特許請求の範囲および/または本明細書において、「〜を含むこと」という用語と共に使用される場合の、「ある(a)」または「ある(an)」という語の使用は、「1つの」を意味しうるが、また、「1または複数の」、「少なくとも1つの」、および「1つまたは1つを超える」という意味とも合致する。

複数の実施形態では、本発明のCD33 CARは、抗原結合性部分と、T細胞膜との分離をもたらすストークドメインを含む。複数の実施形態では、ストークドメインは、エフェクター−標的間の最適な膜間距離を確立する。複数の実施形態では、ストークドメインは、抗原結合性ドメインが、その標的に到達するための可撓性をもたらす。一実施形態では、ストークドメインは、CD8アルファヒンジドメインである。
複数の実施形態では、CARは、CARストークドメインの細胞外ドメインへと融合させた膜貫通ドメインを含む。一実施形態では、天然において、CAR内のドメインのうちの1つと会合する膜貫通ドメインを使用する。複数の実施形態では、膜貫通ドメインは、膜を貫通する、疎水性のアルファヘリックスである。
本明細書で記載されるCARの、細胞内シグナル伝達ドメインとしてもまた公知の、細胞質ドメインは、CARを入れた免疫細胞の、通常のエフェクター機能のうちの少なくとも1つの活性化の一因となる。「エフェクター機能」という用語は、特化された細胞の機能を指す。T細胞のエフェクター機能は、例えば、細胞溶解活性の場合もあり、サイトカインの分泌を含む、ヘルパー活性の場合もある。したがって、「細胞内シグナル伝達ドメイン」という用語は、エフェクター機能シグナルを伝達し、特化された機能を果たすように細胞を方向付ける、タンパク質の部分を指す。通例、細胞内シグナル伝達ドメインの全体を利用しうるが、多くの場合、鎖の全体を使用する必要はない。細胞内シグナル伝達ドメインの切断部分を使用し、それが、エフェクター機能シグナルを伝達する限りにおいて、このような切断部分を、完全鎖の代わりに使用することができる。したがって、細胞内シグナル伝達ドメインという用語は、エフェクター機能シグナルを伝達するのに十分な、細胞内シグナル伝達ドメインの、任意の切断部分を含むことを意図する。
細胞療法は、ヒト疾患の処置に向けて、大きな将来性を有するが、細胞自体またはそれらのトランス遺伝子産物に由来する著明な毒性は、臨床的探索を妨げてきた。本明細書で記載される複数の実施形態では、本明細書で記載されるCARを含む免疫エフェクター細胞であって、哺乳動物対象、例えば、ヒトへと輸注された免疫エフェクター細胞は、それらの使用から傷害作用が生じた場合、このような免疫エフェクター細胞の作用を調節するために、除去することができる。したがって、ある特定の実施形態では、本明細書で記載される治療剤である、CD33特異的キメラ抗原受容体に加えてまた、第2の遺伝子も、本明細書で記載される改変免疫エフェクター細胞へと導入することができる。第2の遺伝子は、事実上、CD33 CAR含有細胞の枯渇を可能とする「キルスイッチ」である。ある特定の実施形態では、「キルスイッチ」とは、少なくともHER1もしくはCD20の抗体結合エピトープ、またはこれらの機能的断片を含み、任意選択で、シグナルポリペプチド配列またはこの断片を含むHER1ポリペプチドまたはCD20ポリペプチドを含むHER1タグまたはCD20タグである。
本明細書で記載される方法および組成物により包含される、例示的なCARおよびヒトCD33受容体オープンリーディングフレームは、表1にある。
一部の実施形態では、本明細書で記載されるCARを、対象へと、サイトカインを含むがこれらに限定されない、1または複数のさらなる治療剤と共に投与する。場合によって、サイトカインは、少なくとも1つのケモカイン、インターフェロン、インターロイキン、リンホカイン、腫瘍壊死因子、またはこれらの変異体もしくは組合せを含む。場合によって、サイトカインは、インターロイキンである。場合によって、インターロイキンは、IL−1、IL−2、IL−3、IL−4、IL−5、IL−6、IL−7、IL−8、IL−9、IL−10、IL−11、IL−12、IL−13、IL−14、IL−15、IL−16、IL−17、IL−18、IL−19、IL−20、IL−21、IL−22、IL−23、IL−24、IL−25、IL−26、IL−27、IL−28、IL−29、IL−30、IL−31、IL−32、IL−33、およびこれらの機能的な変異体および断片のうちの少なくとも1つである。一部の実施形態では、サイトカインは、膜結合型の場合もあり、分泌型の場合もある。複数の実施形態では、サイトカインは、可溶性IL−15、可溶性IL−15/IL−15Rα複合体(例えば、ALT−803)である。ある特定の場合には、インターロイキンは、膜結合型IL−15(mbIL−15)、またはIL−15と、IL−15Rαとの融合体を含みうる。一部の実施形態では、mbIL−15は、本明細書で記載される改変免疫エフェクター細胞と共に共発現させることができる、膜結合型キメラIL−15である。一部の実施形態では、mbIL−15は、全長IL−15Rα、またはこれらの機能的な断片もしくは変異体とインフレームで融合させた、全長IL−15(例えば、天然IL−15ポリペプチド)、またはこれらの断片もしくは変異体を含む。場合によって、IL−15を、IL−15Rαへと、リンカーを介して、間接的に連結する。場合によって、mbIL−15は、Hurton et al., “Tethered IL-15 augments antitumor activity and promotes a stem-cell memory subset in tumor-specific T cells,” PNAS 2016において記載されている通りである。場合によって、サイトカインを、CARと同じ免疫エフェクター細胞内で発現させる。一部の実施形態では、上記で記載したサイトカインは、遺伝子転写のための誘導性プロモーターの制御下にありうる。一態様では、誘導性プロモーターは、遺伝子スイッチリガンドの誘導性プロモーターでありうる。場合によって、誘導性プロモーターは、RHEOSWITCH(登録商標)遺伝子スイッチなど、低分子リガンド誘導性の、2つのポリペプチドによる、エクジソン受容体ベースの遺伝子スイッチでありうる。
本発明はまた、本発明の核酸を挿入したウイルスベースの系などの送達系も提供する。代表的なウイルス発現ベクターは、アデノウイルスベースのベクター(例えば、Crucell,Inc.(Leiden、Netherlands)から市販されている、アデノウイルスベースのPer.C6系)、レンチウイルスベースのベクター(例えば、Life Technologies(Carlsbad、Calif.)製の、レンチウイルスベースのpLPI)、およびレトロウイルスベクター(例えば、pCFB−EGSHを付加したpFB−ERV)、ヘルペスウイルスを含むがこれらに限定されない。ある実施形態では、ウイルスベクターは、レンチウイルスベクターである。レンチウイルスなどのレトロウイルスに由来するベクターは、トランス遺伝子の、娘細胞における、長期にわたる、安定的な組込みおよびその繁殖を可能とするので、長期にわたる遺伝子導入を達成するのに適するツールである。レンチウイルスベクターは、肝細胞などの非増殖性細胞に形質導入しうるという点で、マウス白血病ウイルスなど、がんレトロウイルスに由来するベクターに優る追加の利点を有する。レンチウイルスベクターはまた、低免疫原性という追加の利点も有する。一般に、かつ、複数の実施形態では、適切なベクターは、少なくとも1つの生物において機能的な複製起点、プロモーター配列、好都合な制限エンドヌクレアーゼ部位、および1または複数の選択用マーカー(例えば、国際公開第01/96584号パンフレット、国際公開第01/29058号パンフレット、および米国特許第6,326,193号明細書)を含有する。
本明細書で記載されるCARをコードする核酸はまた、免疫エフェクター細胞へと、DNA配列を、脊椎動物の染色体へと導入するための合成DNAトランスポゾン系を指す「Sleeping Beauty(SB)トランスポゾン系」など、非ウイルスベースの送達系を使用して導入することもできる。例示的なSBトランスポゾン系については、例えば、米国特許第6,489,458号明細書および同第8,227,432号明細書において記載されており、図3に例示されている。Sleeping Beautyトランスポゾン系は、Sleeping Beauty(SB)トランスポザーゼおよびSBトランスポゾンから構成される。
本明細書では、本明細書で記載されるCARを発現する改変細胞が提示される。ある特定の実施形態では、本明細書で記載される改変細胞は、免疫エフェクター細胞である。本明細書の複数の実施形態では、骨格と、(1)切断型表皮増殖因子受容体(HER1tまたはHER1t−1)、ならびに(2)(a)CD33抗原結合性ドメイン、(b)ストークドメイン、(c)膜貫通ドメイン、(d)4−1BBもしくはCD28、またはこれらの両方を含む共刺激シグナル伝達ドメイン、および(e)CD3ゼータシグナル伝達ドメインを含むキメラ抗原受容体(CAR)をコードする核酸配列とを含むベクターを含む免疫エフェクター細胞が提示される。
本明細書で記載される、1または複数の異種遺伝子または異種ポリペプチドを発現するように修飾された免疫エフェクター細胞が提示される。本明細書で記載されるCD33 CAR、ならびにHER1tタグ、HER1t−1タグ、CD20タグ、およびCD20t−1タグのうちの少なくとも1つを発現するように修飾された免疫エフェクター細胞が提示される。場合によって、本明細書で開示されるCD33 CAR、mbIL−15、ならびにHER1tタグ、HER1t−1タグ、CD20タグ、およびCD20t−1タグのうちの少なくとも1つを発現するように修飾された免疫エフェクター細胞が提示される。

ある特定の態様では、本明細書で記載される実施形態は、リダイレクトされた、抗原特異的免疫エフェクター細胞(例えば、T細胞、NK細胞、またはNK T細胞)を作り出し、かつ/またはこれを拡大増殖させる方法であって、細胞に、CARをコードするDNA(またはRNA)構築物を含有する発現ベクターによりトランスフェクトし、次いで、任意選択で、細胞を、フィーダー細胞、組換え抗原、または受容体に対する抗体で刺激して、細胞を増殖させるステップを含む方法を含む。ある特定の態様では、CARを発現するように改変された細胞(または細胞集団)は、幹細胞、iPS細胞、免疫エフェクター細胞、またはこれらの細胞の前駆細胞である。
本明細書で記載されるCARを発現するように、T細胞を改変する前であれ、この後であれ、T細胞は、例えば、米国特許第6,352,694号明細書、同第6,534,055号明細書、同第6,905,680号明細書、同第6,692,964号明細書、同第5,858,358号明細書、同第6,887,466号明細書、同第6,905,681号明細書、同第7,144,575号明細書、同第7,067,318号明細書、同第7,172,869号明細書、同第7,232,566号明細書、同第7,175,843号明細書、同第5,883,223号明細書、同第6,905,874号明細書、同第6,797,514号明細書、同第6,867,041号明細書、および米国特許出願公開第20060121005号明細書において記載されている方法を一般に使用して、活性化させ、拡大増殖させることができる。
本明細書の複数の実施形態では、レンチウイルスベクター(LV)により形質導入された免疫エフェクター細胞(例えば、T細胞)について記載される。例えば、LVは、CD33の抗原認識ドメインを、CD8アルファヒンジのストークドメインおよびこの変異体、CD3ゼータの細胞内ドメイン、CD28、4−1BB、またはこれらの任意の組合せとCD3ゼータの細胞内ドメインを組み合わせるCARをコードする。したがって、場合によって、形質導入されたT細胞は、CAR媒介性T細胞応答を誘発しうる。
本開示の一態様は、本発明のCD33特異的CARをコードするコード領域と、任意選択で、安全性の理由で組み入れられる遺伝子、例えば、HER1tまたはHER1t−1、およびこれらの機能的変異体、またはCD20またはCD20t−1、およびこれらの機能的変異体とを含む第1のベクターを含む、キットおよび組成物に関する。これらのキットおよび組成物は、各々が異なるタンパク質またはタンパク質のサブセットをコードする、複数のベクターを含みうる。これらのベクターは、ウイルスベクターの場合もあり、非ウイルスベクターの場合もあり、エピソームベクターの場合もあり、組込みベクターの場合もある。一部の実施形態では、ベクターは、トランスポゾン、例えば、sleeping beautyトランスポゾンである。
表2には、本明細書で提示される実施形態に含まれる、ある特定の配列についての代表的なリストを提示する。
レンチウイルスベクターを介する、CD33 CARの送達:CD33 CARレンチウイルスベクターは、HIV−1由来のベクター骨格に基づき構築した。HER1t遺伝子を、CD33 CARへと、3’末端において、インフレームの、自己切断型トセア・アシグナ(Thosea asigna)ウイルス2Aペプチドペプチド(T2A)を介して遺伝子融合させた。いずれの遺伝子も、下記で記載されるpFUGWレンチウイルスプラスミド骨格へとクローニングした。
LV−CD33 CARベクターは、トランス遺伝子の発現を駆動する、ヒト伸長因子1アルファ1(EF1A1)プロモーターおよびウシ成長ホルモンポリA配列、ならびにKozakリボソーム開始配列を含有した。ベクターは、ウッドチャック肝炎ウイルス転写後調節エレメント(WPRE)のほか、pFUGWプラスミドに由来するLTR配列も使用した。複製起点は、pUC19プラスミドに基づいた。図2は、CD33 CARのレンチウイルスベクターマップである。
CD33 CARベクターを作り出す方法
LV−CD33 CARベクターを、3つの供給源からアセンブルした:
(1)以下を、既に構築されたベクターから得、制限酵素であるPacIおよびBlpIで消化した:
(a)pFUGW骨格構成要素
(b)ヒト伸長因子1アルファ1プロモーター(EF1A1)プロモーター、およびGM−CSFRアルファシグナルペプチドを伴う抗CD33 scFvを駆動するエンハンサー
(c)CD8αヒンジ領域および膜貫通領域
(2)4−1BBシグナル伝達ドメインモジュールは、合成遺伝子として得た;
(3)CD3ζ、T2A転写リンカー、GM−CSFRアルファシグナルペプチド、HER1t枯渇マーカー、ウシ成長ホルモンポリA、およびフランキングのクローニング部位を含有する領域を、PCRにより増幅した。
送達系の効率
健常ドナーに由来するT細胞への、LV−CD33 CARレンチウイルスベクターによる形質導入は、典型的に、形質導入の12〜14日後において、CD33 CARおよびHER1tを安定的に共発現する、25〜50%の細胞を結果としてもたらした。図4は、CAR−T細胞の作出のための、典型的なレンチウイルスによる形質導入の概略について記載する。図5は、レンチウイルスによる形質導入の12日後における、健常ドナーのT細胞1個からの、CD33 CARおよびHER1tの発現についての代表的なデータを示す。Dynabeads(登録商標)Human T−Activator CD3/CD28ビーズを使用して、精製ヒトCD3+ T細胞を活性化させてから、感染多重度(MOI)を5として、CD33 CARレンチウイルスベクターによる形質導入を行った。細胞は、IL−2を含有する培地中で、形質導入後12日間にわたり増殖させた。形質導入の12日後に、フローサイトメトリー解析を実施して、それぞれ、プロテインLおよびセツキシマブmAbにより測定される、CD33 CARおよびHER1tの発現を評価した。
遺伝子素材の組込み
送達された遺伝子素材は、プロウイルスゲノムの形態で組み込まれた。レンチウイルスによる形質導入を使用して、T細胞内に導入されたトランス遺伝子は、研究期間にわたり安定的であった。図8は、レンチウイルスによる形質導入後における、CD33−CAR−T細胞についてのコピー数評価である。5のMOIを使用して、ヒトSUPT1 T細胞または健常なドナー270169(D270169)に由来するT細胞への、レンチウイルスによる形質導入後において、宿主ゲノムへと組み込まれた遺伝子コピーの数を決定するのに、ドロップレットデジタルPCR(ddPCR)を用いた。形質導入の少なくとも14日間後において、ゲノムDNA(gDNA)を得た。ddPCR反応は、Bio−Rad社製のddPCRシステム(QX200TM AutoDG ddPCRシステム)を、CD33 CARを検出するための、Taqmanベースのプライマーおよびプローブセットと共に使用して準備した。あらゆる細胞は、2コピーずつのRNaseP遺伝子を有するという仮定に基づき、RNasePを、コピー数変異体(CNV)を評価するための、標準化基準遺伝子として使用した。SUPT1/CD33 CAR−T細胞上およびD270169/CD33 CAR−T細胞上のCD33 CARの発現は、フローサイトメトリーにより確認され、それぞれ、約80%および約30%(データは示さない)であると報告された。D270169/CD33 CAR−T細胞について、アッセイにおいて測定された試料(「測定」と称する)は、混合集団に由来するが、さらなる計算を実施して、CARを発現するT細胞の、インサートのコピー数(「逆算」と称する)を決定した。示されるデータは、2つの実験による、細胞1個当たりの平均値コピー数±SDである。
遺伝子導入系の有効性について評価する、動物モデルおよび培養細胞モデル
導入された遺伝子の発現により測定される、レンチウイルス媒介性遺伝子導入の効率を、形質導入細胞のフローサイトメトリーにより査定した(図5)。形質導入T細胞の拡大増殖は、妥当な用量を達成するように、形質導入後12〜14日間にわたり、ex vivoの培養物培地中で実行した(図7)。
サイトカイン解析:マウスに由来する血漿試料を、プロトコール内の特異的時点、またはマウスが瀕死となり、人道的理由で安楽死させられた時点において回収した。血漿を得るために、血液を、回収用EDTA試験管内に回収し、次いで、室温、5,000×gで、15分間にわたりスピンし、次いで、結果として得られる血漿を、きれいな回収用試験管へと導入した。血漿を凍結させ、アッセイまで、−80℃で保存した。ProcartaPlex(登録商標)ヒトサイトカイン/ケモカイン/増殖因子パネル1キット(eBioscience)は、以下の解析物:脳由来神経栄養因子(BDNF)、表皮増殖因子(EGF)、エオタキシン、線維芽細胞増殖因子2(FGF−2)、顆粒球/マクロファージ−コロニー刺激因子(GM−CSF)、GROアルファ(CXCL1)、肝細胞増殖因子(HGF)、インターフェロン(IFN)アルファ、IFNガンマ(IFNγ)、インターロイキン(IL)−1受容体アルファ(IL−1RA)、IL−1アルファ、IL−1ベータ、IL−2、IL−4、IL−5、IL−6、IL−7、IL−8、IL−9、IL−10、IL−12p70、IL−13、IL−15、IL−17A、IL−18、IL−21、IL−22、IL−23、IL−27、IL−31、インターフェロンガンマ誘導性タンパク質10(IP−10;CXCL10としてもまた公知である)、白血病阻害因子(LIF)、単球走化性タンパク質−1(MCP−1;CCL2として公知である)、マクロファージ炎症性タンパク質1アルファ(MIP−1アルファ;CCL3としてもまた公知である)、MIP−1ベータ(CCL4としてもまた公知である)、ベータ−神経増殖因子(NGFベータ)、RANTES(CCL5としてもまた公知のRegulated upon Activation Normal T cell Expressed and Secreted)、血小板由来増殖因子BB(PDGF−BB)、胎盤増殖因子1(PIGF−1)、幹細胞因子(SCF)、間質由来因子1アルファ(CXCL12アルファとしてもまた公知のSDF−1アルファ)、腫瘍壊死因子アルファ(TNFアルファ)、TNFベータ(リンホトキシンアルファとしてもまた公知である)、血管内皮増殖因子アルファ(VEGF−A)、および血管内皮増殖因子D(VEGF−D)のマルチプレックス検出のための、45プレックスの磁気ビーズベースのキットである。アッセイは、製造元の特殊なプロトコールに従い実施した。データは、Bio−Plex Managerソフトウェアversion 6.1を稼働するBio−Rad Bio−Plex(登録商標)200測定器(Bio−Rad、Hercules、CA)上で収集した。
多様なシグナル伝達ドメインを伴うレンチウイルスCD33 CARを構築し、発現について調べた。CD3ζシグナル伝達ドメインだけを伴うCD33 CAR、CD28−CD3z(CD28zと称する)シグナル伝達ドメインを伴うCD33 CAR、またはwithHER1tの共発現を伴うCD28−CD3zドメインを伴うCD33 CARを伴うレンチウイルス粒子を、図4に概括された概略図に従い、あらかじめ活性化させたヒトT細胞へと形質導入した。フローサイトメトリー解析は、形質導入の12〜14日後に、T細胞について実施した。CD33 CARの発現は、プロテインL染色を介して検出し、HER1tは、セツキシマブ染色を介して検出した(図15Aおよび15B)。
Sleeping BeautyによるCD33 CAR
様々なCD33 CAR構築物を、Sleeping Beautyトランスポザーゼ−トランスポゾン系において、構築および作出した(図3)。例えば、CD33−CD8a−CD28TM−CD3z構築物を、多様なEF1aプロモーター長(短、中、および長)で作り出した。また、HER1t細胞タグを伴うCD33 CAR構築物も作りだし、調べた。
リソソーム膜タンパク質1(LAMP−1)としてもまた公知のCD107aは、細胞の後期エンドソーム−リソソーム内では、構成的に発現するが、脱顆粒化細胞の細胞表面上では、一過性に発現する。CD33の発現を伴うかまたは伴わない、異なる標的細胞を認識するCD33 CAR Tの能力を、細胞1個当たりの細胞内IFNγの共時的な検出により評価する、脱顆粒化アッセイを確立した。略述すると、CD33 CAR T細胞を、96ウェルプレート内のE:T比を10:1として、標的細胞と共に共培養した。標的細胞は、K562/CD19(内因性のCD33発現を有する)、THP−1およびEL−4(マウス細胞株)を含んだ。共培養の開始時に、蛍光コンジュゲートCD107a抗体または蛍光コンジュゲートアイソタイプ抗体を、Transport Inhibitor Cocktail(モネンシンおよびブレフェルジンを含有する;1倍濃度;eBioscience)と共に添加し、37℃で、4時間にわたりインキュベートした。インキュベーション時間の終了時に、細胞を、プレート内でペレット化させ、細胞表面抗原を、CAR発現およびT細胞マーカーの検出のために染色した。細胞表面染色の後、細胞はまた、製造元の指示書に従い、Fixable Cell viability dye(eBioscience)によっても染色し、次いで、洗浄するのに続き、Fix/Perm Solution(BD Biosciences)による固定を行った。試料を固定した後で、細胞を、Perm/Wash solution(BD Biosciences)中で洗浄し、次いで、蛍光コンジュゲート抗ヒトIFNγ抗体で細胞内染色した。試料を洗浄し、次いで、適切な染色緩衝液中に再懸濁させ、LSR IIフローサイトメーター(BD Biosciences)により、データを収集した。図18に示した通り、CD107a脱顆粒化の著明な発現が、K562由来細胞およびCD33を発現するTHP−1細胞株だけにおいて観察されたのに対し、免疫エフェクター細胞だけおよびEL−4との共培養により観察された脱顆粒化は、最小限であった。細胞内IFNγ発現についても、同様のパターンを観察した。
ゲノムへの構築物の組込みを媒介するように、Sleeping Beautyベースのトランスポゾン系を使用して、CAR構築物を、細胞へと、電気穿孔を介して導入した。0日目に、PBMC2000万個を、15μgのトランスポゾン(CD33 CAR−CD8−CD28zに対して)および5μgのトランスポザーゼ(pKan−CMV−SB11)と混合された100μLのAmaxa Human T cell Nucleofector solution(型番:VPA−1002; Lonza、Basel、Switzerland)中に再懸濁させ、Program U−14を使用して、電気穿孔した。翌日(1日目)、細胞をカウントし、セツキシマブを使用する、プロテインL染色およびHER1t染色により、CARの発現について表面染色した。細胞を、比を1:1とする、γ照射(100Gy)またはマイトマイシンC処理されたAaPCで刺激した。使用されるAaPC細胞は、CD64−CD8641BBL−CD19−mbIL−15/IL15Ra−ROR1抗原を発現するK562−AaPCであった。CAR T細胞を、比を1:1とするAaPCで刺激した。培養物に、刺激の第1のラウンドには、IL−21(30ng/ml)だけを補充し、その後、残りの刺激には、組換えヒトIL−2(50IU/ml)およびIL−21(30ng/ml)(Pepro−Tech)を補充した。T細胞培養物は、典型的に、7日間にわたり続いた各刺激サイクルの終了時において表現型解析にかけた。プロテインL染色を用いて、またはマルチパラメータフローサイトメトリーにより検出される、組換えCD33/Fcタンパク質染色により、培養物を、CAR T細胞の発現について、表現型解析にかけた。培養物はまた、NK細胞(CD3− CD56+集団として規定される)の増殖についても、緊密にモニタリングし、百分率が、全細胞集団のうちの10%を超えたら、製造元の指示書に従い、CD56についての磁気ビーズ(Stem Cell Technologiesおよび/またはMiltenyi Biotec)を使用して、CAR T細胞培養物から除去した。フローサイトメトリーを使用して、AaPCによる刺激後における、多様なドナーPBMCからの、CD33 CARの発現について査定した。代表的なドナーによるデータを、CD33 CAR−CD8−CD28zについては、表3にまとめ、HER1t細胞タグを伴うかまたは伴わないCD33−CAR−CD8−4−1BBzについては、表4にまとめる。図19は、HER1tタグを伴う、Sleeping Beauty CD33 CAR−CD28z構築物の発現をさらに裏付ける。
レンチウイルスによる、NK細胞への形質導入はまた、多様なHER1tタグ(配列番号32および54)およびC20tタグへとカップリングさせたCD33 CARによっても実施した。図25〜27は、CD33 CAR、ならびにHER1tタグおよびCD20t−1タグの、NK細胞上の発現を裏付ける。図28〜29はまた、CD33 CAR NK細胞が、NK抵抗性CD33+ AML細胞を、異なるE:T比で、効率的に溶解させることも裏付けた。異なる標的細胞と共に共培養したところ、CD33 CAR NK細胞により、サイトカイン(例えば、IFNγ、TNF−a、IL−6、RANTES、GM−CSF、IL−10、MIP−1a、およびMIP−1b)が産生された(データは示さない)。
Claims (73)
- キメラ抗原受容体(CAR)をコードする単離核酸であって、前記CARが、
(a)CD33抗原結合性ドメイン、
(b)ストークドメイン、
(c)膜貫通ドメイン、
(d)4−1BBもしくはCD28、またはこれらの両方を含む共刺激シグナル伝達ドメイン、
(e)CD3ゼータシグナル伝達ドメイン
を含む、単離核酸。 - 前記CD33抗原結合性ドメインが、
(a)配列番号8のアミノ酸配列(hM195scFv)との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチド、
(b)配列番号9および10のアミノ酸配列(M2H12)との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチド、
(c)配列番号11および12のアミノ酸配列(DRB2)との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチド、ならびに
(d)配列番号14および15のアミノ酸配列(My9−6)との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチド
のうちの少なくとも1つを含む、請求項1に記載の単離核酸。 - 前記CD33抗原結合性ドメインが、配列番号8のアミノ酸配列(hM195scFv)との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチドである、請求項1または2に記載の単離核酸。
- 前記ストークドメインが、配列番号22のアミノ酸配列(CD8アルファヒンジ)との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチドである、請求項1または2に記載の単離核酸。
- 前記共刺激シグナル伝達ドメインが、4−1BBを含む、請求項1から4のいずれかに記載の単離核酸。
- 4−1BBの共刺激シグナル伝達ドメインが、配列番号24のアミノ酸配列との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチドを含む、請求項5に記載の単離核酸。
- 前記共刺激シグナル伝達ドメインが、CD28を含む、請求項1から5のいずれかに記載の単離核酸。
- CD28の共刺激シグナル伝達ドメインが、配列番号28のアミノ酸配列との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチドを含む、請求項7に記載の単離核酸。
- 前記CD3ゼータシグナル伝達ドメインが、配列番号26のアミノ酸配列との、少なくとも90%、91%、92%、93%、94%、95%、96%、97%、98%、99%、または100%の同一性を有するポリペプチドを含む、請求項1から8のいずれかに記載の単離核酸。

- 請求項1から13のいずれか一項に記載のヌクレオチドを含む免疫エフェクター細胞。
- 骨格と、(1)切断型表皮増殖因子受容体(HER1t)、ならびに(2)(a)CD33抗原結合性ドメイン、(b)ストークドメイン、(c)膜貫通ドメイン、(d)4−1BBもしくはCD28、またはこれらの両方を含む共刺激シグナル伝達ドメイン、および(e)CD3ゼータシグナル伝達ドメインを含むキメラ抗原受容体(CAR)をコードする核酸配列とを含むベクターを含む免疫エフェクター細胞。
- (1)キルスイッチ、選択マーカー、バイオマーカー、またはこれらの組合せとしての使用のための細胞タグ、ならびに(2)(a)CD33抗原結合性ドメイン、(b)ストークドメイン、(c)膜貫通ドメイン、(d)4−1BBもしくはCD28、またはこれらの両方を含む共刺激シグナル伝達ドメイン、および(e)CD3ゼータシグナル伝達ドメインを含むキメラ抗原受容体(CAR)を含む免疫エフェクター細胞。

- T細胞、ナチュラルキラー(NK)細胞、細胞傷害性Tリンパ球(CTL)、または調節性T細胞である、請求項35から41のいずれか一項に記載の免疫エフェクター細胞。
- 前記CD33抗原結合性ドメインが、CD33に結合すると、抗腫瘍免疫を呈する、請求項42に記載の免疫エフェクター細胞。

(b)前記細胞に、請求項1から13および47から48のいずれか一項に記載の単離核酸をコードする、1または複数のベクター、ならびにトランスポザーゼをコードするベクターをトランスフェクトして、改変CD33 CAR発現T細胞の集団をもたらすステップと、
(c)任意選択で、CD33 CAR T細胞の前記集団を、ex vivoにおいて、2日間またはこれ未満にわたり培養するステップと
