Abstract
Description
本発明は環状デプシペプチド類、またはその誘導体に関する。
カリクレイン7は、キモトリプシン様活性を示すカリクレイン遺伝子ファミリーのS1セリンプロテアーゼである。ヒトカリクレイン7(hK7、KLK7または角質層キモトリプシン酵素(SCCE)、Swissprot P49862)は皮膚生理学(1、2、3)において重要な役割を有する。それは主に皮膚で発現され、皮膚生理学において重要な役割を有することが報告されている。hK7は、剥離の過程における角化扁平上皮の細胞間接着構造の分解に関与する。剥離過程はよく制御され、角質層の一定の厚みを維持するために角質細胞のデノボ製造と繊細なバランスが取られ、皮膚の最外側層は、皮膚バリア機能に必須に関与する。この点に関して、hK7はコルネオデスモソーム(corneodesmosomal)タンパク質であるコルネオデスモシンおよびデスモコリン1を開裂できることが報告されている(4、5、6)。両方のコルネオデスモソームの分解は剥離に必要である。加えて、つい最近、2種の脂質処理酵素β−グルコセレブロシダーゼおよび酸性スフィンゴミエリナーゼがhK7により分解され得ることが示された(7)。両方の脂質処理酵素は、それらの基質グルコシルセラミド類およびスフィンゴミエリンと共分泌され、これらの極性脂質前駆体をそれらのあまり極性ではない生成物、例えばセラミドに処理し、これが続いて細胞外層状膜に統合される。この層状膜構造は、機能的皮膚バリアに必須である。最後に、hK7がインターロイキン−1β(IL−1β)前駆体をその活性形態にインビトロで活性化することが示されている(8)。ケラチン生成細胞がIL−1βを発現するが、活性形態の特異的IL−1β変換酵素(ICEまたはカスパーゼ1)を発現しないため、ヒト表皮におけるIL−1β活性化は他のプロテアーゼを介して起こることが提唱され、その可能性のある候補がhK7である。

実施例1.1 式(II)−(VII)、(XI)−(XIV)および(XVII)の化合物の製造
株:コンドロマイセス・クロカタス株を、我々の研究室の環境サンプルである、腐ったクルミ材から単離した。
この株は、子実体の形態学ならびに16S−RNA遺伝子の部分配列に基づき、疑いなくコンドロマイセス・クロカタスとして同定された。コンドロマイセス・クロカタスは、DSMZ(DSMZ(2007))により生物学的リスクグループ1に分類された。コンドロマイセスは、デルタプロテオバクテリア綱内のミクソコッカス目に属する、粘液細菌科の属である。ミクソコッカス目細菌は、ミクソバクテリアとも呼ばれるが、ほとんどの他の細菌と区別される2個の特徴を有するグラム陰性桿状細菌。それらは飢餓により能動的滑走機構を使用して固体表面上を遊走し、凝集して子実体を形成する(Kaiser(2003))。
本発明のコンドロマイセス・クロカタス株は、受託番号19329としてDSMZに寄託されている。
前培養を、本発明のコンドロマイセス・クロカタス株の液体培養からの5ml(=10%)を、200mlのバッフル付振盪フラスコ中の50mlの培地MD1(Bode et al. 2003に従い適合、表6参照)に接種することにより開始した。11日間、ロータリー・シェーカー上で、30℃で120rpmのインキュベーション後、1回目の中間培養を、500mlのバッフル付振盪フラスコ中、前培養物由来の各10ml(=10%)を5×100mlの培地MD1に接種することにより開始した。7日間、ロータリー・シェーカー上で、30℃で120rpmのインキュベーション後、2回目の中間培養を、2Lのバッフルが付いていない振盪フラスコ中、19×500mlの培地MD1に1回目の中間培養物からの各25ml(=5%)を接種することにより開始した。6日間、ロータリー・シェーカー上で、30℃で150rpmのインキュベーション後、2回目の中間培養物全部(9.5リットル=9.5%)を使用して、100リットルの製造培地POL1に接種した(Kunze et al. 1995に従い適合、表7参照)。
この100L主培養を、100L規模スチールタンク醗酵槽で行った。温度を30℃に、通気を20l/分(=0.2vvm)に、そして撹拌速度を50rpmに制御した。0.5バールのわずかな加圧を醗酵槽容器内部で維持した。培養pHを、3N H2SO4または3N NaOHの制御された添加により6.9−7.1に維持した。約1日の遅滞期後、酸素消費が約4日間加速し、培養物の指数増殖を示した。最後の2日間、酸素消費はわずかに減少し、培養の静止期を示した。7日間後、培養を、5.3mg/lの、式IIの環状デプシペプチドと共に回収した。
培養濾液を200l酢酸エチルで抽出した。1時間のturax処理を含む2時間の接触時間後、有機相を分離し、20lの水で洗浄し、最後に蒸発させて、‘培養濾液抽出物’と呼ぶ乾燥抽出物を得た。
他の式(II)の環状デプシペプチドも細胞抽出物中に低濃度で発見されている。とりわけ、他の環状デプシペプチド類は式(IV)、(V)および(XI)−(XV)および(XVII)のものである。

前培養を、本発明のコンドロマイセス・ロブスタス株の液体培養由来の各20ml(=20%)を、500mlのバッフル付振盪フラスコ中、6×100mlの培地MD1(Bode et al. 2003に従い適合)に接種することにより開始した。1日間、ロータリー・シェーカー上で、30℃で120rpmのインキュベーション後、前培養由来の各100ml(=25%)を1回目の中間培養を、2Lのバッフル付振盪フラスコ中、6×400mlの培地MD1にに接種することにより開始した。3日間ロータリー・シェーカー上で、30℃で120rpmのインキュベーション後、2回目の中間培養を、15リットルの培地MD1を含む20Lスチールタンク発酵槽に1回目の中間培養由来の3リットル(=20%)を接種することにより開始した。温度を30℃に、通気を20l/分(=1.0vvm)および撹拌速度を80rpmに制御した。0.5バールのわずかな加圧を醗酵槽容器内部で維持した。pHを制御しなかったが、培養のpHは、開始時のpH6.95から7日目のpH6.88にわずかに低下しただけであった。7日後、2回目の中間培養(18リットル=20%)を使用して、90リットルの製造培地POL1(Kunze et al. 1995に従い適合)(出発体積=108リットル)に接種した。主培養を、100L規模スチールタンク醗酵槽で行った。温度を30℃に、通気を30l/分(=0.3vvm)および撹拌速度を開始時50rpmに、そして4日後80rpmに制御した。0.5バールのわずかな加圧を醗酵槽容器内部で維持した。培養pHを、2N H2SO4または1.5N NaOHの制御された添加により6.8−7.2に維持した。14日後、培養物を回収し、3mg/lの力価であった。
培養濾液を200l酢酸エチルで抽出した。1時間のturax処理を含む2時間の接触時間後、有機相を分離し、20lの水で洗浄し、最後に蒸発させて、‘培養濾液抽出物’と呼ぶ12.5gの乾燥抽出物を得た。

株:コンドロマイセス・アピクラタス株を土壌サンプルから単離した。本発明のコンドロマイセス株は、16S−RNA遺伝子の部分的配列に基づき、コンドロマイセス・アピクラタスと同定された。コンドロマイセス・アピクラタスは、DSMZ(DSMZ(2007))により生物学的リスクグループ1に分類された。コンドロマイセスは、デルタプロテオバクテリア綱内のミクソコッカス目に属する、粘液細菌科の属である。ミクソコッカス目細菌は、ミクソバクテリアとも呼ばれるが、ほとんどの他の細菌と区別される2個の特徴を有するグラム陰性桿状細菌。それらは飢餓により能動的滑走機構を使用して固体表面上を遊走し、凝集して子実体を形成する(Kaiser(2003))。

蛍光強度(FI)測定のために、Ultra Evolutionリーダー(TECAN, Maennedorf, Switzerland)を使用する。この装置は、350nm(20nmバンド幅)および500nm(25nmバンド幅)帯域フィルターを、各々蛍光励起および放出獲得のために供える。シグナル:バックグラウンド比を高めるために、適当な二色ミラーを使用する。光学フィルターおよび二色ミラーはTECANから購入する。各ウェルのフルオロフォアを、測定あたり3回のフラッシュで励起させる。
IC50値測定のために、アッセイを室温で384ウェルプレートで行う。全ての最終アッセイ体積は、30μlであった。試験化合物を90%(v/v)DMSO/水に溶解し、水(0.05%(w/v)CHAPS含有)で望むアッセイ濃度の3倍に希釈する。11個の最終化合物濃度は:0.3nM、1nM、3nM、10nM、30nM、100nM、300nM、1μM、3μM、10μMおよび30μMである。各アッセイについて、10μl 水/CHAPS(±試験化合物)を各ウェルに添加し、続いて10μl プロテアーゼ溶液(1.5×アッセイ緩衝液で希釈)を添加する。最終アッセイ溶液中のプロテアーゼ濃度は0.2nMである(Bradford法に従い測定した酵素濃度による)。1時間の室温でのインキュベーション後、反応を10μl 基質溶液(1.5×アッセイ緩衝液に溶解した基質、最終濃度は2μM)の添加により開始する。化合物の酵素作用に対する作用を線形プログレス曲線から得て、1回目は基質直後に取り、2回目は1時間後である2回の読みとりから測定する。IC50値を、非線形回帰分析ソフトウェア(XLfit, Vers. 4.0; ID Business Solution Ltd., Guildford, Surrey, UK)を使用して、阻害パーセンテージ対インヒビター濃度のプロットから計算する。
環状デプシペプチド類は、hカリクレイン7を表11に示すIC50値で阻害する。
材料および緩衝液
ヒト好中球エラスターゼ(cat. no. SE563)をElastin Products Company, Inc. (EPC, Owensville, USA)から購入する。乾燥粉末(純度は供給者によると>95%)を20mM 酢酸ナトリウム緩衝液、pH5.0、50%(v/v)グリセロールに溶解し、数等分して−80℃に冷却する。

加えて、環状デプシペプチド類はヒトキモトリプシンを0.001μM〜0.02μMの範囲のIC50で阻害する。
式(VIII)の環状デプシペプチドの生物学的活性をカリクレイン7で測定した。本発明の環状デプシペプチドは、ヒトカリクレイン7を3nM未満のIC50で阻害する。この環状デプシペプチドは、ヒトキモトリプシンおよびヒト好中球エラスターゼを、各々約0.004μMおよび約0.0025μMのIC50で阻害した。
A) マウスの皮膚バリア破壊の回復試験
方法:皮膚バリア破壊を、無毛SKH1マウス群の皮膚を繰り返しS-Sqame(登録商標)皮膚サンプリングディスクで剥ぐことにより達成した。≧40mg/cm2/時間の経表皮水分喪失(TEWL)が達成されたとき、この方法を終えた。TEWLを、Tewameter TM210(Courage Khazaka, Cologne, DE)で指定した。バリア破壊直後、30μl試験化合物を10mM濃度で適用した。対照動物を溶媒(エタノール/プロピレングリコール、3/7(v/v))のみで同様に処置した。TEWLをバリア破壊前、直後、および3時間後に測定した。各動物において、回復パーセントを次の式を使用して計算した:(1−[3時間目のTEWL−ベースラインTEWL]/[剥いだ直後のTEWL−ベースラインTEWL])×100%。
方法:Crl:NMRIマウスの、毛を剃った腹部を50μlの2%オキサゾロンで1日目に感作し、8日目に右耳の内側表面を10μl オキサゾロンで攻撃した。攻撃していない左耳を対照として使用し、皮膚炎を9日目の耳介重量(炎症性腫脹の指標として取る)の差異により評価した。動物を、攻撃後30分に10μl 試験化合物または溶媒のみで局所的に処置した。処置効果を、媒体のみで処置した動物と比較した炎症性耳介腫脹の阻害パーセントとして計算した。
ACD誘発前8日間、500μlの10%2,4−ジニトロフルオロベンゼン(DNFB、DMSO/アセトン/オリーブ油[1/5/3、v/v/v]に溶解)を、両耳の根元および両鼠蹊(100μl/部位)の皮膚上に感作のために適用した。攻撃反応を、毛を剃った背側外側背面の対側性試験部位(各々7cm2サイズ)上への15μlのDNFB(1.0%)で誘発させた。処置のために、試験化合物およびプラセボ(溶媒のみ)を、攻撃0.5および6時間後に各動物の2試験部位に対側性に適用した。試験部位を、炎症がピークに達したときである攻撃24時間後に臨床的に試験した。攻撃を0から4のスケールで採点し(表15)、指定部位あたり組み合わせて最大12点とした。皮膚発赤を、反射率測定によりA*値を使用して測定した。
a)一工程法:2mLのジクロロメタン/アセトニトリル(1:1)中の20mgの式(II)の環状デプシペプチドおよび0.027mL トリエチルシランの溶液に、−50℃で、0.014mLの三フッ素化ホウ素エーテラートをゆっくり添加した。反応混合物を−5℃に温め、その温度にさらに30分維持し、飽和NaHCO3溶液に注ぎ、EtOAcで抽出した。有機層を硫酸ナトリウムで乾燥させ、溶媒を真空で除去した。残渣のHPLC(XTerra [5cm];アセトニトリル/炭酸アンモニウム緩衝液pH10勾配)での精製により、9.8mgの式(II)の環状デプシペプチドの誘導体(式中、Ahpが3−アミノ−ピペリジン−2−オンに置換されている)を得た。ESI MS:935.36 [M+Na]+。
粗物質を400mLの2−プロパノールに溶解し、115mgのPd/C(10%)を添加し、混合物を、出発物質が消費されるまで(HPLC)大気圧下で水素化した。得られた残渣をクロマトグラフィー(SiO2;cHex/EtOAc(1:1)+10%MeOH)で精製し、684mgの式(II)の環状デプシペプチド(式中、Ahpが3−アミノ−ピペリジン−2−オンに置換されている)を得た。
5mLの1−PrOH中の75mg(0.081mmol)の式(II)の環状デプシペプチドの溶液に、30μLの硫酸を添加し、反応混合物をr.t.で48時間撹拌した。後処理のために、反応混合物をジクロロメタンで希釈し、飽和ビカーボネート溶液で洗浄した。有機層の硫酸ナトリウムでの乾燥後、溶媒を除去し、得られた残渣をシリカゲルクロマトグラフィー(cHex/EtOAc(1:1)+10%MeOH)で精製した。収量:65mg(83%)の式(II)の環状デプシペプチドの誘導体(式中、Ahpが3−アミノ−6−プロポキシ−ピペリジン−2−オンに置換されている)。ESI MS:993.37 [M+Na]+。
2mLのジクロロメタン中の25mg(0.027mmol)の式(II)の環状デプシペプチドの溶液を0℃に冷却した。次いでDIPEAおよびトリフルオロ酢酸無水物(TFAA)を添加した。反応混合物を室温にゆっくり温め、さらに4時間撹拌した。後処理のために、反応混合物をジクロロメタンで希釈し、塩酸および飽和ビカーボネート溶液で千手下。硫酸ナトリウムで乾燥後、溶媒を除去し、得られた残渣をシリカゲルクロマトグラフィー(cHex/EtOAc(1:1)+10%MeOH)で精製した。収量:14mg(57%)の式(II)の環状デプシペプチドの誘導体(式中、A1のアミド側鎖がニトリルに置換されている)。ESI MS:933.30 [M+Na]+。

2mLの乾燥アセトン中の200mg(0.21mmol)の実施例5の環状デプシペプチド、57.5mg(0.41mmol)のK2CO3の混合物に、60.5mg(0.31mmol)の(E)−3−フェニル−2−プロペニルブロマイドを添加し、一夜超音波処理した(温度は約50℃に上昇)。後処理のために、反応混合物をジクロロメタンで希釈し、水で洗浄した。有機層の硫酸ナトリウムでの乾燥後、溶媒を除去し、得られた残渣をHPLC(15cm Zorbax;アセトニトリル/水性NH4OAc緩衝液:20→95%)で精製した。収量:100mg(45%)のA6におけるO−((E)−3−フェニル−2−プロペン−1−イル)L−チロシンを特徴とする実施例5の環状デプシペプチドの誘導体。ESI MS:1109.37 [M+Na]+。
2mLの乾燥アセトン中の200mg(0.21mmol)の実施例5の環状デプシペプチド、46.5mg(0.31mmol)のヨウ化ナトリウム、および57.5mg(0.41mmol)のK2CO3の混合物に、44mg(0.31mmol)の3−(クロロメチル)−1,5−ジメチル−1H−ピラゾールを添加し、一夜超音波処理した(温度は約50℃に上昇)。後処理のために、反応混合物をジクロロメタンで希釈し、水で洗浄した。有機層の硫酸ナトリウムでの乾燥後、溶媒を除去し、得られた残渣をHPLC(15cm Zorbax;アセトニトリル/水性NH4OAc緩衝液:20→95%)で精製した。収量:90mg(40.5%)のA6におけるO−(1,5−ジメチル−1H−ピラゾール−3−イル)メチル−L−チロシンを特徴とする実施例5の環状デプシペプチドの誘導体。ESI MS:1101.39 [M+Na]+。
同様に、式(II)の環状デプシペプチドの3−(クロロメチル)−1,5−ジメチル−1H−ピラゾールでの処理は、A6におけるO−(1,5−ジメチル−1H−ピラゾール−3−イル)メチル−L−チロシンを特徴とする化合物を提供する。ESI MS:1059.19 [M+Na]+。
20mgの前記酸、6.81mg(0.031mmol)の8−アミノ−3,6−ジオキサオクタン酸tert−ブチルエステル、および15.8mg(0.041mmol)のHATUを2mLの乾燥DMFに溶解し、11μLのDIEPAを添加し、r.t.で一夜撹拌した。後処理のために、反応混合物をEtOAcで希釈し、飽和NaHSO4およびNaHCO3溶液および塩水せ洗浄した。有機層の硫酸ナトリウムでの乾燥後、溶媒を除去し、得られた残渣をHPLC(15cm Zorbax;アセトニトリル/水性NH4OAc緩衝液:20→95%)で精製した。収量:7mg(29%)の表題化合物。ESI MS:1194.32 [M+Na]+。

- 式(I):

- A5とA6の間のアミド結合の窒素原子がメチルで置換されており、チロシンのOH基がORであり、ここで、Rは水素、非置換であるか、またはアリール、アリールアルキル、アリールアルケニルまたはアリールアルキニル、ヘテロシクリルおよびヘテロシクリルアルキルでさらに置換された(C1−12)アルキル、(C2−12)アルケニル、(C2−12)アルキニル、ハロ(C1−12)アルキル、ハロ(C2−12)アルケニル、ハロ(C2−12)アルキニル、(C1−12)アルコキシカルボニル、(C1−12)アルコキシ−カルボニル(C1−12)アルキル、(C1−12)アルキルアミノカルボニルから成る群から選択される、請求項1に記載のデプシペプチド。
- A4がAhp誘導体3−アミノ−ピペリジン−2−オン、Ahp−IまたはAhp−IIであり、ここでRが(C1−12)アルキル、(C2−12)アルケニル、(C2−12)アルキニル、ハロ(C1−12)アルキル、(C1−12)アルコキシ(C1−12)アルキル、(C1−12)アルコキシ(C1−12)アルコキシ(C1−12)アルキル、ヒドロキシ(C1−12)アルキル、フェニルおよびフェニル(C1−6)アルキルから成る群から選択される請求項1に記載のデプシペプチド。
- アシル残基XがCH3CH2CH(CH3)COまたは(CH3)2CHCOであり、
A1がグルタミン、グルタミン酸、またはその誘導体であり、
A2がスレオニンであり、
A3がロイシンであり、
A4がAhp、3−アミノ−ピペリジン−2−オン、プロリン、5−ヒドロキシ−プロリンまたはその誘導体であり、
A5がイソロイシンであり、
A6がチロシン、N−Me−チロシンまたはその誘導体であり、
A7がイソロイシンまたはバリン、好ましくはイソロイシンである、
請求項1に記載のデプシペプチド。 - A4がAhp、Ahp−I、3−アミノ−ピペリジン−2−オン、プロリン、または5−ヒドロキシ−プロリン、好ましくはAhp、Ahp−I、3−アミノ−ピペリジン−2−オン、または5−ヒドロキシ−プロリン、また好ましくはAhp、Ahp−Iまたは5−ヒドロキシ−プロリン、より好ましくはAhpである、請求項1から5のいずれかに記載のデプシペプチド。
- A1、A2、A3、A5、A6およびA7がL−アミノ酸である、請求項1から7のいずれかに記載の環状デプシペプチド。
- A4が3S,6R Ahpである、請求項1から8のいずれかに記載の環状デプシペプチド。
- A1がグルタミン、オルニチン、または実施例のいずれかに記載のグルタミン誘導体であり、例えばグルタミンニトリル、グルタミン酸エステル、例えばC1−12アルキルエステル(例えばグルタミン酸メチルエステル)または例えばC6−24アリールエステル(例えばグルタミン酸フェニルまたはベンジルエステル)から選択される、請求項1から9のいずれかに記載の環状デプシペプチド。
- 請求項1から10のいずれかに記載の環状デプシペプチドを薬学的に許容される担体および/または成分と共に含む、医薬組成物。
- 医薬として使用するための、特に、請求項13から15のいずれかに記載の通り患者の処置方法に使用するための医薬として使用するための、請求項1から11のいずれかに記載の環状デプシペプチド、および該方法請求項において記載の疾患または障害の処置用薬の製造における、該デプシペプチド類の使用。
- 炎症性および/または過増殖性およびそう痒性皮膚疾患、例えばケロイド、肥厚性瘢痕、アクネ、アトピー性皮膚炎、乾癬、膿疱性乾癬、酒さ、ネザートン症候群または他のそう痒性皮膚疾患、例えば結節性痒疹、高齢者の不特定掻痒ならびに上皮性バリア機能不全を伴う他の疾患、例えば高齢者の皮膚、炎症性腸疾患およびクローン病、ならびに膵炎、または癌、特に卵巣癌、嚢胞性線維症(CF)、慢性閉塞性肺疾患(COPD)、肺線維症、成人呼吸窮迫症候群、慢性気管支炎、遺伝性気腫、リウマチ性関節炎、IBD、乾癬、喘息を有する対象の処置方法であって、該対象に治療的有効量の請求項1−10のいずれかに記載の環状デプシペプチド、またはその誘導体を投与することを含む、方法。
- 対象がケロイド、肥厚性瘢痕、アクネ、アトピー性皮膚炎、乾癬、膿疱性乾癬、酒さ、ネザートン症候群または他のそう痒性皮膚疾患、例えば結節性痒疹、高齢者の不特定掻痒ならびに上皮性バリア機能不全を伴う他の疾患、例えば高齢者の皮膚、炎症性腸疾患およびクローン病、ならびに膵炎、または癌、特に卵巣癌を有する、請求項13に記載の方法。
- 対象がケロイド、肥厚性瘢痕、アクネ、アトピー性皮膚炎、乾癬、膿疱性乾癬、酒さ、ネザートン症候群または他のそう痒性皮膚疾患、例えば結節性痒疹、高齢者の不特定掻痒ならびに上皮性バリア機能不全を伴う他の疾患、例えば高齢者の皮膚を有する、請求項14に記載の方法。
- 対象が嚢胞性線維症(CF)、慢性閉塞性肺疾患(COPD)、肺線維症、成人呼吸窮迫症候群、慢性気管支炎、遺伝性気腫、リウマチ性関節炎、IBD、乾癬、喘息を有する、請求項13に記載の方法。
- コンドロマイセス株、その変異株または突然変異株を、適当な培地で培養し、所望によりそのようにして製造した環状デプシペプチドを化学誘導体化することを含む、請求項1から10のいずれかに記載の環状デプシペプチド、またはその誘導体の製造方法。
- コンドロマイセス株、その変異株または突然変異株の生合成遺伝子を異種微生物宿主株で発現させ、所望によりそのようにして製造した環状デプシペプチドを化学誘導体化することを含む、請求項1から10のいずれかに記載の環状デプシペプチド、またはその誘導体の製造方法。
- 株がコンドロマイセス・クロカタス(DSM 19329)またはコンドロマイセス・ロブスタス(DSM 19330)またはコンドロマイセス・アピクラタス(DSM 21595)である、請求項17または18に記載の方法。
- DSM 19329またはDSM 19330またはDSM 21595の下に寄託された、請求項1から10のいずれかに記載の環状デプシペプチド、またはその誘導体を製造する、単離されたコンドロマイセス微生物。
- 請求項20に記載の単離されたコンドロマイセス微生物により製造された、または請求項17から18の方法により得られた環状デプシペプチド、またはその誘導体。
- 請求項1に記載の環状デプシペプチドの誘導体、またはその誘導体の製造方法であって、択一的に
