Description

キットは、
(i)構成的に活性なプロモーターの制御下にある第1の核酸配列、および
(ii)それが発現される細胞の分化/消耗状態、またはそれが発現される細胞の微小環境における環境代謝物の存在
のいずれかに応じて、選択的に活性なプロモーターの制御下にある第2の核酸配列
を含み得る。
(i)核酸配列が発現される細胞のある特定の分化/消耗状態において転写物分解を引き起こす、特異的miRNA標的配列を含む第1の核酸配列、および
(ii)特異的miRNA標的配列を欠く第2の核酸配列
を含み得る。
(i)構成的に活性なプロモーターの制御下にある第1の核酸配列、および

マイクロRNA(miRNA)は、小型の非コードRNA分子(約22ヌクレオチドを含む)であり、遺伝子発現のRNAサイレンシングおよび転写後制御において機能する。miRNAは、mRNA分子における相補的配列による塩基対形成を介して機能する。結果として、このようなmRNA分子は、次のプロセスのうちの1つまたは複数によりサイレンシングされる:(i)mRNA鎖の2つの断片への切断、(ii)ポリ(A)尾部の短縮によるmRNAの不安定化、およびリボソームによるmRNAのタンパク質へのより効率的ではない翻訳。

環境毒素に対する細胞応答は、大部分は、アリール炭化水素受容体(AHR)の活性化により媒介される。AHR活性化は、毒素がPAS(Per−Arnt−Sim)ドメインに結合した後に発生する。これにより、構造変化が始まって細胞シャペロンの放出が生じ、これによりARNT転写因子との二量体形成が可能となる。生じるAHR/ARNTヘテロ二量体が特異的DNA配列(XRE−生体異物認識エレメント)に結合すると、細胞傷害に対する応答に必要とされる遺伝子の上方制御が生じる(図6)。

本発明は、キメラ抗原受容体(CAR)および選択的に発現されるNOIを含む細胞を提供する。

膜貫通ドメインは、膜を横断する、古典的CARの配列である。これは、疎水性アルファヘリックスを含み得る。膜貫通ドメインは、CD28に由来し、良好な受容体安定性をもたらし得る。

国際公開第2016/193696号では、免疫細胞活性化後にシグナル伝達経路を調節する、様々な融合タンパク質および切断型タンパク質について記載している。
(i)リン酸化免疫受容体活性化チロシンモチーフ(ITAM)に結合するタンパク質由来のSH2ドメインを含むが、キナーゼドメインを欠く、切断型タンパク質、

代替の実施形態では、作用物質は、ホスファターゼ「減弱因子」であり、CARまたはTCRエンドドメインの脱リン酸化を引き起こし、ある特定の転写状態における活性化に対する閾値を上昇させ得る。

作用物質は、サイトカインまたはケモカインであり得る。例えば、IL12、flexiIL12、GM−CSF、IL7、IL15、IL21、IL2およびCCL19から選択される。特に、作用物質は、IL−12であり得る。
細胞接着分子(CAM)は、細胞接着において他の細胞または細胞外基質(ECM)との結合に関与する細胞表面上に位置するタンパク質である。
本発明の作用物質は、CARまたはTCRを発現する細胞の活性を調節する転写因子であってもよいし、またはそれを含んでいてもよい。
NOIは、標的細胞、例えば、腫瘍細胞の活性を調節する作用物質をコードし得る。

AP1/SRE/STAT3/STAT5応答性プロモーターをレポーター遺伝子eGFPのコード配列の上流にクローニングした、自己不活型レトロウイルスベクターを構築した。この第1のオープンリーディングフレームの後には、PGKプロモーターおよびRQR8細胞表面マーカーをコードする第2のコード配列を配置する。正常ドナー由来の初代ヒトT細胞に、レトロウイルスベクターを形質導入し、3μg/mLのPHAおよび50U/mLのIL−2で72時間刺激するか、または刺激せずに放置した。細胞のメモリー表現型について、フローサイトメトリーにより解析した。結果を図9に示す。種々のメモリーコンパートメント(ナイーブ、セントラルメモリー、エフェクターメモリーおよびエフェクター)は、PHAおよびIL−2による刺激後に形質導入した種々のT細胞間で異ならない。
CREB応答性プロモーターをレポーター遺伝子eGFPのコード配列の上流にクローニングした、自己不活型レトロウイルスベクターを構築した。この第1のオープンリーディングフレームの後には、PGKプロモーターおよびRQR8細胞表面マーカーをコードする第2のコード配列を配置する。正常ドナー由来の初代ヒトT細胞に、レトロウイルスベクターを形質導入し、PHAで24時間刺激するか、または刺激せずに放置した。細胞のメモリー表現型およびeGFP発現について、フローサイトメトリーにより解析した。結果を図11および12に示す。CREB応答性プロモーターは、エフェクターメモリーおよびエフェクター細胞サブセットにおいてeGFPの上方制御を誘導した。
自己不活型レトロウイルスベクターを構築し、これにより、CD4+T細胞に特異的な初期プロモーターを第1のCARのコード配列の上流にクローニングする。この第1のCARは、fmc63由来の抗CD19scFv、CD8スペーサーおよびCD28−CD3Zエンドドメインを使用して構築する。PGKプロモーターを、この第1のコード配列から下流にクローニングする。第2のCARをコードする第2のコード配列を、PGKプロモーターから下流にクローニングする。この第2のCARは、hd37由来の抗CD19scFv、CD8スペーサーおよび41BB−CD3Zエンドドメインを使用して構築する。このレトロウイルスカセットにより、hd37/41BB−CD3Z CARの発現をすべての細胞において生じるはずであるが、加えて、fmc63/CD28−CD3Z CARが、CD4+T細胞において選択的に発現されるはずである。T細胞にレトロウイルスベクターを形質導入する。正常ドナー由来の初代ヒトT細胞に、このレトロウイルスベクターを形質導入する。2つのCARの示差的発現を、フローサイトメトリーにより決定する。CD19に対する2つの異なるscFvの使用によって、2つの異なる抗イディオタイプ抗体を使用したフローサイトメトリーによる、独立した発現の検証が可能となる。このようなT細胞の性能は、41BB−CD3ZまたはCD28−ZのいずれかのCARを発現する単純なベクターを形質導入したT細胞に対して、in vitroでの共培養において、およびNALM6のNSGマウスへの異種移植モデルにおいても比較する。

キヌレニンは、アミノ酸トリプトファンから酵素IDOの作用により合成される免疫抑制代謝物である。腫瘍細胞により発現されるIDOは、固形腫瘍の微小環境において高レベルのキヌレニンをもたらすことが多く、次いで、このキヌレニンは、高度に免疫抑制性の環境を生成し、これにより、腫瘍反応性CAR T細胞の機能が阻害されて腫瘍拒絶が妨げられる。CAR T細胞がキヌレニンの存在に反応可能な機構を、望ましい導入遺伝子を発現させることによりデザインすると、このようなT細胞が、キヌレニンにより媒介される免疫抑制を克服することが可能となる。

- 前記阻害性分子が、1つまたは複数のITAM結合ドメインを含むが、キナーゼドメインを欠く、切断型ZAP70を含む、請求項43に記載の核酸構築物。
- 前記第1の核酸配列が、CD28共刺激性ドメインを含むCARまたはCAR成分をコードし、前記第2の核酸配列が、OX40または41BB共刺激性ドメインを含むCARまたはCAR成分をコードし、ここで前記核酸構築物がT細胞において発現されると、第1のCARまたはCAR成分が構成的に発現されるが、前記細胞がエフェクターメモリーまたはエフェクター状態にあると、第2のCARまたはCAR成分が選択的に発現される、請求項38に記載の核酸構築物。
- 前記第1の核酸配列が、キメラ抗原受容体(CAR)、CAR成分または改変T細胞受容体(TCR)をコードし、前記第2の核酸配列が、サイトカインをコードし、ここで前記核酸構築物がT細胞において発現されると、前記CAR、CAR成分またはTCRが構成的に発現されるが、前記T細胞の微小環境における環境代謝物の存在下で前記サイトカインが選択的に発現される、請求項38に記載の核酸構築物。
- 請求項28から31のいずれかに記載の核酸配列、請求項32から36のいずれかに記載の核酸配列のキット、または請求項37から46のいずれかに記載の核酸構築物を含む、ベクター。
- 請求項1から25のいずれかに記載の細胞を生成するための方法であって、請求項28から31のいずれかに記載の核酸配列、請求項32から36のいずれかに記載の核酸配列のキット、請求項37から46のいずれかに記載の核酸構築物、または請求項47に記載のベクターを細胞に導入するステップを含む、方法。
- 前記細胞が、被験体から単離した試料に由来する、請求項48に記載の方法。
- 請求項1から25のいずれかに記載の複数の細胞を含む医薬組成物。
- 疾患の処置および/または予防における使用のための、請求項50に記載の医薬組成物。
- 請求項50に記載の医薬組成物を被験体に投与するステップを含む、疾患を処置および/または予防するための方法。
- 次のステップ:
(i)細胞を含む試料の単離、
(ii)請求項28から31のいずれかに記載の核酸配列、請求項32から36のいずれかに記載の核酸配列のキット、請求項37から46のいずれかに記載の核酸構築物、または請求項47に記載のベクターの前記細胞への形質導入またはトランスフェクション、および
(iii)(ii)の細胞の被験体への投与
を含む、請求項52に記載の方法。 - 疾患の処置および/または予防のための医薬の製造における、請求項50に記載の医薬組成物の使用。
- 前記疾患が、がんである、請求項51に記載の使用のための医薬組成物、請求項52もしくは53に記載の方法、または請求項54に記載の使用。
