿を生じさせるための貧溶媒をメタノールからヘキサンに変更した以外は実施例2と同様にして、p(ADGMA−ran−BMA)を合成し、更に脱保護することによりp(DGMA−ran−BMA)を得た。1H NMRにより、得られた化合物を同定した。

(1)共重合反応
GPC装置のポンプとしては日本分光社製の「PU−980」を、脱気装置としては日本分光社製の「DG−980−50」を、カラムとしては昭和電工社製の「Asahipak GF−510HQ(7.5mm I.D.×300mm)」を、カラムオーブンとしては日本分光社製の「CO−965」を、RI検出器としては昭和電工社製の「RI−101」を用いた。溶離液としては、PBMA、PEGMA5−BMA95、およびPEGMA10−BMA90用にはアセトン(ナカライテスク社製,GR特級)を用い、その他の共重合体用にはメタノール(ナカライテスク社製,GR特級)を用いた。流速は0.6mL/minに、温度は35℃に設定した。標準試料としては、ポリエチレンオキシド(「TSK標準ポリエチレンオキシド」東ソー社製)を用いた。結果を表6に示す。
また、得られた共重合体中の親水性モノマー成分のモル分率を1H NMRスペクトルより算出し、使用した親水性モノマー成分のモル分率と比較した結果を表6に示す。
また、GPCにより各共重合体とPBMAの分子量を算出したところ、いずれの重合体も105オーダーの分子量であることから、コーティングを行うのに十分な分子量を有するものあることが確認された。
マイクロチューブ内に数mgの各共重合体を取り、各溶媒を1mL加えて超音波処理に付し、且つボルテックスミキサーを用いて攪拌し、目視で状態を確認し、以下の基準で溶解性を評価した。結果を表7に示す。
〇: 不溶物がなくなった
×: 不溶物がなくならなかった
親水性モノマーとしてPEGMAを用いた場合は、メタノールに対する高い溶解性を得るのに10%より多い添加量を要したのに対して、DGMAでは5%の添加量でメタノールに対して高い溶解性を示したことから、DGMAが少量で高い極性を付与する性質を持つことが示された。
アセトン及びクロロホルムに対しても、DGMAが5%および10%の共重合体(DGMA5−BMA95とDGMA10−BMA90)のいずれもが高い溶解性を示したことから、幅広い極性の溶媒に溶解することが示唆された。
(1)コーティング
ペレット状のポリウレタン(「Tecoflex(R)」Lubrizol社製)を70℃で3時間減圧することで予備乾燥し、10gをAl板で挟み、ゴンノ油圧機製作所社製の「メルトプレス」を用い、170℃、0MPaにて3分間加熱し、4MPaにて1分間加圧した後、室温に冷却することでシート状に加工した。このポリウレタンシートを1cm×1cmに切断し、エタノール中で30秒間超音波処理をしてからコーティングに用いた。
表7に示した各重合体のうち、PBMA、PEGMA5−BMA95、およびPEGMA10−BMA90はアセトンに、その他の共重合体はエタノールに溶解して0.5質量%溶液とした。この共重合体溶液に上記ポリウレタンシートを3秒間浸漬し、1秒で引き上げる操作を2回繰り返した後、40℃で2時間真空乾燥することでディップコーティングを行った。また、ポリウレタンシートをエタノールに対して同様の浸漬、引き上げ、真空乾燥を行い、対照試料(未コーティングポリウレタンシート)とした。
各ポリウレタンシートをATR−IR測定により分析したところ、未コーティングのポリウレタンシートでは1684cm-1付近にウレタン結合由来のC=O伸縮のみが観察されるのに対して、共重合体によるコーティングを施したポリウレタンシートでは1725cm-1付近に共重合体のメタクリル酸エステル部位由来のC=O伸縮のショルダーピークが観察されたことと、未コーティングでは1095cm-1付近にウレタン結合由来のC−O伸縮ピークが観察されるのに対して、コーティングを施したものでは1147cm-1付近に共重合体のC−O伸縮が観察されることから、ポリウレタンシート表面に共重合体がコーティングされたことが確認された。
未コーティングポリウレタンシート、並びに、PBMA、DGMA10−BMA90、GMA70−BMA30、およびPEGMA20−BMA80によってコーティングを施したポリウレタンシートに白金を蒸着し、20keVにてSEMによりシート表面を観察した。結果を図1に示す。
バイオマテリアルにおける表面の粗さは、表面が粗いほど生体との接触面積が大きくなり、タンパク質や細胞の吸着、それに続く生体反応が起こる場が増大してしまうという点で重要な性質である。共重合体によるコーティングを施したポリウレタンシート表面をSEMにより観察したところ、図1の通り、いずれの共重合体によるコーティングも、未コーティングのポリウレタンシートと比較して平滑になる傾向が見られた。
従って、本発明に係る共重合体のコーティングは、表面の粗さという点で基板となるバイオマテリアルの機能を損なうことはないことが示唆された。
未コーティングポリウレタンシートおよびコーティングポリウレタンシート上に100μLの純水を乗せ、そのうち96μLを吸い取ることで4μLを残し、水滴とポリウレタンシートの接触面の水平方向から写真を撮影した。この水滴の接触角をθ/2法によって算出した。同様の測定を、ポリウレタンシート上の3点で行った。結果を図2に示す。
図2に示される結果の通り、いずれの共重合体によるコーティングもPBMAのみによるコーティングより小さな接触角を示し、親水性モノマーの組成比が増加するに従って接触角が減少した。更にDGMAを10mol%、GMAを50mol%以上、およびPEGMAを20mol%含む共重合体コーティングシートでは、未コーティングシート(bare)よりも有意に小さな接触角であったことから、DGMA、GMAおよびPEGMAが表面に親水性を付与する性質を持つことが示された。特にDGMAは10mol%の組成でGMAが30mol%の共重合体と、PEGMAが20mol%の共重合体よりも接触角が有意に小さかったことから、デンドリティックグリセロールが少量で高い表面親水性を付与することが示された。
図3のグラフにおいて、DGMAのプロットがGMAとPEGMAに対してグラフの左下に存在することから、モノマーの質量分率で比較してもデンドリティックグリセロールが少量で高い表面親水性を付与することが示された。デンドリティックグリセロールとポリエチレングリコールは同様のポリエーテル構造を有するが、デンドリティックグリセロールは枝分かれ構造によって同じ分子量でも末端に多数の水酸基を持つ。従って高い水酸基密度を達成できるので、直鎖のポリエチレングリコールと比較して質量分率に対する親水性が高いと考えられる。
次に、水酸基数について考察した。1分子中の水酸基数を比較するとDGMAは8つ、GMAは2つであり両者に差がある。そこで水酸基数を考慮して比較するために、共重合体の単位重量あたりの水酸基数[mmol/g]に対して接触角をプロットしたグラフを図4に示す。DGMAのプロットがGMAに対してグラフの左下に存在することから、同量の水酸基数でもDGMAが高い表面親水性を付与することが明らかになった。
この理由としては、DGMAのデンドリティックグリセロール構造に存在するエーテル構造の極性も表面の親水性に寄与しているということと、デンドリティックグリセロール構造がGMAと比較して主鎖から長距離まで広がっており柔軟な構造を持つことで、よりコーティング表面に露出しやすいということが考えられる。
未コーティングポリウレタンシートおよびコーティングポリウレタンシートをPBS(12mM,pH7.4)中に1週間浸漬した後、1mg/mLのフィブリノーゲン(Fib)を含むPBS溶液1mL中に37℃で2時間浸漬した。各ポリウレタンシートを取り出し、両面をそれぞれ1mLのPBSで洗浄し、次いで1質量%ドデシル硫酸ナトリウム水溶液0.2mLに浸漬して10分間超音波処理した。この溶液から0.15mLずつをビオラモ96ウェルプレート(アズワン社製)に取り、Thermo Scientific Micro BCATM Protein Assay Kitの蛍光試薬0.15mLを加えて30秒間振とうし、37℃で2時間静置した後、マイクロプレートリーダーを用いて562nmの吸光を測定した。測定は1種類の重合体と未コーティングに対して3回行った。Fibの2.5,5.0,10,20,40,および200μg/mL PBS溶液を用いた較正曲線によって、各ポリウレタンシートに吸着したフィブリノーゲン量を定量した。結果を図5に示す。
図5に示される結果の通り、Fibの吸着量について、PBMAによるコーティングが未コーティングと有意差が見られないのに対し、各親水性モノマー共重合体によるコーティングではいずれも吸着量が有意に減少しており、親水性モノマーの組成比が増加するに従って吸着量が減少していることから、親水性モノマーがFib吸着抑制効果を持つことが示された。特にDGMAが10mol%の共重合体が、GMAが30mol%の共重合体、およびPEGMAが10mol%の共重合体よりも有意に低い吸着量を示した。タンパク質吸着の高い抑制効果が報告されているポリエチレングリコールよりも低い吸着量であったことから、デンドリティックグリセロールが少量で高いFib吸着抑制効果を発現することが明らかになった。
図6に示される結果の通り、PEGMA共重合体はGMA共重合体に比べてグラフの右下にプロットがあり、表面の親水性がそれほど高くない割りにFib吸着が抑制されているといえる。GMAは低分子であるため排除体積効果を無視でき、親水性がFib吸着抑制の主な要素だと仮定すると、GMA共重合体とPEGMA共重合体のこの差異がPEGMAの排除体積効果の因子だと考えることができる。この前提でDGMA共重合体に着目すると、DGMAを5mol%含む共重合体(接触角:47°)はGMAとPEGMAの間のPEGMAに近いところに存在することから、デンドリティックグリセロールが接触角測定で観察された高い親水性だけでなく、ポリエチレングリコールと同様の排除体積効果によってもタンパク吸着を抑制している可能性が示唆された。
一方で、DGMAを10mol%含む共重合体(接触角:15°)はGMAの各共重合体と同じ領域に存在し、排除体積効果によると考えられる差異は観察されなかった。この理由としては、DGMAの組成が多くなるとデンドリティックグリセロール同士の距離が近くなり互いの立体反発で運動性が低下したことが考えられる。
表6に示す各重合体を溶媒に溶解して0.5質量%溶液とした。溶媒として、PBMA、PEGMA5−BMA95、およびPEGMA10−BMA90にはアセトン、その他の共重合体にはエタノールを用いた。得られた共重合体溶液に、2cm×2cmに切断したAl板を3秒間浸漬し1秒で引き上げる操作を2回繰り返した後、40℃で2時間真空乾燥することでディップコーティングを行った。重合体がAl板表面にコーティングされたことをRAS−IR測定により確認した。
いずれの共重合体も1700cm-1付近にC=O伸縮のピークが、1150cm-1付近にC−O伸縮のピークが観察されたことから、Al基板に対してもコーティングを施すことができることが示された。
従って、本発明に係る共重合体が様々な基板に対するコーティング材料として利用できる可能性が示された。
表6に示す各重合体を溶媒に溶解して10質量%溶液とした。PBMA、PEGMA5−BMA95、およびPEGMA10−BMA90にはアセトン、その他の共重合体にはエタノールを用いた。得られた共重合体溶液をポリエチレンフィルム上に流して室温で静置して乾燥し、流動性がなくなった後に40℃で2時間真空乾燥することで各重合体のフィルムを作製した。いずれの重合体からも、無色透明のフィルムが得られた。
実施例4で作製した各共重合体フィルムを数mg切り取り、重量を測定した。次いで、25℃で純水に浸漬し、各時間経過後にフィルムを取り出して表面の水を拭き取った後、重量を測定した。重量の有意な増加が見られなくなった状態を膨潤平衡とした。含水率を以下の式で算出した。各共重合体につき4つ以上のサンプルで含水率を測定した。結果を図7に示す。

各共重合体フィルムの膨潤平衡含水率を測定したところ、図7に示される結果の通り、いずれの共重合体も親水性モノマーの組成比が増加するにしたがって膨潤平衡含水率が増加した。
係る結果より、ポリエーテル構造が水和に大きく寄与していることが示唆された。
純水に浸漬し膨潤平衡に達した各共重合体フィルムを、DSC用アルミパンに入れて密閉した。各フィルムを30℃から−100℃まで5℃/minの速度で冷却し、−100℃で5分間保持した後、30℃まで5℃/minの速度で昇温した。中間水の低温結晶化のピークが全てのフィルムにおいて観察されなかったことから、以下の2式によってWfとWnfを求めた。

[式中、Wtotalは乾燥したフィルム単位重量当たりの全ての水和水の重量を示し、Wfは自由水の重量を示し、Wnfは不凍水の重量を示し、m0は乾燥したフィルムの重量を示し、ΔHmは昇温曲線における吸熱ピークの面積から求めた融解エンタルピーを示し、ΔHwaterは水の融解エンタルピーを示す。]
DSC曲線のピークの面積から水を定量する際には、3回測定を行った平均値を用いた。
膨潤平衡時のフィルム単位重量当たりの全ての水和水と、DSC測定により求めた水和水の状態の内訳を図9のグラフに示す。いずれの共重合体フィルムも中間水が存在すると観察される低温結晶化ピークは観察されず、自由水と不凍水のみで構成されることが分かった。中間水を持つことが報告されているポリ(2−メトキシエチル アクリレート)と類似の構造であり水酸基を有する点が異なるポリ(2−ヒドロキシエチル メタクリレート)や、メトキシエチル基がエチル基である点が異なるポリ(エチル アクリレート)や、主鎖がアクリレートではなくメタクリレートであるポリ(2−メトキシエチル メタクリレート)がいずれも中間水を持たないことから、中間水はメトキシ基、水酸基、エチル基といった親水性の違いによる水和状況の違いや、少しの主鎖の違いによっても大きく左右されるものであることが考えられる。本発明に係る共重合体においては、疎水性の高いブチル基をベースとすることやメタクリレートモノマーを用いていること等が、中間水が観察されない理由として考えられる。
Fib吸着量と全ての水和水量との相関係数が−0.44であるのに対して、Fib吸着量と不凍水量との相関係数が−0.90であることから、本発明に係る共重合体が示したFib吸着抑制効果は、水和水全体ではなく不凍水の量が大きく寄与したものだと考えられる。この実験結果は、表面に水和した水が存在することでタンパク質が表面に吸着するためのエネルギーが大きくなり吸着抑制効果を生み出すという過去の報告と一致している。
従って、DGMA−BMAが同じ又はそれ以上の組成のGMA−BMAやPEGMA−BMAよりも高いFib吸着抑制効果を示したのは、デンドリティックグリセロールが多くの強固に水和した水である不凍水を持ち、Fibが吸着するためのエネルギー障壁を大きくしたことが寄与しているということがこの実験結果から示唆された。
従って、上に述べた水和状態の解析においてPEGMA5−BMA95に水和した水の89.1%というほとんどの水が自由水ではあったが、その自由水として計算された水和水は、他の共重合体の自由水と比べると高分子と強い相互作用をしていると考えられる。しかし、図5に示したようにFib吸着試験の結果では大きな吸着抑制効果は見られず、図10(2)に示したFib吸着量と不凍水との関係において大きく外れた挙動を示していないことから、Fib吸着量との相関性という点ではあくまで不凍水の量が重要であると考えられる。
またDGMA10−BMA90とPEGMA10−BMA90において吸熱ピークが観察されなかった理由としては、ピークがブロード化した可能性が考えられる。高分子鎖と水分子との相互作用が強く、水分子がうまく配列できず結晶化が長時間にわたった場合はピークがブロード化すると考えられ、DGMA10−BMA90とPEGMA10−BMA90に水和した水の中の自由水についても、他の共重合体と比較して高分子と強く相互作用している可能性が示唆された。
更にDGMA10−BMA90については、自由水として計算した昇温曲線上の吸熱ピークの低温側に伸びるブロードなショルダーピークが観測された。このショルダーピークは、中間水が存在する系において−40℃付近で低温結晶化した中間水が低温で融解を始めることで観測されると報告されているピークと類似している。一般的には、−40℃付近で低温結晶化する挙動が見られ、これが融解する挙動がショルダーピークとして現れるが、DGMA10−BMA90については低温結晶化に伴う発熱ピークが明確には見られなかったことから、中間水の存在は認められたものの、その量はDSC測定において測定可能な量ではなかったものと考えられる。
以上の結果から、DGMA10−BMA90に水和した水は、僅かな中間水であり、デンドリティックグリセロールと強く相互作用をしているということが示唆された。
Claims (8)
- X1がカルボニル基であり、且つX2がエステル基である請求項1に記載のグリセロールデンドロン共重合体。
- R2〜R5が上記式(III)で表される基である請求項1または2に記載のグリセロールデンドロン共重合体。
- R8とR9がHである請求項1〜3のいずれかに記載のグリセロールデンドロン共重合体。
- 請求項1〜3のいずれかに記載のグリセロールデンドロン共重合体の溶液を用いて基材をコーティングする工程を含むことを特徴とするコーティング方法。
- 上記溶液に上記基材を浸漬する請求項5に記載のコーティング方法。
- 上記基材の少なくとも表面が疎水性高分子で構成されている請求項5または6に記載のコーティング方法。
