Abstract
【解決手段】本発明は有効量のテトラサイクリン誘導体を投与することを含む、哺乳類の細胞及び組織中の内在性インターロイキン-10生成の増進方法である。また、その方法は有効量のテトラサイクリン誘導体を哺乳類に投与することによりインターロイキン-10生成を増進することを含む。好ましいテトラサイクリン化合物はそれらの抗菌活性を低下又は排除するように修飾されたテトラサイクリン化合物である。その方法は過度のIL-1及びTNFα生成を特徴とする哺乳類の医療症状を治療するのに使用し得る。

Description
本発明は国立歯科研究協会により授与された、R37 DE-03987のもとに政府の支持によりなされた。政府は本発明に或る種の権利を有する。
本発明は哺乳類の細胞又は組織中の内在性インターロイキン-10(以下、IL-10と称する)生成の増進方法に関する。
インターロイキン、インターフェロン、コロニー刺激因子及びTNFαはサイトカインと称される多様な多機能性タンパク質のグループの例である。サイトカインは種々の細胞中に非常に低い濃度で通常存在する分泌可溶性タンパク質のクラスである。リンパ系細胞、炎症造血細胞及び結合組織細胞の如きその他の細胞(例えば、繊維芽細胞、造骨細胞)は細胞増殖、分化及びエフェクター機能を調節することにより免疫応答、炎症応答、修復応答及び急性期応答を調節する種々のサイトカインを分泌する。サイトカインの作用は種々の細胞型の高アフィニティー受容体への結合により媒介される(Collierら, Trends in Pharmacol. Sci. 10: 427-431 (1989))。
例えば、サイトカインインターロイキン-1(IL-1)はマクロファージ、滑膜細胞、ケラチノサイト、軟骨細胞及び多形核白血球の如き細胞型中で生成される。それは多種の症状、特に炎症により伴われる症状に役割を果たすことが知られている。
過度のIL-1生成は発熱、筋肉消耗及び嗜眠状態を生じ得る。IL-1の生物活性の総説について、Larrickら, Immunology Today 10:61-66(1989)を参照のこと。こうして、IL-1の特定の生物活性を抑制することが治療上望ましい。
IL-1活性を抑制する一つの方法は全身遺伝子療法の使用による。米国特許第5,766,585号明細書はIL-1アンタゴニストをコードする組換えベクターを投与することによる哺乳類の慢性関節リウマチ炎症及びその他の自己免疫疾患の治療方法を開示している。
別の重要なサイトカインはヘルパーT細胞、B細胞、単球、マクロファージ及びその他の細胞型により生成される35-40 kDaペプチドであるIL-10である。in vitroで、IL-10はIL-1及びTNFαを含むサイトカイン生成を抑制するその能力により証明されるように免疫抑制性を示した(総説について、Fiorentinoら, Journal of Immunology 147: 3815 (1991)を参照のこと)。
また、IL-10はその他の炎症性サイトカインの活性化を抑制し、それ故、強力な抗炎症活性を有する。また、IL-10はマスト細胞及び胸腺細胞の増殖を刺激することが知られている(Mooreら“Interleukin-10”, Annual Review in Immunology 11: 165-190 (1993))。
例えば、精製IL-10は或る型のウイルス感染症を抑制することがin vitroで示された。米国特許第5,665,345号明細書はIL-10を投与することによるヒト細胞中のヒト免疫不全ウイルス、レトロウイルス、及びカポージ肉腫の複製の抑制方法を開示している。
同様に、IL-10は重度の複合免疫不全マウスの骨髄転移を抑制することが示されていた(Stearnsら, Invasion Metastasis 17(2):62-74 (1997))。
過度のIL-1及びTNFα生成を特徴とする症状を治療するための上記の通常のアプローチは外在性の精製又は組換えIL-10を静脈内投与することに制限されていた。IL-10はタンパク質であるので、哺乳類に静脈内注入することが困難である。何とならば、タンパク質がしばしば溶液から滲出し、静脈内投与セットに使用されるプラスチック又はガラスに結合するからである。また、タンパク質はしばしば不適合性であり、またデキストロース又は生理塩類溶液の如き生理溶液と混合された時に沈殿する。加えて、経口経路及び局所経路がIL-10投与に利用できない。タンパク質が胃腸道中で分解されるので、経口経路が利用できない。
化合物、テトラサイクリンは下記の一般構造:

テトラサイクリンは、それらの抗生物質の性質に加えて、幾つかのその他の用途を有すると記載されていた。例えば、テトラサイクリンはまたコラーゲン分解酵素、例えば、哺乳類コラゲナーゼ、ゲラチナーゼ、マクロファージエラスターゼ及び細菌コラゲナーゼの活性を抑制することが知られている(Golubら, J. Periodont. Res. 20:12-23 (1985); Golubら, Crit. Revs. Oral Biol. Med. 2: 297-322 (1991);米国特許第4,666,897号;同第4,704,383号;同第4,935,411号;同第4,935,412号)。加えて、テトラサイクリンは哺乳類骨格筋中の消耗及びタンパク質分解を抑制することが知られていた(米国特許第5,045,538号)。
同様に、Golubらの米国特許第5,532,227号はテトラサイクリンがタンパク質の過度のグリコシル化を改善し得ることを開示している。特に、テトラサイクリンは糖尿病患者のコラーゲンの過度のグリコシル化から生じる過度のコラーゲン架橋を抑制する。
これらの性質はテトラサイクリンが幾つかの疾患を治療するのに有益にする。
例えば、非抗菌性テトラサイクリンを含む、テトラサイクリンは関節炎を治療するのに有効であるという幾つかの示唆があった。例えば、Greenwaldら,“テトラサイクリンはアジュバント関節炎でメタロプロテイナーゼ活性を抑制し、またフルビプロフェンと組み合わせて、骨損傷を改善する”, Journal of Rheumatology 19:927-938(1992); Greenwaldら,“MMPインヒビターによる分解関節炎障害の治療:マトリックスメタロプロテイナーゼの抑制におけるテトラサイクリンの潜在的な役割:治療潜在性”, Annals of the New York Academy of Sciences 732: 181-198 (1994); Kloppenburgら,“活性慢性関節リウマチにおけるミノサイクリン”, Arthritis Rheum 37:629-636 (1994); Ryanら,“骨関節炎における軟骨分解を改良するテトラサイクリンの潜在性”, Current Opinion in Rheumatology 8: 238-247 (1996); O'Dellら,“ミノサイクリン又は偽薬による初期の慢性関節リウマチの治療”, Arthritis Rheum 40:842-848 (1997)を参照のこと。
皮膚障害におけるテトラサイクリンの有効性がまたElewskiら, Journal of the American Academy of Dermatology 8:807-812 (1983)により研究されていた。
Elewskiらはテトラサイクリン抗生物質が皮膚疾患で抗炎症活性を有し得ることを開示していた。
同様に、Plewigら, Journal of Investigative Dermatology 65:532 (1975)は抗菌剤が炎症性皮膚病を治療するのに有効であるという仮説を試験するために設計された実験を開示している。Plewigらの実験はテトラサイクリンがヨウ化カリウムパッチにより誘発されるプステルを治療する際に抗炎症性を有することを証明している。
以上に基づいて、テトラサイクリンは異なる治療に有効であることが判明された。しかしながら、テトラサイクリンがIL-10の内在的生成を増進するのに使用し得ることは全く示唆されていなかった。
これらの目的及びその他の目的が有効量のテトラサイクリン誘導体を哺乳類の細胞に投与することによる哺乳類の細胞又は組織中の内在性インターロイキン-10生成の増進方法を提供する本発明により達成し得ることが今発見された。
本発明の好ましい実施態様が説明及び記載の目的のために選ばれたが、本発明の範囲を何ら限定するものではないことが意図されている。本発明の或る局面の好ましい実施態様が添付図面に示される。
本発明の方法は内在性IL-10生成を増進するのに有益であり,これはIL-1及びTNFα生成を抑制し、又はダウンレギュレートすることが知られている。本明細書に使用される、内在性IL-10生成の増進は哺乳類内のin vivo又は哺乳類の細胞もしくは組織内のin vitroの通常のレベルより実質的に上にIL-10サイトカインレベルを増大し、又はアップレギュレートすることと定義される。内在性IL-10生成は通常のレベルより少なくとも約10%から約1600%まで増進されることが好ましい。内在性IL-10をアップレギュレートすることはサイトカインIL-1及びTNFαのダウンレギュレーションをもたらす。
本発明の方法は、例えば、生きている哺乳類だけでなく、培養された組織、臓器又は細胞系中でin vivo、in vitro、及びex vivoで使用し得る。哺乳類として、例えば、ヒト、並びにペット動物、例えば、イヌ及びネコ、実験動物、例えば、ラット及びマウス、並びに牧場動物、例えば、ウマ及びウシが挙げられる。本明細書に使用される組織は一緒になって或る種の特別な機能を行なう同様に特殊化された細胞の集合である。培養された細胞系はIL-10を生成するあらゆる細胞、例えば、ヒト末梢血単球細胞又は滑膜繊維芽細胞様細胞を含む。
本明細書に使用されるテトラサイクリン誘導体は下記の一般構造:

テトラサイクリン抗生物質の使用は、一般に感染症を治療するのに有効であるが、望ましくない副作用をもたらし得る。例えば、抗生物質テトラサイクリンの長期投与は健康なフローラ、例えば、腸フローラを減少又は排除することがあり、また抗生物質耐性生物の生産又は酵母及び真菌の過剰増殖をもたらし得る。これらの重大な不利は典型的にはこれらの化合物の慢性投与を必要とする治療レジメを排除する。
抗生物質テトラサイクリンに構造上関連しているが、それらの抗生物質活性が化学修飾により実質的に排除され、又は完全に排除された化合物のクラスが特定された。抗生物質活性の実質的な排除は抗生物質活性がテトラサイクリンのそれよりも実質的に小さい場合に生じる。抗生物質活性がテトラサイクリンのそれよりも少なくとも約10倍小さいことが好ましく、テトラサイクリンのそれよりも少なくとも約5倍小さいことが更に好ましい。
しかしながら、環系の基本構造の変化、又は1-4位及び10-12位にある置換基の置換は実質的に小さい抗菌活性を有し、又は抗菌活性を実質的に全く有しない合成テトラサイクリンを一般にもたらす。
化学修飾テトラサイクリン(CMT)誘導体として、例えば、4-デジメチルアミノテトラサイクリン(CMT-1)、テトラサイクリノニトリル(CMT-2)、6-デジメチル-6-デオキシ-4-デジメチルアミノテトラサイクリン(CMT-3)、7-クロロ-4-デジメチルアミノテトラサイクリン(CMT-4)、テトラサイクリノピラゾール(CMT-5)、4-ヒドロキシ-4-デジメチルアミノテトラサイクリン(CMT-6)、12α-デオキシ-4-デジメチルアミノテトラサイクリン(CMT-7)、5-ヒドロキシ-6-α-デオキシ-4-デジメチルアミノテトラサイクリン(CMT-8)、4-デジメチルアミノ-12-α-デオキシアンヒドロテトラサイクリン(CMT-9)、及び7-ジメチルアミノ-6-デメチル-6-デオキシ-4-デジメチルアミノテトラサイクリン(CMT-10)が挙げられる。全てがIL-10の内在性生成を増進する非抗菌性テトラサイクリンとして有益である。
また、抗菌活性を有するテトラサイクリン誘導体が本発明において意図されている。しかしながら、このような化合物は抗菌活性を実質的に有しないが、哺乳類の細胞又は組織中でIL-10の内在性生成を増進するのに有効である量で使用されることが好ましい。この型の好ましい化合物として、テトラサイクリン、ドキシサイクリン、デメクロサイクリン、及びミノサイクリンが挙げられる。
本発明の方法において、有効量のテトラサイクリン誘導体が投与される。本明細書に使用される有効量は内在性IL-10生成を増進するという特定の結果を得るのに有効な量である。テトラサイクリン誘導体は抗菌活性を殆ど有しないか、又は全く有しない量で与えられることが好ましい。テトラサイクリン誘導体は、それが微生物の増殖を有意に阻止しない場合に有効に抗菌性ではない。それ故、その方法はその抗菌性を低下又は排除するように化学修飾されたテトラサイクリン誘導体を有益に使用し得る。このような化学修飾テトラサイクリンの使用が本発明において好ましい。何とならば、それらが抗菌性テトラサイクリンよりも高レベルで使用し得るとともに、或る種の不利、例えば、有益な微生物の無差別の死滅、及び耐性微生物の出現(これはしばしば抗微生物量又は抗菌量のこのような化合物の使用を伴う)を回避することができるからである。
被験者についての最大投薬量は望ましくない副作用又は耐えられない副作用を生じない最高投薬量である。例えば、テトラサイクリン化合物は約0.1mg/kg/日から約30mg/kg/日まで、好ましくは約1mg/kg/日から約18mg/kg/日までの量で投与し得る。本発明の目的のために、副作用として、臨床上重大な抗微生物活性又は抗菌活性だけでなく、毒性作用が挙げられる。例えば、約50mg/kg/日を超える投薬量はおそらくヒトを含む殆どの哺乳類で副作用を生じるであろう。いずれにしても、当業者は当分野の技能及び知識により導かれ、本発明は記載された効果を得るのに有効である投薬量を無制限に含む。
テトラサイクリン誘導体の投与は種々の方法で行ない得る。培養された細胞系又は組織系中で、テトラサイクリン誘導体は細胞又は組織を有効量のテトラサイクリン誘導体と直接接触させることにより投与し得る。
生きている哺乳類では、本発明のテトラサイクリン誘導体は非経口経路及び腸経路(これはまた徐放送達系を含む)により全身投与し得る。例えば、本発明のテトラサイクリン誘導体は送出の好ましい経路である静脈内で容易に投与し得る(例えば、静脈内注射)。静脈内投与は当業者により理解されるようにテトラサイクリン誘導体を好適な担体(ビヒクル)又は賦形剤中で混合することにより行ない得る。
経口使用又は腸使用がまた意図され、錠剤、カプセル、ピル、トローチ、エリキシル剤、懸濁液、シロップ、ウェハ、チューインガム等の如き製剤がテトラサイクリン誘導体を与えるのに使用し得る。
また、テトラサイクリン誘導体の組み合わされ、又は協調された局所かつ全身の投与が本発明に意図されている。例えば、非吸収性テトラサイクリン化合物、例えば、CMT-2又はCMT-6が局所投与でき、一方、被験者中で実質的な吸収及び有効な全身分布の可能なテトラサイクリン化合物、例えば、CMT-1、CMT-3、CMT-7、又はCMT-8が全身投与し得る。
当業者は哺乳類中の内在性インターロイキン-10生成の増進方法を実証するための下記の実施例の能力を認めるであろう。それ故、以下に示される結果は或る種の化学修飾テトラサイクリン誘導体が哺乳類の細胞又は組織中で内在性IL-10生成を増進することができることを明らかに示す。しかしながら、更に一般に、これらの誘導体は内在性IL-10を増進することが望ましいその他の生物系、及びその他の疾患又は症状に利用し得る。
以下の実施例は本発明の更なる理解を助けるために示される。使用される特別な方法及び条件は本発明の更なる例示であることが意図され、本発明の妥当な範囲を限定しない。
実施例1
LPS刺激ヒト末梢血マクロファージに関するテトラサイクリンの効果
健康なヒト血液ドナーから得られた末梢血単球細胞(PBMNC)をリンフォプレプ(ナイコムド、オスロ、ノルウェー)で密度勾配遠心分離により白血球濃厚液から分離した。Levy及びEdgington, Journal of Experimental Medicine, 151:1232 (1980)の操作を使用して、細胞をその後に塗布し、ヒト血清被覆プラスチック皿に付着させた。次いで細胞をヒト血清アルブミン及びEDTAを含むパック生理食塩水で脱着し、改良Wright-Giemsa染色剤で染色されたサイトスピン製剤の顕微鏡試験により90%以上の単核食細胞であることを確かめた。
次いで細胞をテフロン(登録商標)ビーカー中で非付着条件下で10%ヒトAB血清(NABI)、2ミリモル/Lのグルタミン、1ミリモル/Lのピルベート、25ミリモル/LのHEPES、100μg/mlのストレプトマイシン、及び20μg/mlのセフォタキシムを補給したRPMI1640培地(ギブコ、英国)を含む培地中で最低7日間にわたって培養した。Liaoら, Blood, 83(8):2294-2304 (1994)の操作を使用して、マクロファージ(単球由来マクロファージ)に分化したPBMNCを培養液から回収した。単球由来マクロファージを1x106細胞/mlで無血清培地中で再懸濁させ、種々の実験のためにウェルプレートに塗布した。ならし培地を回収し、その後の分析のために約-80℃で凍結した。
ならし培地
ならし培地は下記の添加剤とともにマクロファージ培養物を含んでいた:(a)細菌LPSなし;(b)0.2μg/mlの細菌LPS(これはIL-1、TNFα、及びIL-10の生成を増進することが知られている);(c)0.2μg/mlの細菌LPS+5μMのCMT-3(高レベルの抗コラゲナーゼ活性を有するテトラサイクリン誘導体);(d)0.2μg/mlの細菌LPS+10μMのCMT-5(認められる抗コラゲナーゼ活性を有しないテトラサイクリン誘導体);(e)0.2μg/mlの細菌LPS+10μMのCMT-3;(f)0.2μg/mlの細菌LPS+20μMのCMT-3。
結果を図1にグラフで示す。LPSを使用しないマクロファージの培養は検出できる内在性IL-10の生成をもたらさなかった。しかしながら、培養液中のマクロファージへの0.2μg/mlのLPSの投与は内在性IL-10生成を6時間後に約180pg/ml(ピコグラク/ml)に、また24時間の終了時に約230pg/mlに著しく刺激した。
同様に、10μMのCMT-3及び20μMのCMT-3は24時間の終了の時点で内在性IL-10生成を夫々約380pg/ml及び約400pg/mlに増大し続けた。10μMの投与量のCMT-5は8時間の期間で10μMのCMT-3と同じ内在性IL-10生成の増大を生じた。これはLPS単独により生じるIL-10のレベルを超えて内在性IL-10生成の約50%から約100%の増大に相当する。しかしながら、10μMのCMT-5は24時間の長いインキュベート期間ではCMT-3と同様に内在性IL-10生成を増大し続けなかった。
IL-1刺激ヒト末梢血単球細胞に関するテトラサイクリンの効果
健康なヒト血液ドナーから得られたヒトPBMNCをフィコール-ペーク(ファーマシア、米国)で密度勾配遠心分離により白血球濃厚液から新たに分離した。
培地
次いで細胞を24ウェル組織培養プレート中で1%FBS(ウシ胎児血清)、2ミリモル/Lのグルタミン、100μg/mlのストレプトマイシン、及び100単位のペニシリンを補給したRPMI1640培地(ギブコ、英国)を含む培地中で培養した。PBMNCを1x106細胞/mlで無血清培地中で再懸濁させ、種々の実験のためにウェルプレートに塗布した。ならし培地を回収し、その後の分析のために約-20℃で凍結した。
ならし培地は下記の添加剤とともにPBMNC培養物を含んでいた:(a)IL-1なし;(b)1ng/mlのIL-1;(c)1ng/mlのIL-1+5μg/mlのCMT-5(テトラサイクリンピラゾール);(d)1ng/mlのIL-1+5μg/mlのCMT-3;及び(e)1ng/mlのIL-1+5μg/mlのCMT-8。
ヒトIL-10を結合するモノクローナル抗体を含むELISAキット(エンドゲン社、ウォバーン、MA)を使用して、ならし培地を48時間のインキュベーション期間後に内在性IL-10生成について分析した。
図2はヒトPBMNCによるIL-10生成の棒グラフである。IL-1を使用しないで、IL-1を使用して、又はIL-1+CMT-5を使用して単核細胞を培養すると、検出できる内在性IL-10の生成をもたらさなかった。しかしながら、1ng/mlのIL-1+5μg/mlのCMT-3を用いる培養は48時間の終了時に約160pg/mlまでの量の内在性IL-10を生じた。同様に、単核細胞に添加されたIL-1+5μg/mlのCMT-8を用いる培養はIL-10の内在性生成を約180pg/mlまで増大した。これは内在性IL-10生成の約16倍までの増大に相当する。
IL-1刺激ヒト滑膜繊維芽細胞様細胞に関するテトラサイクリンの効果
ヒト滑膜繊維芽細胞様細胞(SF)を150cm2の組織培養フラスコ中で2ミリモル/Lのグルタミン、100μg/mlのストレプトマイシン、及び100単位のペニシリンを補給したDMEM培地中に1%のFBSを含む培地中で培養した。培養液は更に(a)1ng/mlのIL-1;(b)1ng/mlのIL-1+5μg/mlのCMT-5;(c)1ng/mlのIL-1+5μg/mlのCMT-3;及び(d)5μg/mlのCMT-5である対照を含んでいた。細胞を48時間の期間にわたってインキュベートした。メッセンジャーRNAを単離し、IL-10生成をノーザンブロット分析により分析した。
図3を参照して、ノーザンブロット分析を使用して、内在性IL-10mRNA発現又は生成を吸収単位に基づいてグラフで示す。CMT-3は吸収単位の増大により表されるSF細胞中の内在性IL-10生成を著しく増進した。図4を参照して、SF細胞中の内在性IL-10の生成は約25pg/mlまでであった。内在性IL-10生成の増大はIL-1+CMT-5で刺激された細胞について観察されなかった。これらの結果は或る種の化学修飾テトラサイクリン誘導体がSF細胞中で内在性IL-10生成を増進することができることを明らかに示す。
IL-1刺激皮膚繊維芽細胞様細胞中のIL-10生成に関するテトラサイクリンの効果
皮膚繊維芽細胞様細胞(DF)を150cm2の組織培養フラスコ中で2ミリモル/Lのグルタミン、100μg/mlのストレプトマイシン、及び100単位のペニシリンを補給したDMEM培地中に1%のFBSを含む培地中で培養した。培養液は更に(a)添加剤なし;(b)1ng/mlのIL-1;(c)1ng/mlのIL-1+5μg/mlのCMT-5;及び(d)1ng/mlのIL-1+5μg/mlのCMT-3を含んでいた。ヒトIL-10を結合するモノクローナル抗体を含むELISA(エンドゲン社、ウォバーン、MA)を使用して、内在性IL-10生成を測定した。培養液を48時間インキュベートした。
IL-1及びCMT-3を添加した場合、DF細胞はIL-10の内在性生成を示さなかった。
IL-1+CMT-5を添加した場合、同様の結果がDF細胞により示された。これらのデータはDF細胞が内在性IL-10を生成しないことを示す。これらの結果が図4にグラフで示され、これはIL-1、IL-1+CMT-5及びIL-1+CMT-3で刺激されたヒトDF細胞、ヒトSF細胞、及びヒトPBMNC中の内在性IL-10生成を要約する三次元のグラフである。
次の実験を設計して、生成された内在性IL-10が生物活性であることを測定した。内在性IL-10の生物活性を、2種のインジケーター細胞系U937(ATCC、受理番号CRL1593、ロックビル、Md)及びHL-60(ATCC、受理番号CCL240、ロックビル、Md)中でサイトカインTNF-α及びIL-1生成を抑制するその能力により測定した。これらの細胞系を1mlの平底ウェル中で培養した(1x106細胞/ml)。実験に使用した上澄みを生成するために、PBMNCを1ng/mlのIL-1+5μg/mlのCMT-8で刺激し、48時間インキュベートした。この上澄みは1.23pg/mlのIL-10を含んでいた。

Claims (21)
- 哺乳類の細胞に有効量のテトラサイクリン誘導体を投与することを特徴とする、哺乳類の細胞又は組織中の内在性インターロイキン-10生成の増進方法。
- 前記テトラサイクリン誘導体が有効な抗菌活性を実質的に有しない請求の範囲第1項記載の方法。
- 前記テトラサイクリン誘導体が4-デジメチルアミノテトラサイクリンである請求の範囲第1項記載の方法。
- 前記哺乳類の細胞又は組織が哺乳類中にある請求の範囲第1項記載の方法。
- 前記テトラサイクリン誘導体が6-デメチル-6-デオキシ-4-デジメチルアミノテトラサイクリンである請求の範囲第3項記載の方法。
- 前記テトラサイクリン誘導体がテトラサイクリノ-ピラゾールである請求の範囲第3項記載の方法。
- 前記テトラサイクリン誘導体が7-クロロ-4-デジメチルアミノテトラサイクリンである請求の範囲第3項記載の方法。
- 前記テトラサイクリン誘導体が4-ヒドロキシ-4-デジメチルアミノテトラサイクリンである請求の範囲第3項記載の方法。
- 前記テトラサイクリン誘導体が12α-デオキシ-4-デジメチルアミノテトラサイクリンである請求の範囲第3項記載の方法。
- 前記テトラサイクリン誘導体が5-ヒドロキシ-6-α-デオキシ-4-デジメチルアミノテトラサイクリンである請求の範囲第3項記載の方法。
