Abstract

発明の分野
分子生物学および医薬の分野における本発明は、光受容体変性網膜において内網膜神経細胞を光感受性細胞へ変換し、それによって視覚認知および視覚の種々の局面を回復する、ライト・ゲートの(light−gated)陽イオン選択性膜チャネル、チャネルロドプシン−2(Chop2)のような、微生物型のロドプシンの使用に関する。

(b)視覚的に局所の物体を検出し、かつ訓練なしよりも有効にとりわけこの物体を動かすためにこの被験体を訓練することによって特徴付けられる方向付けおよび移動度の訓練、
のうちの1つ以上によって達成される。

本発明は、ヒト、他の哺乳動物または他の動物被験体での眼の障害を処置するための方法を提供する。詳細には、眼の障害は、網膜色素上皮細胞または光受容体細胞における変異遺伝子または遺伝子欠損を含む障害である。本発明の方法は、眼の細胞における遺伝子の産生の発現を指向するプロモーター配列に作動可能に連結されるか、またはそのプロモーター配列の制御下にある眼の細胞に特異的な正常遺伝子をコードする核酸配列を担持する、組み換えウイルスの有効量の硝子体内または網膜下注射によるこの被験体への投与の工程を包含し、そして変異された遺伝子または欠失された遺伝子の発現の欠失または不正確な発現を置き換える。
本発明の方法が考慮され、かつ視覚の1つ以上のパラメーターを改善するために用いられ得る眼科の障害としては、限定はしないが、眼の前部および後部の両方に影響する発達性の異常が挙げられる。前眼部障害としては、緑内障、白内障、角膜ジストロフィー、円錐角膜が挙げられる。後眼部障害としては、光受容体の機能不全によって生じる盲目障害、および/または網膜のジストロフィーおよび変性によって生じる死が挙げられる。網膜の障害としては、先天性定常夜盲症、加齢性黄斑変性症、先天性錐体ジストロフィー、および大グループの網膜色素変性症(RP)−関連障害が挙げられる。これらの障害としては、種々の年齢で生じる網膜における光受容体細胞、桿体および錐体の遺伝子的に死滅しやすい傾向が挙げられる。とりわけ、重篤な網膜症、例えば、年齢とともに進行し、小児期および青年期早期に盲目を生じるRP自体のサブタイプ、ならびにRP関連の疾患、例えば、LCAの遺伝的サブタイプ(人生の初年度程度の早期に小児期に視覚喪失を高頻度に生じる)である。後者の障害は、光受容体細胞、桿体および錐体の重篤な減少、およびしばしば完全な喪失によって遺伝的に特徴付けられる。(Trabulsi,EI,編,Genetic Diseases of the Eye,Oxford University Press,NY,1998)。
神経節細胞またはON型双極細胞に標的される。Dr.J.G.Flanneryのグループからの研究は、AAV−2ウイルスベクターを用いることによってON型網膜神経節細胞においてGFPを標的する、ヒトギャップ接合性タンパク質(コネキシン−36)プロモーターのフラグメントを特定した(Greenberg KPら、2007,In vivo Transgene Expression in ON−Type Retinal Ganglion Cells:Applications to Retinal Disease.ARVO abstract,2007)。mGluR6プロモーターの容易にパッケージ可能なバージョン(<2.5kb)によって、ON型双極細胞(桿体双極細胞およびON型錐体双極細胞の両方)に対するChR2の標的化が可能になる。

アデノ随伴ウイルスは、小型の一本鎖DNAウイルスであって、効率的な複製のためにヘルパーウイルスを要する(Berns,KI,Parvoviridae:the viruses and their replication,p.1007〜1041(第2巻),Fields,BNら、Fundamental Virology,第3版,(Lippincott−Raven Publishers,Philadelphia(1995))。AAVの4.7kbのゲノムは、2つの逆方向末端反復(ITR)、ならびにそれぞれRepタンパク質およびCapタンパク質をコードする2つのオープンリーディングフレーム(ORF)を有する。Repリーディングフレームは、分子量78、68、52および40kDaの4つのタンパク質をコードする。これらのタンパク質は主に、AAV複製を調節すること、ならびにAAVのレスキューおよび宿主細胞染色体への組み込みに機能する。Capのリーディングフレームは、分子量85(VP1)、72(VP2)および61(VP3)kDaの3つの構造的タンパク質をコードし、これらのタンパク質がビリオンカプシドを形成する(Berns,前出)。VP3は、総AAVビリオンタンパク質の80%より多くを含む。

本発明のミニ遺伝子または導入遺伝子は、コード配列またはORFに対して作動可能に連結された適切な配列を含み、標的された宿主細胞でその発現を促進する。「作動可能に連結された(operably linked)」配列としては、コード配列と接触しているプロモーターのような発現制御配列、およびポリペプチド産物の発現をトランスでまたは遠位で制御するように作用する発現制御配列の両方が挙げられる。
本発明に用いられるrAAVは、本明細書に記載される材料および方法、ならびに当該分野で周知の材料および方法を用いて構築および生成され得る。本発明の任意の構築物を生成するために好ましい方法は、構成的および誘導性の遺伝子操作、組み換え操作、および合成的技術、例えば、上記で引用される引用文献に示されるものである。
Chop2導入遺伝子および上記されるような標的眼細胞における使用のための細胞特異的プロモーターを含むrAAVは、従来の方法を用いて混入について評価され、そして例えば、網膜下注射による投与のための滅菌または無菌の薬学的組成物中に処方されるべきである。
本発明の種々のパラメーターを評価するためのインビトロおよびインビボの両方の研究を用いてもよく、これには、失明するヒトの眼科障害の認識された動物モデルを含む。ヒト網膜症、例えば、小児失明の大きい動物モデルが有用である。本明細書に提供される実施例によって、この方法が網膜疾患の範囲を処置するのに同様に用いられ得るということを当業者は容易に予想できるようになる。

従って「視覚(vision)」としては、光の存在を単に検出する能力が挙げられる。これは、本発明に従って処置されている罹患した被験体を訓練して光を検出する可能性を開き、これによって、個体は彼の環境から離れて応答することが可能になり、ただし相互作用はそのままであり得る。一例では、シグナルのアレイを生成して、これに対して視力低下したヒトが応答し得、これによってこの人は、離れて電気的方法によって連絡するか、または毎日のタスクを行う能力が強化される。さらにこのような人の移動は、「光検出」から生じる光のこのような新たな検知を用いるように訓練された場合劇的に増強する。光知覚が完全に欠損すれば、その人は、その人から離れたなんらかの物体について見分ける方法(聴覚および臭いは別として)がなくなる。

方法
Chop2−GFPキメラは、発現されたタンパク質がChop2領域のC末端に対して連結されたGFPを有するように、チャネルオプシン−2(Cho2)の活性フラグメント(配列番号3)をコードする核酸(配列番号2)に対して、緑色蛍光タンパク質(GFP)をコードする核酸(下に示されるとおり配列番号1の一部)を連結することによって作成した。これらの配列の両方とも、上記で考察されるような「導入遺伝子(transgene)」を構成する。Chop2−GFPのDNAを、CMVプロモーターの制御下でHEK293細胞中にトランスフェクトした。
明るいGFP蛍光は、トランスフェクション後にHEK細胞中で18〜24時間内に検出した。その蛍光は、原形質膜に優先的に位置した。Chop2−GFPキメラの機能の保存は、パッチクランプ記録によって確認した。かなりの光ゲート電流がまた、内因性のオール・トランスレチナールの添加なしにChop2−GFP−発現HEK細胞で観察され、このことは、多数の機能的なChop2−GFPチャネルが、発色団について内因性の前駆体のみを用いてHEK細胞で形成されたということを示している。注射の3〜4ヵ月後、GFP蛍光が、注射された眼の網膜神経細胞で観察された。明るいGFP蛍光が多くの神経節細胞および水平細胞、いくつかのアマクリン細胞で、そして時には双極細胞で、注射後少なくとも1つの10週間にわたって観察された。Chop2−GFP−発現網膜神経細胞は、光刺激に応答して強固な膜脱分極を示し、そしてオール・トランスレチナールの外因性の供給源は必要としなかった。
進行性のインビトロおよびインビボの結果によって、Chop2の異所性発現が、「盲目の(blind)」網膜に対する光感受性の回復についての治療ストラテジーであることが示される。直接光ゲートされた膜チャネルChop2の機能的な発現を、インビボでラットの網膜ニューロンで実証した。従って、第二または第三次の網膜ニューロンにおける光ゲートの膜チャネルの発現は、光受容体生成後の光知覚の回復のための有用なストラテジーである。
材料および方法
DNAおよびウイルスベクター構築物
ChoP2のN末端フラグメント(Met1−Lys315)をコードするDNAフラグメントを(Nagelら、2003)、Chop2コードフラグメントの3’末端でインフレームに挿入された、ポリアデニル化シグナル(mP1)およびGFPを含むマウスプロタミン1遺伝子の最後のエキソンを含むpBluescriptベクター(Stratagene)中にクローニングして、Chop2−GFP融合タンパク質を生成した。Chop2−GFPキメラの機能は、トランスフェクトされたHEK293細胞中で確認した。

C.reinhardtiiの全長Chop2タンパク質をコードする天然の核酸配列(GenBank Accession #AF461397)は、以下のヌクレオチド配列を有する(配列番号4)。このコード配列は、ヌクレオチド28で始まるATGコドンで開始することに注目のこと。
配列番号4のコード部分は、737個のアミノ酸のポリペプチドをコードする737個の三つ組コドン(プラス終止コドン)として組織化された配列番号5として下に示される。ATG開始コドンおよびTAA終止コドンは強調される。
本発明に有用な別の有用なChop2配列は、310アミノ酸のポリペプチド(全長のネイティブなChoP2の生物学的に活性なフラグメント)をコードする933ヌクレオチド(終止コドンを含む)の核酸であり、Chlamydomonas reinhardtii由来の合成構築物である(EF474017およびZhangら,2007,Nature(印刷中)を参照のこと)。この配列は、ヒト発現についてコドン最適化される。下に示されるnt配列は配列番号7であり、示されるコードされたアミノ酸配列は、配列番号8である。アミノ酸配列である配列番号8を有するポリペプチドは、C末端で短縮され、310でProがAsnを置き換えている配列番号6のフラグメントである。
AAVベクター注射
全ての動物実験は、施設のレベルで行い、そして実験動物の管理と使用についてのNIHのガイド(NIH Guide for Care and Use of Laboratory Animals)に従った。
動物をベクター注射後、種々の時点で屠殺した。Chop2−GFP蛍光の発現を、平坦なホール・マウントの網膜、垂直の網膜の、そして冠状の脳切片で検査した。切開した網膜および脳を、PBS中の4%パラホルムアルデヒドを用いて、それぞれ0.5〜2時間室温で、そして24時間4℃で固定した。この固定された網膜(3%アガロースに包埋)および脳を、ビブラトームを用いることによって切断した。この網膜および脳の切片または網膜のホール・マウントを、スライド上にマウントして、Vectashield培地(Vector Laboratories)でカバーした。GFP蛍光は、それぞれ、465〜495nm、505nm、および515〜555nmの励起フィルター、干渉フィルターおよび放射フィルターを装備した蛍光顕微鏡下で可視化し、そしてほとんどの画像はデジタルカメラで得た(Axiocam,Zeiss)。いくつかの画像を共焦点顕微鏡(TCS SP2,Leica)で得た。半薄垂直網膜切片の光学顕微鏡のために、眼を除核して、PBS中でリンスして、1%の四酸化オスミウム、2.5%のグルタルアルデヒドおよび0.2MのSorensonのリン酸緩衝液(pH7.4)中で、4℃で3時間固定した。次いで眼を段階的エタノールで脱水して、プラスチックに包埋して、1μmの切片中に切断して、メチレンブルー/アズール混合物で染色した。
解離した網膜細胞および網膜スライスを、以前に記載のとおり調製した(Pan,2000およびCuiら、2003)。ホール・セルの配置中でパッチ電極での記録を、EPC−9増幅器およびPULSEソフトウェア(Heka Electronik,Lambrecht,Germany)によって行った。記録は、以下を(mMとして)含むハンクス溶液中で行った:NaCl、138;NaHCO3,1;Na2HPO4、0.3;KCl、5;KH2PO4、0.3;CaCl2,1.25;MgSO4,0.5;MgCl2,0.5;HEPES−NaOH,5;グルコース,22.2;フェノールレッドを、0.001%(v/v)で;pHを7.2に、0.3NのNaOHを用いて調節した。
多重電極アレイ記録は、TianおよびCopenhagen(2003)によって報告された手順に基づいた。要するに、網膜を解剖して、ニトロセルロース濾紙ストリップ(Millipore Corp.,Bedford,MA)上に光受容体を横倒しに置いた。マウントした網膜を、200μm離れた30μmの直径の電極のMEA−60多重電極アレイ記録チャンバに入れ(Multi Channel System MCS GmbH,Reutlingen,Germany)、ここでは神経節細胞層が記録電極に面している。網膜は、全ての実験の間34℃で酸素化された細胞外溶液中で連続してパルスした。この細胞外溶液は、以下を(mMとして)含んだ:NaCl、124;KCl、2.5;CaCl2、2;MgCl2、2;NaH2PO4、1.25;NaHCO3,26;およびグルコース,22(95%のO2および5%のCO2でpH7.35)。記録は通常、網膜を記録チャンバに入れた後60分で開始した。各々の光刺激のオンセットの間の間隔は10〜15秒であった。シグナルは、200Hz(低カットオフ)と20kHz(高カットオフ)との間でフィルタリングした。個体の神経細胞由来の応答は、Offline Sorterソフトウェア(Plexon,Inc.,Dallas,TX)を用いて分析した。
視覚誘発性電位の記録は、野生型マウスのC57BL/6および129/Sv株の4〜6か月齢で、そしてrdl/rdlマウスで6〜11ヵ月齢で行った。記録は、ウイルスベクター注射後2〜6か月で行った。
解離した細胞および網膜のスライス記録のために、光刺激を、150Wのキセノンランプベースの走査単色光分光器によって、10nm(TILL Photonics,Germany)のバンド幅で生成して、光ファイバーで顕微鏡に伝えた。多重電極アレイ記録のために、光応答は、単色光分光器または175Wのキセノンランプベースのイルミネーター(Lambda LS,Sutter Instrument)(400〜580nmの帯域通過フィルターを備える)によって誘発させ、液体光ガイダー(liquid
light guider)を通して記録チャンバの底に投射した。視覚誘発電位のために、光刺激を単色光分光器によって生成して、光ファイバーを通して眼に投射した。光の強度は、中性密度フィルターによって減弱させた。光エネルギーは、薄型センサー(TQ82017)および光強度測定器(Model:TQ8210)(Advantest,Tokyo,Japan)によって測定した。
インビボにおける網膜神経細胞でのChop2の発現
Chop2タンパク質の発現および局在化を直接可視化するために、Chop2チャネルのC末端部分を、GFPで置き換えて、Chop2−GFPキメラを作成した。アデノ随伴ウイルス(AAV)ベクターを選択して、網膜神経細胞中へのChop2−GFP融合タンパク質の発現を標的した。この理由はAAVベクターが、内網膜神経細胞を含む非分裂細胞へ導入遺伝子を送達し(Harvey ら、,2002 and Martinら、2003)、そして宿主ゲノムへ導入遺伝子を組み込む能力(Flotte,2004)である。
ChR2発現内網膜神経細胞の光誘発性電流の特性
Chop2チャネルの機能的な特性を、ホール・セルパッチクランプ記録を用いることによって内網膜神経細胞で検査した。これらの記録は、光受容体媒介性の光応答が確実に排除されるように、急性分離した細胞で行った。Chop2−GFP陽性細胞は、GFP蛍光によって特定した(図2A)。Chop2発色団、オール・トランスレチナールの前駆体は、添加されなかった。なぜなら、それは細胞中で普遍的に存在し得るからである(Kimら、1992、ならびにThompsonおよびGal,2003)。光誘発性の応答は、全ての記録されたGFP蛍光細胞で観察され(n=34)、このことは、機能的なChR2(発色団が結合したChop2)は、細胞中に既に存在する網膜発色団を有する網膜神経細胞で形成され得る。結果として、機能的なChR2 チャネルの発現はまた、外因性網膜発色団の供給なしに、培養された海馬神経細胞で近年報告されている(Boydenら、2005;ただしLiら、2005を参照のこと)。
光受容体欠損のrd1/rd1マウスにおけるChop2の発現
野性型網膜におけるChR2の発現および機能が達成されているが、本発明者らは、ChR2の発現が、光受容体変性後の網膜における光応答を回復し得るか否かに取り組んだ。この目的を達するために、この実験は、ヒトでの網膜色素変性症のいくつかの形態と同様である(McLaughlinら、1993)、サイクリックGMOホスホジエステラーゼPDE6中でヌル変異を有する光受容体変性モデルである、ホモ接合性のrdl(rdl/rdl)マウスで(Bowesら、1990)で行った。Chop2−GFPのウイルスベクターを、2〜12カ月齢で新生(P1)または成体マウスの眼に硝子体内注射した。野性型動物で観察された結果と同様、明るいGFPシグナルが、Chop2−GFP−注射された網膜で、主に網膜神経節細胞で観察された(図4Aおよび4B)。記録実験の時点で(他に示さない限り4ヶ月以上の年齢)、光受容体細胞は存在しなかった(図4C)。Chop2−GFPの発現は、15月齢のrdl/rdlマウスから図4Aで示された例と同様、rdl/rdlマウスで、16ヶ月までの年齢(ウイルス注射の3〜6カ月後)で観察された。これらの結果によって、この光受容体欠損モデルにおける内網膜神経細胞は、光受容体の完全な死滅後長期生存するだけでなく、Chop2−GFPの安定な発現の能力を保持していることが示される。
rdl/rdlマウスのChR2−発現生存内網膜神経細胞の光誘発性応答
rdl/rdlマウスにおけるChR2−発現網膜神経細胞の光応答特性を、網膜スライスにおけるホール・セル(whole−cell)パッチクランプ記録によって検査した。記録は、神経節細胞層に位置するGFP陽性の細胞から作成した。光誘発性の電流は、GFP陽性の細胞で観察された。電流の大きさは、ここでも光強度に依存性であった(図4Dおよび4Eにおける上のトレース;図4Fに示される光強度および電流の関係も参照のこと)。2群のChR2−発現網膜神経細胞が、それらの応答特性に基づいて観察された:一群の一過性スパイク発射神経細胞(図4D)および一群の徐放性スパイク発射神経細胞(図4E)。膜脱分極および/またはスパイク頻度はまた、光強度に依存した(図4Dおよび4Eにおける下部のトレース)。さらに、光は、より高い強度で、拡大時間スケールで第二のトレース(黒)および第四のトレース(赤)を重ね合わせることによって、図4Dおよび図4Eの右パネルで図示されるとおり電圧応答の動態を顕著に加速した。膜脱分極、スパイク発火頻度、および第一スパイクピークまでの時間に対する光強度の関係を、それぞれ図4G、4Hおよび4Iに示す。これらの結果により、ChR2の発現を伴う生存している網膜第三次神経細胞が、膜脱分極および/または作用電圧発火および応答動態で、光強度をコードし得ることが実証される。
ChR2−媒介性の網膜活性の多重電極アレイ記録
rdl/rdlマウスの光受容体欠損網膜のスパイクコード能力を、ホール・マウント網膜由来の多重電極アレイ記録の使用によってChR2の発現後に検査した。図5Aにおけるサンプル記録から示されるとおり、ライト・オンおよびオフに応答した高速動態でのスパイク発火が、Chop2−GFP−発現網膜で観察された(n=11、網膜)。光誘発性スパイク発火は、CNQX(25〜50μM)プラスAPV(25〜50μM)(n=3)の適用によって影響されず、このことは、この応答が、記録された細胞のChR2に由来することを示している。このような光誘発性のスパイク発火は、GFP単独を担持するウイルスベクターを注射された(n=2網膜)か、または注射されていない(n=3)網膜で観察された。後者によって、光受容体由来の光応答がないことが確認された。光誘発性のスパイク発火は、スラミン(100μM)では影響されず(n=2)、これは、メラノプシン受容体媒介性の光電流をブロックし得ることが報告されている(Melyanら、2005およびQiuら、2005)。
視覚誘発性電位
rdl/rdlマウスの網膜におけるChR2媒介性の光応答が、視覚皮質に伝達されたか否かを試験するために研究を行なった。AAV感染によって達成される網膜神経節細胞で、GFPのような導入遺伝子の発現は、脳でのより高次の視覚中心においてそれらの終わりまで拡大され得ることが報告された(Harveyら、2002)。従って、Chop2−GFP−発現網膜神経節細胞の軸索末端の解剖学的突出を最初に検査した。一貫して、網膜神経節細胞のChop2−GFP標識軸索末端は、外側膝状体腹側核(ventral lateral geniculate nucleus)および外側膝状体背側核(dorsal lateral geniculate nucleus)(図6A)、ならびに上丘(superior colliculus)(図6B)を含む脳のいくつかの領域で観察された。これらの結果によって、変性網膜における網膜神経節細胞の中央突出が維持されることが示される。
実施例1〜7の考察
本明細書に提示される結果によって、ChR2の発現に基づいた光受容体欠損げっ歯類網膜における光応答の回復のストラテジーは、機械的にかつ技術的に実現可能であることが実証された。最も重要な事には、この結果によって、ChR2が、網膜神経細胞における光センサーとしてその使用についてのいくつかの主な基準を満たすことが示された。第一に、融合されたChop2−GFPを担持するAAVベクターの送達によって、本発明者らは、網膜神経細胞がChoP2の長期の発現に耐容する能力を示した。今まで、Chop2−GFPタンパク質の発現は、12ヶ月間の非ジストロフィーラットの神経細胞で、そして光受容体欠損rdl/rdlマウスで6ヶ月間インビボにおいてウイルス注射後に達成されている。従って、本発明の結果によって、網膜神経細胞におけるChR2の発現が、正常な光周期条件下で生体適合性であることが示される。
