Abstract
Description
本発明は、複数のキメラ抗原受容体(CAR)を含む細胞に関するものである。該細胞は、標的細胞による2またはそれより多くの抗原の発現(または非発現)パターンに差異がある故に、該標的細胞を特異的に認識することができるものであり得る。
若干の免疫治療剤が、治療用モノクローナル抗体(mAb)、免疫コンジュゲートmAb、放射性コンジュゲートmAbおよび二重特異性T細胞エンゲージャーを含め、がん処置での使用について記載されている。
キメラ抗原受容体は、モノクローナル抗体(mAb)の特異性をT細胞のエフェクター機能に結び付けるタンパク質である。キメラ抗原受容体の通常形態は、抗原認識性アミノ末端、スペーサー、T細胞生存および活性化シグナルを伝達する複合エンドドメインにすべて連結された膜貫通ドメインを有するI型膜貫通ドメインタンパク質の形態である(図1A参照)。
「on−target,off−tumour」毒性の問題に取り組むため、二重抗原特異性(specificty)をもつCAR T細胞が開発された。この「二重標的化」手法では、2つの相補的CARが同じT細胞集団で共発現され、それぞれ離れた腫瘍標的に指向され、相補的シグナルを提供すべく操作が加えられている。
本発明の実施形態において、例えば以下の項目が提供される。
(項目1)
T細胞であって、第1のキメラ抗原受容体(CAR)および第2のCARを該細胞表面で共発現し、各CARが、
(i)抗原結合ドメイン
(ii)スペーサー
(iii)膜貫通ドメイン、および
(iv)エンドドメイン
を含み、該第1のCARおよび該第2のCARの該抗原結合ドメインが異なる抗原に結合し、該第1のCARの該スペーサーが該第2のCARの該スペーサーとは異なり、該第1のCARまたは該第2のCARの一方が活性化エンドドメインを含む活性化CARであり、他方のCARがライゲーション・オフ阻害性エンドドメインを含む阻害性CARのいずれかである、T細胞。
(項目2)
前記第1のCARおよび前記第2のCARがそれらのそれぞれの標的抗原と結合したとき、該第1のCARおよび該第2のCARが前記T細胞膜上で空間的に分離されるように、該第1のCARの前記スペーサーが、該第2のCARの前記スペーサーとは異なる長さおよび/または電荷および/またはサイズおよび/または立体配置および/またはグリコシル化を有する、項目1に記載のT細胞。
(項目3)
第1の前記スペーサーまたは第2の前記スペーサーのいずれかがCD8ストークを含み、他方のスペーサーがIgG1のヒンジ、CH2およびCH3ドメインを含む、項目2に記載のT細胞。
(項目4)
前記第1のCARまたは前記第2のCARの一方が、活性化エンドドメインを含む活性化CARであり、他方のCARが、ライゲーション・オフ阻害性エンドドメインを含む阻害性CARであり、その阻害性CARが、阻害性CARライゲーションの非存在下では該活性化CARによるT細胞活性化を阻害するが、該阻害性CARがライゲーションされたときには、該活性化CARによるT細胞活性化を有意に阻害しない、項目2または3に記載のT細胞。
(項目5)
前記阻害性エンドドメインが、CD148またはCD45からのエンドドメインの全部または一部を含む、項目4に記載のT細胞。
(項目6)
前記第1のCARの前記抗原結合ドメインがCD5と結合し、前記第2のCARの前記抗原結合ドメインがCD19と結合する、項目4または5に記載のT細胞。
(項目7)
2つより多くの抗原の区別可能なパターンを担持する、T細胞などの細胞により特異的に刺激されるような、先行する項目に記載のCARを2つより多く含むT細胞。
(項目8)
項目1〜7のいずれかに記載の前記第1のキメラ抗原受容体(CAR)および前記第2のキメラ抗原受容体(CAR)の両方をコードする核酸配列。

キメラ抗原受容体(CAR)
図1で概略的に示されているCARは、細胞外抗原認識ドメイン(バインダー)を細胞内シグナル伝達ドメイン(エンドドメイン)に結び付けるキメラI型膜貫通タンパク質である。バインダーは、典型的にはモノクローナル抗体(mAb)から誘導された1本鎖可変フラグメント(scFV)であるが、抗体様抗原結合部位を含む他のフォーマットに基づくことも可能である。スペーサードメインは、通常膜からバインダーを単離し、好適な配向をもたせるのに必要である。使用される一般的なスペーサードメインは、IgG1のFcである。より小型のスペーサー、例えば、抗原によっては、CD8αからのストーク、さらにはIgG1ヒンジ単独でも十分であり得る。膜貫通ドメインは、細胞膜にタンパク質をつなぎ止め、スペーサーをエンドドメインに結び付ける。

この実施形態では、本発明の第1のCARおよび第2のCARの抗原結合ドメインは、異なる抗原に結合し、両CARとも、活性化エンドドメインを含む。両CARとも、2種の異なる受容体の交差対合を防止するために異なるスペーサードメインを有する。このため、T細胞に対し、抗原の一方または両方の認識の際に活性化するように操作することができる。このことは、Goldie−Coldmanによる仮説により示されているように腫瘍学の分野で有用である:単一抗原を標的化するだけでは、ほとんどのがんに固有の高い変異率故に前記抗原のモジュレーションにより腫瘍エスケープが起こり得る。2つの抗原を同時に標的化することにより、かかるエスケープの確率(probably)は指数関数的に低減される。
ANDゲートおよびAND NOTゲートを生成するためのCARの後続的対合は、T細胞活性化の動力学的分離モデル(KS)に基づく。これは、いかにしてT細胞受容体による抗原認識が下流の活性化シグナルに変換されるかを説明する、実験データに裏付けられた機能モデルである。簡単に述べると、基底状態では、T細胞膜上のシグナル伝達成分は、動的ホメオスタシスにあり、脱リン酸化ITAMは、リン酸化ITAMより好適である。これは、膜貫通CD45/CD148ホスファターゼの活性が、lckなどの膜連結キナーゼよりも大きいためである。T細胞が、同種抗原のT細胞受容体(またはCAR)認識を通して標的細胞と結び付いたとき、堅固な免疫学的シナプスが形成される。T細胞および標的膜のこの近接した並置により、CD45/CD148は、シナプスへ適合できないそれらの大きなエクトドメイン故に排除される。ホスファターゼの非存在下での、シナプスにおける高濃度のT細胞受容体関連ITAMおよびキナーゼの分離は、リン酸化ITAMの方が有利にはたらく状態をもたらす。ZAP70は、閾値のリン酸化ITAMを認識し、T細胞活性化シグナルを伝搬する。本発明では、T細胞活性化に対するこの進歩した理解を活用する。特に、本発明は、いかにして異なる長さおよび/またはかさおよび/または電荷および/または立体配置および/またはグリコシル化を有するエクトドメインがシナプス形成の際に特異的な分離(differential segregation)をもたらすかについての理解に基づいている。
この実施形態では、1つのCARが活性化エンドドメインを含み、1つのCARが阻害性エンドドメインを含むことにより、阻害性CARは構成的に第1の活性化CARを阻害するが、その同種抗原の認識の際にその活性化CAR阻害を解除する。このように、標的細胞が両同種抗原を発現する場合のみ発動するようにT細胞は操作され得る。この行動は、ITAMドメイン含有活性化エンドドメイン、例えばCD3−ゼータのエンドドメインを含む活性化CAR、およびITAMを脱リン酸化できるホスファターゼ(例えば、CD45またはCD148)からのエンドドメインを含む阻害性CARにより達成される。重大なことに、両CARのスペーサードメインは、サイズおよび/または形状および/または電荷などが有意に異なる。活性化CARのみがライゲーションされたとき、阻害性CARはT細胞表面上で溶解状態にあり、活性化CARを阻害するシナプスの内外で拡散することができる。両CARがライゲーションされたとき、スペーサー特性が異なる故に、活性化CARおよび阻害性CARは特異的に分離され、活性化CARに、阻害性CARにより妨害されないT細胞活性化を誘発させ得る。
この実現形態では、1つのCARが活性化エンドドメインを含み、1つのCARが阻害性エンドドメインを含み、その結果この阻害性CARがその同種抗原を認識したときのみ活性である。このため、このように操作されたT細胞は、第1の抗原の存在のみに応答して活性化されるが、両抗原が存在するときには活性化されない。本発明は、第1のCARと共局在するスペーサーを伴う阻害性CARにより実現されるが、阻害性CARのホスファターゼ活性は、当然阻害性CARが溶解状態で阻害するほど強力ではないか、または阻害性CARがその同種標的を認識したときのみ阻害性エンドドメインは実際にホスファターゼをリクルートするかのいずれかである。かかるエンドドメインは、本明細書では「ライゲーション・オン」または半阻害性と呼ぶ。
NOTゲートは、正常細胞であって、ただし活性化エンドドメインを含むCARにより認識される抗原も発現する正常細胞により発現される抗原の標的化を可能にするため、腫瘍学分野でかなりの有用性がある。かかる抗原の一例は、正常幹細胞および急性骨髄性白血病(AML)細胞により発現されるCD33である。CD34は、幹細胞で発現されるが、典型的にはAML細胞からは発現されない。CD33 AND NOT CD34を認識するT細胞は、白血病細胞の破壊をもたらすが、正常幹細胞を残存させる。
上記成分を伴う動力学的分離モデルにより、複合ゲート、例えば2つより多くの標的抗原のパターンに応答して発動するT細胞が作製され得る。例えば、3つの抗原が存在するときのみ(A AND B AND C)発動するT細胞を作製することが可能である。ここで、細胞は3つのCARを発現し、それぞれ抗原A、BおよびCを認識する。1つのCARは興奮性であり、2つのCARは阻害性であり、各CARは、特異的分離をもたらすスペーサードメインを有する。3つ全てがライゲーションされたときのみ、T細胞は活性化する。さらなる例(A OR B)AND C:ここで、抗原AおよびBを認識するCARは、活性型であり、共局在するスペーサーを有し、抗原Cを認識するCARは阻害性であり、異なる共分離をもたらすスペーサーを有する。さらなる例(A AND NOT B)AND C:ここで、抗原Aに対するCARは、活性化エンドドメインを有し、条件的阻害性エンドドメインを有する抗原Bに対するCARと共局在する。抗原Cに対するCARは、AまたはBとは異なる形で分離するスペーサーを有し、阻害性である。事実、より複合的なブール論理でも、任意の数のCARおよびスペーサーを伴う本発明のこれらの単純な成分でプログラム化することができる。
本発明のT細胞のCARはシグナルペプチドを含み得、その結果、CARがT細胞などの細胞の内側で発現されるとき、新生タンパク質を小胞体、それに続いて細胞表面へと指向させ、そこで発現される。

本発明の第2の実施態様は、本発明の第1の実施態様で記載した第1のCARおよび第2のCARをコードする1つまたはそれより多くの核酸配列(複数も可)に関する。
核酸配列は、以下の配列のうちの1つ、またはその変異型を含み得る:

本発明はまた、1つまたはそれより多くのCARエンコード核酸配列(複数も可)を含むベクターまたはベクターのキットを提供する。かかるベクターは、宿主細胞へ核酸配列(複数も可)を導入するのに使用され得、それにより、その宿主細胞が第1のCARおよび第2のCARを発現する。
本発明はまた、本発明の第1の実施態様によるT細胞またはNK細胞などの複数のCAR発現細胞を含む医薬組成物に関する。医薬組成物は、さらに薬学的に許容し得る担体、希釈剤または賦形剤を含んでいてもよい。医薬組成物は、所望により1種またはそれより多くのさらなる医薬活性ポリペプチドおよび/または化合物を含んでいてもよい。かかる製剤は、例えば、静脈内注入に好適な形態であり得る。
本発明のT細胞は、がん細胞などの標的細胞を殺すことができるものであればよい。標的細胞は、抗原発現の特定パターン、例えば抗原A AND 抗原Bの発現;抗原A OR 抗原Bの発現;または抗原A AND NOT 抗原Bの発現またはこれらのゲートの複合的反復により認識可能であり得る。
本発明の原理を証明する目的で、抗CD19および抗CD33に基づく受容体を任意選択した。レトロウイルスベクターを用いて、CD19およびCD33をクローン化した。これらのタンパク質の先端を切除した結果、それらはシグナルを発さず、長期間にわたって安定して発現させることができた。次に、これらのベクターを用いて、SupT1細胞系に対し単独に、または二重に形質導入を行い、両抗原に陰性の細胞(野生型)、どちらか一方に陽性の細胞および両方に陽性の細胞を確立した。発現データを図3に示す。
ORゲートを構築するため、CD19およびCD33を認識する1対の受容体を共発現させた。異なるスペーサーを用いることにより、交差対合を防止した。両受容体とも、表面安定性を改善するためのCD28由来の膜貫通ドメインおよび単純な活性化シグナルを提供するためのCD3−ゼータのものから誘導されたエンドドメインを有していた。かくして、1対の独立した第一世代CARを共発現させた。配列を共発現させるのに使用されたレトロウイルスベクターカセットは、口蹄疫2A(foot−and−mouth 2A)自己切断性ペプチドを利用することにより、両受容体の1:1共発現を可能にした。カセット設計を図4に、タンパク質構造を図5に示す。相同性領域のヌクレオチド配列をコドンゆらぎ状態としてレトロウイルスベクター逆転写の間の組換えを防止した。
両CARの発現を、Fcに融合させた同種抗原で染色することによりT細胞表面上で試験した。異なる種(CD19にはマウスおよびCD33にはウサギ)のFcドメインを用いることにより、両CARの共発現を、異なるフルオロフォアでコンジュゲートした異なる2次抗体で染色することにより細胞表面上で決定した。これを図6に示す。
ANDゲートでは、単純な活性化受容体を、基本的には活性を阻害するが、一旦受容体がライゲーションされると、その阻害が止められてしまう受容体と組み合わせる。これは、短い/嵩高くないCD8ストークスペーサーおよびCD3−ゼータエンドドメインをもつ標準的第一世代CARを、そのエンドドメインがCD148エンドドメインまたはCD45エンドドメインのいずれかを含む嵩高いFcスペーサーをもつ第2の受容体と組み合わせることにより達成された。両受容体がライゲーションされたとき、スペーサー寸法の差異により、異なる膜区画において異なる受容体が単離され、CD3−ゼータ受容体をCD148またはCD45エンドドメインによる阻害から解放させる。かくして、一旦両受容体が活性化されると、活性化のみが起こる。CD148およびCD45は、本来この様式で機能するため、それらをこのために選択した:例えば、非常に嵩高いCD45エクトドメインは、免疫学的シナプスから受容体全体を排除する。発現カセットを図8に、後続のタンパク質を図9に示す。
観察結果がCD19/CD33および使用されていたそれらのバインダーのいくらかの特異的な特徴の発現ではなかったことを確証するため、ここでは、活性化(ITAM)シグナルがCD19ではなくCD33の認識の際に伝達され、また阻害性(CD148)シグナルがCD33ではなくCD19の認識の際に伝達されるように、2つの標的化scFvを交換した。CD45エンドドメインおよびCD148エンドドメインは、機能的に類似していると考えられるため、CD148エンドドメインを伴うANDゲートに実験を制限した。これにより、当然機能的ANDゲートが依然としてもたらされる。新しい論理ゲートを発現するT細胞を、CD19またはCD33のいずれか一方または両方を担持する標的でチャレンジした。T細胞は、CD19およびCD33の両方を発現する標的に応答したが、これらの抗原の一方のみを発現するかまたはどちらも発現しない標的には応答しなかった。これは、ANDゲートが依然としてこのフォーマットで機能性であることを示す(図18B)。
この目的は、異なるスペーサーにより誘発される特異的分離が、これらの論理CARゲートを生成する能力の背後の中心機構であることを証明することであった。このモデルは、活性化CARのみがライゲーションされれば、強力な阻害性「ライゲーション・オフ」型CARが膜において溶解状態であり、活性化CARを阻害することができるということである。一旦両CARがライゲーションされると、両CARスペーサーが十分に異なれば、それらはシナプス内で分離し、共局在することはない。このため、重要な必要条件は、スペーサーが十分に異なることである。このモデルが正しければ、両スペーサーが、両受容体がライゲーションされたとき、共局在するほど十分に類似していれば、ゲートは、機能し得ない。これを試験するため、元のCARにおける「嵩高い」Fcスペーサーを、本発明者らはネズミCD8スペーサーで置き換えた。これがヒトCD8と類似した長さ、かさおよび電荷を有するが、当然それと交差対合するものではないことが予測された。このため、新しいゲートは、CD19を認識し、ヒトCD8ストークスペーサーおよび活性化エンドドメインを有する第1のCAR;同時に、CD33を認識し、マウスCD8ストークスペーサーおよびCD148エンドドメインを有する第2のCARを有した(図18C)。T細胞に対し、この新しいCARゲートを発現するように形質導入した。次いで、これらのT細胞を、CD19単独、CD33単独またはCD19およびCD33の両方を発現するSupT1細胞でチャレンジした。T細胞は、元のANDゲートのとおりにいずれかの抗原のみを発現するSupT1細胞には応答しなかった。しかしながら、CAR T細胞は、両抗原を発現するSupT1細胞には応答し得なかったことから、このモデルが確認された(図18C)。機能的ANDゲートは、両CARが免疫学的シナプス内で共局在することがないようにそれらが十分に異なるスペーサーを有することを要求する(図23AおよびB)。
CD45およびCD148などのホスファターゼは、非常に強力であるため、免疫学的シナプスに少量入っただけでも、ITAM活性化を阻害することができる。これは、論理ANDゲートの阻害の基礎である。他のクラスのホスファターゼ、例えばPTPN6および関連ホスファターゼは、それほど強力ではない。少量のPTPN6が拡散によりシナプスに入っても活性化を阻害することはないと予測された。加えて、阻害性CARが活性化CARと十分に類似したスペーサーを有するならば、両CARがライゲーションされると、シナプス内で共局在することができると予測された。この場合、大量の阻害性エンドドメインであれば、両抗原が存在するときITAMが活性化するのを止めるのに十分である。かくして、AND NOTゲートを作製することができた。
AND NOTゲートのモデルは、両CARで使用されるスペーサーの性質が、ゲートの正確な機能にとって重要であるという事実を軸として展開する。PTPN6を伴う機能的AND NOTゲートでは、両CARスペーサーは十分に類似しており、両CARがライゲーションされたとき、両方がシナプス内で共局在し、高濃度であるため弱いPTPN6でさえ活性化を阻害するのに十分である。スペーサーが異なったならば、シナプスにおける分離は、活性化を可能にするITAMからPTPN6を離すことになり、AND NOTゲートを破壊する。これを試験するため、ネズミCD8ストークスペーサーをFcのそれで置き換えて対照を生成した。この場合、試験ゲートは2つのCARで構成された。第1のCARは、ヒトCD8ストークスペーサーおよびITAMエンドドメインを有してCD19を認識し;一方、第2のCARは、FcスペーサーおよびPTPN6からのホスファターゼを含むエンドドメインを有してCD33を認識する。このゲートは、CD19に応答して活性化するが、同じくCD19およびCD33の両方に応答して活性化する(図22B、この図では、このゲートの機能を、元のAND NOTゲートおよび実施例6に記載の対照ANDゲート変異型の機能と比較する)。この実験データは、PTPN6を伴う機能的AND NOTゲートの場合、共局在するスペーサーが必要とされるモデルを証明している。
PTPN6に基づくAND NOTゲートと同様に、ITIMに基づくゲートもまた、AND NOTゲートとして機能するために免疫学的シナプスにおける共局在を要求する。この仮説を証明するため、対照ITIMに基づくゲートを以下の通り生成した:2つのCARを共発現させた−第1のCARは、ヒトCD8ストークスペーサーおよび活性化エンドドメインを有してCD19を認識し;FcスペーサーおよびLAIR1のものに由来するITIM含有エンドドメインを有する抗CD33 CARと共発現させる。このゲートの活性を、元のITIMに基づくAND NOTゲートの活性と比較した。この場合、改変されたゲートは、CD19を発現する標的に応答して活性化したが、同じくCD19およびCD33の両方を発現する細胞に応答して活性化した。これらのデータは、ITIMに基づくAND NOTゲートが動力学的分離に基づくモデルに従うこと、および機能ゲートを作製するためには正確なスペーサーが選択されなければならないことを示す(図23B)。
現時点での動力学的分離モデルについての理解および本明細書記載の実験データに基づき、2−CARゲートについてのモデルの概要を図24に提示する。この図は、それぞれ異なる抗原を認識する、2つのCARを発現する細胞を示す。一方または両方のCARが細胞上の標的抗原を認識すると、シナプスが形成され、もともとあったCD45およびCD148は、それらのエクトドメインがかさ高い故にシナプスから排除される。これがT細胞活性化のためのステージを設定する。標的細胞が唯一の同種抗原をもつ場合、同種CARがライゲーションされ、同種CARはシナプスへと分離する。ライゲーションされていないCARはT細胞膜上で溶解状態のままであるため、シナプスの内外へと拡散することができ、その結果、低濃度のライゲーションされていないCARを伴う高局所濃度のライゲーションされたCARの領域が形成する。この場合、ライゲーションされたCARがITAMを有し、ライゲーションされていないCARが、「ライゲーション・オフ」型阻害性エンドドメイン、例えばCD148のそれを有するならば、ライゲーションされていないCARの量は、活性化を阻害するのに十分であり、ゲートはオフである。対照的に、この場合、ライゲーションされたCARがITAMを有し、ライゲーションされていないCARが、PTPN6などの「ライゲーション・オン」型阻害性エンドドメインを有するならば、ライゲーションされていないCARの量は、阻害するには不十分であり、ゲートはオンである。両同種抗原をもつ標的細胞によりチャレンジされると、両同種CARがライゲーションされ、免疫学的シナプスの一部を形成する。重要なことに、CARスペーサーが十分に類似しているならば、両CARはシナプスに共局在するが、CARスペーサーが十分に異なるならば、両CARはシナプス内で分離する。この後者の場合、一方のCARは高濃度で存在するが、他方のCARは存在しない膜の領域が形成される。この場合、分離は完全であるため、阻害性エンドドメインが「ライゲーション・オフ」型であるとしても、ゲートはオンである。前者の場合、両CARが高濃度で共に混ざった膜の領域が形成される。この場合、両エンドドメインが濃縮されるため、阻害性エンドドメインが「ライゲーション・オン」型であるとしても、ゲートはオフである。スペーサーおよびエンドドメインの正しい組み合わせを選択することにより、論理をCAR T細胞にプログラム化することができる。
T細胞を生成するため、(1)各CARが、CARが機能するように単に抗原接近およびシナプス形成を可能にするスペーサーを有し、そして(2)各CARが活性化エンドドメインを有するように、抗Aおよび抗BのCARを生成しなければならない。「抗原A AND NOT B」ゲートCAR T細胞を生成するため、(1)両CARが、交差対合することはないが、標的細胞上で両同種抗原を認識したとき、CARを共分離させるようにするスペーサーを有し、そして(2)一方のCARが活性化エンドドメインを有し、同時に、他方のCARが弱いホスファターゼ(例えば、PTPN6)を含むかまたはリクルートするエンドドメインを有するように、抗Aおよび抗BのCARを生成しなければならない。(3)「抗原A AND 抗原B」ゲートCAR T細胞を生成するため、(1)両CARが標的細胞上で両同種抗原を認識したとき、両CARが共分離することのないように、一方のCARが他方のCARとは十分に異なるスペーサーを有し、(2)一方のCARが活性化エンドドメインを有し、他方のCARが、強力なホスファターゼ(例えば、CD45またはCD148のもの)を含むエンドドメインを有するように、抗Aおよび抗BのCARを生成しなければならない。所望の効果を達成するための正確なスペーサーは、既知サイズ/形状などをもつスペーサーのセットから、ならびに標的抗原のサイズ/形状など(例えば図30参照)および標的抗原上の同種エピトープの場所を考慮に入れて選択され得る。
APRILの天然形態は、分泌II型タンパク質である。CARのためのBCMA結合ドメインとしてのAPRILの使用は、このII型分泌タンパク質のI型膜結合タンパク質への変換を必要とし、このタンパク質が安定しており、この形態でBCMAへの結合性を保持するためにも必要である。候補分子を生成するため、APRILのアミノ末端先端部を欠失させて、プロテオグリカンへの結合性を除去した。次に、シグナルペプチドを付加することにより、新生タンパク質を小胞体、およびそれゆえ細胞表面へと向かわせた。また、使用されるスペーサーの性質は、CARの機能を変更することができるため、3つの異なるスペーサードメインを試験した:(i)Fc結合モチーフを除去するように改変されたヒトIgG1スペーサー;(ii)CD8ストーク;および(iii)IgG1ヒンジ単独を含む、APRILに基づくCARが生成された(図25におけるカートゥーンおよび図26におけるアミノ酸配列)。これらのCARは、マーカータンパク質−先端切除CD34が好都合なマーカー遺伝子として共発現され得るようにバイシストロニックレトロウイルスベクターで発現された(図27A)。
この研究の目的は、構築されたAPRILに基づくCARが細胞表面で発現されるかどうか、およびAPRILが折り畳まれて天然タンパク質を形成するかどうかを試験することであった。T細胞を、これらの異なるCAR構築物で形質導入し、マーカー遺伝子についての染色と一緒に、市販されている抗APRIL mAbを用いて染色し、フローサイトメトリーにより分析した。この実験の結果を、APRIL結合性がマーカー遺伝子の蛍光に対してプロットしている図27Bに示す。これらのデータは、このフォーマットで、APRILに基づくCARが細胞表面で発現され、APRILが抗APRIL mAbにより認識されるほど十分に折り畳まれていることを示す。
正常ドナーからのT細胞を、種々のAPRIL CARで形質導入し、野生型、またはBCMAおよびTACIを発現するように操作されたSupT1細胞のどちらかに対して試験した。幾つかの異なるアッセイを使用して、機能を決定した。古典的クロム放出アッセイを実施した。ここでは、標的細胞(SupT1細胞)を、51Crで標識し、異なる比率でエフェクター(形質導入T細胞)と混合した。共培養上清中の51Crを数えることにより、標的細胞の溶解を決定した(図28Aは、累積データを示す)。
PBMCを血液から単離し、PHAおよびIL−2を用いて刺激した。2日後、細胞を、レトロネクチン被覆プレート上でCD19:CD33 ANDゲート構築物を含むレトロウイルスにより形質導入した。5日目、ANDゲート構築物により翻訳された2つのCARの発現レベルを、フローサイトメトリーにより評価し、細胞からCD56+細胞(主としてNK細胞)を枯渇させた。6日目、PBMCを、1:2のエフェクター対標的細胞比で標的細胞との共培養物に入れた。8日目、上清を集め、IFN−ガンマ分泌についてELISAにより分析した(図29)。
