Description
血管生成抑制ポリペプチドIle-Val-Arg-Arg-Ala-Asp-Arg-Ala-Ala-Val-Pro(ES-2と称す)の両端に、少なくともその一端にインテグリンファミリーに親和性又は結合力を有するポリペプチドが接続してあり、前記した「インテグリンファミリーに親和性又は結合力を有するポリペプチド」とは、Arg-Gly-Asp又はArg-Gly-Asp-Linkerを含む配列を指し、上述したリンカー(Linker)及び本出願すべてのリンカーは、一つか又は複数の異なるアミノ酸のことを指すのである。

Ile-Val-Arg-Arg-Ala-Asp-Arg-Ala-Ala-Val-Proの標的性を高めることができる(表1参照)。
高性能血管生成抑制剤の製造方法であり、そのステップは以下の通りである。
Ala-Cys-Asp-Cys-Arg-Gly-Asp-Cys-Phe-Cys-Linkerを含む配列であり、その中の-Linkerは一つ又は複数の異なるアミノ酸である。
を行い、用いられたPEG分子量範囲が直鎖状(相対分子量が2000-30000Dα)又は分枝状(相対分子量が40000-60000Dα)であり、その中には、(1)PEG-ビニルスルホン酸(PEG-Vinylsulphone)と、(2)PEG-ヨードアセトアミド(PEG-Iodoacetamide)と、(3)PEG-マレアミド(PEG- Maleimide)と、(4)PEG-オルトピリジンジスルフィド
(PEG-Orthopyridyldisulfide)と、(5)SC-mPEG(メトキシ化PEGスクシンイミジルカーボネート)と、又はSS-PEG(スクシンイミジルスクシネート)又はPEG-イソシアン酸エステル(PEG-lsocyanate)(7)(8)と、(6) メトキシ化PEGプロピオンアルデヒド(mPEG-ALD)が含まれる。
ナノ薬物製造が含み、ポリ乳酸(PLA)ナノ粒子やナノ微粒子、又はポリ----ーシアノアクリレートブチル(PBCA)ナノ粒子やナノ微粒子、又はキトサンナノ粒子やナノ微粒子の製造が含まれる。
本発明の有益な効果は下記の通りである。
合成コーデイングされたRGD-EDポリペプチド配列の塩基をテンプレートにする。上流・下流プライマーを設計して、その中、上流プライマーにNdeI酵素切断部位を加え、下流プライマにはArg-Gly-Asp配列とXhoI部位を有する。PCR増幅し、増幅生成物がアガロースゲル電気泳動回収と純化を経て、NdeIとXhoI制限酵素切断を行い、原核生物の発現ベクターpwにクローニングし、PCR陽性クローンをスクリーニングする。ヌクレオチド配列の分析でその配列に設計上著しい変化が発生したのを確認した。
合成のプライマー1:5’GGAATTCCATATG ATCGTGCGCCGTGCCGACCGC3’
合成のプライマー2:5’CCGCTCGAGGCAGAAGCAGTCACCACGGCA3’
うち、プライマー1はNdeI部位とIle-Val-Arg-Arg-Ala-Asp-Arg-Ala-Ala-Val-Pro
の一部の配列をコーデイングする。プライマー2はXhoI部位とArg-Gly-Asp配列を有する遺伝子をコーデイングした。
Ile-Val-Arg-Arg-Ala-Asp-Arg-Ala-Ala-Val-Pro(ED)。
発現型プラスミドを大腸菌に転換させ、組み換え菌が1mMIPTG誘発3時間後細胞を採取し、超音波破砕し、上清と沈澱に遠心分離し、15%SDS-PAGEの電気泳動、クマシーブリリアンドブルー染色のSDS-PAGEスキャン(UVP White/Ultraviolet transilluminator)を行い、その発現結果を分析する。
超音波破砕と遠心分離をし、封入体沈澱を0.1Mtrisとデオキシコール酸ナトリウムにて洗滌して、沈澱をラウリルサルコシン酸ナトリウム(SLS)に溶解する。4℃、10000rpm にて5min遠心分離し、上清液4℃にて透析する。透析液はbufferA(10mM Tris-HCI、0.1mM酸化型グルタチオンと1mM還元型グルタチオン、pH7.4)である。6−8hで一回液体を取り替え、計三回取り替える。最後に一回透析をする。透析液はbuffer B(10mM Tris-HCI、pH7.4)である。サンプルに対して直接SP-Sepharose Fast Flow(Amersham Pharmacia Biotech)-クロマトグラフィーを行う。カラムへの平衡洗滌はbuffer Bを使い、0.6M NaCI、 Tris-HCI、 pH7.4と1M NaCI、Tris-HCI、pH7.4 を使って順番で溶出する。溶出液を混合させ、buffer B透析後、冷凍乾燥して濃縮させる。クロマトグラフィーの結果は図1の示した通りである。
BCE細胞とNIH 3T3細胞を培養する。その方法:培養液DMEMに10%の不活化牛胎児血清(BCS)、1%の抗生物質及び3ng/ml bFGFが含まれる。細胞増殖分析方法は下記の通りである。PBSにて細胞を洗滌してトリプシンを消化する。浮遊細胞培養液を加え、遠心分離して細胞を採取する。細胞の濃度を25,000 cells/mlに調整する。細胞を6オリフィス(0.5ml/well)に移入して24h培養し、培養液を1ml DMEMに交換する。5%BCS、1%抗生物質、1ng/ml bFGF、各ホールに異なる投与量を入れてさらに48h培養する。トリプシンにて細胞を消化してPBSに再浮遊させる。70%の冷たいエタメールにて30min 固定させ、7-AAD染色する。細胞流量計測器で分析を行う。
体内の抗血管生成活性を測定するため、CAM分析をする。すべての実験がクリーンベンチに無菌操作で行われる。消毒した六日間の鶏胚を37℃、90%湿度の条件下で培養する。二日間後、各卵の上部に穴を開けて試薬を滅菌のWhatman濾紙に滴加し、CAMsの血管集中部位に入れる。48時間培養した後、鶏胚とCAMsを観察して写真を撮る。
図2の示したように、RGD-ED体内抗新生血管生成活性を評価するため、用量の異なるRGD-EDを使ってCAM実験を行った。うち、0.05μg、0.1μg と0.2μgのRGD-EDが明らかに新生血管と血管生成を抑制できるのである。0.5μgRGD-EDは完全に血管生成を抑制でき、鶏胚の死亡に至るのである。RGD-EDは血管生成抑制の潜在的作用を有する。
約1−70重量%のポリペプチドを最適約20−50%にして、約20−95重量%のポリエチレングリコールSC-mPEG (メトキシ化PEGスクシンイミジルカーボネート、平均分子量5000)又はSS-PEG(スクシンイミジルスクシネート)又はPEG-イソシアン酸塩(PEG-lsocyanate)の溶液と混合する。約50−93重量%の上記修飾物を取って充分混合し、4℃−40℃の間、最適25℃−37℃のシェーカーで10分以上振とう反応させる。イオン交換クロマトグラフィーでN末端修飾される生成物を分離する。
約1−70重量%のポリペプチドを最適約20−50%にして、約20−95重量%のmPEG-NH2(平均分子量5000)、少量のDCCI(ジシクロヘキシルカルボジイミド)溶液と混合する。最適約50−93重量%の上記修飾物を充分混合し、4℃−40℃の間、最適23℃−37℃のシエーカーで10分以上振とう反応させる。最適90分、カルボキシがmPEG-NH2の中のアミノ酸と結合してアミドを形成する。RP-HPLC分離でN末端の修飾される生成物を得る。
約1−70重量%のポリペプチドを最適約1−30%にして、約20−95重量%のmPEG-MAL(mPEG-マレイミド、平均分子量5000)溶液と混合する。最適約70−94%の上記修飾物を充分混合し、4℃−40℃の間、最適20℃−37℃のシエーカーで10分以上振とう反応させる。最適60分、イオン交換クロマトグラフィーでN末端の修飾される生成物を得る。
その他の修飾方法はすべてPEG修飾を参照する。最良修飾効果はPEG修飾とリポソーム修飾である(図4参照)。
培養したB16F10悪性黒色腫細胞を0.05%のトリプシンで消化し、1000rpm 5分遠心分離し、PBSに再浮遊させる。C57BL/6マウス(6-8週)の外側皮下に5x105細胞0.1mlを接種する。腫瘍の平均体積が200mm3-300mm3に達した際、マウスをランダムに組を分け、一組7匹にする。中の一組はRGD-ED治療を受け、もう一組はRGD-EDの含まない治療を受ける。上記二組の線量は皆5mg/kg/dであり、対照しながらPBSを注射する。治療は腫瘍接種の反対側に皮下注射方法を用いる。毎日ノギスで腫瘍の大きさを測定して腫瘍の体積を計算する。使用公式は腫瘍体積=長さ×幅2×0.52で、治療効果は限定時間内腫瘍の抑制率で表現、即ち、(1-T/C)×100%で、T=治療組の腫瘍体積、C=対照組の腫瘍体積。
プライマー2コーデイングするXhoI部位に有する遺伝子配列Arg-Gly-AspはArg-Gly-Asp-Gly-Gly-Gly-Gly配列に変わる。増幅して Ile-Val-Arg-Arg-Ala-Asp-Arg-Ala-Ala-Val-Pro-Gly-Gly-Gly-Gly-Arg-Gly-Asp
の遺伝子配列を得て、クローニングする。

約1−70重量%のポリペプチドを最適約1−30重量%にして、約20−95重量%のmPEG-MAL(mPEG-マレアミド、平均分子量5000)の溶液と混合する。最適約70−94%の上記修飾物を充分混合し、4℃―40℃の間、最適20℃−37℃のシエーカーで10分以上振とう反応させる。最適60分。イオン交換分離でN末端の修飾される生成物を得る。
培養した人肝細胞癌SGC7901を0.05%のトリプシンで消化し、1000rpmで5分遠心分離してPBSに再浮遊させ、6-8週間の裸マウスの外側皮下に5×105細胞0.1mlを接種する。腫瘍の平均体積が100mm3-200mm3に達した時、マウスをランダムに組を分け、一組7匹にする。うち、一組はRGD-ED治療を受け、もう一組はリポソーム修飾のRGD-EDで治療し、あとの一組はポリエチレングリコール修飾のRGD-EDで治療する。上記三組の線量は皆3mg/kg/dであり、対照しながらPBSを注射する。治療は腫瘍接種の反対側に皮下注射方法を用いる。毎日ノギスで腫瘍の大きさを測定して腫瘍の体積を計算する。使用公式は腫瘍体積=長さ×幅2×0.52で、治療効果は限定時間内腫瘍の抑制率で表現、即ち、(1-T/C)×100% で、T=治療組の腫瘍体積、C=対照組の腫瘍体積。
約10−90重量%のポリペプチドを最適約25−50重量%にして、約10−90重量%の活性化低分子量ヘパリンと混合し、最適約43−75重量%。pH=7-9の緩衝液に4℃、18時間以上軽く振とう反応させ、イオン交換クロマトグラフィーで遊離アノミ基の修飾される生成物を分離する。
80mg−120mgのPEG-PLA と15mg-30mgのポリペプチドを取り、40mlのN,N-ジメチルホルムアミドに溶解して混合物をMWCO 3500の透析袋に移す。3L−4Lの蒸留水に48時間透析し、遠心分離して沈澱を取り除く。上清を0.45μmで膜濾過してポリマーのミセル溶液を得る。
1gのデキストラン(分子量35000)を取って活性化させ、10mg−60mgのポリペプチドを入れる。最適20mg−45mg。4℃のシエーカーで15時間以上ゆっくりと振とう反応させる。イオン交換クロマトグラフィーで遊離アミノ基の修飾される生成物を分離する。
約5−90重量%のポリペプチドを最適約25−60重量%にして、約10−95重量%の活性化パルミチン酸と混合する。最適約43−90重量%、4℃―40℃の間、最適25℃−37℃のシエーカーで30分以上振とう反応させる。イオン交換クロマトグラフィーで遊離アミノ基の修飾される生成物を分離する。
約1−90重量%のポリペプチドを最適約7−50重量%にして、約10−95重量%の活性化ポリシアル酸と混合する。最適約50%−93重量%、4℃―40℃の間、最適25℃−37℃のシエーカーで15時間以上振とう反応させる。イオン交換クロマトグラフィーで遊離アミノ基の修飾される生成物を分離する。
10mg-50mgのキトサンを取って20ml-40mlの水と混合し、20-40分超音波して微粒子分散液を獲得する。3.5mg-6mgのポリペプチドを入れて2ml-5mlの無水メタナールに溶解し、超音波の過程においてゆっくりと微粒子分散液に滴加してポリペプチドを封じておく。遠心分離でナノ微粒子を得る。

- 高性能血管生成抑制剤であって、血管生成を抑制するポリペプチドIle-Val-Arg-Arg-Ala-Asp-Arg-Ala-Ala-Val-Pro の両端に、少なくとも一端はインテグリンファミリーに親和性と結合力を有するポリペプチドが接続され、前記「インテグリンファミリーに親和性と結合力を有するポリペプチ」はArg-Gly-Asp又はArg-Gly-Asp-linkerの配列を含み、その中の-Linkerは一つ又は複数の異なるアミノ酸である、
ことを特徴とする高性能血管生成抑制剤。 - 前記Arg-Gly-Asp又はArg-Gly-Asp-Linkerを含むポリペプチド配列が以下の配列より選ばれたこと、即ち、

- 請求項1に記載のインテグリンファミリーに親和性と結合力を有する高性能血管生成抑制剤の製造方法であって、以下のステップが含まれる、即ち、
血管生成を抑制するポリペプチドIle-Val-Arg-Arg-Ala-Asp-Arg-Ala-Ala-Val-Pro の両端に、少なくとも一端はインテグリンファミリーに親和性と結合力を有するポリペプチドが接続され、前記「インテグリンファミリーに親和性と結合力を有するポリペプチド」はArg-Gly-Asp又はArg-Gly-Asp-Linker又はAla-Cys-Asp-Cys-Arg-Gly-Asp-Cys-Phe-Cys又はAla-Cys-Asp-Cys-Arg-Gly-Asp-Cys-Phe-Cys-Linkerを含む配列であり、その中の-Linkerは一つ又は複数の異なるアミノ酸である、
ことを特徴とするインテグリンファミリーに親和性と結合力を有する高性能血管生成抑制剤の製造方法。 - 前記ポリペプチド配列及びこのポリペプチドをコーディングする塩基配列が化学合成法を用い、形成されることを請求項6に記載のインテグリンファミリーに親和性と結合力を有する高性能血管生成抑制剤の製造方法。
- 前記ポリペプチド配列及びこのポリペプチド配列をコーディングする塩基配列が以下の方式である遺伝工学法を用い、形成されること、即ち、
Arg-Gly-Aspインテグリン結合配列ペプチドを含む血管生成抑制剤遺伝子配列を合成し、この配列をテンプレートにし、上流・下流プライマーを設計し、プライマー配列の5’末端と3’末端にクローニングに適切する制限酵素切断部位を付加し、PCR増幅し、RGD-ED遺伝子を獲得し、遺伝子をベクターにクローニングし、陽性クローンをスクリーニングし、ヌクレオチド配列の解析と鑑定を行い、
RGD-ED遺伝子は原核生物発現ベクターと組換え、発現型プラスミドを形成し、大腸菌に転化させ、IPTGで発現RGD-EDを誘導し、発現生成物は封入体として存在し、
封入体のタンパク分離、溶解、活性再生を行い、更にイオン交換クロマトグラフィーでRGD-EDタンパク生成物を分離・純化し、フロースルーを収集し、凍結乾燥し、
すべてのRGD−ED遺伝子に対し、真核生物の発現ベクターと組換え、発現型プラスミドを形成し、真核細胞に転化させ、発現RGD-EDを誘導し、発現生成物を分離・純化する、
こと請求項6に記載のインテグリンファミリーに親和性と結合力を有する高性能血管生成抑制剤の製造方法。 - 請求項1に記載のポリペプチド配列及びこのポリペプチド配列をコーディングする塩基配列に対する修飾方法であって、以下のステップの一つを用いること、即ち、
前記ポリペプチド配列にPEGで修飾する;
又は前記ポリペプチド配列に対し、へパリン修飾をする;
又は前記ポリペプチド配列に対し、デキストラン修飾をする;
又は前記ポリペプチド配列に対し、ポリビニルピロリドン修飾をする;
又は前記ポリペプチド配列に対し、PEG−ポリアミド酸共重合体修飾をする;
又は前記ポリペプチド配列に対し、パルミチン酸修飾をする;
又は前記ポリペプチド配列に対し、ポリシアル酸修飾をする;
又は前記ポリペプチド配列に対し、リポソーム修飾をし、前記リポソームはリポソームREV、乾燥したリポソームDRV、多重ラメラリポソームMvlから選択される;
又は前記ポリペプチド配列に対し、ナノテクノロジー修飾をする、
ことを特徴とするポリペプチド配列及びこのポリペプチド配列をコーディングする塩基配列に対する修飾方法。 - 前記ポリペプチドの修飾ステップは以下の通りであること、即ち、
(1) 1〜70重量%のポリペプチドを用いて、20〜95重量%のPEG又はへパリン又はデキストラン又はポリビニルピロリドン又はPEG−ポリアミド酸共重合体又はパルミチン酸又はポリシアル酸又はリポソーム又はナノ粒子溶液と混合する;
(2)4℃〜40℃の間、好ましくは25℃〜37℃であり、シェーカーで10分以上振とう反応させる;
(3)修飾された生成物を分離する、
ことを特徴とする高性能血管生成抑制剤の修飾方法。 - 前記ステップ(1)中のポリエチレングリコール(PEG)は用いられたPEG分子量範囲が直鎖状であり、相対分子量が2000-30000Dαであり、又は分枝状であり、相対分子量が40000-60000Dαであり、その中には(1)PEG-ビニルスルホン酸と、(2)PEG-ヨードアセトアミドと、(3)PEG ‐マレアミドと、(4)PEG-オルトピリジンジスルフィドと、(5)SC-mPEG(メトキシ化PEGスクシンイミジルカーボネート)又はSS-PEG (スクシンイミジルスクシネート)又はPEG-イソシアン酸エステルと、(6)メトキシ化PEGプロピオンアルデヒドとが含まれる、
ことを特徴とする請求項10に記載の高性能血管生成抑制剤の修飾方法。 - 請求項1〜5の何れか一項に記載の高性能血管生成抑制剤であって、当該高性能血管生成抑制剤及び修飾生成物を、抗腫瘍薬物を製造するために用いることを特徴とする高性能血管生成抑制剤の用途。
