Abstract
Description
本発明の他の目的は、癌組織、癌細胞または癌患者のテロメラーゼの活性抑制を介して癌の予防または治療方法を提供するところにある。
(a)正常の場合、赤血球より小さい大きさ(50〜200nm)の原形ないし楕円形で、運動性がある;
(b)核酸含有;
(c)免疫化学蛍光染色時ミトコンドリアと類似の反応を示す;
(d)フュージョン(fusion)および/またはフィジョン(fission)生態様式を示す;
(e)フュージョンがない場合には大きさが300nmまで成熟して、DNA含有類似ミトコンドリアに成熟して、SEMまたはTEM電子顕微鏡写真上でミトコンドリアと類似する構造を示す;
(f)フュージョンが発生する場合には数千nmまで大きくなる;
(g)癌患者由来の場合、正常由来より大きさ(長径500nm以上)が大きく、形態が不均一な変異ルテリアル発生;および
(h)エキソソームとは異なる光反応を示す。
追加的に、次の特性中一つ以上を有することを特徴とする:
(i)自家蛍光(autofluorescence)を示す;
(j)200〜400nm大きさでATPを産生する;
(k)付着性がある;
(l)二重膜構造;
(m)特定条件で変異ルテリアルはburstingされ、bursting以後にはstemnessを有する;
(n)p53遺伝子およびテロメア調節機能を有する;および
(o)CD39またはCD73の表面抗原を有する。
従って、第二観点において、本発明は、テリオンを有効成分として含有する癌の予防または治療用医薬組成物に関する。
(a)植物に溶媒を添加して、50〜90℃で空気または酸素を利用して間欠的にバブリングしながら振盪する段階;
(b)前記振盪によって気化される水蒸気またはガスを捕集した後、冷却させて凝縮液を収得する段階;
(c)0.8〜1.2μmの空隙を有するフィルターを利用して前記収得された凝縮液をフィルタリングする段階;
(d)前記フィルタリングされた凝縮液を遠心分離する段階;および
(e)前記遠心分離された上澄み液から植物由来ルテリオンを分離する段階;
本発明で使用されるルテリオンは、ルテリオンに水分を添加してIR光線照射下に18〜30℃で培養して使用することができる。
本発明のルテリオン組成物は、任意の適した手段によってヒトまたは動物対象に投与され得る。
また、第四観点において、本発明は、ルテリオンを有効成分として含有する組成物を投与することを特徴とする癌の予防または治療方法に関する。
第五観点において、本発明は、ルテリオンを有効成分として含有する組成物を投与することを特徴とする癌組織、癌細胞または癌患者のテロメラーゼ活性抑制方法に関する。
第六観点において、本発明は、ルテリオンを有効成分として含有する組成物を投与することを特徴とする癌細胞の増殖抑制方法に関する。
第七観点において、本発明は、ルテリオンを有効成分として含有する組成物を癌の予防または治療に用いる用途に関する。
第八観点において、本発明は、ルテリオンを有効成分として含有する組成物を癌組織、癌細胞または癌患者のテロメラーゼ活性抑制に用いる用途に関する。
第九観点において、本発明は、ルテリオンを有効成分として含有する坑癌組成物を製造する用途に関する。
薬用植物である漆(Rhus verniciflua stokes)100gを20−30倍の大きさ、すなわち2−3リットル容器の大きさに合わせて切断して容器に入れて、植物の5〜8倍である500〜800g(好ましくは植物の6倍、すなわち600g)の蒸溜水を容器に投入した後、80℃以下で全湯を行った。約8時間程度全湯をするが、約3時間毎に20〜30分間酸素でバブルリングを加えて植物のルテリオンがかたまらないようにした。前記バブリング後抽出時スチームされる気化した水蒸気をフラスコに収集した。前記収集した水蒸気を冷却して凝縮させて収得した凝縮液にIR光線(波長:3〜1000um内外、好ましくは200〜600um)を1−2時間照射してルテリオンが変異されないようにした。その後、0.8μm以上の径の空隙を備えるフィルターを利用して前記凝縮液をフィルタリングして、フィルターを通過した凝縮液だけ1200〜5000rpmで5〜10分間繰り返し遠心分離した。前記遠心分離を介して収得した上澄み液にはIR光線を照射して運動性を有して集まるルテリオン粒子をピペットを利用して分離して、前記分離したルテリオンを50nm径の空隙を備えるフィルターを通過させてフィルタリングされなかった部分だけPBSで洗浄して漆由来ルテリオンを収得した。前記過程によって長径50−800nmのルテリオンを収得することができ、これは暗視野顕微鏡または共焦点顕微鏡によって観察確認が可能であった。前記収得したルテリオンは大きさに応じて50−200nm(発生期)/200−400nm(成熟期)/400−600nm(分裂期)/600−800nm(過分裂期)に区分した。
同様の方法で表1〜4に記載された薬用植物からルテリオンを収得した。
正常細胞(Fibroblast)および癌細胞(NCl−H1975、MDA−MA−468、WiDr)1×106細胞/mlをそれぞれ60mmプレートに接種した後、約12時間後に実施例1で分離されたルテリオンを50μg/mlの濃度で処理して培養した。細胞培養は、PSF(antibiotic−antimycotic)抗生剤がない条件で、1% FBS DMEM培地を使用して37℃、5%CO2培養器で行った。細胞接種48時間後に収穫してテロメラーゼ活性を測定した。
種々のヒト癌細胞株(肺癌(NCI−H1975)、大腸癌(WiDr)、乳癌(MDA−MB−486)、肝臓癌(HCC38)、白血病(AGH−77))および正常細胞株(fibroblast)でルテリオンの癌細胞に対する細胞毒性を測定するためにMTT分析を実施した。

- ルテリオンを有効成分として含有する癌細胞のテロメラーゼ活性抑制用組成物。
