Abstract
Description
T細胞レセプター(TCR)は、CD4+及びCD8+ T細胞により天然に発現される。TCRは、主要組織適合性複合体(MHC)分子(ヒトでは、MHC分子はヒト白血球抗原又はHLAとしても知られる)との複合体で、抗原提示細胞の表面にディスプレイされる短いペプチド抗原を認識するように設計されている(Davisら、Annu Rev Immunol、1998;16:523-44)。CD8+T細胞は、細胞傷害性T細胞とも呼ばれ、MHCクラスI分子に結合するペプチドを特異的に認識するTCRを有する。CD8+ T細胞は、一般には、癌細胞及びウイルス性感染細胞を含む病気の細胞の発見及びその破壊の仲介を担っている。対応する抗原に対する天然レパートリーの癌特異的TCRの親和性は、胸腺選択の結果として一般的に低く、これは癌細胞が頻繁に検出と破壊から逃れることを意味する。T細胞による癌認識を促進することを目的とした新規免疫治療のアプローチは、効果的な抗癌治療の開発のための非常に有望な戦略を提供する。

方法

(1989) New York:Cold spring harbor laboratory pressに記載のような)標準的方法を用いて、別々にpGMT7ベースの発現プラスミドにクローニングした。発現プラスミドを別々に大腸菌株Rosetta(BL21pLysS)、又はT7発現系に形質転換し、単一アンピシリン抵抗性コロニーを、TYP(+アンピシリン100μg/ml)培地中で37℃にて、約0.6〜0.8のOD600まで増殖させた後、0.5mM IPTGでタンパク質発現を誘導した。誘導の3時間後に細胞を遠心分離により採集した。BugBusterタンパク質抽出試薬(Merck Millipore)を製造業者の指示に従って用いて細胞ペレットを溶解させた。封入体ペレットを遠心分離により回収した。ペレットをTriton緩衝液(50mM Tris-HCl pH8.1、0.5%Triton-X100、100mM NaCl、10mM NaEDTA)中で2回洗浄し、最後に界面活性剤フリーの緩衝液(50mM Tris-HCl pH8.1、100mM NaCl、10mM NaEDTA)中に再懸濁した。6Mグアニジン-HClで可溶化し、OD280を測定することによって、封入体タンパク質収量を定量した。次いで、消光係数を用いてタンパク質濃度を算出した。8M尿素で可溶化し、約2μgを還元条件下の4〜20%SDS-PAGEにロードして封入体の純度を測定した。次いで、デンシトメトリーソフトウェア(Chemidoc, Biorad)を用いて純度を推定又は算出した。封入体を、短期保存については+4℃にて、長期保存については−20℃又は−70℃にて保存した。
方法
ImmTACの調製は、TCR β鎖を、抗CD3単鎖抗体にリンカーを介して融合させたこと以外は、実施例1の記載のとおりに行った。加えて、カチオン交換工程を、アニオン交換後の精製の間に行った。この場合、アニオン交換からのピーク画分を20mM MES(pH6.5)中で20倍希釈し、POROS(登録商標) 50HSカチオン交換カラムに適用した。結合タンパク質を、20mM MES中0〜500mM NaClのグラジエントで溶出させた。ピークImmTAC画分をプールし、50mM Tris pH8.1に調整した後、濃縮し、実施例1に記載のとおりにゲル濾過マトリクスに直接適用した。
精製した可溶性TCR及びImmTAC分子の該当のペプチド-HLA複合体に対する結合の分析を、BIAcore 3000若しくはBIAcore T200装置を用いる表面プラズモン共鳴又はForteBio Octet装置を用いる二重層干渉分光法により行った。ビオチン化クラスI HLA-A*02分子を、目的のペプチドと共にリフォールディングし、当業者に公知の方法を用いて精製した(O'Callaghanら(1999),Anal Biochem 266(1):9-15;Garbocziら(1992),Proc Natl Acad Sci USA 89(8):3429-3433)。全ての測定は、0.005% P20を補充したダルベッコのPBS緩衝液中で25℃にて行った。
ビオチン化ペプチド-HLAモノマーをストレプトアビジン共役CM-5センサチップに固定した。平衡結合定数は、約200応答単位(RU)のペプチド-HLA-A*02複合体を被覆したフローセル上に30μl/分の定流速で注入した可溶性TCR/ImmTACの系列希釈物を用いて決定した。平衡応答は、各TCR濃度について、無関係のペプチド-HLAを含有するコントロールフローセルでのバルク緩衝液応答を減算することにより規格化した。KD値は、Prismソフトウェア及びLangmuir結合等温式 結合= C×Max/(C+KD) (式中、「結合」は注入したTCR濃度Cでの平衡結合(RU)であり、Maxは最大結合である)を用いる非線形曲線フィッティングにより得た。
ビオチン化ペプチド-HLAモノマーを、ストレプトアビジンを予め固定した(SA)ストレプトアビジンバイオセンサ(Pall ForteBio)に1nmに捕捉した。センサを遊離ビオチン(2μM)で2分間ブロックした。平衡結合定数は、ロードしたバイオセンサを、96ウェル又は384ウェルサンプルプレート内で系列希釈した可溶性TCR/ImmTAC中に浸漬することにより決定した。プレート振盪は1000rpmに設定した。低親和性相互作用(μM範囲)については、短い結合時間(約2分間)及び短い解離時間(約2分間)を用いた。Octetデータ分析ソフトウェア(Pall ForteBio)を用いて無関係のpHLAをロードした参照バイオセンサの二重参照減算により結合曲線を加工処理した。平衡時の応答(nm)を用いて、等式応答 = Rmax×*conc/(KD + conc) (式中、「応答」は各TCR濃度(conc)での平衡結合(nm)であり、RmaxはpHLA飽和時の最大結合応答である)にフィッティングさせた定常状態のプロットからKD値を推定した。
可溶性天然型TCRを、実施例1に記載の方法に従って調製し、pHLAに対する結合を実施例3に従って分析した。α鎖及びβ鎖のアミノ酸配列は、図2に示したものに対応するものであった。可溶性ビオチン化HLA-A*02を、PRAMEペプチドSLLQHLIGL(配列番号1)を用いて調製し、BIAcoreセンサチップに固定した。
結合はさまざまな濃度で決定され、相互作用のKD値は141μMであると決定された。実施例3の平衡BIAcore法を用いて、14の無関係なペプチドHLA-A*02複合体のパネルに対して交差反応性(特異性)を評価した。14の無関係なpHLAを3つのグループに分け、3つのフローセルの1つにロードして、フローセルごとに各pHLAをおよそ1000RU与えた。30μLの可溶性野生型TCRを、130及び488μMの濃度で20μL / 分の速度で、すべてのフローセルに注入した。いずれの濃度でも、天然型TCRがSLLQHLIGL-HLA-A*02複合体に特異的であることを示す有意な結合は検出されなかった。
図3及び4にそれぞれ示された変異TCRα及びβ可変ドメインのアミノ酸配列(配列番号6〜24)を使用して、ImmTAC分子を調製した。α鎖の開始時にグリシン残基を含めると(配列番号2の番号付けに対して-1位)、大腸菌での生産中にN末端メチオニンの切断効率を改善することがわかった。非効率的な切断は、不均一なタンパク質産物をもたらす可能性があるため、及び/又は開始メチオニンの存在がヒトにおいて免疫原性である可能性があるため、治療にとって有害であり得る。以下のα鎖及びβ鎖を含むImmTAC分子の全長アミノ酸配列を図5に示す。

以下の表にあるデータは、示されたTCR可変ドメイン配列を含むImmTAC分子が、特に適切な親和性及び/又は半減期でSLLQHLIGL-HLA-A*02複合体を認識したことを示している。
実施例5に記載されるように、同じTCR可変ドメイン配列を含むImmTAC分子を、PRAMEポジティブ癌細胞に対するCD3+T細胞の強力かつ特異的な再指向化を仲介する能力について、評価した。T細胞活性化の指標としてインターフェロン-γ(IFNγ)放出を利用した。
以下のα鎖及びβ鎖を含むImmTAC分子の全長アミノ酸配列を図5に示す。

以下の表に示されるα及びβ可変ドメインを含む各ImmTAC分子は、抗原ポジティブMel624細胞の存在下で再志向化T細胞の強力な活性化を証明した。いずれの場合も、EC50値をデータから計算し、以下の表に示す。さらに、各ImmTAC分子は、1nMまでの濃度で、2つの抗原ネガティブ、HLA-A*02ポジティブ細胞の認識を最小限又はまったく示さなかった。ImmTAC分子は、HLA-A*02ネガティブであるPRAMEポジティブ細胞の認識も示さなかった(データは示していない)。図6は、以下の表に収載されている4つのImmTAC分子の代表的なデータを示す。
変異TCR配列を含むImmTAC分子の特異性をさらに実証するために、実施例6に記載されているのと同じELISPOT方法論を用いて、健康なヒト組織に由来する正常細胞のパネルを標的細胞として、さらなる試験を行った。
・正常組織には、心臓血管、腎臓、骨格筋、肺、血管系、肝臓、及び脳が含まれる。いずれの場合も、抗原ポジティブMel624癌細胞をポジティブコントロールとして使用した。

図7(パネルa)に示されたデータは、変異α鎖及びβ鎖a28b50及びa79b46を含むImmTAC分子が、1nMまでの濃度の抗原ポジティブ癌細胞と比較して8つの正常細胞のパネルに対して最小限の反応性を示すことを証明する。同様に、図7(パネルb)のデータは、a28b57及びa79b46を含むImmTAC分子が、1nMまでの濃度の抗原ポジティブ癌細胞と比較して、4つの正常細胞のパネルに対して最小限の反応性を示すことを表している。
変異TCR配列を含むImmTAC分子が、再指向化T細胞による抗原ポジティブ腫瘍細胞の強力な殺傷を仲介する能力を、IncuCyteプラットフォーム(Essen BioScience)を用いて調べた。このアッセイは、アポトーシスのマーカーであるカスパーゼ-3/7の放出の顕微鏡観察によるリアルタイム検出を可能とする。
アッセイは、CellPlayer 96ウェルカスパーゼ-3/7アポトーシスアッセイキット(Essen BioScience, Cat. No.4440)を用いて行い、製造業者のプロトコルに従って行った。簡潔には、標的細胞(Mel624(PRAME+ve HLA-A*02+ve)又はNCI-H1755)を10,000細胞/ウェルでプレートし、一晩インキュベートして細胞を接着させた。ImmTAC分子をさまざまな濃度で調製し、それぞれ25μlを該当するウェルに加えて、最終濃度が1pMと100pMの間になるようにした。エフェクター細胞を、10:1のエフェクター標的細胞比(100,000細胞/ウェル)で使用した。エフェクター細胞のみ、又は標的細胞のみのいずれかを含むサンプルに加えて、ImmTACを含まないコントロールサンプルも調製した。NucViewアッセイ試薬を30μMで作製し、25μlを各ウェルに加え、最終量が150μlになるようにした(最終濃度5μM)。プレートをIncuCyte装置内に配置し、2時間ごと に(ウェルあたり1画像)3日間にわたって撮像した。各画像中のアポトーシス細胞の数を決定し、mm2あたりのアポトーシス細胞として記録した。アッセイは3連で行った。
本実施例で提示されるデータには、以下のTCRα鎖及びβ鎖を含むImmTAC分子が含まれる。

図8及び9に示されたデータは、100 pM以下の濃度における、変異TCR配列を含むImmTAC分子の存在下での抗原ポジティブ癌細胞(図8のメラノーマ細胞株Mel624及び図9の肺癌細胞株NCI-H1755)のリアルタイム殺傷を表す。ImmTAC分子の非存在下では、殺傷は観察されなかった。
Claims (41)
- SLLQHLIGL(配列番号1)-HLA-A*02複合体に対する結合性を有し、FRはフレームワーク領域でありCDRは相補性決定領域であるFR1-CDR1-FR2-CDR2-FR3-CDR3-FR4をそれぞれ含むTCR α鎖可変ドメイン及び/又はTCR β鎖可変ドメインを含んでなり、
(a)α鎖CDRは以下の配列を有し、
CDR1 - TISGTDY(配列番号39)
CDR2 - GLTSN(配列番号40)
CDR3 - CILILGHSGAGSYQLTF(配列番号41)

- 請求項1〜23のいずれか1項に記載のTCR、又は請求項24〜26のいずれか1項に記載のTCR-抗CD3融合分子、もしくは請求項29又は30に記載の細胞を、1又は2以上の医薬的に許容可能な担体又は賦形剤と共に含んでなる医薬組成物。
- 好ましくはヒト対象における医薬に用いるための、請求項1〜23のいずれか1項に記載のTCR、又は請求項24〜26のいずれか1項に記載のTCR-抗CD3融合分子、もしくは請求項27に記載の核酸、請求項31に記載の医薬組成物又は請求項29もしくは30に記載の細胞。
- 好ましくはヒト対象における癌又は腫瘍の治療方法に用いるための、請求項1〜23のいずれか1項に記載のTCR、又は請求項24〜26のいずれか1項に記載のTCR-抗CD3融合分子、もしくは請求項27に記載の核酸、請求項31に記載の医薬組成物又は請求項29もしくは30に記載の細胞。
- ヒト対象がPRAMEを発現する腫瘍を有する、請求項33に記載の使用のためのTCR、TCR抗CD3融合分子、核酸、医薬組成物又は細胞。

- 医薬的有効用量の請求項33に記載の医薬組成物を、必要とする対象に投与することを含んでなる、癌を有するヒト対象の治療方法。
- 抗腫瘍剤を別々に、組合せて、又は逐次に投与することをさらに含んでなる請求項38に記載の方法。
- 請求項1〜23のいずれか1項に記載のTCR、又は請求項24〜26のいずれか1項に記載のTCR-抗CD3融合分子を含んでなる、ヒト対象投与用の注射可能な製剤。
- a)請求項29に記載の細胞を、TCR鎖の発現に最適な条件下に維持し、b)TCR鎖を単離することを含んでなる、請求項1〜23のいずれか1項に記載のTCR、又は請求項24〜26のいずれか1項に記載のTCR-抗CD3融合分子の製造方法。
