対比染色は、顕微鏡下において、構造をより容易に視覚化できるように、1つ以上の標的を検出するために薬剤で既に染色された後に、サンプルを後処理する方法である。例えば、免疫組織化学的染色をより明確にするために、カバースリップをする前に、対比染色が任意選択的に用いられる。対比染色は、一次染色とは色が異なる。ヘマトキシリン、エオシン、メチルグリーン、メチレンブルー、ギムザ、アルシアンブルー、及びNuclear Fast Redなど、多くの対比染色がよく知られている。DAPI(4’,6-ジアミジノ-2-フェニルインドール)は、使用することができる蛍光染色剤である。
開示される実施態様のうちのある特定の態様、又は全ての態様は、自動化することができ、コンピュータ解析及び/又は画像解析システムによって促進することができる。幾つかの用途では、正確な色又は蛍光比が測定される。幾つかの実施態様では、光学顕微鏡が画像分析に利用される。ある特定の開示される実施態様は、デジタル画像の取得に関する。これはデジタルカメラを顕微鏡に接続することによって行うことができる。染色されたサンプルから得られたデジタル画像は、画像解析ソフトウェアを用いて解析される。色又は蛍光はいくつかの異なる方法で測定することができる。例えば、色は、赤、青、及び緑の値として;色相、彩度、明度の値として;及び/又は、分光撮像カメラを使用して、特定の波長又は波長範囲を測定することによって、測定することができる。サンプルはまた、定性的及び半定量的に評価することもできる。定性的評価には、染色強度の評価、陽性染色細胞及び染色に関与する細胞内コンパートメントの識別、及び全体的なサンプル又はスライドの品質の評価が含まれる。別々の評価が、試験サンプルに対して行われ、この分析には、サンプルが異常状態を表すかどうかを決定するために、既知の平均値と比較することが含まれうる。
サンプルは、生物学的成分を含み、かつ、概して1つ以上の目的とする標的分子を含むことが疑われる。標的分子は、細胞の表面上にあってもよく、細胞は懸濁液中又は組織切片中に存在しうる。標的分子はまた、細胞内に存在してもよく、細胞溶解又はプローブによる細胞の浸透の際に検出することができる。当業者は、サンプル中の標的分子を検出する方法が、用いられるサンプル及びプローブの種類に応じて変わることを理解するであろう。サンプルを収集及び調製する方法は当技術分野において知られている。
ヤギ抗マウス(GAM)、ヤギ抗ウサギ(GAR)、及びマウス抗DIGに対するPNAの例示的なコンジュゲーションが、以下に記載される:
PNAコンジュゲート抗体をUV-Vis吸光度で特徴付けた。抗体-PNAコンジュゲートにおける260nmでの吸光度の増加は、PNAの首尾よい取り込みを示唆した(図2参照)。A260対A280の比は、コンジュゲーションの効率を定性的に評価し、かつ、抗体あたりのPNAオリゴマーの数を潜在的に推定するために用いられうる。
短PNA(15塩基のPNA、10塩基のPNA)オリゴのコンジュゲーションが抗体の結合親和性及び特異性に影響を及ぼさないことを証明するために、PNA-コンジュゲート二次抗体を用いて、異なるマーカーについて扁桃組織に対するIHCアッセイを行った(一次及び二次)。IHCアッセイは全て、特定のアッセイに基づいて修正されたプロトコルを使用して、Benchmark XT(Ventana社)で行われた。PNA-コンジュゲート抗体を、滴定として100μLの体積で、手動で加えた。
幾つかの実施態様では、PNA-コンジュゲート抗体は、Gyros技術又はDNA計数に基づくNanoString nCounterプラットフォームなどの方法によるタンパク質発現の多重定量測定を可能にするように設計されている。したがって、PNA配列は、エクスサイチュPNA計数を可能にするために組織に結合した後にPNAコンジュゲート抗体から切断しなければならない。この特定の例では、PNA配列は、ジスルフィド結合を介して抗体に結合し、これは、(例えば、ジスルフィド結合の還元によって)化学的に切断されて、PNA配列の放出をもたらすことができる。
扁桃スライドを一次抗体(ウサギKi67)、二次抗体(GAR-PL-PNA、ビオチンを有する光切断可能なPNAを有するヤギ抗ウサギ抗体)で処理した。その後、スライドを、異なる時間の間、UV光(ハンドヘルドUVランプ、365nm)で照射した。対照スライドはUV処理せず、比較のために使用した。次いで、全てのスライドを検出用にSA-HRP及びDABで処理した。
発色検出の他に、コンジュゲート中のビオチンは、SA-フルオロフォアを用いて蛍光的にも検出されうる。図6は、一次抗体、その後、PNAをコンジュゲートした二次抗体を用いて、Ki67について染色し、SA-FITCで検出した扁桃スライドの蛍光画像を示している。SA-FITCは、PNA配列のビオチンに結合する。蛍光シグナルはKi67マーカーの局在と一致している。この実験によって、PNAコンジュゲート抗体がもたらす検出スキームの多様性が実証された。
特定の理論に縛られることを望むものではないが、NanoString nCounterプラットフォームは、最大800種類の異なるDNA配列を潜在的に検出する能力を有しており、抗体PNAコンジュゲートを使用することで、IHCの非常に大きな多重化能力を可能にすることが報告されている。我々は、検出スキームがレポーター鎖の標的オリゴ(おそらくはDNA又はPNAでありうる)へのハイブリダイゼーションに基づいていることから、PNAはDNAと同様の方式で検出することができると仮定している。しかしながら、PNAの3’末端上のビオチンの存在は、捕捉鎖を使用する必要性を排除するため、PNAオリゴマーは、NanoString nCounterプラットフォームで典型的に用いられる、標準的なDNA標的(典型的には約100塩基の長さ)よりも著しく短くできると考えられる。さらには、DNA対DNAと比較して、DNAに対するPNAのより高い結合親和性は、比較的短いPNA/DNAレポーター二本鎖に対して十分な安定性をもたらすと考えられる。
先の実験では、PNAはビオチン標識に対する染色によって検出された。異なる標識(発色性、蛍光発生性等)を担持する相補的DNA又はPNA配列は、代替的な染色手法として使用することができる。この実験では、扁桃スライドを、ウサギ抗Ki67一次抗体、その後、PNAをコンジュゲートしたGARと共にインキュベートした。その後、スライドを蛍光標識を有するPNAに相補的なDNA配列(例えば、FITC又はローダミン)と共にインキュベートした。図8は、スライドがDNAハイブリダイゼーションによって首尾よく染色されたことを示している。蛍光染色はマーカーの局在と一致し、バックグラウンドシグナルは観察されなかった。DNA配列を、約185nMの濃度及び約100μLの容量の手動滴定として加えた。

この実験では、相補的DNAをハプテン(DIG)で標識した。2つの扁桃スライドを、ウサギ抗Ki67及びマウス抗CD45、その後、それぞれPNAコンジュゲートGAR及びPNAコンジュゲートGAMと共にインキュベートした。DIGで標識した相補的DNAを両方のスライドと共にインキュベートし、その後、HRPを抗DIG抗体とコンジュゲートし、DAB沈着を行った。DNAを、約185nMの濃度及び約100μLの容量で手動の滴定ステップとして加えた。

Gyros技術を使用したPNAの定量化を実証するため、約0.0274nMから約20nMの範囲の濃度のビオチン化PNAオリゴ(Bt-PNA)を試験した(表1)。各濃度について、1:2の比のBt-PNAとのジゴキシゲニンで標識した相補的一本鎖DNA(DNA-DIG)を、最初に室温でハイブリダイズさせた後、二連のGyrosによって分析した。検出は、Ms-抗DIG、その後にAlexaFluor647-GAMを使用して達成した。Gyrosプログラムを使用して四点曲線(表1参照)を当てはめ、0.998を超えるR2を有する標準曲線を生成した。特に、最低PNA濃度のシグナル対バックグラウンド比(S/B)は、約10であり、PNA又はssDNAによる非常に低いバックグラウンドシグナルが示唆された。
