Claims (7)
- 配列番号1のアミノ酸配列で構成されたヘキスロン酸C4−エピメラーゼ(Hexuronate C4−epimerase)のN−末端から125番セリン(S)のアミノ酸残基、267番バリン(V)のアミノ酸残基、268番セリン(S)のアミノ酸残基、306番トリプトファン(W)のアミノ酸残基と386番アルギニン(R)のアミノ酸残基が変異され、185番セリン(S)のアミノ酸残基、272番スレオニン(T)のアミノ酸残基、および403番チロシン(Y)のアミノ酸残基が変異されていないヘキスロン酸C4−エピメラーゼ変異体。
- 請求項1に記載のヘキスロン酸C4−エピメラーゼ変異体をコードする核酸。
- 請求項2に記載の核酸を含む形質転換体。
- 請求項1に記載のヘキスロン酸C4−エピメラーゼ変異体を含む、タガトース生産用の組成物。
- 請求項1に記載のヘキスロン酸C4−エピメラーゼ変異体、請求項3に記載の形質転換体、または請求項4に記載のD−タガトース生産用の組成物とD−フルクトース(D−fructose)を反応させて前記D−フルクトースをエピマー化(epimerization)する工程を備える、D−タガトースの製造方法。
- 前記エピマー化する工程は、金属塩が存在する条件で行われることを特徴とする、請求項5に記載のD−タガトースの製造方法。
- 前記エピマー化する工程の前に、砂糖を酵素で加水分解してD−フルクトースを得る工程またはブドウ糖を酵素で異性化する工程をさらに備えることを特徴とする、請求項5に記載のD−タガトースの製造方法。
