(実施例1 シロイヌナズナ(Arabidopsis thaliana)Atmlo変異体植物の相補)
(材料および方法)
メロンCmKIP遺伝子の完全長cDNAを、標準のクローニング技術を使用して、pBINPLUSまたはGatewayベクターで、35Sプロモーターの制御下でクローン化した。
同定されたCmKIP遺伝子の、ウドンコ病に対する感受性への予測していた関与を確認するために、CmKIP1、CmKIP2、CmKIP3、およびCmKIP4の完全長cDNA配列を、ユビキタス35Sプロモーターを使用して、シロイヌナズナ(Arabidopsis)内で発現させた。さらに、完全長ゲノムフラグメントも、35Sプロモーターを使用して、CmKIP2について発現させた。
Claims (11)
- ウドンコ病抵抗性付与遺伝子であって、該抵抗性付与遺伝子によってコードされるアミノ酸配列が、配列番号2、配列番号4、配列番号6、配列番号8、配列番号10、配列番号12、および配列番号14、ならびに70%超の同一性、好ましくは80%超の同一性、より好ましくは90%超の同一性、最も好ましくは95%超の同一性を有するアミノ酸配列からなる群から選択され、かつ、該抵抗性付与遺伝子が損傷している、ウドンコ病抵抗性付与遺伝子。
- ウドンコ病抵抗性付与遺伝子であって、該抵抗性付与遺伝子から転写されるcDNA配列が、配列番号1、配列番号3、配列番号5、配列番号7、配列番号9、配列番号11、配列番号13、および配列番号17、ならびに70%超の同一性、好ましくは80%超の同一性、より好ましくは90%超の同一性、最も好ましくは95%超の同一性を有するcDNA配列からなる群から選択され、かつ、該抵抗性付与遺伝子が損傷している、ウドンコ病抵抗性付与遺伝子。
- 前記損傷が、タンパク質発現産物の非存在をもたらす、前記遺伝子における1つまたは複数の変異である、請求項1または請求項2に記載のウドンコ病抵抗性付与遺伝子。
- 前記損傷が、非機能性タンパク質発現産物をもたらす、前記遺伝子における1つまたは複数の変異である、請求項1または請求項2に記載のウドンコ病抵抗性付与遺伝子。
- 前記損傷が、遺伝子サイレンシングである、請求項1または請求項2に記載のウドンコ病抵抗性付与遺伝子。
- メロンから得られるものである、請求項1〜5のいずれかに記載のウドンコ病抵抗性付与遺伝子。
- 請求項1〜6のいずれかにおいて定義される損傷したウドンコ病抵抗性付与遺伝子をそのゲノム内に含むメロン植物であって、前記損傷がウドンコ病抵抗性をもたらすものである、メロン植物。
- 請求項1〜6のいずれかにおいて定義される損傷したウドンコ病抵抗性付与遺伝子をそのゲノム内に含む、請求項7に記載の植物の種子、植物部分または繁殖材料であって、前記損傷が前記植物におけるウドンコ病抵抗性をもたらすものである、種子、植物部分または繁殖材料。
- 配列番号1、配列番号3、配列番号5、配列番号7、配列番号9、配列番号11、および配列番号13、ならびに70%超の同一性、好ましくは80%超の同一性、より好ましくは90%超の同一性、最も好ましくは95%超の同一性を有するヌクレオチド配列からなる群から選択される、単離されたヌクレオチド配列。
- 配列番号2、配列番号4、配列番号6、配列番号8、配列番号10、配列番号12、および配列番号14、ならびに70%超の同一性、好ましくは80%超の同一性、より好ましくは90%超の同一性、最も好ましくは95%超の同一性を有するアミノ酸配列からなる群から選択される、単離されたアミノ酸配列。
- ウドンコ病抵抗性のメロン植物を提供するための、請求項1〜6のいずれかにおいて定義されるウドンコ病抵抗性付与遺伝子、請求項9に記載の単離されたヌクレオチド配列、または請求項10に記載の単離されたアミノ酸配列の使用。
