本明細書では、生体試料中の一又は複数の標的を検出するために開示コンジュゲートを使用する方法の実施態様を開示する。特定の開示実施態様では、コンジュゲートの一又は複数を、インサイツハイブリダイゼーション(ISH)、免疫細胞化学、及び免疫組織化学(IHC)検出スキームなどの標準的アッセイに使用する。特定の開示実施態様では、これらのアッセイのいずれか1つをシグナル増幅と組み合わせてもよい、及び/又はアッセイが複数の異なる標的を検出することができる多重化に関連してもよい。実施態様はまた、1種又は複数の増強剤を含んでもよい。本方法の実施態様を組み合わせてもよい。例えば、IHC検出スキームを使用する方法をISH検出スキームと組み合わせてもよい。開示方法の代表的な実施態様を、細胞クローン性(例えば、細胞が2つのバイオマーカーの両方ではなくいずれか1つを発現している)を決定するため、がん患者のがん治療への応答性を予測するため(例えば、特定の患者が治療に応答するかどうかを決定するための予測バイオマーカーを検出する)、アッセイ性能及び試料完全性を監視するためのバイオマーカー発現及び内部標準遺伝子発現の同時分析のため、並びにこれらの組み合わせのために使用してもよい。
実例となる実施態様では、生体試料中の標的を検出する方法は、生体試料を検出プローブと接触させることと、生体試料をラベリングコンジュゲートと接触させることと、生体試料をシグナリングコンジュゲートと接触させることとを含む。図1は、本開示による方法の一実施態様のステップを提供する流れ図である。特に、本方法は、試料を検出プローブ(複数可)と接触させるステップ1を含む。このステップは、単一の検出プローブ又は複数の異なる標的に特異的な複数の検出プローブのいずれかを含むことができる。その後のステップ2は、試料をラベリングコンジュゲートと接触させることを含む。さらにその後のステップ7は、試料をシグナリングコンジュゲートと接触させることを含む。試料を増幅コンジュゲートと接触させるステップ3及び試料を二次ラベリングコンジュゲートと接触させるステップ5への破線は、これらのステップが任意であることを表している。試料を酵素阻害剤と接触させるステップ10への破線は、任意のループを使用して多重化手法により複数の標的を検出することができることを示している。特定の開示実施態様では、酵素阻害剤を生体試料に添加する一又は複数のステップを使用してもよい。例えば、2種以上のシグナリングコンジュゲートを試料に添加する実施態様では、1つのシグナリングコンジュゲートを共有結合的に沈着させた後及び第2の異なるシグナリングコンジュゲートを添加する前に任意の酵素活性を防ぐために、酵素阻害剤(例えば、ペルオキシダーゼ阻害剤)を添加することができる。
歴史的に、FISHを使用して分離分析が行われてきた。しかしながら、本開示は、発色ISHを使用した三色分離アッセイを提供する。発色検出と蛍光検出との間の違いを図5(A)及び図5(B)に絵を用いて示す。図5(A)は、赤色発色体例51、青色発色体例53及び赤色と青色の多重化発色体例52を示している。発色体が典型的には白色光である光に露光される(すなわち、P0の入射電力を有する光に露光される)と、発色体が種々の波長を吸収する。透過光は、発色体の吸光度及び存在する発色体の量に応じて特定の電力(図5(A)P1、P2及びP3)を有する。良好な検出事象は、多くの発色体の沈着をもたらし、これがより多くの光を吸収し、観察されるシグナルをより小さくする。着色発色体については、光が透過されないので、透過光の減少によって最終的に発色体が黒色に見える。赤色及び青色の多重化発色体例52に示すように、多重化はこの効果を悪化させる。伝統的な赤色発色体及び青色発色体が空間で重複すると、吸光度が広くなり、P3シグナルがP1及びP2よりも小さいことにより示されるように、検出事象が黒っぽく、暗くなる。本質的に、重複シグナルによる発色検出は、減算効果をもたらす。これは、図5(B)に示す蛍光と対照的である。図5(B)を参照すると、紫色蛍光例61、緑色蛍光例63及び紫色と緑色の多重化蛍光例62が示されている。励起光(図中でλexとして示されている)は、3つの例を通して同じであってよく、61はλf1(紫色蛍光)を示し、63はλf2(緑色蛍光)を示し、62はλf1(紫色蛍光)及びλf2(緑色蛍光)を示す。より多くの蛍光が試料上に沈着するので、より強力な蛍光シグナルが産生される。同様に、多重化シナリオでは、発蛍光団について加算効果がある一方で、発色団では減算効果が起こる。この減算対加算が、化合物に発色体を使用した多重化の困難性を著しく特徴付ける。よって、伝統的な発色体を用いた多重化は広く受け入れられていない。本開示は、従来は不可能であった色の組み合わせを可能にする狭い波長吸光度バンドを有するシグナリングコンジュゲートを提供する。よって、本開示は、蛍光多重化と比べて発色多重化が有する固有の欠点にもかかわらず、前例のない発色多重化を提供する。
シグナリングコンジュゲートは、検出可能なシグナルの提供を促進する種々の特徴を提供するように構成される。特定の開示実施態様では、シグナリングコンジュゲートは、明視野シグナルを提供する適当な発色団部分を含む。例えば、本明細書に開示される発色団は、本明細書に開示される標的を検出するのに適した光シグナルを産生するよう選択され得る。特定の開示実施態様では、発色団は、シグナリングコンジュゲートが所望のシグナルを提供するように構成されることを可能にする、以下に論じるものなどの光学特性を有する。
生体試料中の標的を認識するための一定範囲の新規な色を提供することが単独で有用であるが、本開示のシグナリングコンジュゲートは、多重化アッセイ並びに転座プローブを使用したアッセイに特に有用である。図8(A)は非小細胞肺がんに関連するALK再配列について試験する肺組織切片における2つの遺伝子プローブの二重染色の顕微鏡写真であり、図8(B)は酢酸エチル溶液中のfast red及びfast blueのUV−Visスペクトルである。fast redを使用して3’プローブを検出し、fast blueを使用して5’プローブを検出した。図9(A)及び図9(B)は、図8(B)のトレースを別々に示している。図8(B)は、fast red及びfast blueが広い明確なスペクトル吸収特性を有することを示している。fast redは約475nmと約560nmとの間に強い吸収を示す。この範囲を色相環と比較すると、スペクトル吸収特性に対応した予想される色は赤又は橙のいずれかであるだろう。吸収の範囲があまりに大きいので、この範囲は赤又は橙色をもたらすと予想される波長の全てを本質的に網羅する。fast blueは、約525nmと約625nmとの間の強い吸収、fast redよりもさらに広い範囲を示す。再び図6(A)の色相環を参照すると、525〜625nmの吸収は、青、藍及び紫が相補的な、色相環の半分近くを網羅する。
特定の開示実施態様では、伝統的な白色源及びフィルターシステム、例えば、当業者により典型的に使用されているものを使用することができる。他の開示実施態様では、より狭い照明光を産生するためにLED光源を検出ステップで使用することができる。1種又は複数の異なるシグナリングコンジュゲートを使用する実施態様、特に、3種以上の異なるコンジュゲートを使用する場合にこのような光源を使用することができる。
本明細書に記載するチラミドシグナル増幅及びシグナリングコンジュゲートは、試料から入手可能なチロシン残基並びに/又は標的を検出及び標識するために使用する分子/コンジュゲートと反応する。検出すべきバイオマーカーを囲むタンパク質の量は、組織試料間の自然変動に基づいて可変性である。高レベルで存在するバイオマーカーを検出する場合又は複数のバイオマーカーの共局在化を検出する場合、チラミド分子が付着することができるタンパク質の量は、沈着プロセスでの限界反応物質となり得る。組織中のタンパク質量が不十分であると、チラミド系検出が拡散する、バイオマーカーの発現レベルを過小予測する(under−call)可能性、及び共局在化バイオマーカーを検出することができないことになり得る。これらの課題の1つの解決策は、タンパク質性溶液で組織をコーティングし、ホルマリン又は他の固定液を使用してタンパク質を組織と持続的に架橋することにより、より多くのタンパク質結合部位(すなわち、チロシン)を得ることである。
対比染色は、顕微鏡下で構造をより容易に可視化することができるように、試料を一又は複数の標的を検出するために薬剤で染色した後に後処理する方法である。例えば、対比染色は、場合により免疫組織化学染色をより明確にするためにカバーガラスで覆う前に使用される。対比染色は一次染色とは色が異なる。ヘマトキシリン、エオシン、メチルグリーン、メチレンブルー、ギムザ、アルシアンブルー及びNuclear Fast Redなどの多数の対比染色が周知である。いくつかの例では、2種以上の染色を混合して対比染色をすることができる。これにより柔軟性及び染色を選択する能力が提供される。例えば、第1の染色を特定の属性を有するが異なる所望の属性を有さない混合物のために選択することができる。所望の属性を欠いていることを示す混合物に第2の染色を添加することができる。例えば、トルイジンブルー、DAPI及びポンタミンスカイブルーを混合して対比染色を形成することができる。本開示の一態様は、染色した試料が読み取り装置で容易に解釈されるように、当技術分野で既知の対比染色法を開示方法及び組成物と組み合わせることができるものである。
本明細書では開示方法に使用するのに適した種々の異なるコンジュゲートを開示する。本開示により熟慮される種々のクラスのコンジュゲートを以下に記載する。
本開示は、試料、例えば、生体試料中の標的を検出するために使用され得る特定の検出プローブに関する。検出プローブは、標的と特異的に結合することができる特異的結合部分を含む。検出プローブは、ラベリングコンジュゲートを通した検出を可能にする一又は複数の特徴を含む。代表的な検出プローブには核酸プローブ及び一次抗体プローブが含まれる。
実例となる実施態様では、ラベリングコンジュゲートは検出プローブに特異的に結合し、酵素により標的を標識するように構成されている。上記のように、第2の種から又はハプテンを含むように構成された検出プローブは、抗種抗体又は抗ハプテン抗体のいずれかにより検出することができる。ラベリングコンジュゲートを構成する1つの手法は、酵素を抗種抗体又は抗ハプテン抗体に直接カップリングすることであった。この種のコンジュゲート(種々のリンカーを含んでいても含んでいなくてもよい)も米国特許第7695929号に記載されている。ラベリングコンジュゲートは1種又は複数の酵素を含む。代表的な酵素には、酸化還元酵素又はペルオキシダーゼが含まれる。シグナリングコンジュゲートは潜在的反応性部分及び発色部分を含む。酵素は潜在的反応性部分の反応性部分への変換を触媒し、反応性部分が生体試料と、標的の近位で又は標的上で直接共有結合する。
本明細書に開示される別の型のコンジュゲートはシグナリングコンジュゲートである。シグナリングコンジュゲートは、本明細書に開示される方法にしたがって、標的を検出するために使用する検出可能なシグナルを提供する。特定の開示実施態様では、シグナリングコンジュゲートは、潜在的反応性部分及び発色団部分を含む。
開示するシグナリングコンジュゲートは、典型的には本明細書に記載する潜在的反応性部分を含む。例えば、潜在的反応性部分は、開示する増幅コンジュゲートのものと同じであっても異なっていてもよいが、各潜在的反応性部分が反応性ラジカル種を形成することができ、本明細書で提供する一般式を有する。式1に示すように、シグナリングコンジュゲートは任意のリンカーを含んでもよい。リンカーを使用する場合、これを本明細書に開示されるリンカーのいずれかから選択することができる。特定の開示実施態様では、シグナリングコンジュゲートの親水性溶液溶解度を改善する及び/又は生体試料上のコンジュゲート官能性を改善するようにリンカーを選択する。特定の開示実施態様では、リンカーはポリエチレングリコールリンカーなどのアルキレンオキシドリンカーであるが、本明細書に開示されるリンカーのいずれをシグナリングコンジュゲートに使用してもよい。

潜在的反応性部分は、触媒活性化を受けて、試料又は他の検出成分と共有結合することができる反応性種を形成するように構成される。触媒活性化は、1種又は複数の酵素(例えば、酸化還元酵素及びペルオキシダーゼ酵素、例えば、西洋ワサビペルオキシダーゼ)により駆動される。これらの酵素は、過酸化物の存在下で、反応性種の形成を触媒することができる。これらの反応性種、例えば、フリーラジカルは、その産生に近位の、すなわち、酵素の近くのフェノール化合物と反応することができる。試料中の利用可能なフェノール化合物は、ほとんどの場合タンパク質内のチロシル残基である。よって、潜在的反応性部分を、ラジカル形成を触媒し、その後反応性部分に生体試料との共有結合を形成させることができるペルオキシダーゼ酵素及び過酸化物(例えば、過酸化水素)の存在下でタンパク質含有試料に添加することができる。
本明細書では本明細書に開示される方法を実施することにより得られるシグナルを増幅するのに適したコンジュゲートも開示する。増幅コンジュゲートは、典型的には潜在的反応性部分、検出可能な標識及び任意のリンカーを含む。
本開示による実例となる組成物は、生体試料及び複数のシグナリングコンジュゲートを含む。特定の開示実施態様では、組成物は、一又は複数の酵素ラベリング標的を含む生体試料を含む。標的を標識するために使用する酵素は、酵素コンジュゲートなどのラベリングコンジュゲートに由来し得る。組成物はまた、1種又は複数の検出プローブをさらに含んでもよい。複数のシグナリングコンジュゲートは本明細書に開示する通りであり、明視野シグナルを提供するように構成される。複数のシグナリングコンジュゲートは、一又は複数の標的の近位で又は標的上で直接共有結合している。特定の開示実施態様では、入射光の約5%以上を吸収することができるシグナリングコンジュゲートのために特定の発色部分を選択することを含む明視野シグナルを提供するように構成される。特定の開示実施態様では、入射光の約20%が吸収され得る。
本明細書では、本明細書に開示するシグナリングコンジュゲートを含むキットの実施態様も開示する。別の実施態様では、キットは検出プローブを含む。別の実施態様では、キットはラベリングコンジュゲートを含む。別の実施態様では、キットは増幅コンジュゲート及び二次ラベリングコンジュゲートを含む。別の実施態様では、キットは過酸化物溶液をさらに含んでもよい。実例となる実施態様では、キットは検出プローブを含む。実例となる実施態様では、キットの試薬を自動化スライド染色プラットホーム上で使用するように構成された容器に入れる。例えば、容器は、使用するように構成されたディスペンサー及びBENCHMARK Series自動化スライド染色器であってよい。
一般的手順及び調製
全ISH検出をVentana Benchmark XTで行った。DNP又はDIGラベリング(0.25ng/ml最終濃度)プローブを1〜3時間ホルムアミド含有緩衝液中でハイブリダイズし、引き続いて2×SSCでストリンジェンシー洗浄(stringency washing)した。西洋ワサビペルオキシダーゼと結合させた抗DNP又は抗DIGモノクローナル抗体(2.5ng/ml最終濃度)によりプローブ検出を媒介した。H2O2(0.003%の最終百分率)を添加してシグナリングコンジュゲート(12.5μM最終濃度)の沈着を触媒した。
各チラミド染料溶液を、Superfrostスライド上にマウントしたホルマリン固定パラフィン包埋Calu−3、ZR75−1及びMCF−7異種移植片組織上のHer2タンパク質に対して、免疫組織化学モデルを使用してマイクロモルからミリモル濃度の範囲で機能性について試験した。Benchmark XT Ventana自動化スライド染色装置を使用して組織を染色した。試験に必要な試薬には、VMSI Her2(4B5)Primary Antibody VMSI製品番号790−2991、UltraMap抗Rb HRP番号760−4315、TSA番号760−121のAmpMap Detection Kit、Hematoxylin II番号790−2208及びBluing Reagent番号760−2037が含まれる。スライドを脱パラフィンし、次いで、細胞調整1溶液(番号950−124)を使用して抗原を回収し、引き続いて一次抗体を37℃で16分間、二次抗体を37℃で16分間添加し、TSA−H2O2(VMSI番号760−4141)を添加してTSA Diluent(番号60900)又はリン酸緩衝食塩水中の単一チラミド溶液を使用及び20分間反応物をインキュベートして増幅した。Hematoxylinの4分間のインキュベーション、引き続いてBluing溶液の4分間のインキュベーションにより各スライドを対比染色し、勾配アルコールを使用して脱水し、カバーガラスで覆った。
明視野白色光顕微鏡を使用して、チラミドシグナルの評価を可視化した。各スライドは、Her2タンパク質高発現についての陽性対照(Calu−3異種移植片)、中間タンパク質レベル対照(ZR75−1異種移植片)及びHer2タンパク質発現についての陰性対照(MCF7異種移植片)で構成されていた。組織染色をヌクレオチド標的に対して行う前に、上記アッセイで特異的染色を有するチラミド溶液を最適染料強度についてさらに試験した。
患者の腫瘍再発の可能性を分類し、特定の療法から利益を得ることができる患者を同定するために、PCR又はマイクロアレイを使用した組織切片の遺伝子発現の照合がうまく使用されてきた。しかしながら、組織ベースのアッセイの値を改善する組織特異性及び細胞状況は、mRNA抽出中に失われてしまう。さらに、切片中に「混入」非腫瘍細胞が存在するために、偽陽性又は陰性結果が生じるおそれがある。よって、組織状況及び特異性並びに細胞−細胞関係を保護しながら、バイオマーカー発現の堅牢で再現性のある評価を可能にするmRNAを標的にする自動化インサイツハイブリダイゼーションアッセイ(mRNA−ISH)が必要とされている。細胞が2つのバイオマーカーのいずれか一方を発現しているが、両方は決して発現していない細胞クローン性を照合する試験に、状況の保護及び細胞−細胞核酸(RNA)混入を最小化する能力が望まれている。
生体試料(例えば、ホルマリン固定パラフィン包埋組織、「FFPE組織」)におけるmRNA−ISHアッセイ有用性の障害には、試料mRNA完全性/到達性及びアッセイ性能に影響を及ぼす試料調製(例えば、組織固定)の変動が含まれる。本開示の一態様は、アッセイ性能及び試料完全性を監視するためにバイオマーカー発現と内部対照遺伝子発現の同時分析を可能にする、FFPE試料のための自動化mRNA−ISHアッセイを開発したというものである。1つの具体的な例によると、臨床的乳がんFFPE組織塊を、INFORM HER2 Dual ISH及びIHCアッセイ(Ventana Medical Systems,Inc.)をそれぞれ使用してHER2遺伝子コピー数及びHer2タンパク質発現について特徴づけた。既知の方法にしたがってqPCRを使用して、ACTB(β−アクチン)に対するHER2 mRNA発現レベルを決定した。遺伝子コピー、タンパク質発現及びqPCR分析の結果を、FFPE試料においてHER2及びACTB mRNAのmRNA−ISH検出を通して得られた結果(図27)と比較した。様々な組織回収条件を使用して、mRNA完全性が損なわれる、試料を同定するための内部mRNA標準の有用性を試験した。
DNP又はDIGラベリング(0.25ng/ml最終濃度)PTEN DNA ISHプローブを1〜3時間ホルムアミド含有緩衝液中でハイブリダイズし、引き続いて2×SSC中でストリンジェンシー洗浄した。西洋ワサビペルオキシダーゼと結合させた抗DNP又は抗DIGモノクローナル抗体(2.5ng/ml最終濃度)によりプローブ検出を媒介した。H2O2(0.003%の最終百分率)を添加することにより、ローダミン−チラミド(12.5μM最終濃度)の沈着を触媒した。図28(A〜B)は、VCAP異種移植片腫瘍細胞中のPTEN DNA ISHプローブを検出するためにこの実施態様を使用して得られた結果を示している。図28(A)は40倍倍率で撮影した画像であり、図28(B)は63倍倍率で撮影した組織の別の領域の画像である。

DNP又はDIGラベリング(0.25ng/ml最終濃度)ERG3’及びERG5’DNA ISHプローブを1〜3時間ホルムアミド含有緩衝液中でハイブリダイズし、引き続いて2×SSC中でストリンジェンシー洗浄した。西洋ワサビペルオキシダーゼと結合させた抗DNP又は抗DIGモノクローナル抗体(2.5ng/ml最終濃度)によりプローブ検出を媒介した。H2O2(0.003%の最終百分率)を添加することにより、ローダミン−チラミド及びDabsyl−チラミドコンジュゲート(12.5μM最終濃度)の沈着を触媒した。
この実施態様は、複数のシグナリングコンジュゲートを使用して前立腺癌細胞におけるERG遺伝子再配列を検出することに関する。
この実施態様は、複数のシグナリングコンジュゲートを使用してCARPUS癌細胞におけるALK遺伝子再配列を検出することに関する。
この実施態様は、複数のシグナリングコンジュゲートを使用してヒト肺がん組織におけるALK遺伝子再配列を検出することに関する。
この実施態様は、第3の色を作成するために、同時に2つの異なる色のシグナリングコンジュゲートを使用して18S RNA標的を検出することに関する。図35(A〜C)は、mRNA ISHアッセイを使用したCalu−3細胞中の遺伝子標的の直接検出を示す顕微鏡写真である。図35(A)はローダミン−チラミドコンジュゲートを使用した18S RNA標的の検出を示している。図35(B)はDABSYL−チラミドコンジュゲートの直接沈着を使用した18S RNA標的の検出を示している。図35(C)はDABSYL−チラミドコンジュゲートとRhod−チラミドコンジュゲートの両方を使用した検出を示している。図35(A)で観察されるシグナルは紫に見え、図35(B)のシグナルは橙に見え、図35(C)のシグナルは赤に見える。図36は、多重化IHCアッセイを使用してCalu−3細胞中のHER2及びP53タンパク質を直接検出することを示す顕微鏡写真である。HER2はDABSYL−チラミドコンジュゲートの直接沈着により検出される。P53はローダミン−チラミドコンジュゲートの直接沈着により検出される。図35(A〜B)に示す2つのシグナリングコンジュゲートを共に使用して第3の組み合わせ、色を産生することができるが、これらの2つの発色体を、各色を特定の標的に割り当てることができる多重化フォーマットで使用することもできる。
Claims (28)
- 生体試料を第1の検出プローブと接触させることと;
前記生体試料を、第1の酵素を含む第1のラベリングコンジュゲートと接触させることと;
前記生体試料を、第1の潜在的反応性部分及び第1の発色部分を含む第1のシグナリングコンジュゲートと接触させることと;
前記生体試料に光を照射することと;
前記第1のシグナリングコンジュゲートの前記第1の発色部分による前記光の吸光度を通して第1の標的を検出することと
を含む、生体試料中の第1の標的を検出する方法であって、
前記酵素が、前記第1の標的の近位で又は前記第1の標的上で直接的に前記生体試料と共有結合する第1の反応性部分への前記第1の潜在的反応性部分の変換を触媒する、方法。 - 前記第1の検出プローブがオリゴヌクレオチド断片又は抗体を含む、請求項1又は2に記載の方法。
- 前記第1のラベリングコンジュゲートが前記酵素とカップリングした抗体を含み、前記酵素が酸化還元酵素又はペルオキシダーゼである、請求項1に記載の方法。
- 前記抗体が抗種抗体又は抗ハプテン抗体である、請求項3に記載の方法。
- 前記第1の反応性部分が、前記生体試料のチロシン残基、前記第1のラベリングコンジュゲート、前記第1の検出プローブ、又はこれらの組み合わせと反応する、請求項1ないし4のいずれか一項に記載の方法。
- 前記第1の検出プローブが、オキサゾールハプテン、ピラゾールハプテン、チアゾールハプテン、ニトロアリールハプテン、ベンゾフランハプテン、トリテルペンハプテン、尿素ハプテン、チオ尿素ハプテン、ロテノイドハプテン、クマリンハプテン、シクロリグナンハプテン、ジニトロフェニルハプテン、ビオチンハプテン、ジゴキシゲニンハプテン、フルオレセインハプテン、及びローダミンハプテンからなる群から選択されるハプテンを含む、請求項2に記載の方法。
- 前記シグナリングコンジュゲートによる前記光の吸光度が、入射光の少なくとも約5%又は入射光の少なくとも約20%の吸光度を含む、請求項1ないし6のいずれか一項に記載の方法。
- 前記第1のシグナリングコンジュゲートによる前記光の吸光度を通して前記標的を検出することが、赤、橙、黄、緑、藍又は紫から選択される第1の着色シグナルを検出することを含み、前記第1の着色シグナルが前記第1のシグナリングコンジュゲートの前記第1の発色部分に関連するスペクトル吸光度に関連する、請求項1ないし7のいずれか一項に記載の方法。
- 前記第1のシグナリング化合物の前記第1の発色部分に関連する前記スペクトル吸光度が、約30nm〜約250nmの間、約30nm〜約150nmの間、約30nm〜約100nmの間又は約20nm〜約60nmの間の半値全幅(FWHM)を有する、請求項8に記載の方法。
- 検出が、明視野顕微鏡又は同等のデジタルスキャナーを使用することを含む、請求項1ないし9のいずれか一項に記載の方法。
- 前記生体試料に光を照射することが、前記生体試料にスペクトルが狭い光源を照射することを含み、前記スペクトルが狭い光源が約30nm〜約250nmの間、約30nm〜約150nmの間、約30nm〜約100nmの間又は約20nm〜約60nmの間の第2の半値全幅(FWHM)を有するスペクトル放射を有する、請求項1ないし10のいずれか一項に記載の方法。
