Description
従って、本発明の課題は優れたセマフォリン発現増強作用を有するとともに簡便に使用可能且つ安全性が高いセマフォリン発現増強剤を提供することにある。
(アスナロエキスの製法)
アスナロの乾燥枝葉(約1kg)を細切し、エタノール(10リットル)に浸漬して、室温で時々攪拌しながら7日間抽出を行い、得られた抽出液をろ過し、ろ液を5℃で3日間静置した後、再度ろ過して上澄みを得る。これに9倍量の50%1,3−ブチレングリコール溶液を加え、ろ過する。そのろ液を濃縮し、これに更に1,3−ブチレングリコールおよび精製水を加えて、ろ過して製品(約100L)とする。得られたアスナロエキスの1,3−ブチレングリコールの含有量は54.75%、含水量は45%、アスナロ固形物0.25%であった。
ヒト皮膚再構築モデルとしてテストスキンLSE−High(東洋紡社製)を用い、培養した。培地に参考例1のアスナロエキスの1%(V/V)1,3−ブチレングリコール:エタノール=1:1溶液もしくはコントロール溶媒(1,3−ブチレングリコール:エタノール=1:1)を2mg/cm2の量となるように添加して、72時間培養後、テストスキン表皮部分をピンセットで剥ぎ取り、RNAを回収し、RT-PCRでセマフォリン3A及びG3PDHの発現を調べた。
その結果を図1に示す。図1から明らかなとおり、アスナロエキス添加群で有意にセマフォリン3A発現度合い(SEMA3A/G3PDH)が上昇した。
Claims (1)
- アスナロを、一価アルコール、多価アルコール及び水性アルコールから選ばれる一種以上の溶媒で抽出して得られるアスナロ抽出物を有効成分とするセマフォリン発現増強剤。
