(v)抗原結合部位は、「Specificity Determining Residue Usage」(SDRU)(Almagro,Mol Recognit.17:132−43,2004)に基づいてもまた描写されることができ、ここでは、SDRは、抗原接触に直接関与する免疫グロブリンのアミノ酸残基を意味する。
一般的な一態様において、本発明は、PHF−タウに結合する単離モノクローナル抗体又はその抗原結合断片に関する。このような抗PHF−タウ抗体は、PHF−タウにてリン酸化エピトープを結合させる特性、又はPHF−タウにて非リン酸化エピトープを結合させる特性を有することができる。抗PHF−タウ抗体は治療薬として有用である場合があり、また、例えば組織又は細胞中での、生体サンプルにおけるPHF−タウを検出するための調査又は診断試薬としても有用である場合がある。

本発明の抗PHF−タウ抗体、又は本発明のその断片は、脳内でのタウの病理学的凝集を伴う神経変性病、又はタウ異常症を有する患者、例えばADを患う患者における症状の治療、低減、又は予防に使用することができる。
本発明のモノクローナル抗PHF−タウ抗体を、対象におけるAD又は他のタウ異常症の診断方法において使用することができる。

PT3、及びAD脳からの、PHF−タウが富化した(ePHF−タウ)のBalb/cマウスの免疫付与に由来する1組の抗体を、直接酵素結合免疫吸着測定法(ELISA)、ウェスタンブロット、及び免疫組織化学(IHC)で、ホスホ−タウ対非ホスホ−タウでの標的選択性について試験した。PT3は、配列番号:25の重鎖可変領域アミノ酸配列、及び配列番号:30の軽鎖可変領域アミノ酸配列を有するマウス−ハイブリドーマ由来の抗体である。PT3のVH及びVLドメインについてのCDR配列は、米国特許第9,371,376号に示されている。ヒトフレームワーク適応(HFA)法(実施例4を参照)を用いてPT3をヒト化し、本発明のヒト化抗−ホスホ−タウ抗体を生成した(表1及び2を参照)。ヒト化B296は、PT3と同じCDR配列を有する。ヒト化mAb B296は親和性成熟しており、本発明の更なる抗体を生成した(表3を参照)。
マウスIgG1(mIgG1)としての組み換えPT3を、ELISAフォーマットにおいて、富化PHF−タウ及び組み換えヒト野生型タウへの結合について評価した。この組み換えに由来するPT3を、ハイブリドーマ由来の精製PT3と比較した。結果は、精製ハイブリドーマ由来のPT3抗体バッチ、及び組み換え由来のPT3抗体のバッチの両方の間で相当の結合力価曲線を示した(図1)。強力な結合がPHF−タウには存在し、最小の結合がより高濃度にて可溶性タウに存在した。
PT3を用いて、精製非リン酸化組み換えヒトタウ、及びAD脳から調製したサルコシル不溶性PHF−タウに対してウェスタンブロット分析を実施した。PT3は、ホスホ選択性参照抗体AT8 pS202/pT205/pS208と同様に、PHF−タウとの選択交差性を示した(Mercken et al.,Acta Neuropathol.84(3):265−72,1992;Malia et al.,Proteins.84:427−434,2016)and AT100 pT212/pS214(Mercken et al.,1992,Id.;Hoffmann et al.,Biochemistry.36(26):8114−24,1997)(図2)。ホスホとは無関係の参照抗体HT7(Mercken,Ph.D.Thesis:University of Antwerp,Wilrijk−Antwerp,1991)を、組み換えタウ及びPHF−タウの両方と反応させた。ホスホ選択性であるエピトープに向けられるBT2は、S199/S202においてリン酸化タウではなく、組み換えタウのみと反応した(Mercken,1991,Id.)。他のウェスタンブロット実験では、PT3は、より高濃度にてブロットした際でも、組み換えタウとは弱い反応性を示した。
AD及び対照脳の、ホルマリン固定したパラフィン埋め込み切片で免疫組織化学分析を実施し、in situでタウ異常症との反応性を確認した。PT3は、参照タウ異常症特異的診断抗体陽性対照AT8と同様の、しかしより強力な反応性パターンを示した(図3)。これらの実験条件下においては、対照脳の通常のタウとは、有意な反応が検出されなかった(図4)。
IHC分析を、野生型及びノックアウトマウス脳のPT3で実施した。野生型マウス脳のPT3でのIHC分析では、最適なエピトープ保存条件下にて、野生型タウの選択的プールとの反応性を観察することが可能であることが示される。PT3の染色パターンにより、細胞体樹状突起の局在性(図5、矢印)が明らかとなり、ラット及びヒト生検由来組織の抗ホスホ−タウ抗体について、文献に記載された染色が想起される(Matsuo et al.,Neuron.13(4):989−1002,1994)。タウ−1抗体で観察された、タウの典型的な非ホスホ軸索染色パターン(図6、矢印)は存在せず、これは、PT3が微小管結合タウの生理学的に重要なプールとは限定的な反応性を有することを示している。タウノックアウト動物での反応性の欠如により、PT3染色パターンのタウ特異性が確認される。
PT1及びPT3抗PHF−タウ抗体について、ProteOn(Bio−Rad,Hercules,CA)及びBiacore(Biacore,Uppsala,Sweden)機器にてSPRを行うことにより、PHF−タウ及び組み換えタウとの相互作用を評価した。全てのタウ抗体HT7を陽性対照として試験し、AT8を参照抗PHF−タウ抗体として試験した。
hyb、ハイブリドーマにより発現したmAb;rec、組み換えmAb;
括弧内のaKD値は、75nMの注射したmAb濃度を除外することにより得られた。
a Sigma−Aldrich(St.Louis,Mo)製の組み換えタウ(5倍希釈液にて0.12〜75nM)によりProteOnにて試験をし、その後凝集したと判断した。他の全ての試験サンプルについて、インハウスで生成した組み換えタウをBiacoreで使用した。
表6に示すホスホペプチドのパネルを用いた表面プラズモン共鳴(ProteOn)により、PT3のエピトープを決定した。

X線結晶構造解析により、2つのタウホスホペプチドを有するPT3 Fab(B187)の共構造を測定し、これにより、タウエピトープ及びPT3パラトープの同定を行った。
ヒトフレームワーク適合(HFA)法(Fransson et al.,J Mol Biol.398(2):214−31,2010)を使用して、抗タウマウス抗体PT3をヒト化した。Martin(Martin and Thornton,J Mol Biol.263(5):800−15,1996)に従い、ヒトフレームワーク適合のため、CDRを規定した。ヒト化HC及びLCの最も良い組み合わせを発見するために、いくつかのヒト重鎖V領域配列及び軽鎖V領域配列を試験のために選択した。4つのヒトフレームワーク適合PT3重鎖可変領域、及び4つのヒトフレームワーク適合PT3軽鎖可変領域を設計し、完全ヒト重鎖IgG1及びヒト軽鎖κ分子として生成した(図10)。フレームワーク領域(FR)のみの中のマウスPT3 VH及びVLに対する配列類似性に基づいて、ヒト生殖細胞系V遺伝子(VHに関して4つ:IGHV3−23*01、IGHV3−33*01、IGHV3−11*01、及びIGHV1−3*01;VLに関して4つ:IGVK1−16*01、IGVK1−16*01+、IGKV1−39*01、及びIGKV2−24*01)を、ヒトフレームワーク適合VH及びVL多様体を製造するために選択した。VL78(IGVK1−16*01+)はVL77(IGVK1−16*01)の単一点変異体であり、潜在的な異性化のリスクを排除するために、D56S変異を含有する。4つのHFA HC及び4つのHFA LC分子を組み合わせて得られる、16個のHFA多様体モノクローナル抗体の名称を、表9に示す。
生物物理学的特性決定、及びPHFへのELISA結合に基づいて選択したHFA−PT3モノクローナル抗体多様体のサブセットを、以下のホスホペプチド:pT212/pT217−タウペプチド(ペプチド−2、配列番号:48)及びpT212−タウペプチド(ペプチド−8、配列番号:54)への結合に関して、ProteOn XPR36で表面プラズモン共鳴(SPR)により分析した。全ての実験は、25℃にて、ランニング緩衝液及びサンプル希釈緩衝液の両方としてPBST(pH7.4)(Bio−Rad カタログ番号176−2720)を用いて実施した。
アルツハイマー病の脳から単離したPHF−タウに対する結合について、HFA−PT3モノクローナル抗体のサブセットを試験した。3mMのEDTA、及び0.005%のTween20を補充したPBS(pH7.4)をランニング又はシステム緩衝液として使用して、25℃にて全ての相互作用を検討した。マウス抗タウ抗体であるHT7(Pierce、カタログ番号MN1000)を陽性対照として使用した。

X線結晶構造解析により、pT212/pT217−タウペプチド(配列番号:62)を有するB324の共構造を決定し、これにより、タウエピトープ並びにB324(及びB296)パラトープの同定を行った。
2種類の細胞アッセイ:同時培養アッセイ及び枯渇アッセイにおける、タウシーディングの阻害についてPT3を試験した。両方のアッセイ種において、例えば凝集により非常に近接した際にシグナルを生成する、2つの発色団タグK18タウフラグメントを発現するHEK細胞を使用する。細胞を、凝集してリン酸化した、異なる源由来の完全長タウのシードで処理した場合、生物発光共鳴エネルギー移動(BRET)比の変化(すなわち、BRETアッセイ)により、又は、蛍光活性化細胞選別(FACS)を用いた、蛍光共鳴エネルギー移動(FRET)陽性細胞の数を数えること(すなわち、FRETアッセイ;図14)により、定量化可能なK18アグリゲートが生じる(Holmes et al.,2014,PNAS 111(41):E4376−85)。
同時培養用のタウシードを含有するホモジェネートを、K18により誘発された、凝集GFPタグ完全長タウを含有する安定したGFP−タウP301L過剰発現HEK細胞株から生成した。レシピエント細胞は、K18/P301L−NanoLuc及びK18/P301L−HaloTagを安定して発現するHEK細胞であった。タウシードを、試験抗体、及び受容する発色団−K18含有HEK細胞と、72時間同時培養した。K18アグリゲート形成を、BRET比(590nm/450nm)の変化により測定した。PT3は、凝集の誘発を300nMにおいて46.97%、30nMにおいて18.02%、及び3nMにおいて12.57%ブロックした(図15)。
凝集トランスジェニックヒトタウを含有する、週齢22〜23のP301Sの脊髄から、同時培養用のタウシードを含有するホモジェネートを生成した。感度の増加のために、このアッセイで使用したレシピエント細胞は、K18/P301S−YFP及びK18/P301S−CFPを安定して発現するHEK細胞であった。タウシードを、試験抗体、及び受容する発色団−K18含有HEK細胞と、72時間同時培養した。K18アグリゲート形成を、FACSによりFRET陽性細胞の数を数えることにより測定した。PT3は、凝集の誘発を300nMにおいて34.03%、30nMにおいて37.02%、及び3nMにおいて30.68%ブロックした(図16)。
最大阻害割合(%)の値が、シードのエピトープの密度、又はPT3エピトープを含有するシードの数に関係するか否かを調査するために、免疫枯渇アッセイを実施した。免疫枯渇アッセイにおいて、タウシードを試験抗体により培養(インキュベーション)し、Gタンパク質のビーズを用いて溶液から除去した。枯渇した上清を、発色団−K18含有HEK細胞内での残留シーディング能力について試験し、上述したようにFACSにより分析した(Holmes et al.,Proc Natl Acad Sci U S A.111(41):E4376−85,2014)。
PT3は、HEK細胞可溶化物、及びTgP301S脊髄溶解物の両方に由来するタウシードを阻害した。アッセイで得た最大阻害は、異なる抗ホスホ−タウ抗体において、及び異なるシードにおいて変化した(表16)。観察された、300nMにおけるPT3に対する阻害値は、HEK細胞シードに対しては46.97±5.87%であり、TgP301S脊髄抽出物に対しては34.03±2.05%であった。異なる細胞アッセイにおける、ホスホ−タウ抗体に対する異なる最大阻害値により、使用したタウシードのリン酸化状態の違いを示すことができる。TgP301S脊髄で生成したタウシードは神経細胞由来のものであり、HEK細胞由来のタウシードよりもPHF−タウに対して類似性が高いことが予想される。このことにより、脊髄抽出物対HEK細胞可溶化物に対するホスホ−タウ抗体で観察された、全般的に高い効能を説明することができる。
インビボでのタウ抗体の効能を評価するために、脳でタウの病状を示すマウスを、必須のモデルシステムとする(Julien et al.,Methods Mol Biol.849:473−91,2012)。これらのモデルのいくつかが記載されており、これらは一般的に、3つの群に分けることができる:1)株に応じて、5〜9ヶ月後に重篤な病状を示す変異体により、WT又は変異体(例えば、P301L若しくはP301S)を過剰発現するタウトランスジェニックマウス(Allen et al.,J Neurosci.22(21):9340−51,2002;Scattoni et al.,Behav Brain Res.208(1):250−7,2010;Terwel et al.,J Biol Chem.280(5):3963−73,2005;Yoshiyama et al.,Neuron.53(3):337−51,2007);2)変異体タウ(例えば、P301L)を空間時間的に制御して発現するマウス(Liu et al.,Brain Imaging Behav.6(4):610−20,2012)、又は凝集後フラグメント(例えば、K18)を有するマウス(Mocanu et al.,J Neurosci.28(3):737−48,2008);並びに3)プラーク及びタウの病状の両方を示す、変異体タウ及びAPPの両方を発現するマウス(Oddo et al.,J Neurochem.102(4):1053−63,2007)。
注射の検討のために、P301L変異を有する最長のヒトタウアイソフォーム(タウ−4R/2N−P301L)(Terwel et al.,2005,Id.)を発現するトランスジェニックタウ−P301Lマウスを、月齢3ヶ月にて手術に使用した。現地の倫理委員会が認可した手順を遵守して、全ての実験を実施した。定位的手術のために、マウスは、モノクローナル抗体の存在下又は不存在下にて、毛皮質(AP+2.0、前頂からML+2.0、DV、硬膜から2.7mm)、又は海馬(AP−2.0、ML+2.0(前頂から)、DV 1.8mm(硬膜から))に、3μL(速度:0.25μL/分)で、死後AD組織(富化した対らせん状細線維、ePHF)からサルコシル不溶性prepの一側性(右半球)注射を受けた。抗体又は生理食塩水の腹腔内(IP)注射の場合、治療(20mg/kg、2×/週)は頭蓋内注射の1週間前に開始し、マウスを解離のために犠牲にする(頭蓋内注射の2ヶ月後)まで継続した。
注射した半球のマウス組織の重量を測定し、6体積の均質化緩衝液(10mM Tris HCl(pH7.6))中で均質化した。27000×gにて20分間、ホモジェネートを遠心分離にかけ、得られた上清(全ホモジェネート)からアリコートを取り出した後、1%のN−ラウロイルサルコシンを添加した。90分後(900rpm、37℃)、溶液を再び、184000×gにて1時間遠心分離にかけた。上清はサルコシル可溶性画分として保持したが、サルコシル不溶性材料を含有するペレットは、均質化緩衝液中に懸濁させた。
被覆抗体(抗AT8又は全タウ抗体のいずれか)をPBSに希釈(1μg/mL)し、MSDプレート(ウェル当たり30μL)(L15XA,Mesoscale Discoveries)にアリコートとし、これらを終夜4℃でインキュベーションした。5×200μLのPBS/0.5%のTween−20で洗浄した後、プレートをPBS中の0.1%のカゼインでブロックし、5×200μLのPBS/0.5%のTween−20で再び洗浄した。サンプル及び標準(共に、PBS中の0.1%のカゼインに希釈)を添加した後、プレートを終夜4℃でインキュベーションした。その後、プレートを5×200μLのPBS/0.5%のTween−20で洗浄し、SULFO−TAG(商標)がコンジュゲートした検出抗体(PBS中の0.1%のカゼイン中)を添加し、600rpmで振盪しながら室温で2時間インキュベーションした。最後の洗浄(5×200μLのPBS/0.5%のTween−20)の後、150μLの2×緩衝液Tを添加し、プレートをMSDイメージャで読み取った。死後のAD脳(ePHF)のサルコシル不溶性prepの16個の希釈物からなる検量線に対してそのままのシグナルを正規化し、任意の単位(AU)ePHFとして表した。統計分析(Bonferroni事後テストと合わせたANOVA)を、GraphPadプリズムソフトウェアで行った。
皮質同時注射モデルにおけるマウスPT3の活性(図19)が、4つの独立した検討にて確認された。マウスを表17に従い末梢投与し、結果を図20に示す。モデルの更なる改善(図19D)により、表18に示すように、ePHF−タウの用量及び同時注射される抗体の用量の低下が可能となった(結果を図21に示す)。ePHF−タウのこの低下させた用量を用いることにより、末梢投与した際に、PT3は凝集したタウを低下させる有意な効果を有することもまた、発見された(P<0.0001;図21)。
進行性核上性麻痺(PSP)は、パーキンソン症候群、体位の不安定性及び転倒、核上性注視麻痺、並びに認知症を特徴とする、希少かつ致死性の神経変性疾患である(Steele et al.,1964,Archives of Neurology 10:333−359)。病理学的には、脳幹及び大脳基底核、並びに他の脳領域に、4リピート(4R)タウの選択的蓄積が存在する(Dickson DW.Handbook of Clinical Neurology 2008;89:487−491;Williams & Lees,2009,The Lancet Neurology 8:270−279)。アミロイドなどの他の病状の不存在を考慮すると、PSPは一次性タウ異常症であると考えられ、動物モデルデータは、PSPタウが、ADにて生じていると仮説が立てられているものに類似しているシーディングを受け得ることを示唆している(Clavaguera et al..2013,PNAS 110:9535−9540;Sanders et al.,2014,Neuron 82:1271−1288)。そのため、PSPは本発明の抗体で治療することができる。一連の実験を行い、PSPタウとAD PHFタウとの類似性を特性決定した。
ヒト脳組織:臨床診断を受けたPSP(n=5)患者(核尾状核=CAU、及び被殻=PUT)の、典型的には非常に影響を受けている2つの脳領域、並びに、さほど影響を受けていない脳領域(上前頭回=GFS)、並びに、2つの対照(=タウ異常症を有しない)患者の同一の脳領域の凍結保存組織を、Netherland Brain Bankから入手した。後述する凝集アッセイ及び免疫組織化学染色の両方を用いる分析のために、組織を使用した。9名の散発性AD患者の凍結保存組織をUnivulershity of Pennsylvaniaから入手し、凝集アッセイを用いる分析のために使用した。1名のAD患者の凍結保存組織をUnivulershity of Newcastleから入手し、免疫組織化学染色に使用した。
凝集アッセイ:凝集アッセイを実施して、PSPタウのリン酸化度を特定決定した。PT3反応性アグリゲートはPSP脳の中に存在したが、凝集度はAD脳よりも低かった(図23)。参照抗体AT8を用いて得た結果は、PT3を用いて観察されたものに類似していた。これらの結果は、様々なホスホ−タウ抗体を使用して評価した全てのリン酸化部位がPSPタウに存在するものの、ADと比較して、PSPにはよりわずかしかタウ凝集が存在していないことを示唆している。
利用可能なデータは、PT3がPSPのタウに結合することを示唆している。
親和性成熟した抗体の,PHF−タウへのSPR結合の特性決定
親和性成熟したモノクローナル抗体を、アルツハイマー病の脳から単離したPHF−タウへの結合について試験した。3mMのEDTA、及び0.005%のTween 20をランニング又はシステム緩衝液として補充したPBS(pH7.4)を用いて、25℃にてProteOn XPR36 system(Bio Rad,Hercules,CA)を使用して、結合反応速度及び親和性検討を実施した。
ペプチド(10ng/mL)をプレートに終夜直接コーティングしたELISAにより、タウホスホペプチドの結合を分析した。プレートを洗浄し、PBS中の0.1%のカゼインでブロックした後、異なる濃度のHFA−PT3(B296)並びにHFA−PT3の親和性成熟多様体(B809、B333、及びB711)mAbで、プレートをインキュベーションした(図26A)。抗体とのインキュベーションの後、プレートを洗浄して、ウェル当たり50μLのHRPOで、抗Fab抗体を標識した(Jackson Immunoresearch laboratories)(ブロッキング緩衝液中に1:10000に希釈)。別の洗浄ステップの後、メーカーの取扱説明書に従い、「ワンステップ」TMB(Thermo Scientific)を用いて、検出を実施した。EnVision(登録商標)2102 Multilabel Reader(Perkin Elmer,Waltham,MA,USA)でプレートを分析した。結合曲線を、GraphPad Prism7.0ソフトウェアを使用して生成した。図26Aの結合曲線から、B296は最も低い親和性を示した一方で、B711はB296と、またB333及びB809とも比較して、最も強い結合を示したことを知ることができる。これにより、B711は、pT217ペプチドに対して最も強い親和性を有するヒト化PT3であることが示唆される。Fabを用いる同様の実験(図26B)により、M333(B711のFab)は、親PT3分子のFabであるB187と比較して、同様のペプチド結合を有したことが示された。ここでも、M324(B296のFab、HFA−PT3)は、PT3−HFAの親Fab及び親和性成熟した多様体と比較して弱い結合を示した。
