Abstract
一种抗疲劳、提高免疫力的复方中药口服液及其制备方法,属于中药复方制剂技术领域。是由以下药物按重量份数制备而成,玛咖20~30g,迷迭香5~10g,何首乌10~30g,龟甲10~30g,猴头菇5~10g,女贞子5~15g,五味子10~30g,三七15~35g,金钱草20~30g,山药5~15g,秋葵20~30g,萝藦叶15~35g。本发明所述制备方法,利用超声波辅助萃取及酶法制备方法,实现有效物质的最优化提取,制备方法合理,成品质量稳定,口感适宜。通过动物实验及临床实验,进一步证实本发明安全高效,无副作用,应用范围广。适用于体倦无力、阴虚血少、神衰气虚、阴阳失调、肝肾亏损、气滞湿阻、头晕目眩、元气不足等症状,标本兼治,效果显著。

技术领域
本发明属于中药复方制剂技术领域,具体涉及一种抗疲劳、提高免疫力的复方中药口服液及其制备方法。
背景技术
免疫力是指机体抵抗外来病毒侵袭、维持机体内环境稳定性的能力。现代医学科学证明,提高机体免疫力,有利于预防疾病,减缓衰老,提高记忆力,增强体力,改善机体亚健康状态。而近年来,随着人们生活质量的提高和中药草本的开发,人们越来越意识到增强体质、缓解疲劳、提高免疫力和适应力对健康的重要性,抗疲劳、提高机体免疫力的药品也因此受到了更多广泛的关注。
目前,市场上销售的抗疲劳、提高机体免疫力的药品种类繁多,药物作用效果参差不齐。因此亟待研发纯天然无毒、具有独特功效及协同作用的药品。
为了有效提高机体免疫力,从根本上改善人体脏器内部环境,本发明利用草本中药的天然活性及药物之间的科学配伍,效果显著,且无毒副作用,对体倦无力、阴虚血少、神衰气虚、阴阳失调、肝肾亏损、气滞湿阻、头晕目眩、元气不足等症状均有治疗功效。
发明内容
本发明的目的是提供一种抗疲劳、提高免疫力的复方中药口服液,利用中药草本的天然活性及药物之间的科学配伍,标本共治,效果显著,无毒副作用,有效的解决当前抗疲劳、提高免疫力药品的不足。
本发明进一步提供了一种抗疲劳、提高免疫力的复方中药口服液的制备方法,利用超声波辅助萃取及酶法制备方法,实现有效物质的最优化提取,制备方法合理,成品质量稳定,口感适宜。
通过动物实验及临床实验,进一步证实本发明安全高效,无副作用,应用范围广。适用于体倦无力、阴虚血少、神衰气虚、阴阳失调、肝肾亏损、气滞湿阻、头晕目眩、元气不足等症状,标本兼治,效果显著。
本发明所述的一种抗疲劳、提高免疫力的复方中药口服液,其特征在于,是由以下药物按重量份数制备而成:
玛咖20~30g,迷迭香5~10g,何首乌10~30g,龟甲10~30g,猴头菇5~10g,女贞子5~15g,五味子10~30g,三七15~35g,金钱草20~30g,山药5~15g,秋葵20~30g,萝藦叶15~35g。
本发明所述的一种抗疲劳、提高免疫力的复方中药口服液,其优选配方为:
玛咖25g,迷迭香8g,何首乌20g,龟甲20g,猴头菇8g,女贞子10g,五味子20g,三七20g,金钱草25g,山药10g,秋葵25g,萝藦叶20g。
本发明所述的一种抗疲劳、提高免疫力的复方中药口服液的制备方法,其步骤如下:
(1)取20~30g玛咖、5~10g迷迭香、5~15g女贞子分别粉碎成100~300目粗粉后混合,置于反应容器中,加入粗粉3~8倍质量的质量分数40~60%的乙醇水溶液浸泡35~45分钟,加热回流提取,每次回流提取时间为1~2小时,再将反应容器于50~70℃、250W条件下超声提取20~40分钟后过滤,滤渣回收并依次经浸泡、回流提取、超声提取、过滤2~4次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.02~1.10清膏,作为提取液A;
(2)取10~30g五味子、15~35g萝藦叶、15~35g三七分别粉碎成100~300目粗粉后混合,置于反应容器中,加入粗粉3~8倍质量的质量分数50~70%的乙醇水溶液浸泡35~45分钟,加热回流提取,每次回流提取时间为1~2小时,将反应容器于50~70℃、250W条件下超声提取20~40分钟后过滤,滤渣回收并依次经浸泡、回流提取、超声提取、过滤2~4次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.02~1.10清膏,作为提取液B;
(3)取20~30g秋葵鲜果洗净、干燥、粉碎成100~300目粗粉,与5~10g、100~300目猴头菇子实体干粉混合,置于反应容器中,加入混合物3~8倍质量的水浸泡30~50分钟,再加入1.8IU/g纤维素酶和4IU/g果胶酶,在50~70℃水浴、pH=5.0的条件下酶解1~2小时,然后温度升至80~85℃灭酶,并继续保温浸提1~2小时,过滤;将滤渣加水浸泡、酶解、灭酶、浸提共1~2次后弃去,合并过滤液,室温下静置24~48h后取上清液作为提取液C;
(4)取10~30g何首乌、10~30g龟甲、20~30g金钱草、5~15g山药分别粉碎成100~300目粗粉后混合,置于反应容器中,加入粗粉3~8倍质量的水浸泡50~70分钟,加热微沸煎煮提取1~2小时,过滤;将滤渣加水浸泡、煎煮提取共2~4次后弃去,合并过滤液,室温下静置24~48h后取上清液作为提取液D;
(5)将提取液A~D混合,加水稀释,搅拌至溶解,于室温静置24~48小时,过滤3~5次,滤液加入矫味剂(100~120g蜂蜜)和防腐剂(8~12g质量比为1:1的羟苯甲酯和羟苯丙酯),搅拌溶解混合均匀后于高压蒸汽锅中灭菌,分装后得到本发明所述的口服液。
选项应符合《2010版中国药典》液体制剂项下。
【规格】:每支装10mL
【用法用量】:口服。一次10mL(1支),一日三次。
【适应症】:适用于体倦无力、阴虚血少、神衰气虚、阴阳失调、肝肾亏损、气滞湿阻、头晕目眩、元气不足等症状,标本兼治,效果显著。

为使本发明更加容易理解,下面结合具体实施例,进一步阐述本发明。这些实施例只用于说明本发明而不用于限制本发明的范围。
实施例1:
处方:玛咖20g,迷迭香5g,何首乌10g,龟甲10g,猴头菇5g,女贞子5g,五味子10g,三七15g,金钱草20g,山药5g,秋葵20g,萝藦叶15g。
取玛咖20g、迷迭香5g、女贞子5g于药用粉碎机中分别粉碎成200目粗粉后混合共30g,置于反应器中,加入粗粉5倍质量的质量分数50%乙醇水溶液浸泡40分钟,加热回流提取,每次提取2小时,将反应容器于60℃、250W超声仪中提取30分钟后过滤,滤渣回收并依次浸泡、回流提取、超声提取、过滤共3次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.05清膏,作为提取液A;
取五味子10g、萝藦叶15g、三七15g于药用粉碎机中分别粉碎成200目粗粉后混合共40g,置于反应器中,加入粗粉5倍质量的质量分数70%乙醇水溶液浸泡40分钟,加热回流提取,每次提取2小时,将反应容器于60℃、250W超声仪中提取30分钟后过滤,滤渣回收并依次浸泡、回流提取、超声提取,过滤共3次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.05清膏,作为提取液B;
取秋葵20g鲜果洗净、干燥、粉碎成200目粗粉,与5g、200目猴头菇子实体干粉混合共25g,加入粗粉5倍质量的水浸泡40分钟,加入1.8IU/g纤维素酶和4IU/g果胶酶(45IU的纤维素酶和100IU的果胶酶),在50℃水浴、PH=5.0的条件下酶解2小时,然后温度升至80℃灭酶,并继续保温浸提2小时,过滤;将滤渣加水浸泡、酶解、灭酶、提取共2次后弃去,合并过滤液,室温下静置24h后,取上清液,作为提取液C;
取何首乌10g、龟甲10g、金钱草20g、山药5g于药用粉碎机分别粉碎成200目粗粉后混合共45g,加入5倍质量水浸泡60分钟,加热微沸煎煮2小时,过滤;将滤渣加水浸泡、煎煮提取共3次后弃去,合并过滤液,室温下静置24h后,取上清液,作为提取液D。
所有提取液(A-D)混合,加水稀释至2000mL,搅拌至溶解,于室温静置24小时后,过滤3次,滤液加入矫味剂(110g蜂蜜),防腐剂(10g质量比为1:1的羟苯甲酯和羟苯丙酯),搅拌溶解混合均匀后,于高压蒸汽锅中灭菌,最后分装入口服液瓶中(10mL/支)。此后口服液试剂作为实验组进行动物实验,以评价该复方中药口服液的抗疲劳、提高免疫力作用。
实施例2:
处方:玛咖25g,迷迭香8g,何首乌20g,龟甲20g,猴头菇8g,女贞子10g,五味子20g,三七20g,金钱草25g,山药10g,秋葵25g,萝藦叶20g
取玛咖25g、迷迭香8g、女贞子10g于药用粉碎机中分别粉碎成200目粗粉后混合共43g,置于反应器中,加入粗粉5倍质量的质量分数50%乙醇水溶液浸泡40分钟,加热回流提取,每次提取2小时,将反应容器于60℃、250W超声仪中提取30分钟后过滤,滤渣回收并依次浸泡、回流提取、超声提取、过滤共3次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.05清膏,作为提取液A;
取五味子20g、萝藦叶20g、三七20g于药用粉碎机中分别粉碎成200目粗粉后混合共60g,置于反应器中,加入粗粉5倍质量的质量分数70%乙醇水溶液浸泡40分钟,加热回流提取,每次提取2小时,将反应容器于60℃、250W超声仪中提取30分钟后过滤,滤渣回收并依次浸泡、回流提取、超声提取,过滤共3次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.05清膏,作为提取液B;
取秋葵25g鲜果洗净、干燥、粉碎成200目粗粉,与8g、200目猴头菇子实体干粉混合共33g,加入粗粉5倍质量的水浸泡40分钟,加入1.8IU/g纤维素酶和4IU/g果胶酶(59.4IU的纤维素酶和132IU的果胶酶),在50℃水浴、PH=5.0的条件下酶解2小时,然后温度升至80℃灭酶,并继续保温浸提2小时,过滤;将滤渣加水浸泡、酶解、灭酶、提取共2次后弃去,合并过滤液,室温下静置24h后,取上清液,作为提取液C;
取何首乌20g、龟甲20g、金钱草25g、山药10g于药用粉碎机分别粉碎成200目粗粉后混合共75g,加入5倍质量水浸泡60分钟,加热微沸煎煮2小时,过滤;将滤渣加水浸泡、煎煮提取共3次后弃去,合并过滤液,室温下静置24h后,取上清液,作为提取液D。
所有提取液(A-D)混合,加水稀释至2000mL,搅拌至溶解,于室温静置24小时后,过滤3次,滤液加入矫味剂(110g蜂蜜),防腐剂(10g质量比为1:1的羟苯甲酯和羟苯丙酯),搅拌溶解混合均匀后,于高压蒸汽锅中灭菌,最后分装入口服液瓶中(10mL/支)。此后口服液试剂作为实验组进行动物实验,以评价该复方中药口服液的抗疲劳、提高免疫力作用。
实施例3:
处方:玛咖30g,迷迭香10g,何首乌30g,龟甲30g,猴头菇10g,女贞子15g,五味子30g,三七35g,金钱草30g,山药15g,秋葵30g,萝藦叶35g
取玛咖30g、迷迭香10g、女贞子15g于药用粉碎机中分别粉碎成200目粗粉后混合共55g,置于反应器中,加入粗粉5倍质量的质量分数50%乙醇水溶液浸泡40分钟,加热回流提取,每次提取2小时,将反应容器于60℃、250W超声仪中提取30分钟后过滤,滤渣回收并依次浸泡、回流提取、超声提取、过滤共3次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.05清膏,作为提取液A;
取五味子30g、萝藦叶35g、三七35g于药用粉碎机中分别粉碎成200目粗粉后混合共100g,置于反应器中,加入粗粉5倍质量的质量分数70%乙醇水溶液浸泡40分钟,加热回流提取,每次提取2小时,将反应容器于60℃、250W超声仪中提取30分钟后过滤,滤渣回收并依次浸泡、回流提取、超声提取,过滤共3次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.05清膏,作为提取液B;
取秋葵30g鲜果洗净、干燥、粉碎成200目粗粉,与10g、200目猴头菇子实体干粉混合共40g,加入粗粉5倍质量的水浸泡40分钟,加入1.8IU/g纤维素酶和4IU/g果胶酶(72IU的纤维素酶和160IU的果胶酶),在50℃水浴、PH=5.0的条件下酶解2小时,然后温度升至80℃灭酶,并继续保温浸提2小时,过滤;将滤渣加水浸泡、酶解、灭酶、提取共2次后弃去,合并过滤液,室温下静置24h后,取上清液,作为提取液C;
取何首乌30g、龟甲30g、金钱草30g、山药15g于药用粉碎机分别粉碎成200目粗粉后混合共105g,加入5倍质量水浸泡60分钟,加热微沸煎煮2小时,过滤;将滤渣加水浸泡、煎煮提取共3次后弃去,合并过滤液,室温下静置24h后,取上清液,作为提取液D。
所有提取液(A-D)混合,加水稀释至2000mL,搅拌至溶解,于室温静置24小时后,过滤3次,滤液加入矫味剂(110g蜂蜜),防腐剂(10g质量比为1:1的羟苯甲酯和羟苯丙酯),搅拌溶解混合均匀后,于高压蒸汽锅中灭菌,最后分装入口服液瓶中(10mL/支)。此后口服液试剂作为实验组进行动物实验,以评价该复方中药口服液的抗疲劳、提高免疫力作用。
实验例:
1.以下为本发明对小鼠免疫功能进行试验:
1.1实验准备
实验动物由吉林大学实验中心提供的健康昆明种雌性小鼠共160只,体重18-22g,待小鼠适应环境后随机分为四组,每组40只。对照组给予生理盐水10mL/kg,实验组给予不同实验例药物25mL/kg,用药灌胃给予药物,1d/2次,共30天后进行实验。
1.2实验方法
1.2.1负重游泳时间测定:对照组给予生理盐水,实验组给予不同实验例药物。每组随机选取10只小鼠,小鼠给药30天,末次给药结束后,小鼠尾部负重相当于体重5%的铅片后,置于游泳箱中,水温(27±3)℃,水深35cm,记录小鼠开始游
泳至死亡时的时间作为小鼠游泳时间,结果如表1所示。

本实验结果说明,比对照组相比,本处方实验例均可以显著提高负重小鼠在水中的游泳时间,且P<0.05,说明本处方口服液具有显著的抗疲劳,提升体力的功效。
1.2.2肝糖原含量测定;对照组给予生理盐水,实验组给予不同实验例药物。每组随机选取10只小鼠,小鼠给药30天,末次给药结束后,将小鼠放入水温(27±3)℃的游泳箱中游泳90分钟后,立即拔眼球取血,取肝脏用蒽酮法测定肝糖原含量,结果如表2所示。

本实验结果说明,比对照组相比,本处方实验例均可以显著提高小鼠肝糖原含量,说明本处方口服液具有增加小鼠糖原储备或减少糖原储备的作用。
1.2.3小鼠免疫器官重量测定:对照组给予生理盐水,实验组给予不同实验例药物。每组随机选取10只小鼠,小鼠给药30天,末次给药结束后,取小鼠器官,称重,结果如表3所示。

本实验结果说明,比对照组相比,本处方实验例均显著提升小鼠免疫器官重量,且P<0.05,说明本处方口服液均能显著提高免疫力,肠道消化吸收系统的功效。
1.2.4ConA诱导小鼠淋巴细胞转化实验:对照组给予生理盐水,实验组给予不同实验例药物。每组随机选取10只小鼠,小鼠给药30天,末次给药结束后,取小鼠脾脏,制成单细胞悬液,将所得细胞悬液以3*106个/孔(1mL)的密度加入到24孔板上,每一组设置2孔,其中一孔中加入75μL ConA液(刀豆蛋白A),另一孔加入同等量的生理盐水,于37℃,CO2培养箱中培养。24小时后更换新鲜RPMI1640培养基,并加入50μL MTT/孔,继续培养4小时。培养结束后,加入1mL异丙醇,待结晶溶解完全后,分装到96孔板上,每孔设置3个对照,于酶标仪上570nm测定光密度,实验结果如表4所示。

本实验结果说明,本处方三种实验例的吸光度值均与对照值存在显著性差异(P<0.01),而实验例之间吸光度值均不存在显著性差异,说明本处方口服液具有显著的抗病毒,提高免疫力的功效。
1.2.5血乳酸含量测定:对照组给予生理盐水,实验组给予不同实验例药物。每组随机选取10只小鼠,小鼠给药30天,末次给药结束后30分钟,对小鼠进行眼眶采血20μL,加入到5mL离心管中(事先用生理盐水配置1%肝素钠的潤洗),再加入1.5mL蛋白沉淀剂混匀,于4℃冰箱静止15分钟,取出后离心1000rpm/分钟,5分钟,取上清液,使用全血乳酸(LD)测试试剂盒测定乳酸含量。动物采血后不负重,将小鼠放入水温(27±3)℃的游泳箱中游泳15分钟后,立即采血20μL,休息30分钟后,再采血20μL,使用上述方法测定乳酸含量,结果如表5所示。

本实验结果说明,比对照组相比,本处方实验例小鼠在游泳后乳酸积累量显著降低,同时在休息后,小鼠的乳酸快速代谢,说明本处方口服液具有显著提高体力,抗疲劳的功效。
以下为本发明对临床试验的进一步说明:
使用本发明药物治疗易疲劳、免疫力低下者共150例,其中男性88例,女性62例,涉及年龄段为25-66岁。根据病情,每次服用口服液1瓶,每日三次,早、中、晚口服,一个月为一个疗程,连续服用一到两个疗程,并观察治疗效果。在服药期间不服用与其功效相同的药物。
评定标准:
全部缓解:脸色红润,精力旺盛,四肢有力,体力、免疫力提升,抗病毒能力增强,头疼、疲惫、健忘等症状消失。
部分缓解:精神和脸色都所有好转,四肢渐渐有力,体力、免疫力也有所恢复,头疼、疲惫、健忘等症状逐渐消失。
不缓解:症状无任何改善

经动物试验和临床验证,该抗疲劳、提高人体免疫力复方中药口服制剂,具有延缓老化、调经益血、滋补五脏、温补肾阳、速解疲劳、恢复体力,抗寒冷、抗病毒、增加机体免疫力等功效,适用于体倦无力、阴虚血少、神衰气虚、阴阳失调、肝肾亏损、气滞湿阻、头晕目眩、元气不足等症状,标本兼治,效果显著。此外,本发明利用超声波辅助萃取及酶法制备方法,实现有效物质的最优化提取,制备方法合理,成品质量稳定,口感适宜。
Claims (4)
1.一种抗疲劳、提高免疫力的复方中药口服液的制备方法,其步骤如下:
(1)取20~30g玛咖、5~10g迷迭香、5~15g女贞子分别粉碎成100~300目粗粉后混合,置于反应容器中,加入粗粉3~8倍质量的质量分数40~60%的乙醇水溶液浸泡35~45分钟,加热回流提取,每次回流提取时间为1~2小时,再将反应容器于50~70℃、250W条件下超声提取20~40分钟后过滤,滤渣回收并依次经浸泡、回流提取、超声提取、过滤2~4次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.02~1.10清膏,作为提取液A;
(2)取10~30g五味子、15~35g萝藦叶、15~35g三七分别粉碎成100~300目粗粉后混合,置于反应容器中,加入粗粉3~8倍质量的质量分数50~70%的乙醇水溶液浸泡35~45分钟,加热回流提取,每次回流提取时间为1~2小时,将反应容器于50~70℃、250W条件下超声提取20~40分钟后过滤,滤渣回收并依次经浸泡、回流提取、超声提取、过滤2~4次后弃去,最终滤液合并后减压回收乙醇并浓缩至相对密度为1.02~1.10清膏,作为提取液B;
(3)取20~30g秋葵鲜果洗净、干燥、粉碎成100~300目粗粉,与5~10g、100~300目猴头菇子实体干粉混合,置于反应容器中,加入混合物3~8倍质量的水浸泡30~50分钟,再加入1.8IU/g纤维素酶和4IU/g果胶酶,在50~70℃水浴、pH=5.0的条件下酶解1~2小时,然后温度升至80~85℃灭酶,并继续保温浸提1~2小时,过滤;将滤渣加水浸泡、酶解、灭酶、浸提共1~2次后弃去,合并过滤液,室温下静置24~48h后取上清液作为提取液C;
(4)取10~30g何首乌、10~30g龟甲、20~30g金钱草、5~15g山药分别粉碎成100~300目粗粉后混合,置于反应容器中,加入粗粉3~8倍质量的水浸泡50~70分钟,加热至微沸煎煮提取1~2小时,过滤;将滤渣加水浸泡、煎煮提取共2~4次后弃去,合并过滤液,室温下静置24~48h后取上清液作为提取液D;
(5)将提取液A~D混合,加水稀释,搅拌至溶解,于室温静置24~48小时,过滤3~5次,滤液加入100~120g矫味剂和8~12g防腐剂,搅拌溶解混合均匀后于高压蒸汽锅中灭菌,分装后得到复方中药口服液。
2.如权利要求1所述的一种抗疲劳、提高免疫力的复方中药口服液的制备方法,其特征在于:矫味剂为蜂蜜,防腐剂为质量比为1:1的羟苯甲酯和羟苯丙酯。
3.一种抗疲劳、提高免疫力的复方中药口服液,其特征在于:是由权利要求1或2所述方法制备得到。
4.如权利要求3所述的一种抗疲劳、提高免疫力的复方中药口服液,其特征在于:配方为玛咖25g,迷迭香8g,何首乌20g,龟甲20g,猴头菇8g,女贞子10g,五味子20g,三七20g,金钱草25g,山药10g,秋葵25g,萝藦叶20g。
