Abstract
Description
マメ科植物(大豆(白太、Glycine Max.))の生育期間中、初期(VE)時期からV4時期まで、低分子水溶性キトサンまたは麹菌(Aspergillus oryzae)を土壌およびマメ科植物に処理して栽培した。低分子水溶性キトサンは、水に容易に溶解するため土壌に処理することが容易であり、他の誘導因子と比較して食用が可能な成分であって、人体に無害な成分である。低分子水溶性キトサンは、(株)YoungChi Pharmで生産する水溶性キトサン(YWCTS‐100)を購入した。キトサン処理は、水に0.3%の濃度で溶かした後、じょうろを利用して根にも接触し得るように、マメの茎や土壌を中心に、5日間隔で散布した。
キトサンと麹菌をそれぞれ処理した実験群と何も処理せずに栽培したマメの根を生長時期別(VE〜V4時期、計6時期)に採取した後、精製水で洗浄し、乾燥させてから細粉化した。前記マメの根の粉100gを70%エタノール水溶液1リットル(L)に入れ、室温で24時間抽出した後、300メッシュ(mesh)の濾布で濾過した。濾液を、冷却コンデンサの付いた蒸留装置を利用して50℃で減圧濃縮し、乾燥してマメの根抽出物を作製した。
前記実験例2で得られた各生長時期別マメの根抽出物におけるクメストロール(coumestrol)含量の定量分析のために、HPLCを利用した。マメの根抽出物のクメストロールの含量を比較するために、基準物質としてIndofine Chemical社から購入したクメストロールを使用した。
前記実験例1で行われたキトサン処理されたマメの根に、クメストロールの配糖体形態であるクメストリンが生成されたということを確認した。キトサン処理されたマメの根抽出物からクメストリンの糖を除去することにより、抽出物内におけるクメストロールの含量をさらに増加させることができる。
Claims (9)
- マメ科植物の生育期間中、マメ科植物または土壌にキトサンを処理することを含む、クメストリン(Coumestrin)またはクメストロール(Coumestrol)の生産方法。
- 前記キトサン処理は、マメ科植物の根に処理する、請求項1に記載のクメストリンまたはクメストロールの生産方法。
- 前記キトサンを、マメ科植物の生育期間中、VE時期からV4時期までのうちいずれか一つの時期以上の期間中に処理する、請求項1に記載のクメストリンまたはクメストロールの生産方法。
- 前記キトサンは、低分子水溶性キトサンである、請求項1に記載のクメストリンまたはクメストロールの生産方法。
- 前記キトサン処理を4日〜6日間隔で行う、請求項1に記載のクメストリンまたはクメストロールの生産方法。
- 前記方法は、前記栽培されたマメ科植物の抽出物を製造することをさらに含む、請求項1に記載のクメストリンまたはクメストロールの生産方法。
- 前記マメ科植物の抽出物は、根部分の抽出物である、請求項6に記載のクメストリンまたはクメストロールの生産方法。
- 請求項1〜7のいずれか一項によりクメストリンおよびクメストロールを生産すること;および
前記クメストリンに、酸、塩基、酵素および微生物からなる群より選択された一つ以上を処理して、クメストリンから糖を除去することをさらに含む、クメストロールの生産方法。 - 前記酸、塩基、酵素および微生物からなる群より選択された一つ以上の処理は、クメストリンを含むマメ科植物の抽出物に対して処理する、請求項8に記載のクメストロールの生産方法。
