Abstract
【解決手段】抗老化剤に、檸条及び/又は油茶からの抽出物を有効成分として含有せしめる。また、ラミニン5産生促進剤又は紫外線(UV−B)照射によるダメージ回復作用剤に、檸条からの抽出物を有効成分として含有せしめる。さらに、I型コラーゲン産生促進剤、プロフィラグリン産生促進剤又はフィラグリン産生促進剤に、油茶からの抽出物を有効成分として含有せしめる。さらにまた、皮膚化粧料に、檸条及び/又は油茶からの抽出物を配合する。

Description
以下、本発明について説明する。
本発明の抗老化剤は、檸条及び/又は油茶からの抽出物を有効成分として含有する。また、本発明のラミニン5産生促進剤及び紫外線(UV−B)照射によるダメージ回復作用剤は、檸条からの抽出物を有効成分として含有する。さらに、本発明のI型コラーゲン産生促進剤、プロフィラグリン産生促進剤及びフィラグリン産生促進剤は、油茶からの抽出物を有効成分として含有する。
檸条(学名:Caragana chamlagu)は、中国北部から朝鮮半島等に分布するマメ科ムレスズメ属に属する落葉低木であり、これらの地域から容易に入手することができる。抽出原料として使用し得る檸条の構成部位としては、例えば、葉部、花部、枝部、茎部、種子部、根部等が挙げられるが、好ましくは種子部である。
檸条からの抽出物は、ラミニン5産生促進作用又は紫外線(UV−B)照射によるダメージ回復作用を有しており、油茶からの抽出物は、I型コラーゲン産生促進作用、プロフィラグリン産生促進作用又はフィラグリン産生促進作用を有しており、それぞれ皮膚に適用した場合の使用感と安全性とに優れている。そのため、檸条及び/又は油茶からの抽出物は、皮膚化粧料に配合するのに好適であり、特に、檸条及び/又は油茶からの抽出物が抗老化作用を有していることから、老化防止用皮膚化粧料に配合するのに好適である。
檸条の種子部100gを抽出溶媒1000mLに投入し、穏やかに攪拌しながら2時間、80℃にて加熱抽出した後、熱時濾過した。得られた濾液を40℃で減圧下にて濃縮し、さらに減圧乾燥機で乾燥させ、檸条抽出物(試料1〜3)を得た。抽出溶媒として水、50容量%エタノール(水とエタノールとの容量比=1:1)及び80容量%エタノール(水とエタノールとの容量比=1:4)を用いたときの各抽出率を表1に示す。
油茶の葉部100gを抽出溶媒1000mLに投入し、穏やかに攪拌しながら2時間、80℃にて加熱抽出した後、熱時濾過した。得られた濾液を40℃で減圧下にて濃縮し、さらに減圧乾燥機で乾燥させ、油茶葉部抽出物(試料4〜6)を得た。抽出溶媒として水、50容量%エタノール(水とエタノールとの容量比=1:1)及び80容量%エタノール(水とエタノールとの容量比=1:4)を用いたときの各抽出率を表2に示す。
製造例1で得られた檸条抽出物(試料2)について、以下のようにしてラミニン5産生促進作用を試験した。
式中、Aは「試料添加時の波長405nmにおける吸光度」を表し、Bは「試料無添加時の波長405nmにおける吸光度」を表す。
製造例1で得られた檸条抽出物(試料2)について、以下のようにして紫外線(UV−B)照射によるダメージ回復作用を試験した。
上記式において、Ntは「UV−Bを照射していない細胞での吸光度」を表し、Cは「UV−Bを照射し試料溶液を添加していない細胞での吸光度」を表し、Saは「UV−Bを照射し試料溶液を添加した細胞」での吸光度を表す。
製造例2で得られた油茶抽出物(試料5)について、以下のようにしてI型コラーゲン産生促進作用を試験した。
式中、Aは「試料添加時のI型コラーゲン量」を表し、Bは「試料無添加時のI型コラーゲン量」を表す。
製造例2で得られた油茶抽出物(試料5)について、以下のようにしてプロフィラグリン・フィラグリン産生促進作用を試験した。
10μg/列に調整したサンプルをSDS−PAGEにより展開し、PVDF膜に転写した。5%スキムミルクを含むPBS(−)でブロッキングを行った後、抗ヒトフィラグリンモノクローナル抗体(Harbor Bio-Products社製)、ビオチン標識抗マウスIg(Whole Ab,Amersham Biosciences社製)及びストレプトアビジンーペルオキシダーゼ複合体(CALBIOCHEM社製)を、0.1%Tween20、0.3%スキムミルクを含むPBS(−)で1000倍に希釈して順次反応させ、ECL Western blotting detection reagents and analysis system(Amersham Biosciences社製)の発光によりプロフィラグリン及びフィラグリンを検出した。検出したバンドをKODAK 1D Image Analysis Software EDAS290 Version3.5にて定量的に測定した。
上記式において、Aは「試料添加時のNet intensity(プロフィラグリン及びフィラグリンの合計値)」を、Bは「試料無添加時(コントロール)のNet intensity」を表す。
下記組成の乳液を常法により製造した。
