Abstract
Description
本出願は、その全開示を本明細書に参照により組み入れる、2010年6月17日出願の米国仮出願第61/397,940号「受容体、作用剤、疾患の治療および症状」に関する。
米国政府は、本発明において米国国立補完代替医療センター(NCCAM)との間に契約番号(1 P01 AT003960)による権利を有する。
GEnSeMBLEおよびGenDock法およびシステムを用い、苦味物質受容体の3D構造の予測を行った。
TAS2R38受容体の配列に図1に示すシチメンチョウβ1アドレナリン受容体(thβ1AR)とのアラインメントを行った。次にhβ1ARの残基をTAS2R38受容体のアラインメントを行った配列に変異させ、TAS2R38受容体の初期3D構造を生成した。
タンパク質の予測TMドメインを抽出し、7つの単一へリックスを形成し、dreiding力場[Mayo、1990年]を用いて最小化した後、合わせてテンプレートと一致する7つのへリックスのバンドルを形成した。
7つのTMドメインのそれぞれを表す最適へリックスを、x線テンプレートを用い、7へリックスバンドルに配置する。各実験テンプレートは42の自由度:7つのTMへリックスそれぞれのx、y、z、θ、φおよびη値(合計6×7=42)を有する。疎水中心は脂質二重層の中心を通る面として定義されるz=0の残基である。これはタンパク質の疎水プロファイルから、または相同性により計算される。最適化された自由度はヘリックスの傾斜角度θ、へリックスの掃引角度φ、およびヘリックスのヘリックス軸まわりの回転角度ηである。
へリックスの最小化後、構成されたすべての考えられる7ヘリックスバンドルを、7つのへリックスのそれぞれにそれらの軸のまわり12方位(30°間隔)を取らせることにより考慮し、GPCRの7つのへリックスの127=35,000,000パッキングをもたらした。BiHelix法[Goddard、2010年]は、12セットの最近接二重へリックス相互作用、TM1−TM2、TM1−TM7、TM2−TM3、TM2−TM4、TM2−TM7、TM3−TM4、TM3−TM5、TM3−TM6、TM3−TM7、TM4−TM5、T5−TM6およびTM6−TM7を考慮することにより構成される平均場を用い、これら3500万パッキングのエネルギーを推定する。ここでSCREAMを用い、各例について側鎖を最適化した[Kam、2008年]。
次に、7へリックスバンドルをBiHelixからのもっとも良好な1000個の結果の中に構築し、SCREAMを用いて側鎖を再度最適化した。各バンドルを非明示的な膜に浸漬させ、荷電残基を脂質に曝露するヘリックス回転に不利となり得る膜溶媒和効果を計算する。この膜溶媒和については[Goddard、2010年]に記載されている。
ステップEからの回転角(η)の最適セットについて、ここで傾斜(θ、φ)の範囲をηと同時にサンプリングし、最適な7へリックスバンドルを得た。同様に、12対の強く相互作用するヘリックスを考慮し、傾斜の効果を説明する。7へリックスバンドルを、図1に示すように、3つの四重へリックスバンドルに分割する。各四重へリックスについてもっとも低いエネルギーを有する2000個の構造を、エネルギーを増加させることにより選択する。最後に、各個別ヘリックス配座リストから、各ヘリックスにとってもっとも良好な36個の配座を用い、367≒8×1010のバンドル全体のエネルギーを計算し、この方法からもっとも低いエネルギーを有すると推定される1000個の組み合わせを得る。
SuperBiHelixからのこれらの上位1000個のヘリックスバンドルを7ヘリックスバンドル中に構築し、側鎖を再度最適化する。次に、構造を10ステップ最小化する。この方法は、低位のバンドルのアンサンブルをもたらす。低位構造の試験はどのヘリックスが柔軟であるかを示し、活性化に対する洞察をもたらすことができる。
苦味受容体のMD解析に用いる3つのECおよびICループの初期構造を提供するため、苦味受容体のtβ1ARとのアラインメントを用い、これらのループを結晶構造に相同性スレッディングした。次に、最小化および動力学を、固定ヘリックスバンドル原子を用いたループで行った。これらのループはかなり柔軟であり、前ステップにおいて非明示的にのみ処理された溶媒により強く影響されると予想された。
以下の方法およびシステムを用い、ドッキングリガンドおよび予測タンパク質構造を同定した。
BiHelixにおける脂質および水は脂質二重層の不十分な層で非明示的であるので、苦味受容体の予測構造の分子動力学(MD)シミュレーションは、リガンドの有無にかかわらず10ns間、明示的な脂質二重層および水において行った。明示的な水および周期的に無限の脂質を含むNAMD[Phillips、2005年]を用いるMDシミュレーションを行い、タンパク質の脂質および水との相互作用を測定した。予測タンパク質構造を脂質分子から取り出し、1−パルミトイル−2−オレオイル−sn−グリセロ−3−ホスファチジルコリン(POPC)の周期構造に挿入した。このプロセスでは、脂質分子はタンパク質の5Å以内で除去した。次に、これを水分子のボックスに挿入し、水を脂質およびタンパク質の5Å以内で除去した。塩化物イオンを添加し、系の荷電を中和した。膜および水分子を固定化タンパク質で最小化した後、NPTシミュレーションにおいて500ps間平衡化した。最後に、系全体を最小化した後、10nsのNPTシミュレーションを行った。すべてのNPTシミュレーションは、1ps−1の減衰係数および310Kの浴温でランジュバン動力学を用いて行った。圧力は、1atmの標的圧力ならびに200fsのバロスタット振動および減衰時間で、能勢・フーバー・ランジュバンのピストン圧力制御により一定に維持した。ステップサイズは、周期境界条件を適用して、1fsだった。完全系(図3)は、周期的な細胞1個当たり合計41570個の原子について、予測タンパク質、101個の脂質分子、7528個の水分子、および19個の塩化物イオンを含有する。ボックスサイズは75Å×75Å×85Åである。NAMDプログラムを次に用い、310Kの浴温で10nsのNPTMDを行った。

タンパク質構造予測およびMDシミュレーション結果は、W108(3)とA(またはV)262(6)との間のH結合がテイスターhTAS2R38PAV、hTAS2R38AAIおよびhTAS2R38PVVを安定させると予想され、W108(3)とA261(6)との間のH結合が非テイスターhTAS2R38AVIを安定させると予想されることを示す。出願人はさらに、PTCおよびPTUの両方が水素結合によりテイスター(hTAS2R38PAV、hTAS2R38AAIおよびhTAS2R38PVV)中のA(またはV)262と相互作用することができるが、リガンドと非テイスター(hTAS2R38AVI)との間にはH結合がまったくないことを見出す。出願人はここで、アゴニスト結合テイスターとアゴニスト結合非テイスターとの間の違いを概説する。主な違いは苦味受容体中の位置262を含む。残基262はTM3−TM6相互作用だけでなく、アゴニストの受容体との結合にも関与する。
出願人は、[Pronin、2004年]による研究において試験したリガンドのファミリーと複合させたhTAS2R47の予備3−D構造を得た。これらの結果を、方法の妥当性確認をもたらす別のGPCR(ヒトプロスタグランジンDP)の構造予想結果とともに、以下に示す。
出願人は前世代のMembStruk方法を用い、DPプロスタグランジン受容体の構造を予測した後、この予測構造をHierDockで用い、リガンドの4つのファミリーの結合部位を予測した。以下に要約するこの実験は、予測GPCR構造が薬剤開発に用いるのに十分に正確であることを示す。
GI系における苦味受容体の持続的活性化について、苦味受容体リガンド(例えば、PTU誘導体)はGI系に残り、吸収されないことが望ましい。これは吸収、分布、代謝、排泄および毒性のような薬物動力学特性を変えたリガンドを必要とする。
適切な補完分子と結合した活性剤は、既知の方法を用いて調製することができる。以下の実施例における適切な補完分子との結合の例となる反応スキームは、PTU−セルロースの化学合成を示す。
出願人は、5mlの食塩水または5mlの食塩水中の5mMのPTCを、空腹で意識のあるオスのスプラーグ−ドーリーラット(重さ250〜300gm)の胃の中に、標準的な経胃強制給餌により投与した。ラットを次に45分後に安楽死させ、心臓内穿刺によって血液を取り出し、CURE抗体9153を用いる最近修正された放射免疫アッセイ(カリフォルニア大学ロサンゼルス校、J Reeve氏より寄贈)により血液中のPYYを測定した。結果は以下のようにPYYレベルの倍化を示す:食塩水を強制給餌したラット(n=5)71+/−35pM、PTCを強制給餌したラット(n=5)154+/−44pM。ラットの死亡時の興味深い観察は、PTC処理ラットの胃に残った液体の体積は1〜2mlの範囲内だったが、食塩水処理ラットの胃には0.5ml未満しかなかったことだった。この観察は、PTCがTAS2R38との相互作用によって胃排出を調節することを示す。
特定の苦味受容体のリガンドの選択性を調査するための細胞ベースのシステムの実現可能性を決定するため、出願人は、STC−5細胞を選択し、これらの細胞からGLP−1に対するPTCの効果を測定した。出願人は、STC−5細胞がいくつかの他の苦味受容体の中でもTAS2R38を有することを示した。
考えられる各種リガンドにより誘導されるGLP−1の放出を試験するのにNCI−H716細胞の内分泌細胞系列を用いた。

Claims (39)
- 個体に、PTU、PTC、安息香酸デナトニウム、グリチルリチン酸アンモニウム塩、没食子酸エピガロカテキン、ハイパーフォリン、塩化ベルベリン、塩化コプチシン、硫化アリルメチル、ロットレリン、クルクミン、エラグ酸、エンベリンおよび/またはそれらの誘導体から選択される1つ以上のGI苦味受容体リガンドを、個体において1つ以上のGI苦味受容体リガンドの1つ以上のGI苦味受容体との結合を可能にする有効量で投与し、該結合が代謝ホルモンGLP−1、PYYおよび/またはCCKの放出の調節をもたらすステップを含む、個体において代謝ホルモンの放出を調節する方法。
- 前記1つ以上のGI苦味受容体リガンドがPTU、PTC、安息香酸デナトニウム、グリチルリチン酸アンモニウム塩、没食子酸エピガロカテキン、ハイパーフォリン、塩化ベルベリン、塩化コプチシン、硫化アリルメチル、ロットレリン、クルクミン、エラグ酸、およびエンベリンならびに/またはそれらのアゴニスト誘導体を含み、前記結合がGLP−1、PYYおよび/またはCCKの放出の増加をもたらす、請求項1に記載の方法。
- 前記1つ以上のGI苦味受容体リガンドが安息香酸デナトニウムまたはそのアゴニスト誘導体を含み、前記結合がCCKの放出の増加をもたらす、請求項1または2に記載の方法。
- 前記1つ以上のGI苦味受容体リガンドが、前記1つ以上のGI苦味受容体リガンドの全身吸収および放出に干渉するように構成される補完分子と組み合わせて投与される、請求項1〜3のいずれか1項に記載の方法。
- 前記1つ以上のGI苦味受容体リガンドの少なくとも1つが前記補完分子と結合し、苦味物質受容体との結合に対して活性な部分を提供するように構成される苦味物質作用剤をもたらす、請求項4に記載の方法。
- 前記補完分子がセルロース、PEG、単糖、オリゴ糖、アミノ酸、および/またはペプチドである、請求項4または5に記載の方法。
- 前記1つ以上のGI苦味受容体リガンドの前記投与が、前記1つ以上のGI苦味受容体リガンドのGI上皮にわたる全身吸収を最小化するように製剤される組成物で行われる、請求項1〜6のいずれか1項に記載の方法。
- 前記投与が経腸投与により行われる、請求項1〜7のいずれか1項に記載の方法。
- PTU、PTC、安息香酸デナトニウム、グリチルリチン酸アンモニウム塩、没食子酸エピガロカテキン、ハイパーフォリン、塩化ベルベリン、塩化コプチシン、硫化アリルメチル、ロットレリン、クルクミン、エラグ酸、エンベリンおよび/またはそれら誘導体から選択される1つ以上のGI苦味受容体リガンドを適切な媒体とともに含む、個体において代謝ホルモンの放出を調節する組成物。
- 前記1つ以上のGI苦味受容体リガンドが前記1つ以上の苦味受容体リガンドの全身吸収および放出に干渉するように構成される補完分子との組み合わせで含まれる、請求項9に記載の組成物。
- 前記1つ以上のGI苦味受容体リガンドの少なくとも1つが前記補完分子と結合し、苦味物質受容体との結合に対して活性な部分を提供するように構成される苦味物質作用剤をもたらす、請求項10に記載の組成物。
- 前記1つ以上GI苦味受容体リガンドが約1マイクロモル〜約1.25マイクロモルの量で含まれる、請求項9〜11のいずれか1項に記載の組成物。
- 前記組成物が医薬組成物であり、前記適切な媒体が医薬的に許容可能な媒体である、請求項9〜12のいずれか1項に記載の組成物。
- 請求項1〜8のいずれか1項に記載の方法において同時に、組み合わせて、または連続して用いる1つ以上の標的GI苦味受容体を調節することができる少なくとも2つの苦味受容体リガンドを含む、個体において代謝ホルモンの放出を調節するシステム。
- 苦味受容体リガンドの全身吸収および放出に干渉するように構成される補完分子と結合した苦味受容体リガンドを含む、標的苦味物質受容体を調節する苦味物質作用剤において、
該苦味物質受容体リガンドは第1部分および第2部分を含み、該第1部分は該苦味受容体リガンドの該苦味受容体との結合に対して活性であり、該第2部分は該苦味受容体リガンドの該苦味受容体との結合に対して不活性であり、
該補完分子は該苦味受容体リガンドの該第2部分に付着し、該苦味受容体との結合のための該苦味受容体リガンドの該第1部分を提供するように構成される該苦味物質作用剤をもたらす、作用剤。 - 前記苦味受容体リガンドがPTU、PTC、安息香酸デナトニウム、グリチルリチン酸アンモニウム塩、没食子酸エピガロカテキン、ハイパーフォリン、塩化ベルベリン、塩化コプチシン、硫化アリルメチル、ロットレリン、クルクミン、エラグ酸、エンベリンおよびそれらの誘導体から選択される、請求項15に記載の苦味物質作用剤。
- 前記補完分子がセルロース、PEG、単糖、オリゴ糖、アミノ酸、および/またはペプチドである、請求項15または16に記載の苦味物質作用剤。
- 補完分子と結合した苦味物質受容体リガンドを含む苦味物質作用剤であって、該補完分子が該苦味物質受容体リガンドの全身放出に干渉するように構成される、作用剤。
- 個体に有効量の請求項18に記載の1つ以上の苦味物質作用剤を投与するステップを含む、個体において標的苦味受容体を活性化する方法。
- 請求項18に記載の1つ以上の苦味物質作用剤および適切な媒体を含む、個体において標的苦味受容体を活性化する組成物。
- 1つ以上の苦味受容体リガンドおよび該1つ以上の苦味受容体リガンドの全身放出に干渉するように構成される少なくとも1つの補完分子を含む、苦味物質作用剤をもたらすシステムにおいて、
該1つ以上の苦味受容体リガンドは該少なくとも1つの補完分子と結合し、請求項18に記載の苦味物質作用剤をもたらすことができる、システム。 - 個体に有効量の1つ以上の作用剤を投与し、個体において1つ以上の苦味受容体を活性化するステップを含む、個体において代謝ホルモンの放出およびその関連する生物学的プロセスを調節する方法。
- 細胞をPTU、PTC、安息香酸デナトニウム、グリチルリチン酸アンモニウム塩、没食子酸エピガロカテキン、ハイパーフォリン、塩化ベルベリン、塩化コプチシン、硫化アリルメチル、ロットレリン、クルクミン、エラグ酸、エンベリンおよびそれらの誘導体から選択される1つ以上の苦味物質受容体リガンドと接触させ、該苦味物質受容体リガンドの苦味物質受容体との結合を可能にするステップ;ならびに
該接触後、細胞中の生物学的応答を検出するステップ
を含む、細胞中の苦味物質受容体の活性化に関連する生物学的応答を同定する方法。 - 検出される生物学的応答を対照生物学的応答と比較し、該生物学的応答を特徴づけるステップをさらに含む、請求項23に記載の方法。
- GI系における標的細胞中に発現するGI苦味受容体を同定するステップ;
該GI苦味物質受容体の構造を予測するステップ;
予測されるGI苦味物質受容体構造に基づき該GI苦味物質受容体と結合する候補リガンドを同定するステップ;
同定される候補リガンドを苦味受容体活性化アッセイにおいて試験するステップ;および
GI系に関連する代謝ホルモンの少なくとも1つの調節に対する該候補リガンドの効果を試験するステップ
を含む、GI系に関連する代謝ホルモンの放出を調節することができるGI苦味受容体リガンドを同定する方法。 - 前記候補リガンドが前記GI苦味受容体のモデリングによって同定される、請求項25に記載の方法。
- 前記モデリングが相同性モデリングである、請求項26に記載の方法。
- 前記モデリングがヘリックス間水素結合の構造を予測するステップを含む、請求項26または27に記載の方法。
- GI苦味受容体リガンドを結合させてGI苦味物質受容体を特定配座に誘導するステップであって、該特定配座が該GI苦味物質受容体の複数の活性配座の少なくとも1つである、ステップ;
該誘導後、細胞からのホルモン放出の増加または減少を検出するステップ;および
該増加または減少を該GI苦味受容体リガンドの作用によって調節するステップ
を含む、細胞からのホルモン放出を調節する方法。 - 前記誘導ステップ前に、請求項25に記載の方法による前記GI苦味受容体リガンドを同定するステップが行われる、請求項29に記載の方法。
- 個体に、PTU、PTC、安息香酸デナトニウム、グリチルリチン酸アンモニウム塩、没食子酸エピガロカテキン、ハイパーフォリン、塩化ベルベリン、塩化コプチシン、硫化アリルメチル、ロットレリン、クルクミン、エラグ酸、エンベリンおよびそれらの誘導体から選択される1つ以上のGI苦味受容体リガンドを、代謝ホルモンGLP−1、PYYおよび/またはCCKを調節する治療有効量で投与するステップを含む、個体において代謝ホルモンに関連する症状を治療または予防する方法。
- 前記1つ以上のGI苦味受容体リガンドが、前記1つ以上のGI苦味受容体リガンドの全身吸収および放出に干渉するように構成される補完分子と組み合わせて投与される、請求項31に記載の方法。
- 前記1つ以上のGI苦味受容体リガンドの少なくとも1つが前記補完分子と結合し、前記苦味物質受容体との結合に対して活性な部分を提供するように構成される苦味物質作用剤をもたらす、請求項32に記載の方法。
- 前記補完分子がセルロース、PEG、単糖、オリゴ糖、アミノ酸、および/またはペプチドである請求項32または33に記載の方法。
- 前記1つ以上のGI苦味受容体リガンドの前記投与が、前記1つ以上のGI苦味受容体リガンドのGI上皮による全身吸収を最小化するように製剤される組成物で行われる、請求項31〜34のいずれか1項に記載の方法。
- 前記投与が経腸投与により行われる、請求項31〜35のいずれか1項に記載の方法。
- 前記治療有効量が約1マイクロモル〜約1.25ミリモルである、請求項31〜36のいずれか1項に記載の方法。
- 前記症状が肥満、糖尿病、肝臓疾患および/または心血管疾患である、請求項31〜37のいずれか1項に記載の方法。
- 請求項31〜38のいずれか1項に記載の方法において同時に、組み合わせて、または連続して用いるPTU、PTC、安息香酸デナトニウム、グリチルリチン酸アンモニウム塩、没食子酸エピガロカテキン、ハイパーフォリン、塩化ベルベリン、塩化コプチシン、硫化アリルメチル、ロットレリン、クルクミン、エラグ酸、エンベリンおよびそれらの誘導体から選択される少なくとも2つの苦味受容体リガンドを含む、個体において代謝ホルモンに関連する症状を治療または予防するシステム。
