Abstract
Description
本発明は、薬及び免疫学の分野に関する。特に、B型肝炎ウイルス感染及び/又はB型肝炎関連疾患若しくは症状の治療及び/又は予防に使用することができる新規ペプチドに関する。
B型肝炎ウイルス(HBV)による慢性感染は、主要な世界的な健康問題である。HBVは、ヘパドナウイルス科(Hepadnaviridae)ファミリーの原型メンバーであり、肝細胞に感染する強い選好を有する(Ganemら、2004年)。この30年、B型肝炎からの保護のための有効な予防ワクチンが利用可能であったにもかかわらず、推定20億人がそれでもなおHBVに感染しており、2億4千万人超が現在慢性(長期)B型肝炎に感染しており、地理的には西ヨーロッパ及び北アメリカ以外の地域で優性である(世界保健機構、2013年7月)。
本発明者らは、有効な治療ワクチン接種に使用されるHBV抗原の選択を同定した。HBVタンパク質由来ペプチドのHLAクラスI及びクラスII結合能並びにプロテアソームによって切断されることによるHBVタンパク質由来のHLAクラスI結合ペプチドの生成の分析に基づいて、世界的なB型肝炎患者集団において可能性のある全てのT細胞エピトープの非常に高いパーセンテージを網羅する最も免疫原性の高い領域が、HBVポリメラーゼタンパク質、コアタンパク質、Xタンパク質及び大表面タンパク質(large surface protein)中に発見された。これらの領域はT細胞エピトープを多く含有し、化学合成された長鎖ペプチドとしてか又は遺伝子的手法によるかいずれかでB型肝炎患者に投与された場合、そのようなワクチン接種は、活発なT細胞応答を誘発し、HBV感染を治癒させると想定される。
免疫系の活性化若しくは誘導及び/若しくは末梢血中のHBV特異的活性化型CD4+及び/若しくはCD8+T細胞の増加、若しくは治療の少なくとも1週間後にこれらT細胞によって産生されるサイトカインの増加;並びに/又は
HBV感染の増殖の阻害若しくはHBV感染細胞の検出可能な減少若しくはHBV感染細胞の細胞生存度の減少;並びに/又は
HBV感染性細胞死の誘導若しくは誘導の増加;並びに/又は
HBV感染細胞の増加の阻害若しくは予防により好ましくは検出可能である、対象におけるHBV関連疾患若しくは症状の予防、部分的クリアランス及び/若しくは治療又は完全なクリアランスに効果的に使用できるペプチドに関することが好ましい。

本発明において、慢性的なHBV感染患者を治療するための合成長鎖ペプチドからなる最適なT細胞誘導ワクチン組成物を開発した。最も高い推定のT細胞免疫誘導能力を持つHBVポリメラーゼ、コアタンパク質、表面抗原及びXタンパク質の領域を包含する37個の長鎖ペプチド(表3;30〜39アミノ酸の配列)の選択を行った。この目的のために、HLAクラスI及びIIペプチド結合並びにHBVタンパク質に含有される全てのHLAクラスI結合短鎖ペプチド(CTLエピトープの長さ;8〜12aa)のプロテアソームによるC末端生成を予測するアルゴリズムを使用して、第1の推定のHLAクラスI拘束性細胞傷害性T細胞(CTL)エピトープ及び推定のHLAクラスII拘束性ヘルパーTエピトープをこれらのタンパク質において同定した。数値を、免疫原性品質を反映している推定のCTLエピトープ及びヘルパーTエピトープ全てに指定した。プロテアソームによる供給源タンパク質からのそのC末端遊離の可能性と関連するエピトープのHLAクラスI結合能力との組み合わせを組み込んだいわゆるクラスI−BCIスコア(クラスI−結合−切断−免疫原性スコア)を使用して、任意のCTLエピトープの品質を評価した。推定のヘルパーTエピトープを、推定のヘルパーTエピトープの結合能力を反映するいわゆるクラスII−Bスコア(クラスII−結合スコア)を使用して評価し、したがって免疫原性品質を予測した。全てのCTLエピトープ及びヘルパーTエピトープの累積量及び質を反映する統合累積値、すなわち累積クラスI−BCIスコアと累積クラスII−Bスコアの合計を、可能性のある全ての長鎖ペプチドについて算出し、T細胞局所的免疫原性評価(TRIA)スコアで表す。したがって、TRIAスコアにより、HBVタンパク質中の可能性のあるあらゆる長鎖ペプチド(長さ30〜39aa)の全体のT細胞免疫原性の評価が可能になった。最も高いTRIAスコアを持つ37個のHBV由来長鎖ペプチドを、選択した。次に、ナイーブドナー及び過去にHBV感染が除去されたドナーが、37個の長鎖ペプチドの広い組から選択した、様々なTRIAスコアを持つ13個の長鎖ペプチドの1つ又は複数のサブセットに応答できるかどうか評価した。
ペプチド合成
ペプチドを、PTI Preludeペプチド合成装置のよって固相Fmoc/tBu化学を使用して合成し、Gilson分取HPLCシステムによって純度>95%に精製した。ペプチドの同一性及び純度を、Waters Acquity UPLC/TQDシステムによってUPLC−MSで確認した。
SLP当たりの推定のCTL免疫誘導容量を、SLP当たりの累積クラスI−BCIスコアを算出することにより予測した。下で詳述する通り、累積BCIスコアは、HLAクラスIペプチド結合の尺度であるクラスI−Bスコア及びプロテアソームエピトープ遊離の尺度であるCスコアに基づく。
HBVポリメラーゼ由来ペプチド、HBVコアタンパク質由来ペプチド、HBV表面抗原由来ペプチド及びHBV Xタンパク質由来ペプチドの50個のHLAクラスI分子(テキストを参照のこと)に結合するペプチドを、独自のアルゴリズムを使用してin silicoで評価した。各HLAクラスI分子に対して予測した結合ペプチドの上位1.5パーセンタイルを選択した。「クラスI−結合スコア(クラスIBスコア)」は、ペプチドの予測した結合親和性の順位から得られる。簡潔には、最も良く結合すると予測されたペプチドがスコア100になるように、順位を最初に逆転させ、その後100に対して正規化した。例:5つのペプチドを選択した(1.5パーセンタイル内の5つ)。ペプチドに、「逆順位スコア」5〜1(最良の結合ペプチドに対して5)を最初に割り当てた。その後、最良の結合が100を記録するように、各逆順位スコアを、上位1.5パーセンタイル内のペプチドの数に対して正規化する。その目的で、各ペプチドに対する順位スコアを100/5(=20)で乗算する。次いで最良の結合が、クラスI−Bスコアを求める:5x20=100、2番目に良い結合は、4×20=80のクラスI−Bスコアを有するなどである。一般に、順位スコアは、100/n(n=1.5パーセンタイル内のペプチドの数)で乗算される。結果として、選択される1.5パーセンタイル内のペプチドの正確な数に関係なく、予測された最良の結合(特定のHLAクラスI分子に対する)は、常に100を記録する。
HBVポリメラーゼ、HBVコアタンパク質、HBV表面抗原及びHBV Xタンパク質(各HLAクラスI分子ための)の予測された高親和性結合ペプチドの上位1.5パーセンタイルの、プロテアソームによるC末端生成を、2つの独自のアルゴリズムを使用して評価した。これらのアルゴリズムは、HBVポリメラーゼ、HBVコアタンパク質、HBV表面抗原及びHBV Xタンパク質それぞれにおいて特定のアミノ酸位置の後ろにおけるプロテアソーム切断の尤度を予測し、0から1までの間を記録することができ、より高い値は、アミノ酸の後における切断のより高い尤度を表す。値0.5は、任意の閾値として使用され:>0.5は切断が起こる可能性が高く、<0.5は切断が起こる可能性が低い。したがって、1に近い値は、特定の残基の後における切断の高い尤度を示す。異なる両方のアルゴリズムによる予測の間に大きな相違が生じるので、両方の独自のアルゴリズムの予測結果を考慮する切断スコア(Cスコア)を開発した。Cスコアは、両方の方法による別々のスコアの和である。したがって、HBVポリメラーゼにおいて各位置に対するCスコアは、最大2(近く)であり、最小0(近く)であり、2近くは、残基の後における切断が、プロテアソームによって生じることになる両方の方法による非常に高い尤度を反映し、1に近いCスコアは、(両方のネットワーク方法により平均に対して予測される通り)プロテアソームによる切断について中程度の傾向と考えられる。
ペプチドが、HLAクラスI分子に高親和性で結合することになり、C末端で精製されることになる尤度の指標となる尺度であるクラスI−BスコアとCスコアの両方を1つの定量的尺度に組み込むために、クラスI−結合−切断−免疫原性(クラスI−BCI)スコアを、開発した。クラスI−BCIスコアは、Cスコアで乗算したクラスI−Bスコアである。したがって、クラスI−BCIは、最大値200(100x2)(任意単位)を得ることができる。
本発明による各長鎖ペプチドに対する累積クラスI−BCIスコアを(2のうちの1つの)選択基準として使用して、本発明のペプチドを同定する。累積クラスI−BCIスコアは、本発明による長鎖ペプチドに含有されるCD8+細胞傷害性T細胞エピトープの総数と結合能力及びプロテアソームによる細胞内生成の尤度に関して予測されるその質と両方の定量的反映であり、したがって本発明による各ペプチドのCD8+細胞傷害性T細胞誘導力(CD8+T細胞免疫原性)の指標となる。本発明によるペプチドの比較的高い累積クラスI−BCIスコアは、高いCD8+T細胞免疫原性を示す。
HBVポリメラーゼ由来ペプチド、HBVコアタンパク質由来ペプチド、HBV表面抗原由来ペプチド及びHBV Xタンパク質由来ペプチドの13個の一般的なHLAクラスII分子に結合するペプチドを、独自のアルゴリズムを使用してin silicoで評価した。「クラスII−結合スコア(クラスIIBスコア)」は、ペプチドの予測された結合親和性の順位から得られる。簡潔には、最も良く結合すると予測されたペプチドがスコア100になるように、順位を最初に逆転させ、その後100に対して正規化した。リスト内のペプチドの数を減らすために、より低い予測された結合(より低いクラスII−Bスコア)で特定のHLAクラスII分子に結合することが予測されたペプチドの長さバリアントの全てを、リストにおいて破棄する。
本発明による各長鎖ペプチドに対する累積クラスII−Bスコアを第2の選択基準として使用して、本発明のペプチドを同定する。累積クラスII−Bスコアは、本発明による長鎖ペプチドに含有されるCD4+ヘルパーT細胞エピトープの総数と結合能力に関して予測されるその質と両方の定量的反映であり、したがって本発明による各ペプチドのCD4+T細胞誘導力(CD4+T細胞免疫原性)指標となる。本発明によるペプチドの比較的高い累積クラスII−Bスコアは、高いCD4+T細胞免疫原性を示す。
本発明の特定のペプチド(SLP)のTRIAスコアは、本発明の特定の長鎖ペプチド(SLP)の累積クラスI−BCIスコア及び累積クラスII−Bスコアの合計である。
ジチオトレイトール(DTT;Sigma−Aldrich)を、UPLC品質水に新たに溶解し、終濃度2mM DTTになるように2×濃縮プロテアソーム消化緩衝液(60mM Trizmaベース;pH7.5;Sigma−Aldrich、20mM KCl;Sigma−Aldrich、10mM MgCl2;Sigma−Aldrich、10mM NaCl;Sigma−Aldrich)に添加した。次いで、UPLC−品質水130μLを2mM DTTを含有する2×濃縮プロテアソーム消化緩衝液150μL、及び試験しようとするペプチド(貯蔵濃度300nmol/mL)10μLと一緒に3つの反応バイアルに添加した。バイアルをボルテックスした後、バイアル1に水10μL(対照消化モック)、バイアル2に構成的20Sプロテアソーム1μg(10μL)(貯蔵0.1mg/mL;Enzo Life Sciences)、及びバイアル3に免疫20Sプロテアソーム1μg(10μL)(貯蔵0.1mg/mL;Enzo Life Sciences)を添加した。ボルテックス直後にT=0のサンプル50μLを採取し、ギ酸(Sigma−Aldrich)4μLを添加して反応を止めた。反応バイアルをボルテックスし、37℃でインキュベートした。サンプル50μLを、インキュベーション1時間、3時間、6時間及び24時間後に採集した。反応をギ酸4μLで止め、全てのサンプルを20℃で保管した。
エレクトロスプレーイオン化質量分析にオンラインナノエレクトロスプレーインタフェースを備えたQ−TOF1質量分析計(Waters)をおよその流速250nL/分で使用した。ペプチド消化サンプルを、プレカラム(MCA−300−05−C18;Dionex)に捕捉し、70〜90%緩衝液Bの急勾配で10分間にわたって溶出した[緩衝液A、水、アセトニトリル及びギ酸、95:3:1(体積/体積/体積);緩衝液B、水、アセトニトリル及びギ酸10:90:1(体積/体積/体積)]。質量スペクトルを50〜2000ダルトンの質量で記録した。タンデム型質量分析モードにおいて、イオンを3ダルトンのウインドウで選択した。衝突ガスは、アルゴン(4×10−5mbar)であり、衝突電圧は、約30Vであった。精製した構成的プロテアソーム及び免疫プロテアソームによるペプチド消化の場合、質量スペクトルのピークを、BioLynxソフトウェア(Waters)を用いて供給源基質ペプチド中で検索し、特定の消化断片の存在量を、消化されていない基質を含めた全体の合計強度のパーセンテージとして定量的に評価した。
健康なドナー由来末梢血単核細胞(PBMC)を、Ficoll勾配に対する遠心分離で単離した。樹状細胞(DC)を生成するために、およそ50*106個のPBMCを、3*106個細胞/mL完全培地(8% HS、Seralab;ペニシリン/ストレプトマイシン、Lonza;L−グルタミン、Lonzaで補充したIMDM、Lonza)の濃度にし、6ウェルプレート(Corning)に3mL/ウェルで播種した。37℃で1.5時間インキュベーションした後、非接着細胞を、完全培地を使用して3回の洗浄ステップで洗い流した(0日目)。接着細胞を、800U/mL GM−CSF及び500U/mL IL−4(Peprotech)を含有する2mL/ウェルの完全培地中で、37℃で3日間培養した。3日目に、2400U/mL GM−CSF及び1500U/mL IL−4を含有する完全培地1mLを、各ウェルに添加し、37℃で更に3日間培養した。
6日目に、3つのプールに分配した長鎖ペプチドを、3nmol/mL濃度でナイーブドナーの単球由来DCに添加し、37℃で終夜インキュベートした。プール1は、SLP26(配列番号76)、SLP24(配列番号74)、SLP1(配列番号51)及びSLP30(配列番号1142)を含み;プール2は、Abu−SLP 27(配列番号77、アミノ酸1位のシステインが、Abuに置き換えられている)、SLP25(配列番号75)、SLP10(配列番号60)及びSLP34(配列番号1469)を含み;プール3は、SLP5(配列番号55)、SLP13(配列番号63)、SLP14(配列番号64)及びSLP21(配列番号21)を含む。7日目に、細胞を完全培地で2回洗浄してペプチドを除去した。DC及び自己のPBMCを1:10の比で、10ng/mL IL−7及び100pg/mL IL−12p70の存在下で、37℃で10日間共培養した。この過程により生成されるT細胞株を、2〜3日毎に確認し、必要に応じて分割した。
T細胞誘導の3日後(10日目)に、自己のDCの第2のバッチを分化させ、上記のペプチドプールを載せた。T細胞株の開始後10日目(17日目)に、ペプチドを載せたDCを完全培地で2回洗浄し、10ng/mL IL−7及び100pg/mL IL−12p70の存在下で1:10の比(DC:T細胞)でT細胞に添加した。細胞を、7日間共培養した。17日目に、またDCの新たなバッチを分化させ、上記のペプチドプールを載せた。24日目に開始する第2の再刺激の場合、DC及びT細胞を1:10の比で共培養した。再刺激の2日後に、30IU/mL IL−2(Peprotech)及び5ng/mL IL−15(Peprotech)を培地に添加した。31日目に開始する第3の再刺激を、第2の再刺激と同じく実行した。
T細胞活性化及び増殖を測定するために、自己のDCに、上記のT細胞誘導手順の23、30及び37日目に、13個のHBV由来ペプチドのそれぞれを別々に6時間載せた。DCを洗浄し、ペプチドを載せたDC 5000個をT細胞50000個と48時間共培養した。次いで、上清をELISA(IFNγELISA、Diaclone)ために採集し、3Hチミジンを含有する培地を全てのウェルに添加した。放射性3Hチミジンは、新たに形成された(増殖した)細胞のDNAに組み込まれ、インキュベーションの16時間後にMicroBeta液浸計数装置(Wallac/Perkin Elmer)で測定される。
抗原特異性IFNγ産生ヒトT細胞を検出するために、PBMCを、3nmol/mLの指示するペプチドと37℃で72時間最初に予備刺激した。この刺激の間、ELISpot PVDFプレート(Mabtech)を、PBS中の5ug/mL抗ヒトIFNγmAb 1−D1Kコーティング抗体(Mabtech)でコーティングし、4℃で終夜インキュベートした。PBMCの刺激後に、コーティング抗体をプレートから吸い出し、PBSで4回洗浄した。非特異的結合をブロッキングするために、8%FCSを含有するIMDM 100μLを全てのウェルに添加し、37℃で1時間インキュベートした。その間に、刺激しPBMCを採取し、遠心分離し、X vivo15培地(Lonza)に再懸濁し、計数した。全てのPBMCサンプルを、X−vivo15培地中で1.5*106個細胞/mLの濃度にした。PVDFプレートのウェル中の培地を吸い出し、各PBMCサンプル100μLを4組でプレートに添加した。プレートを、37℃で終夜インキュベートした。翌日、上清を破棄し、プレートをPBS/Tween20 0.05%で6回洗浄した。ビオチン化抗ヒトIFNγ7−B6−1 mAb(Mabtech)を、1%FBSを含むPBS中に0.3μg/mLの濃度で全てのウェル(100μL/ウェル)に添加し、RTで2時間インキュベートした。次に、プレートをPBS/Tween20 0.05%を使用して、6回洗浄し、1%FBSを含むPBS中の1μg/mL Extravidin−アルカリホスファターゼ(ALP)(Sigma−Aldrich)を、全てのウェルに添加した(100μL/ウェル)。プレートを、RTで1時間インキュベートした。ALP基質溶液BCIP/NBT−plus(Mabtech)を調製し、プレートをPBS/Tween20 0.05%で4回洗浄した後に、100μL/ウェルを全てのウェルに添加した。比色反応を停止させるために(1〜20分後)、水道水を使用してプレートを徹底的に洗浄した。乾燥後、形成された点をBiosys Bioreader 5000で測定した。
長鎖ペプチドに含有される可能性のある全てのCD8+T細胞エピトープのHLAクラスIペプチド結合予測及び予測されたC末端生成に基づく長鎖ペプチドの選択
ペプチドが結合するHLAクラスI分子に対して予測される高親和性を保有し、プロテアソームのタンパク質分解切断によりペプチドのC末端で生成されるとも予測される両方のペプチドとして、高品質CD8+T細胞エピトープが定義される。本発明によるペプチドは、最適に多数の高品質のCD8+及びCD4+T細胞エピトープを含有するHBVタンパク質領域内で選択された。この目的のために、独自のHLAクラスIペプチド結合アルゴリズム並びにプロテアソームによる切断を予測する2つの独自のアルゴリズムを使用して、最初にHLAクラスI結合及び可能性のある全てのCD8+T細胞エピトープのC末端生成を評価した。その後、各短鎖ペプチド(8〜13アミノ酸)に対して、短鎖ペプチドが結合するHLAクラスI分子に対する短鎖ペプチドの予測される結合親和性とペプチドが細胞内でプロテアソームにより生成される尤度の両方を組み込む単一の定量的尺度、いわゆる結合−切断−免疫原性(BCI)クラスI−スコアを発明した。クラスI−BCIスコアは、(1)上述したアルゴリズムを使用するHLAクラスIペプチド結合のin silico予測の結果から得られる結合クラスI−スコア(クラスI−Bスコア)、及び(2)上述したアルゴリズムを使用する、プロテアソームによるペプチドのプロテアソーム媒介C末端生成のin silico予測の結果から得られる切断スコア(Cスコア)から算出される。表4a、5a、6a及び7aは、これらポリメラーゼ、コアタンパク質、表面抗原又はXタンパク質由来SLPの可能性のある全てのCD8+T細胞エピトープに対するクラスI−BCIスコアをそれぞれ累積BCIクラスIスコアと一緒に示す。表4b、5b、6b及び7bは、これらポリメラーゼ、コアタンパク質、表面抗原又はXタンパク質由来SLPの可能性のある全てのCD4+T細胞エピトープに対するクラスII−Bスコアをそれぞれ累積クラスII−Bスコアと一緒に示す。累積クラスI−BCIスコア(CTLエピトープに対する)と累積BクラスII−スコア(ヘルパーTエピトープに対する)を合わせることにより、1つの定量値、長鎖ワクチンペプチドの全体のT細胞免疫原性を反映するいわゆる総局所的免疫原性評価(TRIA)スコアが得られた(表3)。最も高いTRIAスコア基づいて、HBVポリメラーゼ、コアタンパク質、表面抗原又はXタンパク質に由来する37個のSLPを、更なる評価ための選択した(表3)。37個のSLPから、TRIAスコアの予測能力を検証するために、比較的低い及び高いTRIAスコアを持つSLPを含めて13個のSLPの代表的な組を選んで、in vitro免疫原性を評価した。記述の通り(下記参照)、上記13個のペプチドをT細胞誘導アッセイのために3つのペプチドプールに分けた。
推定のCTLエピトープの同定に重要な構成要素は、プロテアソーム媒介切断によるCTLエピトープのC末端生成の予測である。この予測の信頼性を検証するために、プロテアソーム消化パターンを、機能的に試験した長鎖ワクチンペプチド13個のうち3個を除いた全てについて実験的に評価した(3個の長鎖ペプチドは、技術的な理由により切断されていなかった)。
主要な切断部位:COOH末端として切断部位由来の残基NH2末端を(可能性のある)相補的断片(複数可)と共に含有する断片が、質量スペクトルにおける断片ピークの強度から算出される通り、24時間インキュベーション時に≧7%存在する。
少数の切断部位:COOH末端として切断部位由来の残基NH2末端を(可能性のある)相補的断片(複数可)と共に含有する断片が、24時間インキュベーション時に<7%存在する。累積断片存在量<1%の切断部位は示さない。
選択した13個のペプチドが、ナイーブドナーにおいてT細胞応答を誘導できるかどうか評価するために、PBMCを、HBV感染したことがない6名の健康なドナーから得た白血球層から単離した。これらのPBMCを3つのペプチドプールのいずれかで再刺激してT細胞株を得、13個の選択したペプチドでその後刺激した。IFNγの産生及びT細胞増殖(3Hチミジン取り込み)を、T細胞活性化の読み出しとして測定した。結果を図2に示し、刺激指数(SI)1.5以上を持つ陽性T細胞応答を示すドナーのパーセンテージを示す。SIは、測定したサンプル値を刺激していない対照細胞の値で割ることにより算出される。ナイーブドナーにおいて13個のうち12個の予め選択したペプチドに対して誘導された応答を検出し、11個の予め選択したペプチドが、複数のナイーブドナーにおいて応答を誘導した。
HBV感染を経験し、成功裏にHBVを除去した対象は、HBVに特異的な循環記憶T細胞を保有する。天然に存在するHBV感染の除去に対する選択したワクチンペプチドの妥当性を評価するために、6名のHBV免疫ドナーから得たPBMCにおいて、選択した13個のHBVペプチドに対するT細胞応答の存在を試験した。PBMCの単離後に、細胞を13個のペプチドのそれぞれで刺激し、IFNγ−ELISpotを実行してT細胞応答を検出した。試験したドナー6名のうち3名のPBMCサンプルが、13個のペプチドのうちの1個又は複数に対して陽性IFNγ応答を示した(SI>3)。応答を、13個のペプチドのうち11個(SLP5、10、13、14、18、21、24、25、26、27及び34、それぞれ配列番号55、60、63、64、68、71、74、75、76、77及び1469で表す)に対して観察し、そのうちの6個(SLP10、13、21、24、25及び34、それぞれ配列番号60、63、71、74、75及び1469により表される)は、複数のドナーにおいて応答を誘導した。結果を図3に要約し、ペプチドに対して陽性IFNγ+T細胞応答を示すドナーを示す。試験した各SLPについて、IFNγ応答の平均SIを算出した;SLP1で1.21、SLP5で2.11、SLP10で2.40、SLP13で2.44、SLP14で1.97、SLP18で1.86、SLP21で2.87、SLP24で2.54、SLP25で2.66、SLP26で1.56、Abu−SLP27で1.66、SLP30で1.49及びSLP34で3.02。重要なことに、各ペプチドに対するIFNγ応答の平均SIを、対応するTRIAスコアに対して描画した場合、Abu−SLP27の結果を含む場合(R2=0.37、p=0.028;図4を参照のこと)とAbu−SLP27の結果を除外した場合(R2=0.34、p=0.048;不掲載)の両方で有意な相関(図4)を見る。これは、免疫原性ペプチドの選択ためのTRIAスコアの使用を検証する。
ここで示す実験結果は、選択を検証し、したがって支持し、本発明のHBV由来長鎖ワクチンペプチドの免疫学的妥当性を明白に示す。これらの長鎖ワクチンペプチドは、非近交系集団における高品質のHLAクラスI及びHLAクラスII結合エピトープを最も多数含むHBVタンパク質領域を包含する。好ましい組み合わせのワクチンペプチドを新規のHBV SLPワクチン組成物に組み込んで、慢性的にHBV感染している患者を治療することになる。一般的なHLAクラスI分子全てに対する短鎖ペプチド(8〜12aa)のペプチド結合親和性及びプロテアソームによる切断によるこれら短鎖ペプチドのC末端生成の尤度を予測するアルゴリズムを、推定のヘルパーTエピトープの同定と組み合わせて使用して、最適な37個の長鎖ワクチンペプチドを選択した免疫原性の高い領域を同定した。

- 少なくとも70個の予測されたT細胞エピトープ及び少なくとも3個のプロテアソーム切断部位を含むペプチドであって、HBVタンパク質に由来し、15〜100アミノ酸長であるペプチド。
- HBVポリメラーゼ、HBVコアタンパク質、HBV Xタンパク質及びHBV大表面タンパク質からなる群から選択されるいずれかのタンパク質のアミノ酸配列を含む又は当該アミノ酸配列からなる、請求項1に記載のペプチド。
- 少なくとも70個の予測されたHLAクラスI拘束性CD8+細胞傷害性T細胞エピトープ及び少なくとも1個の予測されたHLAクラスII拘束性CD4+ヘルパーTエピトープを含み、好ましくは配列番号75、51〜74、76〜79、1142〜1145及び1468〜1471からなる群から選択されるペプチドを含む、請求項1又は2に記載のペプチド。
