インテグリンαvβ3をターゲティングするペプチドを触媒抗体38C2に融合することによって、新規な抗体-MRD融合分子を調製した。軽鎖のN末端およびC末端ならびに重鎖のC末端における融合が最も効果的であった。フローサイトメトリーを用いて、この抗体結合体がインテグリンαvβ3を発現するヒト乳癌細胞に効率的に結合することを示した。抗体結合体はまた、メソドール(methodol)およびドキソルビシンプロドラッグ活性化によって測定されるように、親触媒抗体38C2のレトロアルドール活性も保持していた。このことから、細胞ターゲティングおよび触媒抗体能力を選択的化学療法のために効率的に組み合わせられることが実証される。

非触媒抗体へのMRD融合物の別の例は、Herceptin-MRD融合構築物である。Herceptin-MRD融合物は多機能性であり、低分子αvインテグリンアンタゴニストと化学的にプログラムされたインテグリンをターゲティングする抗体の両方が、αvの媒介による細胞接着および増殖の邪魔をすることによって乳癌転移を防止する際に優れた有効性を示す。Herceptin-2×Con4(ErbB2およびang2をターゲティングする)を含むMRD融合物、ならびにHerceptin-V114(ErbB2およびVEGFターゲティングをターゲティングする)を含むMRD融合物、ならびにHerceptin-RGD-4C-2×Con4(ErbB2、ang2、およびインテグリンターゲティングをターゲティングする)を含むMRD融合物が効果的である。
VEGFをターゲティングするMRDを含む抗体を構築した。v114をターゲティングするMRD(SEQ ID NO: 13)を、長いリンカー配列(SEQ ID NO: 2)を用いて38C2のκ鎖のN末端およびHerceptinに融合させた。得られた抗体-MRD融合構築物の発現および試験により、強いVEGF結合が実証された。
IGF-1RをターゲティングするMRD
を、長いリンカー配列を連結物として用いて38C2のκ鎖のN末端およびHerceptinに融合させた物質を研究した。得られた抗体-MRD融合構築物の発現および試験により、強いIGF-1R結合が実証された。IGR-1Rへの強い結合を示すその他のクローンもまた、数回の変異誘発およびスクリーニングの後に同定した。下記に挙げる好ましい配列は、インスリン受容体に対して有意な親和性も結合親和性も示さない(表2を参照されたい)。

Ang-2をターゲティングするMRD(L17)を、肝細胞増殖因子受容体に結合するMet抗体の軽鎖のN末端またはC末端のいずれかに融合させた。短いリンカー配列または長いリンカー配列のいずれかを連結物として使用した。図6は、短いリンカーペプチド(GGGS(SEQ ID NO: 1))によるAng2をターゲティングするMRDとMet抗体のN末端融合物(L17-sL-Met)、長いリンカーペプチド
によるAng2をターゲティングするMRDとMet抗体のN末端融合物(L17-lL-Met)、および長いリンカーペプチドによるAng2をターゲティングするMRDとMet抗体のC末端融合物(Met-iL-L17)を含む構築物を用いたELISAの結果を示す。得られた抗体-MRD融合構築物の発現および試験により、長いリンカーペプチドを使用した場合の強いAng-2結合が実証された。Ang-2をターゲティングするMRDを抗体の軽鎖C末端に融合した物質の方が、抗体の軽鎖N末端へのAng-2ターゲティングの融合物よりも、Ang-2に対して若干強い結合を示した。
ErbB2に結合する抗体Herceptin(Her)の軽鎖に融合されインテグリンαvβ3をターゲティングするMRD(RGD4C)を含む抗体を構築した。短いリンカー配列、長いリンカー配列、または軽鎖定常領域中の4番目のループのいずれかをリンカーとして使用した。図7は、短いリンカーペプチド(GGGS(SEQ ID NO: 1))によるインテグリンαvβ3をターゲティングするMRDとErbB2抗体のN末端融合物(RGD4C-sL-Her)、短いリンカーペプチドによるインテグリンαvβ3をターゲティングするMRDとErbB2抗体のC末端融合物(Her-sL-RGD4C)、軽鎖定常領域中の4番目のループによるインテグリンαvβ3をターゲティングするMRDとErbB2抗体のC末端融合物(Her-lo-RGD4C)、または長いリンカーペプチド

ErbB2に結合する抗体(Her)の軽鎖に融合されインスリン様増殖因子-I受容体をターゲティングするMRD(RP)を含む抗体を構築した。短いリンカーペプチド、長いリンカーペプチド、または軽鎖定常領域中の4番目のループのいずれかをリンカーとして使用した。(Carter et al., Proc Natl Acad Sci U S A. 1992 May 15;89(10):4285-9)。
VEGFをターゲティングするMRD(V114)を、ErbB2に結合する抗体(Her)の軽鎖のN末端に融合させた。中ぐらいの長さのリンカーペプチド
を連結物として使用した。図10は、中ぐらいの長さのリンカーペプチドによるVEGFをターゲティングするMRDとErbB2に結合する抗体のN末端融合物(V114-mL-Her)を含む構築物を用いたELISAの結果を示す。得られた抗体-MRD融合構築物の発現および試験により、強いVEGF結合およびErbB2結合が実証された。
中ぐらいの長さのリンカーペプチドを連結物として用いた、38C2の軽鎖のN末端へのインテグリンαvβ3をターゲティングするMRD(RGD)の融合を研究した。図11により、得られた抗体-MRD融合構築物の発現および試験により、強いインテグリンαvβ3結合があったことが実証される。
短いリンカー配列を連結物として用いた、38C2の軽鎖のC末端へのAng-2をターゲティングするMRD(L17)の融合を研究した。図12により、得られた抗体-MRD融合構築物の発現および試験により、強いAng-2結合があったことが実証される。
インテグリンαvβ3をターゲティングするMRD(RGD4C)を、中ぐらいの長さのリンカーによって、ErbB2をターゲティングする抗体(Her)の軽鎖のN末端に連結し、Ang-2をターゲティングするMRD(L17)を、短いリンカーによって、ErbB2をターゲティングする同じ抗体のC末端に連結した(RGD4C-mL-Her-sL-L17)。図13により、得られた抗体-MRD融合構築物がインテグリン、Ang-2、およびErbB2に結合したことが実証される。
中ぐらいの長さのリンカーを連結物として用いてErbB2に結合する抗体(Her)の重鎖のN末端に融合されたインテグリンαvβ3をターゲティングするMRD(RGD4C)を含む抗体を構築した(RGD4C-mL-her-重鎖)。図14は、この構築物を用いたELISAの結果を示す。この構築物はインテグリンおよびErbB2の両方に結合した。
