Abstract
Description
本発明は、(A)蛋白に結合したメチオニンスルホキシド(MSO)含量がモル基準で蛋白に結合した総メチオニンの8%以下の蛋白質、(B)脂質並びに(C)300センチポアズ(cps)未満のパッケージ粘度及び少なくとも300cpsの消費後誘導粘度をエマルションに付与する誘導粘性繊維系を含有する誘導粘性栄養エマルションに関する。該栄養エマルションは水中油エマルションである。
本発明の栄養エマルションは主に以下に記載する誘導粘性繊維系により消費後に粘度が増加する水中油エマルションである。これらのエマルションは消費前の飲用可能な粘度(本明細書ではパッケージ粘度と言う)と、消費後の増加した粘度(本明細書では誘導粘度と言う)をもつ。
本発明の栄養エマルションは蛋白に結合したメチオニンスルホキシドとしての蛋白に結合したメチオニンがモル基準で8%以下、好ましくは0〜5%、例えば1〜3%の蛋白源を含有する。
本発明の栄養エマルションは誘導粘性繊維系を含有しており、このような系は消費後にエマルションの粘度を増加し、パッケージ粘度と消費後のエマルションの誘導粘度が本明細書に定義する範囲内にある任意系を含む。
誘導粘性繊維系は参照によりその記載内容を本明細書に組込む米国特許出願第20030013679号に記載されているもの等のポリマー制御型誘導粘性繊維系が好ましい。このような系は中性可溶性繊維と、重合度(DP)が少なくとも10の澱粉部分水解物を含む。
本発明で使用する誘導粘性繊維系は参照によりその記載内容を本明細書に組込む米国特許出願第20020193344号に記載されているもの等の酸制御型誘導粘性繊維系を含む。このような系は非水溶性酸溶性多価カチオン源と共に可溶性アニオン性繊維を含む。
本発明の栄養エマルションは脂質と、蛋白質と、糖質を含有する。本明細書に記載するように、蛋白質は低MSO蛋白源でなければならず、糖質は誘導粘性繊維系を含んでいなければならない。脂質成分については以下に記載する。
本発明で使用する蛋白源のメチオニンスルホキシド(MSO)含量は以下の方法に従って測定する。適正な蛋白質選択のためには、蛋白源候補を37℃で24時間酵素加水分解する。次に、得られた酵素消化物のMSOとメチオニン含量を逆相HPLCにより測定する。次に蛋白源におけるMSOの百分率を以下のように決定する。
A.標準調製
a.L−メチオニン100mgとDL−メチオニンスルホキシド(Fluka 64430)50mgを水1000mLに溶かす。これを高標準溶液とする。
b.高標準溶液25.0mLを水で50mLまで希釈する。これを低標準溶液とする。
B.サンプル調製
a.0.1%ナトリウムアジドを添加した0.05M PIPES(pH7.5)25mLにカゼイン酸塩成分90〜95mgを溶かす。
b.カゼイン酸塩溶液3.00mLを1ドラムバイアルにピペッティングする。
c.0.1%ナトリウムアジドを添加した0.05M PIPES(pH7.5)中2mg/mLの濃度でプロナーゼ(Sigma P−5147)150μLを加える。
d.水中2mg/mLの濃度でロイシンアミノペプチダーゼ(Sigma L−0632)60μLを加える。
e.水中2mg/mLの濃度でプロリダーゼ(Sigma P−6675)30μLを加える。
f.バイアルにキャップをし、静かに混合する。クリンプシールHPLCオートサンプラーバイアル(VWR 66020−953)2本に1600μLずつ分配し、バイアルを密閉する。
g.37℃で24時間インキュベートする。
h.MSOとMETを下記HPLCシステムにより試験する。

移動相A:水
移動相B:0.02 KH2PO4(pH2.9)350mL;アセトニトリル650mL
温度:40℃
検出:UV 221nm,214nm
注入量:1μL
溶出プログラム:「C.HPLCシステム」の付表Aの表参照。
各種市販蛋白源のMSO含量を測定し、比較する。次に、誘導粘性栄養エマルションにおける各種蛋白源の物理的安定性を評価する。
Claims (12)
- (A)蛋白に結合したメチオニンスルホキシド含量がモル基準で蛋白に結合した総メチオニンの8%以下である蛋白質、
(B)脂質、ならびに
(C)300cps未満のパッケージ粘度及び少なくとも300cpsの消費後誘導粘度をエマルションに付与する誘導粘性繊維系
を含有し、水中油エマルションである誘導粘性栄養エマルション。 - エマルションが総カロリーの百分率として糖質10〜85%、脂質10〜85%及び蛋白質5〜40%を含有する、請求項1に記載の栄養エマルション。
- 蛋白に結合したメチオニンスルホキシド含量がモル基準で総蛋白に結合したメチオニンの1〜5%である、請求項2に記載の栄養エマルション。
- 誘導粘性繊維系がエマルションの重量を基にして可溶性アニオン性繊維源0.2〜5%及び非水溶性酸溶性多価カチオン200〜9000ppmを含有する、請求項1に記載の栄養エマルション。
- エマルションが可溶性アニオン性繊維0.4〜3重量%及び非水溶性酸溶性多価カチオン源200〜1000重量ppmを含有する、請求項4に記載の栄養エマルション。
- エマルションが可溶性多価カチオンを実質的に含まない、請求項4に記載の栄養エマルション。
- 可溶性アニオン性繊維がアルギン酸塩、低メトキシペクチン、カラゲーナン、キサンタン、ゲランガム及びその組み合わせから構成される群から選択される、請求項4に記載の栄養エマルション。
- 誘導粘性繊維系がエマルションの重量を基にして少なくとも0.4%の中性可溶性繊維及び重合度が少なくとも10の澱粉部分水解物を少なくとも2%含有する、請求項1に記載の栄養エマルション。
- エマルションが中性可溶性繊維0.55〜3.0重量%及び澱粉部分水解物2〜6重量%を含有する、請求項8に記載の栄養エマルション。
- 中性可溶性繊維がグアーガム、高メトキシペクチン、ローカストビーンガム、メチルセルロース、βグルカン、グルコマンナン、コンニャク粉及びその組み合わせから構成される群から選択される、請求項9に記載の栄養エマルション。
- 澱粉部分水解物が重合度約40〜約250である、請求項10に記載の栄養エマルション。
- エマルションがパッケージ粘度40〜250cps及び誘導粘度400〜20,000cpsである、請求項1に記載の栄養エマルション。
