Abstract
Description
本出願は、2007年4月2日出願の米国仮特許出願第60/909,656号、表題「癌バイオマーカーのin vitroおよびin vivo検出のための表面増強ラマン散乱に基づく新種のナノ粒子タグ」に基づく優先権を主張し、参照することによりその全体を本明細書に組み込む。
本発明は、アメリカ合衆国政府の米国国立衛生研究所により付与されたNIH助成金第R01CA108,468号に基づいて政府支援により実施された。政府は、本発明に一定の権利を有する。
本開示は一般的に、表面増強ラマン分光ナノ粒子およびそれらの細胞検出用途に関する。

本明細書で使用したような「ラマン光散乱」という用語は、ある分子が照射されたとき、光子を保持した分子のごく一部が、保持した光子を放出した後、元の振動レベルには戻らないが、異なる振動レベルの基底電子状態まで下降することを意味する。したがって、これらの分子から放射される放射物は、エネルギーが異なり、したがって波長も異なる。このことをラマン散乱という。
本明細書で例示し広く説明したように、本開示の目的に従って、本開示の実施形態は、表面増強ラマン分光(SERS)活性複合体ナノ構造、これらのナノ構造の製造方法、およびこれらのナノ構造の使用方法を包含する。SERS活性複合体ナノ構造は、区別することが可能で、個々に検出することができる。この点に関して、SERS活性複合体ナノ構造は、SERS活性複合体ナノ構造がある種の標的分子と相互作用し、標的分子の検出を可能にするように修飾することができる。さらに、SERS活性複合体ナノ構造は、コード化システムならびに多重化システムで使用することができる。SERS活性複合体ナノ構造は、多くの領域、例えば、フローサイトメトリー、化学アレイシステム、生体分子アレイシステム、バイオセンシング、バイオ標識、高速スクリーニング、遺伝子発現研究、タンパク質研究、医療診断、診断用ライブラリー、およびマイクロ流体システムで使用することができる(がそれらだけに限られない)。
試薬:超純水(18MΩcm−1)が全操作を通じて使用された。以下の化学物質は商用供給源から入手し、さらに精製することなく使用した。1ミリリットル当たり2.6×1010粒子の濃度の60nmクエン酸安定化金粒子(Ted Pella Inc.)、近赤外線放射量子ドット(QD705、Invitrogen)、マラカイトグリーンイソチオシアナート(MGITC)(Invitrogen)、ジエチルチアトリカルボシアニンヨージド(DTTC)(Exciton)、mPEG−SH(MW約5kDa)(Nektar Therapeutics)、HS−PEG−COOH(MW約3kDa)(Rapp Polymers)。ヒト癌細胞系Tu686は、舌の基部の原発性腫瘍から確立された。ヒト癌細胞系NCI−H520は、アメリカ培養細胞系統保存機関(ATCC)から購入した。細胞培養培地、牛胎児血清、血球計数計、および細胞培養用品は、Fisher Scientificから購入した。その他の試薬はすべて、Sigma−Aldrichの最高純度のものを購入した。
合成:約60ナノメートルの標的直径を有する金コロイドは、文献の手順に従って合成された。ガラス製品はすべて厳密に洗浄され、使用前に水で濯いだ。50mLのガラスフラスコ内で、30mLの0.01%HAuCl4水溶液を磁石で撹拌しながら沸騰させた。沸騰したら、180μLの1%クエン酸ナトリウムを迅速に注入した。数分以内に、淡黄色の溶液は濃紫色に変化し、直ちに赤色になった。コロイドは、確実に還元を完了するために約15分間沸騰させ、室温まで冷却し、使用前に30mLに再構成した。
ペグ化SERSナノ粒子の調製:新たに調製したレポーター溶液(3〜4μM)を、迅速に混合している金コロイドに1:6のレポーター溶液/コロイド体積比で滴下し、容易に、金粒子表面にレポーター分子を均等に分布させた。金粒子に対するレポーター分子のモル比は、SERS強度を最大にし、コロイド凝集を最小限にするために最適化した。例えば、最適化された表面被覆値は、60nmの金粒子1個当たりマラカイトグリーンイソチオシアナート14,000分子であり、同じ大きさの金粒子1個当たりクリスタルバイオレット約15,300分子であった。上記のパラメータ(すなわち、レポーター保存溶液濃度、レポーター保存溶液の金ナノ粒子溶液に対する体積比、および金に対するレポーターの添加率)はすべて、生じるタグの凝集状態に影響を及ぼしたことに注意されたい。レポーター溶液を金コロイドに添加したとき、金をレポーターに添加したときよりも高いSERS信号が認められた。
ナノ粒子の特徴付け:UV−Vis吸収スペクトルは、Shimadzu(UV−2401)分光計で、使い捨てポリアクリルキュベットを使用して記録した。透過型電子顕微鏡写真(TEM)は、Hitachi H7500高倍率電子顕微鏡を使用して撮影した。静電気を減少させるためにUV光で前処理した銅の200メッシュの格子にナノ粒子試料(5μl)を滴下した。30分後、濾紙で溶媒を吸い取り、リンタングステン酸染色溶液(1重量%、pH6に調整)を30秒間適用した。新鮮な腫瘍組織標本を、2.5%グルタールアルデヒドを含有する0.1Mカコジル酸緩衝液(pH7.4)を用いて4℃で固定した。組織を0.1Mカコジル酸緩衝液で15分間、3回濯ぎ、1%OsO4緩衝液で後固定し、その後脱水し、樹脂(Epon)に包埋した。超薄切片(約60nm)をウルトラトーム機器で切り出し、TEM画像化のために銅格子に戴置した。
scFvリガンドとの結合:ヒトEGFRに特異的な抗体断片である、scFvB10は、YUAN−FCCCヒト未処理ファージディスプレイライブラリーから、確立された固相バイオパニング法を使用して単離した。大量のscFvが、細菌抽出物から、天然の条件下で、Ni2+NTA−アガロースカラム(Qiagen)を使用して精製された。95%を上回るタンパク質純度は、ドデシル硫酸ナトリウム(SDS)−PAGEを使用して測定した。ヘテロ官能性リンカーHS−PEG−COOH(430μlおよび1μM)は、ポリプロピレン試験管に入れた2.2mlのAu−MGITC(またはAu−DTTCI)溶液に、迅速に混合しながら滴下した。金粒子1個当たりのカルボキシ基の数は、使用したリンカー分子の量を変化させることによって、約5,000個になるように調節した。15分間混合した後、金ナノ粒子を大量のPEG−SH(10μMで1.6ml)に曝露して、ヘテロ官能性PEGによって被覆されていない領域を充填し、これにより、十分に遮蔽され、かつ安定な粒子表面を作製した。カルボン酸官能基にリガンドを共有結合させる前に、金粒子は、遠心分離(1,000g)およびPBSへの再懸濁を3回行うことによって精製した。
細胞SERSの研究:Tu686およびH520細胞はそれぞれ、10%熱不活性化牛胎児血清および抗生物質(ストレプトマイシン、ペニシリンG、およびアンホテリシンB)を添加したDMEM/Ham’s F−12(1:1)およびRPMI−1640で培養し、加湿したインキュベーター内で37℃、5%CO2で維持した。細胞は、35mmの皿で集密になるまで増殖させた。細胞染色法は、卓上結合インキュベーターで、25℃で無菌条件下において実施した。生細胞をscFv結合SERSナノ粒子(15pM、PBS溶液)と30分間穏やかに混合し、次いで穏やかに掻き取って収集した。細胞は、氷冷PBSで4回洗浄を行い、SERSを測定する前に500μlのPBSに再懸濁した。細胞の一部をペグ化対照SERSタグとインキュベーションし、非特異的結合および内部移行を評価した。バックグラウンドの細胞散乱を評価するために、対照SERSタグもEGFR−SERSタグも付加していない細胞の一部をさらに対照として使用した。SERSスペクトルは、コールターカウンターで測定した細胞数に対して正規化した。
腫瘍異種移植片およびin vivoにおけるSERS:50フェムトモルのペグ化SERSナノ粒子を、健康なヌードマウスの2箇所に投与した:(i)皮下注射(皮膚の下1〜2mm)および(ii)筋肉の深部に注射(皮膚の下1cm)。NIRラマン分光計(Inspector Series、DeltaNu)を使用することによって、様々な位置を調べた。皮下SERSスペクトルは3秒で得られ、筋肉スペクトルは21秒で得られ、(注射部位から離れた領域で得られた)対照スペクトルもまた、21秒で得られた。
ペグ化SERSナノタグの設計および特徴付け:図1A〜1Dは、分光学的タグが包埋されたペグ化金ナノ粒子の設計および調製ならびにその概略構造を示す。光吸収スペクトル(図1B)、透過型電子顕微鏡(TEM)構造(図1C)、および流体力学的な大きさのデータ(図1D)も示す。元の金粒子(直径60nm)は、ラマンレポーターでコード化され、チオール−PEGの層で安定化されている。以前の実験で、約60〜80nmの大きさのコアを備えた金ナノ粒子が、赤(630〜650nm)および近赤外線(785nm)の励起でSERSとして最も効率的であることが示された(Krugら、(1999)J.Am.Chem.Soc.121:9208〜9214)。
ペグ化金ナノ粒子の安定性は、濃縮塩(1〜2M)、強酸(0.1M HCl)、強塩基(1M NaOH)、および有機溶媒(メタノール、エタノール、およびジメチルスルホキシドすなわちDMSO)を含む多種多様な条件下で、SERS信号(振動数および強度の両方)を測定することによって研究した。図AおよびB。PEG保護がないと、金ナノ粒子はこのような過酷な条件下では迅速に「壊れる」(すなわち、凝集し、沈殿する)。PEG保護によって、金粒子およびそのSERSスペクトルは完全に安定で、pH1〜2でも(金表面上のレポーター分子のプロトン付加および相対的方向の変化のために)ほんのわずかな相対的強度の変化があるだけである。
癌細胞の分光学的検出:癌細胞検出のために、標的指向金ナノ粒子は、チオール−PEG(約85%)およびヘテロ官能性PEG(SH−PEG−COOH)(約15%)の混合物を使用することによって調製した。図3Aで概略を示したように、ヘテロ官能性PEGは、高い特異性および親和性でEGFRに結合するリガンドであるscFv抗体(MW=25kDa)に共有結合した。UV−Vis吸収および蛍光データによって、約600コピーのscFvリガンドが各金ナノ粒子に結合することが示された。図3Bは、scFv結合金ナノ粒子とヒト癌細胞とをインキュベーションすることによって得られる細胞結合およびSERSスペクトルを示す。ヒト頭頚部癌細胞(Tu686)は、EGFR陽性であり(細胞1個当たり104〜105個の受容体)、強いSERS信号によって検出された。対照的に、ヒト非小細胞肺癌(NCI−H520)はEGFRを発現せず、SERS信号をほとんどまたは全く示さなかった。ターゲティングの特異性を確認するために、Tu686癌細胞を10倍過剰量の遊離scFv EGFR抗体中で予めインキュベーションし、その後競合的結合の研究のためにEGFR標識SERSナノ粒子を添加した。3回洗浄した後、細胞はごくわずかなSERS信号を示した。2部位サンドイッチ様式で第2抗体に結合したSERSナノ粒子の結合特異性もまた、試験して確認した。対照癌細胞(EGFR陰性)および対照ナノ粒子(単純なPEGコーティングナノタグ、および非特異的IgG抗体で機能化したPEGナノタグ)では、スペクトルは図3Bに示したように、弱いが再現性のあるバックグラウンドを示した。バックグラウンドが低いのはおそらく、細胞単離の最中に完全に除去されなかった混合溶液中に残存するSERSナノ粒子が原因であったが、非特異的結合またはナノ粒子内部移行も関与する可能性があった。赤外色素(ジエチルチアトリカルボシアニンすなわちDTTC)は分光学的レポーターとして使用され、785nmの励起で表面増強共鳴ラマン散乱(SERRS)を実現した。この共鳴状態は、吸着した色素が金属粒子への効率的なエネルギー移動によって光分解から保護されているので、光退色を引き起こさなかった。共鳴効果はさらに、表面増強ラマン信号を10から100倍高めることができ、これは、単一癌細胞のラマン分子プロファイリング研究に十分な感度である(図8)。この感受性は、手術によって取り出された癌組織標本、および末梢血試料から捕捉された循環腫瘍細胞の不均一な性質を調べるのに重要である。単一細胞のプロファイリング研究は、EGFRがモノクローナル抗体、およびタンパク質キナーゼをベースにした治療の有効なタンパク質標的であるので、臨床的に非常に重要である。
in vivoにおける腫瘍ターゲティングおよび検出:in vivoにおける光学的画像化および分光法についての重大な難題は、動物組織において光が散乱し吸収されるために、透過深度が限られることである。SERSスペクトルが、動物組織に埋め込まれたペグ化金ナノ粒子から得られるかどうかを測定するために、小用量のナノ粒子を生きた動物の皮下および筋肉深部に注射した。図4に示したように、高い解像度のSERS信号が皮下および筋肉注射によって得られた。
in vivoにおけるナノ粒子の分布および細胞内局在:誘導結合プラズマ質量分析(ICP−MS)を使用した定量的生体分布研究によって、図6に示したように、標的指向金ナノ粒子は非標的指向粒子よりも10倍効率よく腫瘍内に蓄積することが明らかになった。ICP−MSデータによってまた、肝臓および脾臓による非特異的粒子の取り込みが確認されたが、脳、筋肉、またはその他の主要な臓器ではほとんどまたは全く蓄積せず、健康なマウス31に注射した金ナノシェルで報告された生体分布データと同様であった。超微細構造TEM研究によってさらに、SERSナノ粒子がEGFR陽性腫瘍細胞によって取り込まれ、図10および11で示したようにエンドソームおよびリソソームなどの細胞内小器官に局在することが明らかになった。in vivoにおいてエンドサイトーシスされたナノ粒子は、結晶構造およびファセット構造をしており、これは、ほとんど同一のSERSスペクトルがin vitroおよびin vivoにおいてコード化金ナノ粒子から得られたという発見と一致した。ペグ化金粒子は、体循環において、ならびに細胞内小器官に取り込まれた後でも、完全なまま安定であるようである。金粒子注射の2〜3ヶ月後の動物では、毒性もその他の生理学的合併症も認められなかった。
過酷な条件下でのペグ化SERSナノ粒子の安定性:4回の独立した方法によって、濃縮PBS溶液中におけるAu−MGITC−PEG−SHの高い安定性が確認された。PEG−SHコーティングした、およびコーティングしていないAu−MGITC複合体を、図7Aおよび7Bで示したように、UV−vis吸収分光法、TEM、DLS、および視覚的観察によって調べた。コーティングしていないAu−MGITCにPBSを添加すると、UV−vis吸収スペクトルでの劇的なスペクトル変化、TEMにおける大きな凝集、および600〜1,000nmの水力学的直径の粒子の特有の集団の出現からわかるように、コロイドがすぐに凝集して沈殿し、ピンクから透明に明らかに色が変化した。対照的に、PBSで処理したPEG−SHコーティングAu−MGITCは、60nmの金の特徴的なプラズモン共鳴ピークが保存されており、TEMによって過半数が単一粒子であり(溶媒留去のため少数のクラスターを含む)、DLSでは粒子は単一モードの狭いサイズ分布をしており、ピンク色をしていることが示された。生体結合および細胞標識手順で直面する広範囲の条件の影響は、PEG−SHコーティングSERSタグのスペクトル特性に対する影響について調べた。
単一癌細胞のSERSスペクトルおよび相関プラズモン画像化:Tu686およびH520細胞は、8チャンバーガラススライドで集密になるまで増殖させた。15pMの濃度のscFv結合SERSタグを200μLの細胞培養培地に導入し、その後30分間穏やかに混合した。インキュベーション時間後、細胞をPBSで6回徹底的に洗浄して、画像化の前に遊離の金ナノ粒子を除去した。反射型暗視野画像は、ExamineR顕微鏡(DeltaNu、Laramie、Wyoming)で、20×対物レンズを使用して得た。暗視野コンデンサを使用して、白色光の細いビームをタングステンランプから試料に照射した。この方法で、細胞膜にSERSナノタグで染色した細胞は、金ナノ粒子の高い分散特性によって明るい金色を示した。EGFR陰性H520細胞は、大部分が暗いバックグラウンドを示した。Tu686 EGFR陽性細胞は、高レベルのEGFR受容体結合を示したが、一方H520 EGFR陰性細胞は、限られたEGFR発現を有するのみであった。単一細胞SERSスペクトルは、顕微鏡を785nmレーザー励起のラマンモードに変えることによって得られた。20×対物レンズを使用したレーザースポットの大きさは、焦点面で5×10μmであった。図8では、EGFR陽性細胞およびEGFR陰性細胞それぞれについて矢印で示した領域から記録されたSERSスペクトルを示した。各スペクトルは、10秒の曝露時間で得られた。
非標的指向(対照)SERSナノ粒子の生体分布研究:生きた動物におけるSERSナノ構造標的特異的抗体結合プローブの動きを調べるために、以下のこと、すなわち特異的取り込みおよび滞留、バックグラウンドまたは非特異的取り込み、血中クリアランス、ならびに器官分布を調べた。図9に示したように、非特異的ナノ構造の取り込みおよび滞留は主に肝臓および脾臓で起こり、脳、心臓、腎臓、または肺ではSERSナノ粒子の蓄積はほとんどまたは全くなかった。in vivoにおける器官の取り込みおよび分布のこのパターンは、デキストランコーティング磁気酸化鉄ナノ粒子と同様であった。過剰なCOOH基を有するポリマーカプセル化SERSナノ粒子では、腫瘍ターゲティングは認められず、非特異的な器官による取り込みおよび迅速な血中クリアランスが示された。表面PEG基を有するポリマーカプセル化SERSナノ粒子では、器官による取り込みの速度は減少し、血液循環の長さが改善され、腫瘍中のナノ粒子のゆっくりした蓄積が引き起こされる。図6に示したように、PEGによってカプセル化され、かつ抗EGFR抗体を生体結合したナノ粒子では、ナノ粒子は腫瘍の異種移植によって輸送され保持されるが、非特異的な肝臓および脾臓による取り込みがまだ認められた。
透過型電子顕微鏡(TEM)による細胞内における局在の研究:腫瘍、肝臓、脾臓、および腎臓は、細胞および組織の取り込みの後にどこで金ナノ粒子が付着されるかを決定するために、TEMで調べた。図10は、in vivoにおいて腫瘍ターゲティングするためにEGFR標的指向金ナノ粒子を全身的に注射したときの腫瘍組織断片の代表的なTEM画像を示す。このデータは、金ナノ粒子が(たいていが受容体媒介エンドサイトーシスを介して)腫瘍細胞に内部移行し、エンドソームおよびリソソームなどの細胞内小器官に局在することを明らかに示している。

Claims (26)
- コア金属ナノ粒子と、
前記コアの表面に配置されたラマンレポーター分子と、
前記コアおよび前記レポーター分子の表面上に配置されたカプセル化保護層
とを含む表面増強ラマン分光活性複合体ナノ構造であって、カプセル化レポーター分子が測定可能な表面増強ラマン分光特性を有するナノ構造。 - 前記ラマンレポーター分子が、イソチオシアナート色素、多重含硫有機色素、多重ヘテロ含硫有機色素、ベンゾトリアゾール色素、またはそれらの組合せから選択される、請求項1に記載のナノ構造。
