Description
下記の表1に示すように、3つの抗体は、立体構造的なエピトープに結合する‐すなわち、それらは重複するペプチドを結合することによっては、マップしない。特異的な配列をマップする本発明の抗体を表1に示す。測定したオンレート及びオフレートから算出してpMで示した組み換えGaタンパク質及びGbタンパク質に関する標準的なBiacoreアッセイを使用して決定した親和性定数も示す。これら抗体の2つのデータを図6に示す。パネルA、B及びC(図6A、6B、6C)は、3G12の結合データを示し、パネルD、E、およびF(図6D、6E、6F)は、3D3のデータを示す。上段の列は、抗体を用いたバイオセンサーチップのローディングを示し、中段の列は、チップにGaタンパク質を横切って流した後に、バッファーで洗浄することから上昇するシグナルを示し、下段の列は、Gbタンパク質に関する同様の結果を示す。シグナルの増加(上昇)は、オンレートの算出を許容し、一方では、洗浄中の減少は、オフレートの算出を許容する。オフレートに対するオンレートの割合は、親和性定数、Kdである。
