Abstract
【解決手段】インビトロおよびインビボの両方で安定であるジケトピペラジン(DKP)の医学的に許容し得る塩と組み合わせたグルカゴン様ペプチド1(GLP−1)との微小粒子を含有する肺内投与のための乾燥粉末組成物。前記ジケトピペラジンの医薬的に許容し得る塩は、式:2,5−ジケト−3,6−ジ(4−X−アミノブチル)ピペラジンを有し、式中Xは、スクシニル、グルタリル、マレイルおよびフマリルからなる群から選択され、好ましくは、2,5−ジケト−3,6−ジ(4−フマリル−アミノブチル)ピペラジン二ナトリウム塩である。

Description
本出願は、2003年7月22日に出願の米国特許出願第10/632,878号の一部継続出願であり、米国特許法第119条(e)項の規定により、2006年4月14日に出願の米国仮特許出願第60/744,882号に対する利益を請求するものである。上述の各優先権出願は、参照によりその全体が本明細書に組み込まれる。
本発明は医薬製剤の分野に関する。本発明は、グルカゴン様ペプチド1(GLP−1)と組み合わせたジケトピペラジン(DKP)粒子を含む乾燥粉末製剤を開示するものである。本発明は、糖尿病や、癌、肥満等の疾患を治療するための医薬製剤として有用であるが、そのような疾患に限定されない。更に詳しくは、本発明は、肺内送達のための医薬製剤として有用である。
文献にて開示されているように、グルカゴン様ペプチド1(GLP−1)は、30個または31個のアミノ酸からなるインクレチンであり、食事由来の脂肪、炭水化物摂取およびタンパク質に応答して腸内分泌L細胞から放出されるものである。このペプチドホルモンの分泌は2型糖尿病の個体において低下することが見出され、2型糖尿病においては、このペプチドホルモンは、その疾患や他の関連疾患の治療のための候補物質となり得る。
医薬品として使用するための安定した、吸入可能なグルカゴン様ペプチド1(GLP−1)製剤が当技術分野には不足している。当技術分野における不足を解決するために、本発明は、医薬品としてジケトピペラジン(DKP)粒子と組み合わせたGLP−1の製剤を提供するものである。

当技術分野において、医薬品に使用するための安定した吸入可能なグルカゴン様ペプチド1(GLP−1)製剤が不足している。これは、インビボにおけるGLP−1ペプチドの不安定性に起因する。GLP−1化合物は、多くの条件下で溶液中に残存する傾向があり、溶液製剤として投与される場合、比較的短いインビボ半減期を有する。さらに、各種体液(例えば肺液や血液)中に存在することが分かっているジペプチジルペプチダーゼIV(DPP−IV)は、GLP−1分子の生物学的半減期を大きく減少させる。例えば、GLP−1(7−37)の生物学的半減期は、3〜5分であることが示されている(米国特許第5,118,666号を参照のこと)。GLP−1はまた、非経口投与後にインビボにおいて迅速に吸収されることが示されている。同様に、アミドGLP−1(7−36)は、皮下投与される場合、約50分の半減期を有する(やはり米国特許第5,118,666号を参照のこと)。
本発明の特定の態様においては、フマリルジケトピペラジン(FDKP)等のジケトピペラジンと組み合わせた天然のヒトグルカゴン様ペプチド1(GLP−1)を含む最適化された製剤が提供される。本発明のそのようなGLP−1/FDKP製剤は、分解に対して安定的で且つ抵抗性がある。
ジケトピペラジンは、薬物送達および安定化に有用な微小粒子を形成するその能力のためによく知られている。本発明において、ジケトピペラジンは、GLP−1分子の吸収を容易にし、それによって、分解に抵抗性のある安定した製剤を提供するために使用される。
本発明は、治療を必要とする被験体に投与するためのGLP−1/FDKP製剤をさらに提供するものである。本発明で検討される被験体とは、家庭のペットまたはヒトであり得る。特定の態様においては、その治療は、II型糖尿病、肥満、癌またはそれらから生じるあらゆる関連疾患もおよび/または関連症状に対してなされる。ヒトは、特に好ましい被験体である。
以下の実施例は、本発明の特定の態様を示すためのものである。実施例において開示される手法は、本発明を実施する際に十分な機能を果たす代表的手法を説明するものであることは、当業者に認められるはずである。しかしながら、当業者は、本発明の開示に鑑みて、多くの変更は、開示される特定の態様においてなすことが可能であり、本発明の精神と範囲から逸脱することなく、類似または同様の結果をさらに得ることが可能であることを認める。
GLP−1の構造の生物物理学的および解析的な分析
GLP−1の構造および作用を解析するために、多くの生物物理学的および解析的な手法を使用した。これらの手法には、遠紫外円偏光二色性(遠紫外CD)、近紫外円偏光二色性(近紫外CD)、内部蛍光、フーリエ変換赤外分光法(FTIR)、高速液体クロマトグラフィ(HPLC)および質量分析(MS)が含まれ、それらのすべてが当業者によく知られている。GLP−1ペプチドに対する濃度、イオン強度、温度、pH、酸化ストレス、撹拌および複数の凍結融解サイクルの作用を調べるために広範囲にわたる条件が使用され、それらのすべては、さらに下記で詳述されている。また、特定のGLP−1/DKP製剤を実現するために、これらの解析を用いて分解の主要な経路を明らかにし、GLP−1のペプチド構造を操作する条件を確立した。
GLP−1は、American Peptide(米国カリフォルニア州サニーヴェール)またはAnaSpec(米国カリフォルニア州サンノゼ)のいずれかから購入するか、あるいは社内(MannKind Corporation、米国カリフォルニア州バレンシア)で調製した。各種濃度の水性GLP−1試料は、特に明記しない限りpH4.0および20℃で解析した。試料は、各実験前に、一般に新しく調製し、適切な添加剤(あるとすれば、例えば、塩、pH緩衝液、H2O2等)と混合した。さらに、各種条件下のGLP−1の2次構造測定値を、遠紫外CDおよび透過フーリエ変換赤外分光法(FTIR)により収集した。さらに、近紫外CDおよび内部蛍光を用いて、GLP−1の3次構造を、その芳香族残基(すなわちトリプトファン)を囲む環境をモニターすることによって解析した。
円偏光二色性(CD)スペクトルを用いて、タンパク質やペプチド等の分子で表わされるα−ヘリックス、ランダムコイル、β−プリーツシート、β−ターンおよびランダムコイルを解析した。特に、遠紫外CDは、タンパク質およびペプチドの2次構造の種類、例えば純粋なα−ヘリックスやβ−シート等を決定するために用いた。一方で、近紫外CDは、分子の3次構造を解析するために用いた。従って、GLP−1構造に対する濃度の作用を調べるために、遠紫外CDおよび近紫外CDの手法を用いた。
また、GLP−1ペプチドに対するイオン強度の作用を決定するために試験を行った。図2A(遠紫外CD)は、208nmおよび224nmでの極小値により明らかとなる通り、塩の濃度(100mM〜1000mM)を増加させることによって、GLP−1の不規則構造がα−ヘリカル構造に変化することが示される。NaCl濃度を1Mまで上げると、ペプチドの多く(1.0mg/mL)が溶液から析出する(図2A)。しかし、この種の析出物は、水で希釈すると溶解することが明らとなったため、高イオン強度でGLP−1は可逆的に析出する可能性があることが立証された。
また、GLP−1の自己会合した立体配座が温度またはpHのいずれかの変化に対する感受性を有するかどうかを決定するために、試験を行った。図3A(近紫外CD)により、温度が上昇するにつれて、10mg/mLのGLP−1の3次構造が有意に解離することが明らかにされる。一方では、温度は、各種温度およびpH4.0での低濃度(0.05mg/mL)GLP−1に影響を及ぼさない(図3Bおよび3C(遠紫外CD)を参照)。遠紫外CDにより、ペプチドが温度に非感受性であることが示される。それ故、分子運動の増加は、GLP−1の自己会合を有意に妨げる。
撹拌および凍結融解サイクルの作用に加えて、アミド分解および酸化に対するその耐性を決定することによってGLP−1ペプチドの安定性を調べた。
GLP−1/FDKPの吸着試験
吸着試験を実施することによって、GLP−1と懸濁液中のジケトピペラジン(DKP)粒子との相互作用を評価した。吸着試験の変数は、GLP−1/DKP相互作用に対する静電気、水素結合、水構造、タンパク質の柔軟性の作用、および特定の塩結合相互作用について調べるものであった。さらに、いくつかの一般的なタンパク質安定剤を、DKP表面へのGLP−1吸着に対する干渉についてテストした。
FDKP粒子へのGLP−1の結合に対する塩の作用を、HPLC解析によって観察した。GLP−1/FDKP粒子の負荷を、0、25、50、100、250、500、1000および1500mMのNaClの存在下で、5mg/mLのFDKPおよび0.25mg/mLのGLP−1で実施した(図8A)。また、pHおよびNaCl濃度の関数としての、再組成された、FDKPを含まない対照試料中に検出されるGLP−1の量を評価した(図8B)。両方のデータセットのpHを、20mMリン酸塩/20mM酢酸塩混合物により制御した。
FDKP粒子へのGLP−1の結合に対する界面活性剤の作用は、HPLC解析によって観察した。負荷は、界面活性剤の存在下で5mg/mLのFDKPおよび0.25mg/mLのGLP−1で行った(図9A)。また、pHおよび界面活性剤濃度の関数としての、再組成された、FDKPを含まない対照試料において検出されたGLP−1の量を評価した(図9B)。pHおよび対照試料の条件は、上記のイオン強度試験について記載した通りであった。本試験で使用した界面活性剤としては:0.09mMのBrij78、0.01mMのTween80、0.2mMのTritonX、0.12mMのPluronic(プルロニック)F68、0.9mMの、H(CH2)7SO4Na、0.9mMのCHAPS、0.9mMのCetrimide(セトリミド)が含まれた。各界面活性剤の存在下のGLP−1についての負荷曲線は、pHの関数としてGLP−1/FDKPについて示した。
この実験のために、2種の異なるイオンの試験を行ってFDKP粒子へのGLP−1の結合に対するイオンの作用を決定した。両試験において、Cl−はカチオンの対イオンであり、Na+はアニオンの対イオンであった。GLP−1/FDKP粒子の負荷を、前の実験について記載した通りに実施した。前述の通りにpHを制御した。試料は、NaCl(高イオン強度の場合のより優れた評価のために用いた)の存在下および非存在下におけるpH3.0、3.5、4.0または5.0のいずれかのpH緩衝液により調製した。さらなるイオンは、以下の通り、個々の試料中に含まれた:20mMまたは250mMのLiCl、20mMまたは250mMのNH4Cl、20mMまたは250mMのNaF、および20mMまたは250mMのNaCH3COO。
また、FDKP粒子へのGLP−1の結合に対するオスモライトの作用を、HPLC解析により観察した。図11Aは、一般の安定剤(オスモライト)の存在下におけるpHの関数としてのGLP−1/FDKPについての負荷曲線を示す。GLP−1/FDKP粒子の負荷を、前の実験について記載した通りに実施した。同様に、前述で記載した通りにpHを制御した。試料を、pH3.0、且つ20、50、100、150、200または300mMのオスモライト(安定剤)の存在下で調製した。オスモライトは、ヘキシレングリコール(Hex−Gly)、トレハロース、グリシン、PEG、TMAO、マンニトールまたはプロリンであり、「N/A」は、オスモライトが存在しないことを示す。同様の実験においては、試料中のオスモライト(安定剤)の濃度を100mMで一定に維持し、pHを2.0から4.0に変化させた。
水およびタンパク質の構造に影響を及ぼすイオン種を(カオトロープおよびリオトロープ)、これらの因子がFDKPへのGLP−1吸着に果たす役割を決定するために検討した。GLP−1/FDKP粒子の負荷を、前の実験について記載した通りに実施した。同様に、前述の通りにpHを制御した。試料を、pH3.0、且つ0、20、50、100、150、200または300mMの以下のカオトロープまたはリオトロープ:NaSCN、CsCl、Na2SO4、(CH3)3N−HCl、Na2NO3、クエン酸ナトリウムおよびNaClO4の存在下で調製した。同様の実験においては、試料中のカオトロープまたはリオトロープの濃度を、100mMで一定に維持し、pHを2.0から4.0に変化させた。
水素結合の強度を増加させることによって構造化されていないペプチドのヘリカル構造を誘導することが知られているアルコールを、ヘリカル構造がFDKPへのGLP−1吸着において果たす役割を決定するために評価した。GLP−1/FDKP粒子の負荷を、前の実験について記載された通りに実施した。同様に、前述で記載された通りにpHを制御した。各アルコールの作用を、pH2.0、3.0、4.0および5.0で観察した。使用したアルコールは、メタノール(MeOH)、エタノール(EtOH)、トリフルオロエタノール(TFE)またはヘキサフルオロイソプロパノール(HFIP)であった。各アルコールを、5%、10%、15%および20%v/vの濃度で評価した。
各種濃度におけるGLP−1のFDKP粒子表面への吸着を調べた。図14Aは、各種pHでのGLP−1濃度の関数としてのGLP−1/FDKPからの負荷曲線を示すものである。GLP−1濃度は、0.15、0.25、0.4、0.5、0.75、1.0、1.5、2.0、5.0または10mg/mLであった。試料のpHは、2.5、3.0、3.5、4.0、4.5または5.0であった。
全体的に、GLP−1のFDKP粒子との相互作用についての吸着試験によって、GLP−1が、pH4以上で、高吸着で、pHに依存する様式でDKP粒子表面に結合することが示された。DKP粒子表面へのGLP−1の吸着はpHに最も強く影響を受けることが明らかになったが、pH2.0では本質的な吸着は生じず、pH4.0以上で実質的な相互作用が生じた。観察の際、ナトリウムイオンおよびフッ化物イオンによって、低pHで吸着が高まった。界面活性剤等の他の添加剤、および一般の安定剤の、FDKP粒子表面へのGLP−1の吸着に対する作用は、ほんのわずかだった。
GLP−1/FDKP製剤の完全性解析
実施例1および2の実験からの結果に基づいて、表1に記載した特性を有する一連のGLP−1製剤を、本明細書で考察される通りの細胞生存率アッセイのために選択した。大部分の製剤は、GRASの(「一般に安全と認められる」)賦形剤を含有していたが、いくつかは、安定性および吸着の関係を調べることができるように選択した。
肺洗浄液中でインキュベートしたGLP−1の安定性
体液中に見出されるジペプチジルペプチダーゼIV(DPP−IV)がGLP−1を切断および不活性化することから、肺液や血液等の体液中でのGLP−1の安定性を解析した。
アポトーシスおよび細胞増殖に対するGLP−1分子の作用
GLP−1がアポトーシスを抑制するかどうかを検討するために、スクリーニングアッセイを行ってβ細胞死の抑制に対するGLP−1の作用を決定した。ラット膵上皮(ARIP)細胞(膵臓β細胞モデルとして使用;ATCC(米国バージニア州マナッサス)から購入)に、10分間、0、2、5、10、15または20nMの濃度でGLP−1を前もって処理した。次いで、細胞に何も投与しないか、または4.5時間、5μMのスタウロスポリン(アポトーシス誘導物質)を投与した。細胞生存度を、Cell Titer-Glo(登録商標)(プロメガ、米国ウィスコンシン州マディソン)を用いて評価した。細胞死の低下率は、スタウロスポリンが投与された細胞における最高10nMまでのGLP−1濃度の増加によって示した(図19A)。
細胞死に対する候補物質GLP−1/FDKP製剤の作用
細胞に基づくアッセイを行って、GLP−1/FDKP製剤(上記実施例3の表1に開示した通り)が細胞死を抑制する能力を評価した。これらのGLP−1/FDKP粒子製剤は、懸濁液の形態であるか凍結乾燥されている。製剤は、ARIP細胞におけるスタウロスポリンに誘導される細胞死を抑制する能力について解析した。GLP−1試料を前もって投与したARIP細胞を、5μMスタウロスポリンに4時間曝露し、Cell Titer-Glo(登録商標)(プロメガ、米国ウィスコンシン州マディソン)で解析して細胞生存度を決定した。
GLP−1/DKP粒子の肺吸入
GLP−1/FDKPの薬物動態を検討するために、静脈内注射または肺内吸入によりGLP−1/FDKPの各種製剤が投与された雌性Sprague Dawleyラットにおいて血漿中GLP−1濃度を評価した。予備試験において、GLP−1/FDKP粒子製剤の約4%および16%(w/w)のGLP−1を使用した。ラットは、肺内液体滴下注入またはIV注射により投与されたGLP−1溶液の投与を受けた群1、4、7および10を含む12の群に無作為割付けした。群2、5、8、および11は、肺内吸入またはIV注射により投与されたGLP−1/FDKP塩と関連した製剤(表2にて開示)の投与を受けた。群3、6、9、12は、肺内吸入またはIV注射により投与されたGLP−1/FDKP塩と関連した混合製剤の投与を受けた。GLP−1/DKP製剤は、約16%の負荷の塩関連製剤だった。約4%の負荷を達成するために、16%の製剤をDKP粒子と3:1の混合物の形態で混合した。肺内吸入または静脈内注射は、0.08mgのGLP−1総投与量のために0.5mgまたは2.0mgの粒子(それぞれ16%または4%のGLP−1負荷)で行った。
GLP−1/FDKPはラットの食物摂取を減少させる
GLP―1はまた、満腹感の誘発や食物摂取の減少の機能を脳内で果たすことが当技術分野で知られている。満腹感や食物摂取の減少におけるGLP−1のこの役割に基づいて、本発明のGLP−1/FDKP製剤が摂食を減少させ、それにより肥満を制御することが可能になるための作用剤として効果的であったかどうかを決定するために実験を行った。
毒性試験
複数投与が行われた後、GLP−1/DKPの毒性作用および毒物動態学プロファイルの可能性を評価するために、反復投与毒性試験を行った。ラットの14日間の試験、およびサルの28日間の試験を実施した。GLP−1/DKPを、吸入経路により毎日投与する。動物に28日間投与した試験においては、一部の動物を投与計画直後に屠殺するが、一方で他の動物には、屠殺前に1ヵ月間の回復期間を与える。すべての動物は、臨床徴候、各種生理学的パラメーター(GLP−1、グルコース、インスリン、臓器重量、臨床病理学等)および各種臓器の組織病理について評価する。
β−細胞集団に対するGLP−1の作用
GLP−1は、インスリン生合成におけるすべてのステップを促進し、β細胞の成長および生存ならびにβ細胞の分化を直接刺激ことが知られている。これらの作用の組み合わせにより、β細胞集団の増大が生じる。さらに、GLP−1受容体シグナル伝達により、β細胞アポトーシスの減少が生じ、それにより、β細胞集団の増大がさらにもたらされる。GLP−1は、3種の可能な経路(β細胞の細胞増殖の増大、アポトーシスの抑制、導管上皮の推定上の幹細胞(putative stem cell)の分化)によりβ細胞集団を調節することが知られている。
GLP−1/FDKP粒子製剤の調製
GLP−1/FDKP粒子製剤を調製するための代替方法も使用した。製剤は、以下の通りに調製した:1(重量)部のGLP−1を9部の脱イオン水に添加し、次いで少量の氷酢酸を添加して透明な溶液を得ることによって、10重量%のGLP−1原液を調製した。FDKP粒子(約10重量%の粒子)の懸濁原液を三つに分割した。適切な量のGLP−1原液を各懸濁液に添加して、乾燥粉剤中に5重量%および15重量%のGLP−1を有する目的の組成物が得られた。タンパク質溶液の添加後、懸濁液のpHは約3.5だった。次いで懸濁液を約pH4.4〜4.5に調節し、その後、懸濁液を液体窒素中で小球形にして、次いで凍結乾燥して氷を除去した。
各種GLP−1濃度のGLP−1/FDKPの薬物動態
各種GLP−1濃度のGLP−1/FDKPの薬物動態学的特性を評価するために、体重が192.3グラム〜211.5グラムの18体の雌性SDラットを対照群GLP−1(群1、n=3)およびGLP−1/FDKP製剤群(群2〜4、n=5/群)の4つの投与群に分割した。動物は、以下の被験物質の内の1つの投与を受けた。肺内滴下注入による対照(空気)、肺内吸入による、5%のGLP−1(0.12mgのGLP−1)を含有する2.42mgのGLP−1/FDKP、10%のGLP−1(0.19mgのGLP−1)を含有する1.85mgのGLP−1/FDKP、または15%のGLP−1(0.37mgのGLP−1)を含有する2.46mgのGLP−1/FDKP。血液試料は、投与前および投与後の各時点(2、5、10、20、30、40および60分)に、血清FDKPおよび血漿GLP−1濃度のために採集および試験した。
肺内吸入により投与されたGLP−1/FDKPの薬力学的特性
GLP−1/FDKPの薬力学的性質を評価するために、雌性Sprague Dawleyラットを2つの投与群に分割した。動物は、1日1回の肺内吸入により、対照(空気;n=5)または15%GLP−1(0.3mgのGLP−1)を含有する2mgのGLP−1/FDKP(n=10)のいずれかの投与を4日間連続で受けた。
毒物動態学(TK)試験
下記の実施例14〜16は、GLP−1/FDKP吸入粉剤の毒性作用の可能性および毒物動態学プロファイルを評価するためにラットおよびサルにおいて実施された反復投与毒性試験を示すものである。そのデータによって、臨床用に提案されるものより数倍大きな投与量のGLP−1/FDKP吸入粉剤で明らかな毒性はないことが分かる。さらに、各々の種の中の雄性動物と雌性動物との間に差はないと思われる。
肺内吸入により5日間投与されたサルにおけるGLP−1/FDKPの毒物動態学
1日1回(1日につき30分間)、5日間連続のカニクイザル(Macaca fascicularis)への経鼻・経口投与(目的のヒト治療用投与経路)によるGLP−1/FDKPの毒性および毒物動態学プロファイルを決定するために試験を実施した。経鼻・経口投与では、サルの口および鼻をマスクで覆い、30分間試験製剤を吸い込ませた。
肺内吸入によて14日間投与されたラットにおけるGLP−1/FDKPの毒物動態学
この試験は、14日間連続の肺内吸入による毎日の投与後のGLP−1/FDKPの毒性の可能性を評価したものである。ラットは、対照(空気)、10mg/kgのFDKP粒子、1mg/kgのGLP−1/FDKP(0.15mgのGLP−1)、3mg/kgのGLP−1/FDKP(0.45mgのGLP−1)または10mg/kgのGLP−1/FDKP(1.5mgのGLP−1)を、14日間連続で毎日の肺内吸入としてラットに投与した(n=24/性別/群)。動物は、毒性の臨床徴候について毎日観察し、体重および摂食量も記録した。
サルの肺内吸入により28日間投与されたGLP−1/FDKPの毒物動態学
この試験は、少なくとも4週間、吸入により毎日投与されたGLP−1/FDKPの毒性および毒物動態学を評価したものである。作用の可逆性、持続性または遅延性の発現を評価するために、4週間の回復期間を設けた。
エキセンジン/FDKP製剤の調製
エキセンジン−4/FDKPを、酸性エキセンジン−4ペプチド(配列番号3)溶液をFDKP粒子懸濁液と混合することにより調製した。酸性ペプチド溶液は、2%酢酸に10%(w/w)のペプチドが溶解していた。FDKP懸濁液は、約10%(w/w)のFDKP粒子を含有していた。酸性エキセンジン−4ペプチド溶液を、徐々に混合するようにFDKP粒子懸濁液に添加した。エキセンジン−4/FDKP混合物を25%のアンモニア溶液でpH4.50に徐々に滴定した。次いで混合物を液体窒素中で小球形にして、凍結乾燥した。
肺内吸入により投与されたエキセンジン/FDKPの薬物動態
各種濃度のエキセンジン−4/FDKP製剤における、肺内経路を経た複数回投与後のエキセンジン−4(GLP−1類似体)の薬力学的および薬物動態プロファイルを検討するための反復投与予備的毒性試験が進行中である。
肺内吸入により投与されたラットにおけるGLP−1/xDKPの薬物動態
異なるDKPがGLP−1/FDKP製剤の薬物動態プロファイルに影響を及ぼし得るのかどうかを決定するために、各種GLP−1/xDKP製剤を、本明細書と同日付で出願された"Asymmetrical FDKP Analogs for Use as Drug Delivery Agents"という名称の米国仮特許出願(その全体が本明細書に組み込まれる(米国代理人側整理番号51300−00041))に開示されたように調製した。
健康な成人男性被験者におけるGLP−1/FDKP吸入粉剤の第1a相、単回、非盲検、漸増用量、比較対照安全性および忍容性試験
GLP−1は、静脈内(iv)または皮下(sc)注入によって、あるいは複数の皮下注射によって投与される場合、ヒトの高血糖を制御することが示されている。ホルモンの半減期が極めて短いため、連続皮下注入または複数の毎日の皮下注射が必要とされる。これらの経路のいずれも、長期にわたる臨床用途としては実際的ではない。動物の実験により、GLP−1が吸入により投与される場合は治療レベルが達成され得ることが明らかになった。
