Abstract
Description
項1. エステル化度が10~30%、アミド化度が0~25%、且つ遊離酸度が64%以上85%未満であるペクチンと、脂質と、前記ペクチン以外の炭水化物と、タンパク質とを含有する下痢防止用栄養組成物。
項2. 前記ペクチンの重量平均分子量が35万~60万である、項1に記載の下痢防止用栄養組成物。
項3. 前記炭水化物として、前記ペクチン以外の食物繊維、及び/又は糖質を含む、項1又は2に記載の下痢防止用栄養組成物。
項4. 前記炭水化物として、アラビアガム及び/又はガティガムを含む、項1~3のいずれかに記載の下痢防止用栄養組成物。
項5. 更に2価の金属塩を含む、項1~4のいずれかに記載の下痢防止用栄養組成物。
項6. 前記2価の金属塩が、酸性領域で2価の金属イオンを遊離する不溶性塩である、項5に記載の下痢防止用栄養組成物。
項7. 液状である、項1~6のいずれかに記載の下痢防止用栄養組成物。
項8. 前記ペクチンを3~15g/L含む、項7に記載の下痢防止用栄養組成物。
項9. 25℃での粘度が、2~100mPa・sである、項7又は8に記載の下痢防止用栄養組成物。
項10. 下痢防止用栄養組成物の製造のための、
エステル化度が10~30%、アミド化度が0~25%、且つ遊離酸度が64%以上85%未満であるペクチンと、脂質と、前記ペクチン以外の炭水化物と、タンパク質とを含有する組成物の使用。
項11. 下痢の防止が求められる者に、項1~9のいずれかに記載の下痢防止用栄養組成物を投与又は摂取させる工程を含む、下痢防止方法。
本発明の下痢防止用栄養組成物では、エステル化度が10~30%、アミド化度が0~25%、且つ遊離酸度が64%以上85%未満であるペクチンを使用する。このような特定の構造のペクチンを選択することによって、優れた下痢防止効果を奏することが可能になる。また、当該ペクチンは、本発明の下痢防止用栄養組成物に経管投与を行うのに適した粘性を備えさせ、且つ本発明の下痢防止用栄養組成物を液状のまま保管しても粘度安定性を備えさせることができる。更に、当該ペクチンは、酸性領域で2価の金属塩と共存するとゲル化する特性を備えているので、本発明の下痢防止用栄養組成物に2価の金属塩を配合した場合には、摂取又は投与させる際の液状形態において適度な流動性を備えつつ、胃内に入るとゲル化することにより胃食道の逆流を防止することも可能になる。
試料約5gを精密に量り、ビーカーに入れ,塩酸5ml及び60容量%エタノール100mlを加え、10分間かき混ぜた後、ガラスろ過器(1G3)を用いてろ過し,残留物を60vol%エタノール/塩酸混液(20:1)15mlずつで6回洗う。次に、60容量%エタノールで先のガラスろ過器上の残留物を洗液が塩化物の反応を呈さなくなるまで洗う。更にエタノール20mlで洗い、105℃で2.5時間乾燥し、冷後、質量を測定する。この約10分の1に当たる量を精密に量り、その質量をW(mg)とする。これにエタノール2mlを加えて湿らせ、煮沸して冷却した水100mlを加え、時々振り混ぜてよく水和させた後、フェノールフタレイン試液を5滴加え、0.1mol/L水酸化ナトリウム溶液で滴定し、滴定値をV1とする。次に0.5mol/L水酸化ナトリウム溶液20mlを正確に量って加え、よく振り混ぜ、15分間静置する。更に0.5mol/L塩酸20mlを正確に量って加え、液の桃色が消えるまで振り混ぜ、0.1mol/L水酸化ナトリウム溶液で滴定し、滴定値をV2とする。終点は、激しく振り混ぜるとき、液がわずかに桃色を呈するときとする。窒素定量法中のケルダール法の装置に従い,滴定した液を500mlの分解フラスコに移し、しぶき止め及び冷却器を付ける。あらかじめ0.1mol/L塩酸20ml及び新たに煮沸して冷却した水150mlを吸収用フラスコに入れ、冷却器の下端をこの液中に浸す。水酸化ナトリウム(1→10)溶液20mlを分解フラスコに入れ、泡立ち過ぎないように注意しながら加熱し、80~120mlが留出するまで蒸留する。メチルレッド試液を数滴加え、0.1mol/L水酸化ナトリウム溶液で滴定し、滴定値をSとする。別に空試験を行い、滴定値をBとする。そして、下記算出式に従って、エステル化度、アミド化度、及び遊離酸度を算出する。
本発明の下痢防止用栄養組成物には、栄養成分として脂質が含まれる。
本発明の下痢防止用栄養組成物には、栄養成分として、前記ペクチン以外の炭水化物が含まれる。
本発明の下痢防止用栄養組成物には、栄養成分としてタンパク質が含まれる。
本発明の下痢防止用栄養組成物は、液状で提供される場合、基材として水が含まれる。また、本発明の下痢防止用栄養組成物は、固形状の形態で提供される場合は、投与又は摂取を容易ならしめるために、水を添加して液状の形態にした後に投与又は摂取される。
本発明の下痢防止用栄養組成物は、2価の金属塩を含んでいることが望ましい。前述する特定構造のペクチンは、酸性領域で2価の金属塩と共存するとゲル化する特性を備えているので、本発明の下痢防止用栄養組成物に2価の金属塩を配合した場合には、摂取又は投与させる際の液状形態において適度な流動性を備えつつ、胃内に入って酸性環境に晒されると当該ペクチンのゲル化を促進して胃食道の逆流を防止することが可能になる。
本発明の下痢防止用栄養組成物は、更に、必要に応じて、レシチン、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル、ソルビタン脂肪酸エステル等の乳化剤、pH調整剤、ビタミン類、ミネラル、甘味料、酸化防止剤、防腐剤、調味料、着色料、香料等の添加剤が含まれていてもよい。
本発明の下痢防止用栄養組成物を液状で提供する場合、pHとしては、流通、保管時にゲル化や増粘させずに良好な粘性を保持させるという観点から、通常5.5以上9未満、好ましくは6.0~8.0が挙げられる。また、本発明の下痢防止用栄養組成物を固形状で提供する場合であれば、摂取又は投与前に所定量の水を添加して液状にした際にpHが前記範囲になるように、固形状の下痢防止用栄養組成物における組成を適宜設定すればよい。
本発明の下痢防止用栄養組成物は、前記の通り、液状の形態で提供されてもよく、また、粉末状、顆粒状等の固形状の形態で提供されてもよい。
本発明の下痢防止用栄養組成物は、その形態に応じた製法で調製される。例えば、本発明の下痢防止用栄養組成物が液状の場合であれば、水に、前述する特定構造のペクチン、脂質、炭水化物、タンパク質、及び必要に応じて配合される他の成分を所定量添加、混合することにより製造することができる。また、本発明の下痢防止用栄養組成物を乳化状態にする場合であれば、これら混合物に水を混ぜホモジナイザーで乳化を行えばよい。また、本発明の下痢防止用栄養組成物に2価の金属塩を配合する場合には、2価の金属塩の添加時には、2価の金属塩が添加される混合液は2価の金属塩によって前述する特定構造のペクチンのゲル化が生じないpH領域に調整しておくことが望ましい。
本発明の下痢防止用栄養組成物は、下痢の改善又は予防、及び栄養補給の目的で使用される。本発明の下痢防止用栄養組成物の適用対象者は、下痢の症状がありその改善が求められる者、又は下痢を発症するリスクがありその予防が求められる者であればよい。特に、経管栄養補給を受ける患者は、下痢を高頻度に発症しており、本発明の下痢防止用栄養組成物の適用対象として好適である。
1.ペクチンの準備
表2に示すペクチン1~8を準備した。各ペクチンは、それぞれエステル化度とアミド化度が異なるものである。各ペクチンのエステル化度、アミド化度、及び遊離酸度は、前述する方法に従って測定した。
Shodex GPC 101(昭和電工株式会社製)にTSKgel GMPWXL(東ソー株式会社製)を2本連結したものを用い、示差屈折計 RI-71S(昭和電工株式会社製)を装着し、480IIデータステーション GPCプログラム(システムインスツルメンツ株式会社)を用いてデータを解析した。得られたピークの分子量はマルトトリオース及びプルラン標準品(504,1540,6200,21700,48800,200000,348000,2560000の分子量をもつ)の溶出時間及び分子量をもとに作成した検量線により求めた。なお、サイズ排除クロマトグラフィーの設定条件は下記の通りである。

カラム:TSKgel GMPWXL、φ7.8mm×300mm(東ソー株式会社)を2本連結したもの
カラム温度:40℃
流量:1.0ml/min
移動相:0.1mol/l硝酸ナトリウム溶液
試料液注入量:100μl
表1に示す組成の栄養組成物(液状)を調製した。具体的には、ペクチンを除く各配合成分を水に加えてミキサーで混合した後にpH7.0に調整し、その後、ペクチンを加えて高圧ホモジナイザー(LAB-1000,APV社製使用)にて均質化(50MP、2パス)を行った。均質化した栄養組成物(水中油型乳化組成物)をパウチに充填し加熱殺菌処理(121℃、10分)に供した。なお、比較例1はペクチンを含まない栄養組成物である。また、最終的に得られた栄養組成物のpHを表2に示す。
各栄養組成物を摂取させた際の下痢防止効果を評価するために、SD系雄性ラット(9~10週齢)に対して、調製した各栄養組成物を2週間にわたって自由摂取させた。最後の5日間に糞便の性状を観察記録した。糞便の性状は目視により正常便又は下痢(水様便又は泥状便)として判別した。本試験では、各群につき4匹から5匹のラットを用いた。
前記試験例1で使用した栄養組成物の内、比較例5だけは試験期間中に粘度が次第に増している印象を受けた。そこで、前記試験例1で調製した栄養組成物の内、実施例2、実施例4及び比較例5について、常温保管中の粘度推移を検討した。
Claims (11)
- エステル化度が10~30%、アミド化度が0~25%、且つ遊離酸度が64%以上85%未満であるペクチンと、脂質と、前記ペクチン以外の炭水化物と、タンパク質とを含有する下痢防止用栄養組成物。
- 前記ペクチンの重量平均分子量が35万~60万である、請求項1に記載の下痢防止用栄養組成物。
- 前記炭水化物として、前記ペクチン以外の食物繊維、及び/又は糖質を含む、請求項1又は2に記載の下痢防止用栄養組成物。
- 前記炭水化物として、アラビアガム及び/又はガティガムを含む、請求項1~3のいずれかに記載の下痢防止用栄養組成物。
- 更に2価の金属塩を含む、請求項1~4のいずれかに記載の下痢防止用栄養組成物。
- 前記2価の金属塩が、酸性領域で2価の金属イオンを遊離する不溶性塩である、請求項5に記載の下痢防止用栄養組成物。
- 液状である、請求項1~6のいずれかに記載の下痢防止用栄養組成物。
- 前記ペクチンを3~15g/L含む、請求項7に記載の下痢防止用栄養組成物。
- 25℃での粘度が、2~100mPa・sである、請求項7又は8に記載の下痢防止用栄養組成物。
- 下痢防止用栄養組成物の製造のための、
