Abstract
Description
ステップ(1)において、動物の軟骨をダイシングして軟骨基質を作製する。
ステップ(2)において、ステップ(1)の軟骨基質をアルカリ処理する。
ステップ(3)において、ステップ(2)のアルカリ処理された軟骨基質を消毒処理する。
ステップ(4)において、ステップ(3)の消毒された軟骨基質を脱細胞化処理して無細胞軟骨グラフトを作製する。
ステップ(1)において、動物の軟骨をダイシングして複数の軟骨基質を作製する。
ステップ(2)において、ステップ(1)の軟骨基質をアルカリ処理する。
ステップ(3)において、ステップ(2)のアルカリ処理された軟骨基質を消毒処理する。
ステップ(4)において、ステップ(3)の消毒された軟骨基質を脱細胞化処理して無細胞軟骨グラフトを作製する。

Claims (15)
- 以下のステップ(1)〜(4)を含む無細胞軟骨グラフトの調製方法であって、
ステップ(1)において、動物の軟骨をダイシングして、複数の軟骨基質を作製し、
ステップ(2)において、ステップ(1)の軟骨基質をアルカリ処理し、
ステップ(3)において、ステップ(2)のアルカリ処理された軟骨基質を消毒処理し、
ステップ(4)において、ステップ(3)の消毒された軟骨基質を脱細胞化処理して、前記無細胞軟骨グラフトを作製する、無細胞軟骨グラフトの調製方法。 - 前記軟骨は、ブタから得られた鼻軟骨、肋軟骨、耳軟骨、又は膝軟骨である請求項1に記載の方法。
- 各軟骨グラフトは、円盤状、ストリップ状、円錐状、又は粒子状である請求項2に記載の方法。
- 前記ステップ(2)のアルカリ処理は、水酸化ナトリウム溶液で前記軟骨基質を室温で約3-24時間処理することを含む請求項1に記載の方法。
- 前記ステップ(3)の消毒処理は、消毒剤で前記軟骨基質を処理することを含み、前記消毒剤は、アルコール、酸化剤、放射線、フェノール化合物、及び第四級アンモニウム化合物からなる群より選択される請求項1に記載の方法。
- 前記アルコールは、エタノール又はイソプロパノールである請求項5に記載の方法。
- 前記酸化剤は、過酸化水素である請求項5に記載の方法。
- 前記フェノール化合物は、パラベン、安息香酸、又はサリチル酸である請求項5に記載の方法。
- 前記第四級アンモニウム化合物は、塩化ベンザルコニウム、塩化アルキルジメチルベンジルアンモニウム、ジデシルジメチルアンモニウムクロライド、塩化アンモニウムである請求項5に記載の方法。
- 前記ステップ(4)の脱細胞化処理は、超臨界流体(SCF)で前記ステップ(3)の消毒された軟骨基質を約150-350barの圧力、30-50℃の温度で約100分間処理することを含む請求項1に記載の方法。
- 前記SCFは、超臨界二酸化炭素(scCO2)、超臨界亜酸化窒素 (scN2O)、超臨界水(scH2O)、超臨界アルカン、超臨界アルケン、超臨界アルコール又は超臨界アセトンのいずれかである請求項10に記載の方法。
- 前記SCFはscCO2であり、前記温度は約37℃であり、前記圧力は約350barである請求項11に記載の方法。
- 前記無細胞軟骨グラフトの各々は、天然軟骨よりも少なくとも2倍の応力に耐えることができる請求項12に記載の方法。
- 骨軟骨疾患に罹患している被験体を治療するための方法であって、請求項1〜13のいずれか一項に記載の方法によって作製された無細胞軟骨グラフトを前記被験体の病変部位に適用することを含む方法。
- 前記骨軟骨疾患は、先天性欠損、骨折、半月板損傷若しくは欠損、骨/脊柱の変形、骨肉腫、骨髄腫、骨異形成症と脊柱側弯症、骨粗鬆症、歯周病、歯骨損失、骨軟化症、くる病、線維性骨炎、腎性骨ジストロフィー、脊椎固定、脊椎円板の再構築若しくは除去、骨ページェット病、半月板損傷、関節リウマチ、変形性関節症、又は外傷性若しくは外科的な軟骨損傷である請求項14に記載の方法。
