Abstract
Description

アぺリンは、77アミノ酸のプレプロペプチドとして内在的に産生され、いくつかのより短い生物学的に活性な断片またはアぺリンペプチドを生じるように切断される。
受容体スクリーニングアッセイは、本発明のアぺリン融合タンパク質だけではなく、APLNRのアゴニストおよびアンタゴニストを含むいずれの試験化合物も供することができることを理解されたい。APLNRの活性化または不活性化を決定するための多くの受容体スクリーニングアッセイが当該技術分野で周知であり、以下の例は、本発明者が彼らの発明であると見なすものの範囲を限定することを意図するものではない。
融合タンパク質を作製する方法は、当該技術分野で既知である。そのような1つの方法において、DNA発現ベクターが1つの連続した融合ポリペプチドを発現するように、Fcをコードする核酸配列にインフレームで連結されたアぺリンをコードする核酸配列を含有するようにDNA発現ベクターが操作される。アぺリンペプチドは、Fc含有ポリペプチドのC末端またはN末端に連結されてもよい。本発明のアぺリン融合タンパク質は、アぺリンペプチド単独よりも安定であることが予測される。血清安定性タンパク質は、分解に対する耐性を付与するか、または循環からのクリアランスの低下を有するタンパク質を含む。例示的な本発明の血清安定性アぺリン融合タンパク質は、配列番号2、配列番号4、配列番号39、配列番号40、および配列番号41を含む。

実施例1−発現構築物のクローニング
合成遺伝子断片を使用して、アぺリン−13を有するN末端およびC末端のhFc融合物を作製した。得られた融合物、hFc−アぺリン13(配列番号1)およびアぺリン13−hFc(配列番号3)をコードするDNAを、標準的な分子クローニング技術を使用してCMVプロモーターの下流の発現ベクターに挿入した。CHO安定細胞株を作製し、融合タンパク質の生成に使用し、次いで、それをアフィニティー法によって精製した。N末端hFc−アぺリン13およびC末端アぺリン13−hFc融合タンパク質は、それらの予測質量と一致してSDS−PAGEゲル上で泳動する。(図3Aを参照のこと。)抗アぺリン抗体(Abcam、ab59469番)を用いて行ったウエスタンブロット分析を使用して、hFc−アぺリン13およびアぺリン13−hFc上にアぺリンの存在を確認した。(図3Bを参照のこと。)
hAPLNRの活性化を検出するために開発されたバイオアッセイを使用して、非修飾アぺリンペプチド(Bachem、H−4568.0001番)および本発明のアぺリン13融合タンパク質による細胞内cAMPレベルの調節を評価した。ルシフェラーゼレポーター[cAMP応答要素(CRE,4X)−ルシフェラーゼ]とともに、全長ヒトhAPLNR(受入番号NP_005152.1のアミノ酸1〜380)を安定に発現するようにHEK293細胞株をトランスフェクトした。得られた細胞株HEK293/CRE−luc/hAPLNRを、10% FBS、NEAA、ペニシリン/ストレプトマイシン、および100μg/mLハイグロマイシンBを含有するDMEM中に維持した。バイオアッセイのために、0.1% FBSおよびペニシリン/ストレプトマイシン/L−グルタミンを補充した80μLのOPTIMEM中の96ウェルアッセイプレートに、HEK293/CRE−luc/hAPLNR細胞を20,000細胞/ウェルで播種し、5%CO2中、37℃で16時間インキュベートした。翌朝、hAPLNRの活性化を介してフォルスコリンによって誘発されたcAMPの生成を測定するために、非修飾アぺリンペプチドおよびアぺリン13融合タンパク質を段階稀釈し(1:3)、次いで、アッセイ緩衝液(5μMの最終フォルスコリン濃度)中でフォルスコリン(Sigma、F6886番)と混合し、細胞に加えた。5%CO2中、37℃で5時間のインキュベーション後、Victor X機器(Perkin Elmer)を使用して、One Glo試薬(Promega、E6051番)の添加後に発光を測定した。Prism 5ソフトウェア(GraphPad)を用いて、非線形回帰により、データを4パラメータのロジスティック方程式に適合させた。
DiscoverX PathHunter(登録商標)プラットフォームは、関連リガンドを用いた処理に応じたβ−アレスチンのGPCRへの動員に基づいている。このアッセイ形式では、β−アレスチンがβガラクトシダーゼ(β−gal)のN末端欠失変異体に融合されて細胞内で安定に発現される一方で、GPCRがより小さな(42アミノ酸)、弱い競合性β−gal断片に融合される。このアッセイにおけるGPCRのリガンド刺激は、β−アレスチンのGPCRへの動員をもたらし、2つのβ−gal断片の相補性を強制し、基質を検出可能なシグナルに変換する機能的酵素の形成をもたらす(DiscoverX Corporation、Fremont,CA,USA)。
本発明のアぺリン−13融合タンパク質がAPLNRシグナル伝達経路に与える影響を測定するために、アッセイを使用してAPLNR発現細胞株由来のリン酸化ERK1/2(pERK1/2)および全ERKの量を定量化した。ドキシサイクリン誘導性CMVプロモーターの制御下で全長ヒトAPLNR(hAPLNR;受入番号NP_005152.1のアミノ酸1〜380)を安定に発現するように、チャイニーズハムスターの卵巣(CHO)細胞株をトランスフェクトした。得られた細胞株CHO/hAPLNRを、10%FBS、ペニシリン/ストレプトマイシン、L−グルタミン、および250ug/mLハイグロマイシンBを含有するHam’s F12培地中に維持した。
C57/Bl6マウス(群当たりn=3)にhFc(2.5mg/kg)またはアぺリン13−hFc(2.8mg/kg)を皮下(s.c.)投与し(図7A)、1、4、24、および48時間目に血漿を採取した。別個の実験において、C57/Bl6マウス(群当たりn=3)に5mg/kgでhFc−Apein13をs.c.投与し、0、1、2、4、5、6、24時間、および2、3、7、14、21日目に血清を採取した(図7B)。
上の実施例2から6に見られるように、そのN末端にFcが繋ぎ止められたアぺリン−13(hFc−アぺリン13)は、そのC末端にFcが繋ぎ止められたアぺリン−13(アぺリン−hFc)よりも良好な効力を示す。1つ以上のアミノ酸(複数可)がN末端またはC末端から欠失しているかまたはそこに付加されたアぺリン−13ペプチド等の修飾アぺリン−13ペプチドを、APLNRの活性化に関連するそれらの相対的な効力について調べた。
hFcに融合された配列番号42、配列番号43、および配列番号44等の修飾されたアぺリンペプチドを有することを除いて、実施例1と同様に、種々のアぺリン−Fc融合タンパク質を作製した。そのようなhFc−アぺリン13融合タンパク質は、アぺリンペプチド成分のC末端にさらなるC末端アミノ酸を有する。実施例2および実施例7の方法と同様にCREバイオアッセイを使用して、各ペプチドのN末端に繋がれたhFcを有する修飾アぺリン−13ペプチド(hFc−アぺリン13+)と比較して、アぺリン−13ペプチドによるcAMPレベルの調節を評価した(上記参照)。Prism 5ソフトウェア(GraphPad)を用いて、非線形回帰(4パラメータロジスティック)を使用してその結果を分析した。
麻酔下のマウスおよびラットにおける心電図検査によって、本発明のアぺリンFc融合タンパク質の影響、特に、RR間隔(心拍数の指標)、およびイオンチャネル活性の指標としてのQT間隔に与える影響を評価した。

- 前記疾患または状態は、心血管疾患、急性非代償性心不全、うっ血性心不全、心筋梗塞、心筋症、虚血、虚血/再灌流障害、肺高血圧症、糖尿病、肥満、癌、転移性疾患、体液ホメオスタシス、のぼせ症、病的血管新生、網膜症、線維症、およびHIV感染からなる群から選択される、請求項116に記載の方法。
- 少なくとも1つの上記請求項のいずれかに記載のアぺリン融合タンパク質またはポリペプチドを充填した1つ以上の容器を含む、キット。
- 多量体形成成分に融合したアぺリンペプチドを含むポリペプチド。
- 前記多量体形成成分は、ロイシンジッパー、ヘリックスループモチーフ、コイルドコイルモチーフ、Fcドメイン、Fcドメインの断片、またはFcドメインの変異体を含む、請求項119に記載のポリペプチド。
- 前記アぺリンペプチドは、アぺリン42〜77(アぺリン−36)、アぺリン61〜77(アぺリン−17)、アぺリン63〜77(アぺリン−15)、アぺリン64〜77(アぺリン−14)、アぺリン65〜77(アぺリン−13)、アぺリン66〜77(アぺリン−12)、アぺリン67〜77(アぺリン−11)、アぺリン68〜77(アぺリン−10)、アぺリン73〜77(アぺリン−5)、アぺリン61〜76(アぺリン−K16P)、アぺリン61〜75(アぺリン−K15M)、アぺリン61〜74(アぺリン−K14P)、アぺリン−F13A、アぺリン65〜76、アぺリン65〜75、アぺリン66〜76、アぺリン67〜76、アぺリン66〜75、アぺリン67〜75、および[Pyr1]アぺリン−13、またはその断片もしくは誘導体からなる群から選択される、請求項119または120に記載のポリペプチド。
- 前記アぺリンペプチドは、アぺリン−13である、請求項119〜121のいずれか1項に記載のポリペプチド。
- 前記アぺリンペプチドは、1つ以上のペプチドリンカーを介して前記多量体形成ドメインに融合される、請求項119〜122のいずれか1項に記載のポリペプチド。
- 前記多量体形成ドメインは、Fcドメイン、またはその断片であり、前記アぺリンペプチドは、1つ以上のGly−Serリンカーを介してFcドメインまたはFcドメイン断片に融合される、請求項123に記載のポリペプチド。
- 前記アぺリンペプチドは、1つ以上のGly−Serリンカーを介して前記FcドメインのC末端、またはその断片に融合される、請求項124に記載のポリペプチド。
- 前記アぺリンペプチドは、1つ以上のGly−Serリンカーを介して前記FcドメインのN末端、またはその断片に融合される、請求項125に記載のポリペプチド。
- 前記多量体形成ドメインは、IgG1 CH2ドメインおよびIgG1 CH3ドメイン;IgG4 CH2ドメインおよびIgG4 CH3ドメイン;IgG1 CH2ドメインおよびIgG4 CH3ドメイン;ならびにIgG4 CH2ドメインおよびIgG1 CH3ドメインからなる群から選択されるFcドメインである、請求項119〜126のいずれか1項に記載のポリペプチド。
- 前記Fcドメインは、IgGヒンジドメイン、またはその断片もしくは変異体を含む、請求項119〜127のいずれか1項に記載のポリペプチド。
